| 1 | Injectable peptide hydrogel as intraperitoneal triptolide depot for the treatment of orthotopic hepatocellular carcinoma显示文摘Chemotherapy is among the limited choices approved for the treatment of hepatocellular carcinoma(HCC) at intermediate and advanced stages. Preferential and prolonged drug exposure in diseased sites is required to maximize the therapeutic index of the drug. Here, we report an injectable supramolecular peptide hydrogel as an intraperitoneal depot for localized and sustained release of triptolide for the treatment of orthotopic HCC. We chose peptide amphiphile C16-GNNQQNYKD-OH-based nanofibers as gelators and carriers for triptolide. Sustained triptolide release from the hydrogel was achieved over 14 days in vitro, with higher accumulation in and cytotoxicity against human HCC Bel-7402 in comparison with L-02 fetal hepatocytes. After intraperitoneal injection, the hydrogel showed prolonged retention over 13 days and preferential accumulation in the liver, realizing HCC growth inhibition by99.7 ± 0.1% and animal median survival extension from 19 to 43 days, without causing noticeable pathological changes in the major organs. | 2 | Kiwifruit Genome Database(KGD):a comprehensive resource for kiwifruit genomics显示文摘Kiwifruit(Actinidia spp.)plants produce economically important fruits containing abundant,balanced phytonutrients with extraordinarily high vitamin C contents.Since the release of the first kiwifruit reference genome sequence in 2013,large volumes of genome and transcriptome data have been rapidly accumulated for a handful of kiwifruit species.To efficiently store,analyze,integrate,and disseminate these large-scale datasets to the research community,we constructed the Kiwifruit Genome Database(KGD;http://gffzz6fc08c45f9754a68hfffwo95q66uw6cnc.ffgz.tsg.suse.edu.cn/).The database currently contains all publicly available genome and gene sequences,gene annotations,biochemical pathways,transcriptome profiles derived from public RNA-Seq datasets,and comparative genomic analysis results such as syntenic blocks and homologous gene pairs between different kiwifruit genome assemblies.A set of user-friendly query interfaces,analysis tools and visualization modules have been implemented in KGD to facilitate translational and applied research in kiwifruit,which include JBrowse,a popular genome browser,and the NCBI BLAST sequence search tool.Other notable tools developed within KGD include a genome synteny viewer and tools for differential gene expression analysis as well as gene ontology(GO)term and pathway enrichment analysis. | Junyang Yue Jiacheng Liu Wei Tang Ya Qing Wu Xiaofeng Tang Wei Li Ying Yang Lihuan Wang Shengxiong Huang Congbing Fang Kun Zhao Zhangjun Fei Yongsheng Liu Yi Zheng | 2020 | Horticulture Research2020,7,1: | 6 |
| 3 | Grapevine VlbZIP30 improves drought resistance by directly activating VvNAC17 and promoting lignin biosynthesis through the regulation of three peroxidase genes显示文摘Drought stress severely affects grapevine quality and yield,and recent reports have revealed that lignin plays an important role in protection from drought stress.Since little is known about lignin-mediated drought resistance in grapevine,we investigated its significance.Herein,we show that VlbZIP30 mediates drought resistance by activating the expression of lignin biosynthetic genes and increasing lignin deposition.Transgenic grapevine plants overexpressing VlbZIP30 exhibited lignin deposition(mainly G and S monomers)in the stem secondary xylem under control conditions,which resulted from the upregulated expression of VvPRX4 and VvPRX72.Overexpression of VlbZIP30 improves drought tolerance,characterized by a reduction in the water loss rate,maintenance of an effective photosynthesis rate,and increased lignin content(mainly G monomer)in leaves under drought conditions.Electrophoretic mobility shift assay,luciferase reporter assays,and chromatin immunoprecipitation-qPCR assays indicated that VlbZIP30 directly binds to the G-box cis-element in the promoters of lignin biosynthetic(VvPRX N1)and drought-responsive(VvNAC17)genes to regulate their expression.In summary,we report a novel VlbZIP30-mediated mechanism linking lignification and drought tolerance in grapevine.The results of this study may be of value for the development of molecular breeding strategies to produce drought-resistant fruit crops. | Mingxing Tu Xianhang Wang Wuchen Yin Ya Wang Yajuan Li Guofeng Zhang Zhi Li Junyang Song Xiping Wang | 2020 | Horticulture Research2020,7,1: | 5 |
