1Multi-objective regulation in autohydrolysis process of corn stover by liquid hot water pretreatment显示文摘Increasing reducing sugars(xylose and glucose) yield for bioethanol from corn stover depends strongly on optimization of pretreatment conditions. The optimum reaction conditions of two-stage liquid hot water(LHW) pretreatment based on total sugars yield were investigated. Under optimal conditions, the recovery of glucose of corn stover after two-stage LHW pretreatment and 72 h enzymatic digestion, reached 89.55%. In addition, acetic acid-rich spent liquor pretreatment and one-stage LHW pretreatment have been carried out to make comparisons with two-stage LHW treatment. Glucose yield 89.55% is superior to the recovery 83.38% using acetic acidrich spent liquor pretreatment or 80.58% using one-stage LHW pretreatment. The production of total sugars was increased by 7.8% when compared with one-stage pretreatment. Moreover, the structural features of the treated corn stover solid residues were also investigated by XRD and SEM technology in order to clarify the effects of the reaction on corn stover. 2Spatiotemporal variability of ocean chemistry in the early Cambrian,South China显示文摘Following the Ediacaran metazoan radiation,the 'Cambrian Explosion' set up the major framework of todays' animal phyla as well as modern marine ecosystem.Here,we present a preliminary investigation on the temporal and spatial(from shallow to deep waters) variations of the early Cambrian ocean chemistry in South China through analyzing a Fe-S-C systematic dataset integrated from literature.Our investigation indicates that the early Cambrian deep ocean in South China was still anoxic and Fe2+-enriched(i.e.,ferruginous) although its surface was oxic,and in between a metastable euxinic(anoxic and sulfidic) water zone may have dynamically developed in anoxic shelf waters with an increasing weathering sulfate supply.Furthermore,accompanying marine transgression and regression cycles in the early Cambrian,such a 'sandwich' structure in ocean redox chemistry demonstrates five evolutional stages,which can be well correlated to the spatiotemporal patterns of fossil records in South China.The good correlation between metazoan fossil occurrences and water chemistry in South China suggests that early animals possibly possessed ability to inhabit anoxic but generally not euxinic environments as free H2S was fatal to most eukaryotes.This view can well explain why those small shell fauna and sponges disappeared from shelf to slope areas where sulfidic Ni-Mo-rich shales were widely deposited.Thus,we conclude that the spatiotemporal variations of ocean chemistry and its biological effects probably played a key role in the phased animal radiations and 'extinctions' in the early Cambrian.JIN ChengSheng LI Chao PENG XingFang CUI Hao SHI Wei ZHANG ZiHu LUO GenMing XIE ShuCheng 2014Science China Earth Sciences2014,57,4:5

4Effects of Rhizoma Acori Tatarinowii extracts on gamma-aminobutyric acid type A receptor alpha 1 subunit brain expression during development in a recurrent seizure rat model显示文摘Extracts from Rhizoma Acori Tatarinowii (Grassleaf Sweetflag Rhizome,Shichangpu) have been shown to improve learning and memory,reduce anxiety,allay excitement,and suppress seizures.Rhizoma Acori Tatarinowii extracts interact with γ-aminobutyric acid and activate the γ-aminobutyric acid type A receptor,although few studies have addressed the precise effects of γ-aminobutyric acid type A receptor α1 subunit.In the present study,γ-aminobutyric acid type A receptor α1 subunit protein expression in the cerebral cortex and hippocampus,and pathological scores of brain injury,were significantly greater following recurrent seizures,but significantly decreased following treatment with Rhizoma Acori Tatarinowii extracts.These results indicated that Rhizoma Acori Tatarinowii extracts down-regulated γ-aminobutyric acid type A receptor α1 subunit protein expression in the cerebral cortex and hippocampus and protected seizure-induced brain injury during development.Liqun Liu Ding'an Mao Keqiang Chi Xingfang Li Tao Bo Jinming Guo Zhuwen Yi 2011Neural Regeneration Research2011,6,6:4
