1Subcellular daunorubicin distribution and its relation to multidrug resistance phenotype in drug-resistant cell line SMMC-7721/R显示文摘AIM: To investigate the correlation between subcellulardaunorubicin distribution and the multidrug resistancephenotype in drug-resistant cell line SMMC-7721/R.METHODS: The multidrug resistant cell line SMMC-7721/R, a human hepatocellular carcinoma cell line,was established. Antisense oligonudeotides (AS-ODN)were used to obtain different multidrug resistancephenotypes by inhibiting the expression of mdr1 geneand/or multidrug resistance-related protein gene(mrp)using Lipofectamine as delivery agent. Expression ofmdr1 and mrp genes was evaluated by RT-PCR andWestern blotting. Intracellular daunorubicin (DNR)concentration was measured by flow cytometry.Subcellular DNR distribution was analyzed by confocallaser scanning microscopy. Adriamycin (ADM) and DMRsensitivity was examined by MTT method.RESULTS: Low level expression of mdr1 and mrp mRNAsand no expression of P-Glycoprotein(P-gp) andmultidrug resistance-related protein (P190) weredetected in parental sensitive cells SMMC-7721/S, butover-expression of these two genes was observed indrug-resistant cell SMMC-7721/R. The expression ofmdr1 and mrp genes in SMMC-7721/R cells was downregulated to the level in the SMMC-7721/S cells by AS-ODN. Intracellular DNR concentration in SMMC-7721/S cells was 10 times higher than that in SMMC-7721/Rcells. In SMMC7721/S cells intracellular DNRdistributed evenly in the nucleus and cytoplasm, whilein SMMC-7721/R cells DNR distributed in a punctatepattern in the cytoplasm and was reduced in thenucleus. DNR concentration in SMMC-7721/R cells co-transfected with AS-ODNs targeting to mdr1 and mrpmRNAs recovered to 25 percent of that in SMMC7721/Scells. Intracellular DNR distribution pattern in drug-resistant cells treated by AS-ODN was similar to drug-sensitive cell, and the cells resistance index (RI) to DNRand ADM decreased at most from 88.0 and 116.0 to 4.0and 2.3, respectively. Co-Transfection of two AS-ODNsshowed a stronger synergistic effect than separatetransfection.CONCLUSIONS: P-gp and P190 are two membersmediating MDR in cell line SMMC7721/R. Intracellulardrug concentration increase and subcellular distributionchange are two important factors in multidrugresistance (MDR) formation. 2Processing technique of target capsule's micro inflation hole with a scanning probe显示文摘To resolve inflation of inertial confinement fusion (ICF) target and encapsulation of micro inflation hole with adhesive, a scanning probe microscope (SPM) diamond microprobe was used as the cutting tool with SPM in contact mode. Some parameters influencing the quality of micro inflation hole, such as the scanning direction of the diamond tip, the scanning rate and the contact force are discussed. Accurate taper hole was achieved whose dimension and precision could meet the requirements of inflation and encapsulation technique of micro hole. The experimental results show that using SPM diamond microprobe as the cutting tool and with special processing technique, the precision machining of target capsule's taper inflation hole can be realized. 3Research on EM pulse protection property of plasma-microwave absorptive material-plasma sandwich structure显示文摘A plasma-microwave absorptive material (MAM)-plasma sandwich structure is presented to protect the electronic device against high power electromagnetic pulse. The model of electromagnetic wave reflected by and transmitting through the structure is established. Based on the characteristic parameters of plasma generated by discharge and usual MAM, the electromagnetic transmissive properties of the sandwich structure are investigated by the method of finite difference in time domain. The results indicate that in a rather broad frequency range, the electromagnetic attenuations by the structure are obviously better than the sum of attenuations resulted from plasma and MAM respectively. 