| 4 | Analysis of genotypes and phenotypes in Chinese children with tuberous sclerosis complex显示文摘Tuberous sclerosis complex(TSC) is a neurocutaneous syndrome with serious clinical presentations, an autosomal dominant genetic disorder involving multiple organs and systems. We retrospectively investigated the clinical manifestations and genotypes of 20 Chinese children with TSC to enable informed diagnostic and surveillance recommendations in China. A retrospective analysis of clinical manifestations in 20 children(7.00±5.30 years old) with TSC was conducted. A genetic testing of the genes TSC1 and TSC2 was performed in 14 children.The earliest manifestations of TSC were skin lesions(80% of patients) and seizures(75%). Fourteen of the children presented with retinal hamartomas, and 2 of these underwent eye enucleation at other hospitals through misdiagnosis. On magnetic resonance imaging, 18 children exhibited subependymal nodules, and 16 ones showed cortical nodules. 5 cases of non-renal hamartomas, 5 cases of multiple renal cysts, and 5 cases of cardiac rhabdomyomas were observed.The genotyping of TSC1 and TSC2 in 14 children revealed 11 with mutations in TSC2, 2 with mutations in TSC1, and no mutations of either gene in one patient. Eight of these observed mutations are reported here in for the first time. The illness presentations of the TSC2-mutated patients were more severe than that of patients carrying TSC1 mutations.There were differences in the mutations of TSC genes in Chinese children from those reported in other countries. | 5 | Optimizing Building Envelope Dimensions for Passive Solar Houses in the Qinghai-Tibetan Region: Window to Wall Ratio and Depth of Sunspace显示文摘It has been a focus to reduce the energy consumption and improve the space heating performance of high-altitude buildings in winter seasons. In view of the abundant solar energy resources of the high-altitude region, the establishment of passive solar houses should be an effective strategy to deal with the problem of thermal comfort. Both window to wall ratio(WWR) and sunspace depth are of vital importance to determine the thermal comfort level of passive solar houses, while there are limited studies on analyzing their impacts on passive solar houses in high-altitude regions. Therefore, this study is designed to examine how WWR and sunspace depth affect space heating of passive solar houses in the Qinghai-Tibetan region. To be specific, the hourly radiation temperature variations and percentages of dissatisfaction of the residential building with different sunspace depth/WWR(including 0.9 m/33%, 0.9 m/45%, 0.9 m/60%, 1.2 m/33% and 1.5 m/33%) were quantitatively examined. Results indicated that under the condition of 0.9 m/45%, the overall average radiation temperature of the building was approximately 16°C during the entire heating season, which could better satisfy the heating requirements. Meanwhile, the average temperature was higher, and the thermal comfort level was better under the ratio of 45% or the depth of 1.5 m, when only an individual factor in either ratio or depth was considered. | 6 | Diagnosis and treatment of infantile malignant solid tumors in Beijing,China:A multicenter 10-year retrospective study显示文摘Importance:Cancer is the main cause of death by disease in children.Children experience the highest incidence of cancer in the first year of life.However,there is no comprehensive registration system for children with tumors in China.Objective:To summarize the diagnosis and treatment of infant cancer and analyze the status of standardized diagnosis and management among several treatment centers in Beijing,China,thereby providing evidence to guide further clinical research.Methods:From January 1,2010 to December 31,2019,patients with newly diagnosed infantile malignant solid tumors were admitted to six large tertiary pediatric solid tumor diagnosis and treatment centers in Beijing.The epidemiology,clinical features,and therapeutic effects of tumors in these patients were analyzed retrospectively.All patients were followed up until March 31,2020.Results:In total,938 patients were enrolled in this study.There were 530 boys(56.5%)and 408 girls(43.5%);the median age was 6.0 months(range,0-12.0 months).The three most common tumors were retinoblastoma in 366 patients(39.0%),neuroblastoma in 266 patients(28.4%),hepatoblastoma in 133 patients(14.2%),and central nervous system tumors in 52 patients(5.5%).The estimated 5-year overall survival rate was 81.3%±1.8%,and the 5-year event-free survival rate was 71.8%±2.9%.The 5-year overall survival rates of non-rhabdomyosarcoma