5High-resolution cyclostratigraphy of geochemical records from Permo-Triassic boundary section of Dongpan,southwestern Guangxi,South China显示文摘Spectral analysis is applied to geochemical data from a deep-water Permian-Triassic Boundary (PTB) section across PTB in Dongpan, Guangxi for high-resolution cyclostratigraphic research. The characteristics of the Milankovitch Cycles have been well recorded in the strata of the Dongpan section. Spectrum on Ce/La records shows especially that the ratios among its three preponderant cycles in the section are 5:2:1, which well displays the phenomenon of the sedimentary cycles driven by orbital force. 6Glucocorticoid receptor expression in neonatal rat cortex following recurrent seizures The role in developing brain injury显示文摘BACKGROUND: Studies have explored changes in neonatal rat glucocorticoid receptor (GR) expression changes following mature brain injury.OBJECTIVE: To investigate the temporal and special changes of GR during brain development in rats with recurrent seizures.DESIGN,TIME AND SETTING: A randomized,controlled animal experiment was performed at the Department of Pediatrics,Second Xiangya Hospital of Central South University,from February 2008 to March 2009.MATERIALS: Rabbit anti-rat GR monoclonal antibody was purchased from Santa Cruz Biotechnology,USA;goat anti-rabbit IgG was purchased from Zhongshan Goldenbridge Biotechnology,China.METHODS: A total of 48 Sprague-Dawley rats,7 days old,were randomly assigned to control and seizure groups,with 24 animals in each group.Seizures were induced by inhalant flurothyl.MAIN OUTCOME MEASURES: Changes in GR protein expression in the rat cerebral cortex were detected by Western blotting analysis and immunohistochemistry.RESULTS: GR expression in the cerebral cortex of control rats significantly increased with aging (P < 0.05),and varied in the frontal lobe,temporal lobe,and parietal lobe.GR was predominantly expressed in the cytoplasm early and rapidly increased in the nuclei.GR protein expression in the cerebral cortex after seizure was lower in the cytoplasm at 15 days and in nuclear protein at 19 days.CONCLUSION: GR expression displayed temporal and spatial changes in brain development.Recurrent seizures in neonatal rats cause abnormal GR expression and might play an important role in developing brain injury.Tao Bo Lu Yi Tuanmei Wang Jian Li Xingfang Li Dingan Mao 2010Neural Regeneration Research2010,5,2:2
7Molecularly tagged genes and quantitative trait loci in cucumber with recommendations for QTL nomenclature显示文摘Cucumber,Cucumis sativus L.(2n=2x=14),is an important vegetable crop worldwide.It was the first specialty crop with a publicly available draft genome.Its relatively small,diploid genome,short life cycle,and selfcompatible mating system offers advantages for genetic studies.In recent years,significant progress has been made in molecular mapping,and identification of genes and QTL responsible for key phenotypic traits,but a systematic review of the work is lacking.Here,we conducted an extensive literature review on mutants,genes and QTL that have been molecularly mapped or characterized in cucumber.We documented 81 simply inherited trait genes or major-effect QTL that have been cloned or fine mapped.For each gene,detailed information was compiled including chromosome locations,allelic variants and associated polymorphisms,predicted functions,and diagnostic markers that could be used for marker-assisted selection in cucumber breeding.We also documented 322 QTL for 42 quantitative traits,including 109 for disease resistances against seven pathogens.By alignment of these QTL on the latest version of cucumber draft genomes,consensus QTL across multiple studies were inferred,which provided insights into heritable correlations among different traits.Through collaborative efforts among public and private cucumber researchers,we identified 130 quantitative traits and developed a set of recommendations for QTL nomenclature in cucumber.This is the first attempt to systematically summarize,analyze and inventory cucumber mutants,cloned or mapped genes and QTL,which should be a useful resource for the cucurbit research community.Yuhui Wang Kailiang Bo Xingfang Gu Junsong Pan Yuhong Li Jinfeng Chen Changlong Wen Zhonghai Ren Huazhong Ren Xuehao Chen Rebecca Grumet Yiqun Weng 2020Horticulture Research2020,7,1:2

