| 2 | High-throughput screening against thioredoxin glutathione reductase identifies novel inhibitors with potential therapeutic value for schistosomiasis显示文摘Background:Schistosomiasis,a parasitic disease also known as bilharzia and snail fever,is caused by different species of flatworms,such as Schistosoma mansoni(S.mansoni).Thioredoxin glutathione reductase(TGR)from S.mansoni(SmTGR)is a well-characterized drug target for schistosomiasis,yet no anti-SmTGR compounds have reached clinical trials,suggesting that therapeutic development against schistosomiasis might benefit from additional scaffolds targeting this enzyme.Methods:A high-throughput screening(HTS)assay in vitro against SmTGR was developed and applied to a diverse compound library.SmTGR activity was quantified with ThioGlo®,a reagent that fluoresces upon binding to the free sulfhydryl groups of the reaction product GSH(reduced glutathione).Results:We implemented an HTS effort against 59,360 synthetic compounds.In the primary screening,initial hits(928 or 1.56%)showing greater than 90%inhibition on SmTGR activity at a final concentration of 10μM for each compound were identified.Further tests were carried out to confirm the effects of these hits and to explore the concentration-dependent response characteristics.As a result,74 of them(0.12%)representing 17 chemical scaffolds were confirmed and showed a great concentration-dependent inhibitory trend against SmTGR,including structures previously shown to be lethal to schistosomal growth.Of these,two scaffolds displayed a limited structure-activity relationship.When tested in cultured larvae,39 compounds had cidal activity in 48 h,and five of them killed larvae completely at 3.125μM.Of these,three compounds also killed adult worms ex vivo at concentrations between 5μM and 10μM.Conclusion:These confirmed hits may serve as starting points for the development of novel therapeutics to combat schistosomiasis. | Ting Li Peter D.Ziniel Pan-qing He Valerie P.Kommer Gregory J.Crowther Min He Qing Liu Wesley C.Van Voorhis David L.Williams Ming-Wei Wang | 2015 | Infectious Diseases of Poverty2015,4,1: | 3 |

The SIR model has important reference value for thenovel coronavirus epidemic, offering both preventive measures and the ability topredict future trends. Based on an analysis of the classical epidemiological SIRmodel along with key parameters, this paper aims to analyze the patterns ofCOVID-19, to discuss potential anti-COVID-19 measures, and to explain whywe need to conduct appropriate measures against COVID-19. The use of theSIR model can play an important role in public health emergencies. | 9 | Early Mesoproterozoic evolution of midcontinental Laurentia: Defining the geon 14 Baraboo orogeny显示文摘New geochronologic data from midcontinental Laurentia demonstrate that emplacement of the 1476-1470 Ma Wolf River granitic batholith was not an isolated igneous event,but was accompanied by regional metamorphism,deformation,and sedimentation.Evidence for such metamorphism and deformation is best seen in siliciclastic sedimentary rocks of the Baraboo Interval,which were deposited closely following the 1.65-1.63 Ga Mazatzal orogeny.In Baraboo Interval strata,muscovite parallel to slatey cleavage,in hydrothermal veins,in quartzite breccia,and in metamorphosed paleosol yielded ^(40)Ar/^(39)Ar plateau ages of 1493-1465 Ma.In addition,U-Th-total Pb dating of neoblastic overgrowths on detrital monazite gave an age of 1488±20 Ma,and recrystallized hematite in folded metapelite gave a mean U/Th-He age of 1411±39 Ma.Post-Baraboo,arkosic polymictic conglomerate,which contains detrital zircon with a minimum peak age of 1493 Ma,was intruded by a 1470 Ma granite porphyry at the northeastern margin of the Wolf River batholith.This episode of magmatism,regional deformation and metamorphism,and sedimentation,which is designated herein as the Baraboo orogeny,provides a midcontinental link between the Picuris orogeny to the southwest and the Pinware orogeny to the northeast,completing the extent of early Mesoproterozoic(Calymmian)orogenesis for 5000 km along the southern margin of Laurentia.This transcontinental orogen is unique among Precambrian orogenies for its great width(-1600 km),the predominance of ferroan granites derived from partial melting of lower continental crust,and the prevalence of regional high T-P metamorphism related to advective heating by granitic magmas emplaced in the middle to upper crust. | L.Gordon Medaris Jr Brad S.Singer Brian R.Jicha David H.Malone Joshua J.Schwartz Esther K.Stewart Amanda Van Lankvelt Michael L.Williams Peter W.Reiners | 2021 | Geoscience Frontiers2021,12,5: | 0 |