soft tissue sarcoma,neuroblastoma,and retinoblastoma were 100%,88%±2.2%,and 86.9%±2.1%,respectively.The 5-year event-free survival rates were 81.1%±2.7%for neuroblastoma,81.6%±9.8%for non-rhabdomyosarcoma soft tissue sarcoma,and 72.7%±14.1%for extracranial malignant germ cell tumors.Interpretation:The three most common infantile malignant solid tumors were retinoblastoma,neuroblastoma,and hepatoblastoma.Multidisciplinary combined diagnosis and treatment is needed for infantile tumors. | Mei Jin Zhi Tian Yao Xie Zhaoxia Zhang Miao Li YaxiongYu Weiling Zhang Junyang Zhao Huanmin Wang Qi Zeng Long Li Ming Ge Ning Sun Xiaolun Zhang Jian Gong Wanshui Wu Rong Liu Weihong Zhao Dongsheng Huang Xiaoli Ma | 2020 | Pediatric Investigation2020,4,3: | 3 |

| 10 | A functional missense variant in ITIH3 affects protein expression and neurodevelopment and confers schizophrenia risk in the Han Chinese population显示文摘The Psychiatric Genomics Consortium(PGC)has recently identified 10 potential functional coding variants for schizophrenia.However,how these coding variants confer schizophrenia risk remains largely unknown.Here,we investigate the associations between eight potential functional coding variants identified by PGC and schizophrenia in a large Han Chinese sample(n=4022 cases and 9270 controls).Among the eight tested single nucelotide polymorphisms(SNPs),rs3617(a missense variant,p.K315 Q in the ITIH3 gene)showed genome-wide significant association with schizophrenia in the Han Chinese population(P=8.36×10-16),with the same risk allele as in PGC.Interestingly,rs3617 is located in a genomic region that is highly evolutionarily conserved,and its schizophrenia risk allele(C allele)was associated with lower ITIH3 mRNA and protein expression.Intriguingly,mouse neural stem cells stably overexpressing ITIH3 with different alleles of rs3617 exhibited significant differences in proliferation,migration,and differentiation,suggesting the impact of rs3617 on neurodevelopment.Subsequent transcriptome analysis found that the differentially expressed genes in neural stem cells stably overexpressing different alleles of rs3617 were significantly enriched in schizophrenia-related pathways,including cell adhesion,synapse assembly,MAPK and PI3 K-AKT pathways.Our study provides convergent lines of evidence suggesting that rs3617 in ITIH3 likely affects protein function and neurodevelopment and thereby confers risk of schizophrenia. | Kaiqin Li Yifan Li Junyang Wang Yongxia Huo Di Huang Shiwu Li Jiewei Liu Xiaoyan Li Rong Liu Xiaogang Chen Yong-Gang Yao Ceshi Chen Xiao Xiao Ming Li Xiong-Jian Luo | 2020 | Journal of Genetics and Genomics2020,47,5: | 1 |

| 17 | Federated learning-based short-term building energy consumption prediction method for solving the data silos problem显示文摘Transfer learning is an effective method to predict the energy consumption of information-poor buildings by learning transferable knowledge from operational data of information-rich buildings.However,it is not recommended to directly use the operational data without protection due to the risk of leaking occupants’privacy.To address this problem,this study proposes a federated learning-based method to learn transferable knowledge from building operational data without privacy leaking.It trains a transferable federated model based on the operational data from the buildings similar to the target building with limited data.An advanced secure aggregation algorithm is adopted in the training process to ensure that no one can infer private information from the training data.The federated model obtained is evaluated by comparing it with the standalone model without federated learning based on 13 similar office buildings from the Building Data Genome Project.The results show that the federated model outperforms the standalone model concerning the prediction accuracy and training time.On average,the federated model achieves a 25.4%decrease in CV-RMSE when the target building has limited operational data.Even if the target building has no operational data,the federated model still achieves acceptable accuracy(CV-RMSE is 22.2%).Meanwhile,the training time of the federated model is 90%less than that of the standalone model.The research insights can help develop federated learning-based methods for solving the data silos problem in building energy management.The methodology and analysis procedures are reproducible and all codes and data sets are available on Github. | Junyang Li Chaobo Zhang Yang Zhao Weikang Qiu Qi Chen Xuejun Zhang | 2022 | Building Simulation2022,15,6: | 1 |