13Genome-wide association study reveals candidate genes for gummy stem blight resistance in cucumber显示文摘Gummy stem blight(GSB),caused by Didymella bryoniae,is a serious fungal disease that leads to decline in cucumber yield and quality.The molecular mechanism of GSB resistance in cucumber remains unclear.Here,we investigated the GSB resistance of cucumber core germplasms from four geographic groups at the seedling and adult stages.A total of 9 SNPs related to GSB resistance at the seedling stage and 26 SNPs at the adult stage were identified,of which some are co-localized to previously mapped Quantitative trait loci(QTLs)for GSB resistance(gsb3.2/gsb3.3,gsb5.1,and gsb-s6.2).Based on haplotype analysis and expression levels after inoculation,four candidate genes were identified within the region identified by both Genome-wide association study(GWAS)and previous identified QTL mapping,including Csa3G129470 for gsb3.2/gsb3.3,Csa5G606820 and Csa5G606850 for gsb5.1,and Csa6G079730 for gsb-s6.2.The novel GSB resistant accessions,significant SNPs,and candidate genes facilitate the breeding of GSB resistant cucumber cultivars and provide a novel idea for understanding GSB resistance mechanism in cucumber.Jianan Han Shaoyun Dong Yanxia Shi Zhuonan Dai Han Miao Baoju Li Xingfang Gu Shengping Zhang 2023Horticultural Plant Journal2023,9,2:1
14Explorations in Teaching Research by a Tutoring Institution in China显示文摘Purpose:This article focuses on rarely explored organizational practice of teaching research in China’s private tutoring industry.Taking a large private tutoring institution as an example,the article examines how private tutoring institutions understand and engage in educational explorations of standardization and informatization processes.Design/Approach/Methods:The article is based on a case study of one of the biggest tutoring companies in China.The study started with document analysis of the institutional history,supplemented by interviews with personnel who have worked in the institution since it was founded.Next,the researchers collected empirical data using mixed methods.Quantitative data were obtained from the user database owned by the institution.Qualitative data were collected directly by the researchers through interviews and participant observation.Both qualitative and quantitative data were analyzed from multiple perspectives.Findings:Teaching research in private tutoring institutions commonly differs from that in public schooling.In this particular case,it is technology-driven,student-tailored,and processstandardized.Utilized well,it can supplement the mainstream public education system and promote education innovation and equity throughout the country.Originality/Value:It is hoped that this article could give some insights into the possibility of the cooperation between formal schools and tutoring institutions in the areas of teaching research and other in-class and off-campus activities.The article can also draw public attention to the necessity and benefits of adopting technical methods in the teaching process.Yunfeng Bai Xueli Tang Xingfang Li Shunan Fan 2019ECNU Review of Education2019,2,1:1

16Genome-wide analysis of expansins and their role in fruit spine development in cucumber(Cucumis sativus L.)显示文摘Cucumber is one of the most widely consumed vegetables worldwide,and the fruit spine is an important fruit quality trait.Expansins play critical roles in fruit development;however,the regulation of expansins in cucumber fruit spine development has not been reported.In this study,33 expansin genes were identified in the cucumber genome V3;additionally,expansin genes in Citrullus lanatus,Cucumis melo,Cucurbita maxima,Lagenaria siceraria,and Benincasa hispida were also identified.Phylogenetic analysis of expansin proteins in Cucurbitaceae and Arabidopsis showed that they evolved separately in each plant species.Phylogenetic analysis showed that C.maxima was derived earlier than the other five Cucurbitaceae species.The expression of CsEXPA2,CsEXPA14,and CsEXLA3 varied in cucumber lines with different fruit spine densities.A yeast two-hybrid assay showed that a putative auxin transporter encoded by numerous spine gene(ns)interacts with CsEXLA2,which may be involved in the development of the numerous spines in cucumber.These results provide novel insights into the expansins related to plant development and fruit spine development in cucumber.Xiaoping Liu Shaoyun Dong Han Miao Kailiang Bo Caixia Li Yingying Yang Xingfang Gu Shengping Zhang 2022Horticultural Plant Journal2022,8,6:1
