1间接荧光抗体法快速诊断海兰褐蛋鸡J亚群禽白血病的研究显示文摘某地区海兰褐蛋鸡发生肿瘤性传染病 ,经临床症状和病理学检查 ,在肝、脾、肾、卵巢、输卵管、肺、骨髓、腺胃、肠道等可见胞浆内充满球形嗜酸性颗粒的骨髓瘤细胞 ,呈灶状或弥漫性的分布。该变化是禽骨髓细胞瘤病病理学的特征性变化 ,具有证病意义。经用特异性抗 J亚群白血病病毒囊膜蛋白 gp85的单克隆抗体的免疫组化方法 ,在病料组织切片上检出病毒抗原 ,从而确诊为蛋鸡 J亚群禽白血病。本研究是尝试用间接荧光抗体法来快速诊断该病。采用特异性抗 J亚群禽白血病病毒囊膜蛋白 gp85单克隆抗体对组织触片作间接荧光抗体试验 ,检测 J亚群禽白血病病毒抗原。在骨髓、食管、肌肉、输卵管、肾都出现了范围广而且很明亮的荧光 ;肝、肺、脾的荧光较为明亮 ;心脏的荧光较弱 ,但清晰可见。表明在病料组织中存在特异性病毒抗原。徐镔蕊 董卫星 何召庆 冯小宇 余春明 张丽 任艳丽 L F LEE Mao-xiang LI 2002中国兽医杂志2002,38,9:49
2Evolving concepts in bone infection: redefining “biofilm”,“acute vs. chronic osteomyelitis”, “the immune proteome” and “local antibiotic therapy”显示文摘Osteomyelitis is a devastating disease caused by microbial infection of bone. While the frequency of infection following elective orthopedic surgery is low, rates of reinfection are disturbingly high. Staphylococcus aureus is responsible for the majority of chronic osteomyelitis cases and is often considered to be incurable due to bacterial persistence deep within bone. Unfortunately, there is no consensus on clinical classifications of osteomyelitis and the ensuing treatment algorithm. Given the high patient morbidity,mortality, and economic burden caused by osteomyelitis, it is important to elucidate mechanisms of bone infection to inform novel strategies for prevention and curative treatment. Recent discoveries in this field have identified three distinct reservoirs of bacterial biofilm including: Staphylococcal abscess communities in the local soft tissue and bone marrow, glycocalyx formation on implant hardware and necrotic tissue, and colonization of the osteocyte-lacuno canalicular network(OLCN) of cortical bone. In contrast, S.aureus intracellular persistence in bone cells has not been substantiated in vivo, which challenges this mode of chronic osteomyelitis. There have also been major advances in our understanding of the immune proteome against S. aureus, from clinical studies of serum antibodies and media enriched for newly synthesized antibodies(MENSA), which may provide new opportunities for osteomyelitis diagnosis, prognosis, and vaccine development. Finally, novel therapies such as antimicrobial implant coatings and antibiotic impregnated 3D-printed scaffolds represent promising strategies for preventing and managing this devastating disease. 10Reproducibility of thrombus volume quantification in multicenter computed tomography pulmonary angiography studies显示文摘AIM To evaluate reproducibility of pulmonary embolism(PE) clot volume quantification using computed tomography pulmonary angiogram(CTPA) in a multicenter setting.METHODS This study was performed using anonymized data in conformance with HIPAA and IRB Regulations(March 2015-November 2016). Anonymized CTPA data was acquired from 23 scanners from 18 imaging centers using each site's standard PE protocol. Two independent analysts measured PE volumes using a semi-automated region-growing algorithm on an FDA-approved image analysis platform. Total thrombus volume(TTV) was calculated per patient as the primary endpoint. Secondary endpoints were individual thrombus volume(ITV), Qanadli score and modified Qanadli score per patient. Inter-and intra-observer reproducibility were assessed using intra-class correlation coefficient(ICC) and BlandAltman analysis. RESULTS Analyst 1 found 72 emboli in the 23 patients with a mean number of emboli of 3.13 per patient with a range of 0-11 emboli per patient. The clot volumes ranged from 0.0041-47.34 cm3(mean +/-SD, 5.93 +/-10.15 cm3). On the second read, analyst 1 found the same number and distribution of emboli with a range of volumes for read 2 from 0.0041 – 45.52 cm3(mean +/-SD, 5.42 +/-9.53 cm3). Analyst 2 found 73 emboli in the 23 patients with a mean number of emboli of 3.17 per patient with a range of 0-11 emboli per patient. The clot volumes ranged from 0.00459-46.29 cm3(mean +/-SD, 5.91 +/-10.06 cm3). Inter-and intraobserver variability measurements indicated excellent reproducibility of the semi-automated method for quantifying PE volume burden. ICC for all endpoints was greater than 0.95 for inter-and intra-observer analysis. 