4High sensitive detection of presenilin-1 point mutation based on electrochemilumi-nescence显示文摘Electrochemiluminescence (ECL) is a high- sensitive detection method with broad biological applications. Ruthenium (Ⅱ) tris (bipyridyl) 2+3(Ru(bpy)) and tripro-pylamine (TPA) are most commomly used combination of the reactive species in ECL. The redox of 2+3Ru(bpy) with excess TPA at the surface of an electrode produces a highly efficient and stable light emission due to a rapid cycle of the reactions. This rapid, highly sensitive and accurate method has been applied to gene quantification. In this work, an ECL detection system is designed and applied in detection of presenilin-1 (PS-1) gene mutation. The results show that given the same polymerase chain reaction (PCR) cycle num-ber, the ECL intensities from the digested and the undigested wild-type samples are nearly identical, but the difference in ECL intensities between the digested and the undigested mutant samples is distinctive. 5CALCULATION OF 3-D TRANSIENT TEMPERATURE FIELD DURING LASER TRANSFORMATION HARDENING PROCESS显示文摘A three-dimensional transient heat transfer model for laser transformation hardening process has been developed in this paper. The finite size of the laser treated sample, the surface heat loss of the sample, the latent heat of phase transformation and the temperature dependence of thermal properties of materials were considered. The heat source was considered as a moving Gaussian heat flux with a constant velocity. Three-dimension unequally spatial grid explicit finite difference equations, alternating direction implicit finite difference equations and implicit finite difference equations were deduced respectively. Three programs to calculate the temperature field were developed using Fortran language. The transient temperature fields of C22, 42CrMo, C60 steel samples during laser transformation hardening process were calculated using these programs, and the widths and depths of laser transformation hardening zones were also predicted. C22, 42CrMo, C60 steel samples were treated by CO2 laser,the widths and depths of laser transformation hardening zones of these samples were also measured experimentally. 6A mechanism for enhanced photocatalytic activity of nano-size silver particle modified titanium dioxide thin films显示文摘Ag-TiO2 nanostructured thin films with silver volume fraction of 0–20% were deposited on silicon and quartz substrates by RF magnetron sputtering and annealed in ambient air at 950°C for 1 h. The phase structure and surface topography of the films were characterized by X-ray diffractometer and transmission electron microscope. Photocatalytic activity of the films was evaluated by light induced degradation of methyl orange (C14H14N3NaO3S) solution using a high pressure mercury lamp as lamp-house. The relation of photocatalytic activity and silver content was studied in detail. It was found that silver content influences phase structure of TiO2 thin films, and silver in the films is metallic Ag (Ag0). With increasing silver content from 0 to 20 vol%, photocatalytic activity of the films increases first and then decreases. A suitable amount (2.5–5 vol%) silver addition can significantly enhance the photocatalytic activity of TiO2 films. 7Performance Comparison and Analysis of IP and MPLS Networks显示文摘With the increasing need for Quality of Service( QoS ),the traditional IP network is becoming less competitive. In order to give readers quantitative and convictive performance comparisons between IP and MPLS network,we choose Berkeleys NS 2 simulator [1] in our research on network throughput and end to end TCP behavior. Using NS 2 simulator,we analyze the performance of a network with MPLS Traffic Engineering( MPLS TE ).Specifically,the throughput and TCP end to end delay time under two different queuing algorithms are obtained and compared between the MPLS network and traditional IP network. We find that in the IP network using SFQ queuing algorithm will help traffic balance compared to the simple FIFO scheme,but it still can not utilize the available bandwidth,which in turn leads to the poor performance as in the FIFO case.With MPLS TE , the throughput will be higher and the end to end TCP delay time remains almost the same even in heavy load case.We notice that if flows are isolated partially,using SFQ scheme will have a comparable performance as the fully isolation case using FIFO scheme in a MPLS network,and that if flows are isolated fully for FIFO and SFQ schemes,both will have the same result.Finally,we conclude that a better queuing algorithm will help reduce the label space requirement.LIU Er wu, CAO Ming cui, CHEN Chun han, LI Feng (State Key Laboratory of Laser Technology, Huazhong University of Science and Technology, Wuhan 430074, P.R.China) 2001The Journal of China Universities of Posts and Telecommunications2001,8,2:3
