1L-type calcium current in right ventricular outflow tract myocytes of rabbit heart显示文摘The mechanism of idiopathic ventricular tachycardia originating from the right ventricular outflow tract (RVOT) is not clear. Many clinical reports have suggested a mechanism of triggered activity. However, there are few studies investigating this because of the technical difficulties associated with examining this theory. The L-type calcium current (ICa-L), an important inward current of the action potential (AP), plays an important role in arrhythmogenesis. The aim of this study was to explore differences in the APs of right ventricular (RV) and RVOT cardiomyocytes, and differences in electrophysiological characteristics of the ICa-L in these myocytes. Rabbit RVOT and RV myocytes were isolated and their AP and ICa-L were investigated using the patch-clamp technique. RVOT cardiomyocytes had a wider range of AP duration (APD) than RV cardiomyocytes, with some markedly prolonged APDs and markedly shortened APDs. The markedly shortened APDs in RVOT myocytes were abolished by treatment with 4-AP, an inhibitor of the transient outward potassium current, but the markedly prolonged APDs remained, with some myocytes with a long AP plateau not repolarizing to resting potential. In addition, early afterdepolarization (EAD) and second plateau responses were seen in RVOT myocytes but not in RV myocytes. RVOT myocytes had a higher current density for ICa-L than RV myocytes (RVOT (13.16±0.87) pA pF-1, RV (8.59±1.97) pA pF-1; P<0.05). The ICa-L and the prolonged APD were reduced, and the EAD and second plateau response disappeared, after treatment with nifedipine (10 μmol L-1), which blocks the ICa-L. In conclusion, there was a wider range of APDs in RVOT myocytes than in RV myocytes, which is one of the basic factors involved in arrhythmogenesis. The higher current density for ICa-L is one of the factors causing prolongation of the APD in RVOT myocytes. 3Transcriptome and Metabolome Analyses of Sea Cucumbers Apostichopus japonicus in Southern China During the Summer Aestivation Period显示文摘Aestivation is a common strategy of sea cucumbers(Apostichopus japonicus)in response to high-temperature conditions.Previous studies have individually investigated the immune and physiological alterations at the aestivation stage.However,these studies have not evaluated the relationship between immunity and physiology.In this study,we explored the transcriptome and metabolome of A.japonicus during the aestivation stage to study the relationship.The transcriptome analysis of dormant(aestivation)and revived A.japonicus generated 2368 differentially expressed genes,including 927 downregulated genes and 1441 upregulated genes.Based on Gene Ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analyses,the downregulated genes in the dormant group were found to be involved in DNA replication,RNA metabolic process,and protein metabolism,which results in the inhibition of motility,skeletal development,neural activity,cell proliferation,and development of A.japonicus.In contrast,the upregulated genes were found to be associated with fatty acid metabolism,carbohydrate hydrolysis,and phagocytosis.In the metabolome analysis,the downregulated metabolites were found to be associated with fatty acid metabolism,starch and sucrose metabolism,and TCA cycle.This indicates that dormant sea cucumbers consume reserved carbohydrates and fatty acids to maintain low levels of energy supply.The protein-protein interaction network analysis further revealed that carbohydrate hydrolysis promoted phagocytosis activity in the dormant group.This study provides new insights into potential molecular mechanisms of sea cucumber survival in high-temperature conditions,which is critical in aquaculture of sea cucumbers.YANG Qiuhua ZHANG Xusheng LU Zhen HUANG Ruifang TRAN Ngoc Tuan WU Jianshao YANG Fuyuan GE Hui ZHONG Chenhui SUN Qian ZHOU Chen LIN Qi 2021Journal of Ocean University of China2021,20,1:2

5New State Recovery Attacks on the Grain v1 Stream Cipher显示文摘The Grain v1 stream cipher is one of the seven finalists in the final e STREAM portfolio. Though many attacks have been published,no recovery attack better than exhaustive key search on full Grain v1 in the single key setting has been found yet. In this paper,new state recovery attacks on Grain v1 utilizing the weak normality order of the employed keystream output function in the cipher are proposed. These attacks have remarkable advantages in the offline time,online time and memory complexities,which are all better than exhaustive key search. The success probability of each new attack is 0.632. The proposed attack primarily depends on the order of weak normality of the employed keystream output function. 7Comparative transcriptome analysis of Phascolosoma esculenta under different salinities显示文摘Salinity is an important parameter that influences the developmental progress of aquatic organisms.The larvae of Phascolosoma esculenta developed into the early stage trochosphere,the late stage trochosphere and the early stage pelagosphere in the low-salinity group(15),the middle group(22.5)and the high-salinity group(30),respectively,after being incubated for 50 h,according to morphological observation.The results showed that high salinity could accelerate the cellular development of P.esculenta.To elucidate the molecular mechanism of larval development of P.esculenta under different salinities,we performed transcriptome analysis of the larvae in P.esculenta.Nine transcriptome libraries of P.esculenta larvae were constructed,i.e.,the low-salinity group(A1,A2 and A3),the middle group(B1,B2 and B3)and the high-salinity group(C1,C2 and C3),and each group with three repeats.These nine paired-end libraries were sequenced using the Illumina HiSeq 2500 platform.In total,57.63 Gb clean data were obtained and de novo assembled into 320,527 unigenes with mean lengths of 482.62 bp and N50 of 529 bp.A total of 249,162 unigenes were significantly matched to known unique proteins.The gene ontology(GO)annotation and the Kyoto Encyclopedia of Genes and Genomes(KEGG)were used to determine the regulation pathways and candidate genes that are associated with the process of larval development.A total number of 12,811,16,991 and 1735 transcripts were identified as differentially expressed genes(DEGs)from the comparison of A vs.B,A vs.C,and B vs.C,respectively.Of these differentially expressed genes,many DEGs that were upregulated in the high-salinity group were involved in the function that controls the larval development of P.esculenta,such as the neuroactive ligand-receptor interaction and ECM-receptor interaction.In addition,the results of the transcriptome analysis were verified by quantitative real-time PCR(qRT-PCR).After morphological observation and transcriptome analysis,we think high salinity(30)is more suitable than medium(22.5)or low(15)salinity for the development of P.esculenta larvae.This information provides guidance for artificial breeding and paves the way to the further research on the individual development of P.esculenta under different salinities.Xiangyang Lin Suzhen Deng Xiande Liu Chenhui Zhong Kun Qiao Leibin Li 2020Aquaculture and Fisheries2020,5,6:1

9Improved Guess and Determine attack on the MASHA stream cipher显示文摘Dear editor,K2 is a secure and high-performance stream cipher and has been standardized by ISO/IEC 18033-4. The MASHA stream cipher is a successor of K2 with integrated MAC functionality proposed in 2011. Based on optimizing the guess and determination process, we present an improved Guess and Determine attack on MASHA with time complexity of 2224, reducing the time complexity of the existing attack by a factor of 296.