17Preparation of silicon carbide nitride films on Si substrate by pulsed high-energy density plasma显示文摘Thin films of silicon carbide nitride (SiCN) were prepared on (111) oriented silicon substrates by pulsed high-energy density plasma (PHEDP). The evolution of the chemical bonding states between silicon, nitrogen and carbon was investigated as a function of discharge voltage using X-ray photoelectron spectroscopy. With an increase in discharge voltage both the C 1s and N 1s spectra shift to lower binding energy due to the formation of C-Si and N-Si bonds. The Si-C-N bonds were observed in the deconvolved C 1s and N 1s spectra. The X-ray diffractometer (XRD) results show that there were no crystals in the films. 18Natural variation in STAYGREEN contributes to low-temperature tolerance in cucumber显示文摘Low-temperature(LT)stress threatens cucumber production globally;however,the molecular mechanisms underlying LT tolerance in cucumber remain largely unknown.Here,using a genome-wide association study(GWAS),we found a naturally occurring single nucleotide polymorphism(SNP)in the STAYGREEN(CsSGR)coding region at the gLTT5.1 locus associated with LT tolerance.Knockout mutants of CsSGR generated by clustered regularly interspaced palindromic repeats(CRISPR)/CRISPR-associated nuclease 9 exhibit enhanced LT tolerance,in particularly,increased chlorophyll(Chl)content and reduced reactive oxygen species(ROS)accumulation in response to LT.Moreover,the C-repeat Binding Factor 1(CsCBF1)transcription factor can directly activate the expression of CsSGR.We demonstrate that the LT-sensitive haplotype CsSGRHapA,but not the LT-tolerant haplotype CsSGR^(HapG)could interact with NON-YELLOW COLORING 1(CsNYC1)to mediate Chl degradation.Geographic distribution of the CsSGR haplotypes indicated that the CsSGR^(HapG)was selected in cucumber accessions from high latitudes,potentially contributing to LT tolerance during cucumber cold-adaptation in these regions.CsSGR mutants also showed enhanced tolerance to salinity,water deficit,and Pseudoperonospora cubensis,thus CsSGR is an elite target gene for breeding cucumber varieties with broad-spectrum stress tolerance.Collectively,our findings provide new insights into LT tolerance and will ultimately facilitate cucumber molecular breeding.Shaoyun Dong Caixia Li Haojie Tian Weiping Wang Xueyong Yang Diane MBeckles Xiaoping Liu Jiantao Guan Xingfang Gu Jiaqiang Sun Han Miao Shengping Zhang 2023Journal of Integrative Plant Biology2023,65,12:0
19Gamma-aminobutyric acid A receptor gamma 2 subunit following Mg-free-induced seizures in cultured developing neurons显示文摘BACKGROUND:Studies have demonstrated that in vitro cultured cortical neurons from embryonic rats can produce spontaneous recurrent epileptiform discharges following transient Mg2+-free extracellular solution culture. OBJECTIVE: To explore gamma-aminobutyric acid A receptor (GABAAR) γ2 subunit expression follow-ing Mg2+-free-induced seizures in cultured developing neurons. DESIGN,TIME AND SETTING: Cellular and molecular biology. The in vitro experiment was performed at the Department of Pediatrics,Second Xiangya Hospital of Central Southern University between January 2007 and February 2008. MATERIALS: Cortical neurons of Wistar rats on gestational days 16-17 were used. Normal extracellular solution (pH 7.3) consisted of NaCl 145 mmol/L,KCl 2.5 mmol/L,HEPES 10 mmol/L,MgCl2 1 mmol/L,CaCl2 2 mmol/L,glucose 10 mmol/L,and glycine 0.01 mmol/L. In addition,there was no MgCl2 in the Mg2+-free extracellular solution. METHODS: Cortical neurons cultured for 6 days were exposed to normal extracellular solution (control group) and Mg2+-free media (Mg2+-free group) respectively for 3 hours,followed by continuous culture in DMEM solution. MAIN OUTCOME MEASURES: On days 1,7 and 12 after Mg2+-free treatment,real-time RT-PCR,im-munochemistry,and flow cytometry were used to detect GABAAR γ2 subunit expression. RESULTS: Compared with the control group,GABAAR γ2-positive cells decreased significantly on days 1 and 7 after Mg2+-free treatment (P<0.01),but significantly increased on day 12 (P<0.01). GABAAR γ2 subunit mRNA expression decreased significantly at 7 days Mg2+-free treatment when measured by real-time RT-PCR compared with the control group (P<0.05). CONCLUSION: GABAAR γ2 subunit expression is modified following Mg-free-induced seizures in cul-tured developing neurons. 20Formation and Oxidation Resistance of Silicide Coatings for Mo and Mo-Based Alloys显示文摘The forming process of silicide coatings on pure Mo and Mo-base alloys, obtained by the gas- phase deposition method. has been studied by examining the microstructure of coatings and the relationship between coating thickness and process parameters. It was shown that the growth of coatings was diffusion-controlled, the diffusion of silicon to be coated into Mo or Mo-base alloys was mainly responsible for the formation of silicide.