11Increased catabolism and decreased unsaturation of ganglioside in patients with inflammatory bowel disease显示文摘AIM: To investigate whether accelerated catabolism of ganglioside and decreased ganglioside content contribute to the etiology of pro-inflammatory intestinal disease. METHODS: Intestinal mucosa from terminal ileum or colon was obtained from patients with ulcerative colitis or inflammatory Crohn's disease(n = 11) undergoing bowel resection and compared to control samples of normal intestine from patients with benign colon polyps(n = 6) and colorectal cancer(n = 12) in this observational case-control study. Gangliosides and phospholipids of intestinal mucosa were characterized by class and ceramide or fatty acid composition using liquid chromatography triple-quad mass spectrometry. Content and composition of ganglioside classes GM1, GM3, GD3, GD1 a, GT1 and GT3 were compared among subject groups. Content and composition of phospholipid classes phosphatidylcholine(PC) and phosphatidylethanolamine were compared among subject groups. Unsaturation index of individual ganglioside and phospholipid classes was computed and compared among subject groups. Ganglioside catabolism enzymes beta-hexosaminidase A(HEXA) and sialidase-3(NEU3) were measured in intestinal mucosa using western blot and compared among subject groups. RESULTS: Relative GM3 ganglioside content was 2-fold higher(P < 0.05) in intestine from patients with inflammatory bowel disease(IBD) compared to control intestine. The quantity of GM3 and ratio of GM3/GD3 was also higher in IBD intestine than control tissue(P < 0.05). Control intestine exhibited 3-fold higher(P < 0.01) relative GD1 a ganglioside content than IBD intestine. GD3 and GD1 a species of ganglioside containing three unsaturated bonds were present in control intestine, but were not detected in IBD intestine. The relative content of PC containing more than two unsaturated bonds was 30% lower in IBD intestine than control intestine(P < 0.05). The relative content of HEXA in IBD intestine was increased 1.7-fold(P < 0.05) and NEU3 was increased 8.3-fold(P < 0.01) compared to normal intestine. 12丁醇同分异构体/汽油混合燃料的燃烧和非常规污染物特性的试验研究(英文)显示文摘本文研究了丁醇同分异构体与汽油混合燃料在进气道喷射的火花点火发动机中的燃烧和排放特性。试验在发动机转速为1200 r/min、发动机负荷为3 bar BMEP 和5 bar BMEP 以及不同混合气空燃比条件下(φ=0.83~1.25)进行,使用了70%体积比的纯汽油和30%体积比的丁醇同分异构体混合燃料(N30,S30,I30 和T30)。结果表明,相比于纯汽油,所有的丁醇同分异构体/汽油混合燃料表现出更高的缸内压力,其中N30 有最高的缸内压力峰值和最早的缸内压力峰值相位;T30 表现出较高的brake燃油消耗率(BSFC)和较低的制动热效率(BTE);相比于其他燃料,N30 的UHC 排放较低而I30 的CO排放较高。在非常规排放物特性方面,相比于纯汽油,丁醇同分异构体/汽油混合燃料有较高的乙醛排放;相比于其他混合燃料,N30 的1,3-丁二烯排放较高。使用丁醇同分异构体/汽油混合燃料可以有效地抑制单环芳烃类质(BTEX)的排放。李元绪 宁智 闫峻豪 Timothy H LEE Chia-fon F LEE 2019Journal of Central South University2019,26,8:3

16Analyzing the H19-and T65-epitopes in 38 kd phosphory-lated protein of Marek's disease viruses and comparing chicken immunological reactions to viruses point-mutated in the epitopes显示文摘DNA sequencing analysis in 38 kd phosphorylated protein (pp38) ORF of Marek's disease viruses (MDV) indicated that all tested 10 virulent strains with different pathotypes had 'A' at base #320 and glutamine at aa#107 while reacted with monoclonal antibody (Mab) H19 in indirect fluorescence antibody test (IFA). However, vaccine strain CVI988 had 'G' at base#320 and arginine at aa#107 instead, when it was negative in IFA with Mab H19. Some strains were also reactive with Mab T65 in IFA while there was 'G' at base #326 and glycine at aa#109, but the other strains, which had 'A' at base #326 and glutamic acid at aa#109, did not react with Mab T65. By comparison of CVI988 to its point mutants CVI/rpp38(AG) and CVI/rpp38(AA) with 1 or 2 base(s) changes at bases #320 and /or #326 of pp38 gene for their reactivity with Mab H19 and T65, it was confirmed that the glutamine at aa#107 and glycine at aa#109 were critical to epitopes H19 and T65 respectively. Immuno-reactions to MDV were compared in SPF chickens inoculated with cloned CVI988 and its mutant CVI/rpp38(AG). It was found that antibody responses to MDV in chickens inoculated with CVI/rpp38(AG) were delayed and significantly lower than that in chickens inoculated with the native CVI988. 20马立克氏病病毒MEQ蛋白对MDV增殖影响的研究显示文摘在马立克氏病病毒 MDV致病机理的研究中 ,弄清致病基因 meq与病毒增殖之间的关系及其分子机制是十分重要的基础。以重组反转录病毒 (RCAS meq)感染鸡胚成纤维细胞 (CEF) ,使源于 MDV强毒参考株 (v MDV) GA株的 meq基因表达于宿主细胞内 ,然后再用 GA株感染这些细胞。通过利用 MDV强毒株特异的抗 pp3 8单克隆抗体所进行的“黑斑”试验以确定 MDV的增殖水平 ,并与未接种 RCAS meq的CEF进行比较。研究结果发现 ,细胞内表达的 meq基因产物可促进 GA株于体外培养细胞中的感染与增殖(病毒斑数增多 )。根据试验的结果 ,作者认为 meq基因在感染细胞内的表达水平是 MDV增殖以及进而能致病。韦平 Lucy F Lee 2002广西农业生物科学2002,21,1:2
