1Injectable collagen scaffold promotes swine myocardial infarction recovery by long-term local retention of transplanted human umbilical cord mesenchymal stem cells显示文摘Stem cell therapy is an attractive approach for recovery from myocardial infarction(MI)but faces the challenges of rapid diffusion and poor survival after transplantation.Here we developed an injectable collagen scaffold to promote the long-term retention of transplanted cells in chronic MI.Forty-five minipigs underwent left anterior descending artery(LAD)ligation and were equally divided into three groups 2 months later(collagen scaffold loading with human umbilical mesenchymal stem cell(hUMSC)group,hUMSC group,and placebo group(only phosphate-buffered saline(PBS)injection)).Immunofluorescence staining indicated that the retention of transplanted cells was promoted by the collagen scaffold.Echocardiography and cardiac magnetic resonance imaging(CMR)showed much higher left ventricular ejection fraction(LVEF)and lower infarct size percentage in the collagen/hUMSC group than in the hUMSC and placebo groups at 12 months after treatment.There were also higher densities of vWf-,α-sma-,and cTnT-positive cells in the infarct border zone in the collagen/cell group,as revealed by immunohistochemical analysis,suggesting better angiogenesis and more cardiomyocyte survival after MI.Thus,the injectable collagen scaffold was safe and effective on a large animal myocardial model,which is beneficial for constructing a favorable microenvironment for applying stem cells in clinical MI.Qiang Wangn Xiaojun He Bin Wang Jun Pan Chunying Shi Jie Li Liudi Wang Yannan Zhao Jianwu Dai Dongjin Wang 2021Science China(Life Sciences)2021,64,2:5

3Yeast One-hybrid System Used to Identify the Binding Proteins for Rat Glutathione S-transferase P Enhancer I显示文摘Objective To detect the trans-factors specifically binding to the strong enhancerelement (GPEI) in the upstream of rat glutathione S-transferase P (GST-P) gene. MethodsYeast one-hybrid system was used to screen rat lung MATCHMAKER cDNA library toidentify potential trans-factors that can interact with core sequence of GPEI(cGPEI).Electrophoresis mobility shift assay (EMSA) was used to analyze the binding of trans-factors to cGPEI. Results cDNA fragments coding for the C-terminal part of thetranscription factor c-Jun and rat adenine nucleotide translocator (ANT) were isolated. Thebinding of c-Jun and ANT to GPEI core sequence were confirmed. Conclusions Rat c-juntranscriptional factor and ANT may interact with cGPEI. 14Age-Constrained Dynamic Content Replacing and Delivering for UAV-Assisted Context Awareness显示文摘In this work,we employ the cache-enabled UAV to provide context information delivery to end devices that make timely and intelligent decisions.Different from the traditional network traffic,context information varies with time and brings in the ageconstrained requirement.The cached content items should be refreshed timely based on the age status to guarantee the freshness of user-received contents,which however consumes additional transmission resources.The traditional cache methods separate the caching and the transmitting,which are not suitable for the dynamic context information.We jointly design the cache replacing and content delivery based on both the user requests and the content dynamics to maximize the offloaded traffic from the ground network.The problem is formulated based on the Markov Decision Process(MDP).A sufficient condition of cache replacing is found in closed form,whereby a dynamic cache replacing and content delivery scheme is proposed based on the Deep Q-Network(DQN).Extensive simulations have been conducted.Compared with the conventional popularity-based and the modified Least Frequently Used(i.e.,LFU-dynamic)schemes,the UAV can offload around 30%traffic from the ground network by utilizing the proposed scheme in the urban scenario,according to the simulation results.Liudi Wang Shan Zhang Xishuo Li Hongbin Luo 2022China Communications2022,19,7:0