8A systematic investigation of high harmonic generation using mid-infrared driving laser pulses显示文摘We report on a systematic investigation of the influences of gas pressure,focal position and focusing geometry on high harmonic generation by use of mid-infrared femtosecond laser pulses. We also discuss the spatial characteristics of harmonics under different focusing conditions. A program to calculate the temperature field was developed using FORTRAN language. The transient temperature field, the width and depth of laser transformation hardening zone of 42CrMo steel sample during laser transformation hardening process was calculated. The widths and depths of laser transformation hardening zones of 42CrMo steel samples were also measured experimentally. 10The characteristics of arc beam shaping in hybrid plasma and laser deposition manufacturing显示文摘As a new direct metal prototyping technology,the hybrid plasma and laser deposition manufacturing (PLDM) is proposed in this paper. In order to figure out the characteristics of plasma arc beam and mould in the PLDM process of high temperature alloy, the high speed CCD camera is used to obtain the picture around the plasma arc. Afterwards the sketch of picture is clearly obtained. And the effect of laser parameter, such as average power, pulse width, pulse repetition frequency and the angle between laser beam and plasma arc beam on the plasma arc appearance, is studied experimentally. The results show that the modality of plasma arc beam is markedly influenced by laser beam. 12I-V and C-V properties of TiO_2 thin film by pulsed-laser reactive deposition显示文摘TiO-2 thin films have bee deposited on p-Si(111) substrates by pulsed-laser ablation of metallic Ti target in the O-3 ambient. The current-voltage and capacitance-voltage of the Al/TiO-2/Si capacitors are measured. The results show that the dielectric constant of thin film after being annealed at 700℃ is found to be 46, and the border trap density and the interface state density at the TiO-2/p-Si interface are 1.8×10 12 cm -2 and 2×10 12 eV -1·cm -2, respectively. 13Online diagnosis of electron excitation temperature in CH_4+H_2 discharge plasma at atmospheric pressure by optical emission spectra显示文摘Methane coupling under low temperature plasmas at atmospheric pressure is a green process by use of renewable sources of energy. In this study, CH4+H2 dis- charge plasma was on-line diagnosed by optical emission spectra so as to char- acterize the discharge system and to do spade work for the optimization of the technical parameters for future commercial production of methane coupling under plasmas. The study was focused on a calculation method for the online diagnosis of the electron excitation temperature in CH4+H2 discharge plasma at atmospheric pressure. The diagnostic method is easy, efficient and fairly precise. 15Influence of Nitrogen Flow Rate on the Microstructure and Properties of N and Me (Me=Cr,Zr) Co-doped Diamond-like Carbon Films显示文摘In this paper,Me (Me=Cr,Zr) and N co-doped diamond like carbon (DLC-MeN) composite films were prepared on cemented carbide substrates by pulsed bias arc ion plating.The effect of nitrogen flow rates on the microstructure and properties of the films were investigated by X-ray photoelectron spectroscopy (XPS),Raman spectra,grazing incident X-ray diffraction (GIXRD),high resolution transmission electron microscopy (HRTEM) and nano-indentation.Raman,GIXRD and HRTEM results show that the deposited films are nanocomposite films with MeN nanocrystalline phase embedded within DLC amorphous matrix,which are vital for the mechanical properties of the films.The nitrogen flow rate has significant effect on the compositions and structures and hence on the hardness and elastic modulus of the films,and increasing nitrogen flow rate decreases drastically the hardness and elastic modulus of the films.Zhiyun Han,Hongkai Li,Guoqiang Lin and Chuang Dong Key Laboratory for Material Modification by Laser,Ion and Electron Beams,Dalian University of Technology,Dalian 116085,China 2010Journal of Materials Science & Technology2010,26,11:2