15Satellite-observed significant improvement in nearshore transparency of the Bohai Sea during pollution control显示文摘The Bohai Sea(BS)is the unique semi-closed inland sea of China,characterized by degraded water quality due to significant terrestrial pollution input.In order to improve its water quality,a dedicated action named“Uphill Battles for Integrated Bohai Sea Management”(UBIBSM,2018–2020)was implemented by the Chinese government.To evaluate the action effectiveness toward water quality improvement,variability of the satelliteobserved water transparency(Secchi disk depth,Z_(SD))was explored,with special emphasis on the nearshore waters(within 20 km from the coastline)prone to terrestrial influence.(1)Compared to the status before the action began(2011–2017),majority(87.3%)of the nearshore waters turned clear during the action implementation period(2018–2020),characterized by the elevated Z_(SD)by 11.6%±12.1%.(2)Nevertheless,the improvement was not spatially uniform,with higher Z_(SD)improvement in provinces of Hebei,Liaoning,and Shandong(13.2%±16.5%,13.2%±11.6%,10.8%±10.2%,respectively)followed by Tianjin(6.2%±4.7%).(3)Bayesian trend analysis found the abrupt Z_(SD)improvement in April 2018,which coincided with the initiation of UBIBSM,implying the water quality response to pollution control.More importantly,the independent statistics of land-based pollutant discharge also indicated that the significant reduction of terrestrial pollutant input during the UBIBSM action was the main driver of observed Z_(SD)improvement.(4)Compared with previous pollution control actions in the BS,UBIBSM was found to be the most successful one during the past 20 years,in terms of transparency improvement over nearshore waters.The presented results proved the UBIBSM-achieved remarkable water quality improvement,taking the advantage of long-term consistent and objective data record from satellite ocean color observation.Xuyan Li Jinzhao Xiang Liudi Zhu Zhibin Yang Ting Wei Bing Mu Xiaobo Zhang Tingwei Cui 2023Acta Oceanologica Sinica2023,42,12:0
16Cloning and Sequence Analysis on 3′ Coding Region of Wild Boar and Cross Bred Pig Myostatin Gene显示文摘Myostatin, with a highly conservative gene among breeds is a negative regulator of muscle. The 3′ coding regions of wild boar and crossbred pig myostatin were cloned by RT-PCR and sequenced respectively. The homology of the nucleotide sequence between wild boar and crossbred pig was 100% and there was no difference in this region compared with pig myostatin gene of Genbank. 17Three-dimensional bioprinting sodium alginate/gelatin scaffold combined with neural stem cells and oligodendrocytes markedly promoting nerve regeneration after spinal cord injury显示文摘Accumulating research has indicated that the transplantation of combined stem cells and scaffolds is an effective method for spinal cord injury(SCI).The development of three-dimensional(3D)bioprinting technology can make the 3D scaffolds combined with cells more accurate and effective for SCI treatment.However,unmyelinated newborn nerve fibers have no nerve signaling conduction,hampering recovery of motor function.In this study,we designed and printed a type of sodium alginate/gelatin scaffold loaded with neural stem cells and oligodendrocytes,which were involved in the formation of the myelin sheaths of neural cell axons.In order to observe the effectiveness of this 3D bioprinting scaffold,we transplanted it into the completely transected rat spinal cord,and then immunofluorescence staining,hematoxylin–eosin staining and behavioral assessment were performed.The results showed that this 3D bioprinting scaffold markedly improved the hindlimb motor function and promoted nerve regeneration.These findings suggested that this novel 3D bioprinting scaffold was a good carrier for cells transplantation,thereby enhancing spinal cord repair following injury.Shuo Liu Hui Yang Dong Chen Yuanyuan Xie ChenXu Tai Liudi Wang Peng Wang Bin Wang 2022Regenerative Biomaterials2022,9,1:0

19Cloning and Analysis of Telomere-associated Sequences of Ginkgo biloba L..显示文摘Total genomic DNA was extracted from leaves of Ginkgo biloba L. by the method of hot CTAB. After determining quantification of DNA sample by microclorimetric spectrophotography, Arabidopsis-type telomere primer and Sau3AⅠ cassette primer were used to isolate telomere-associated sequences from G. biloba L. by the method of cassette-ligation-mediated polymerase chain reaction (PCR). Using this method, two telomere-associated sequences, TAS1 and TAS2, were isolated. The authors preformed Southern hybridization of EcoRⅠ-treated G. biloba genomic DNA with each clone. The hybridization pattern showed that the clones obtained were derived from G. biloba genomic DNA. 20Study on the Polymorphisms of Porcine Myostatin Gene in Promoter Region by PCR-RFLPS显示文摘In order to further study functions of the porcine myostatin gene, we analyzed the polymorphisms of porcine myostatin gene in promoter region among different breeds including Yorkshire, Landrace, Duroc, Junmu, Min pig and Sanjiang white pig by PCR-RFLPs. The allele T dominated in the imported lean-type pig breeds such as Yorkshire, Landrace and Duroc. No allele A was detected in Junmu and Sanjiang white pig, and the frequencies of three genotypes were about equal in Min pig.
