| 1 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 7 | The effects of nitrogen fertilizer application on methane and nitrous oxide emission/uptake in Chinese croplands显示文摘The application of nitrogen(N) fertilizer to increase crop yields has a significant influence on soil methane(CH_4) and nitrous oxide(N_2O) emission/uptake.A meta-analysis was carried out on the effect of N application on(i) CH_4 emissions in rice paddies,(ii) CH_4 uptake in upland fields and(iii) N_2O emissions.The responses of CH_4 emissions to N application in rice paddies were highly variable and overall no effects were found.CH_4 emissions were stimulated at low N application rates(<100 kg N ha^(-1)) but inhibited at high N rates(>200 kg N ha^(-1)) as compared to no N fertilizer(control).The response of CH_4 uptake to N application in upland fields was 15%lower than control,with a mean CH_4 uptake factor of-0.001 kg CH_4-C kg^(-1) N.The mean N_2O emission factors were 1.00 and 0.94%for maize(Zea mays) and wheat(Triticum aestivum),respectively,but significantly lower for the rice(Oryza sativa)(0.51%).Compared with controls,N addition overall increased global warming potential of CH_4 and N_2O emissions by 78%.Our result revealed that response of CH_4 emission to N input might depend on the CH_4concentration in rice paddy.The critical factors that affected CH_4 uptake and N_2O emission were N fertilizer application rate and the controls of CH_4 uptake and N_2O emission.The influences of application times,cropping systems and measurement frequency should all be considered when assessing CH_4 and N_2O emissions/uptake induced by N fertilizer. | SUN Bin-feng ZHAO Hong LU Yi-zhong Lü Fei WANG Xiao-ke | 2016 | Journal of Integrative Agriculture2016,15,2: | 18 |
| 8 | Formation of complexes of Rubisco-Rubisco activase from La^3+, Ce^3+ treatment spinach显示文摘The complex of Rubisco and Rubisco activase from LaCl3-, CeCl3- treated spinach in vivo is induced. SDS-PAGE result shows that the purified proteins from LaCl3-, CeCl3- treated spinach have not only large and small subunits (55 kD, 14.4 kD) of Rubisco, but also two large subunits of 45 kD and 41 kD near the large subunits of Rubisco. Native-PAGE shows that the purified proteins from LaCl3-, CeCl3- treated spinach have not only a band of Rubisco (560 kD), but also a band of about 1100 kD, about twice distant from Rubisco, which might be a complex of Rubisco and Rubisco activase. The purified enzyme activities from LaCl3-, CeCl3-treated spinach are 1.8 and 2.8 times that of the control,the intensities of absorption and fluorescence are sig- nificantly higher than that of the purified Rubisco from the control, and the total sulfhydryl groups and available sulfhydryl groups are 36—39 –SH per mol enzyme, 14—25 –SH per mol enzyme more than those of the purified Rubisco from the control, respectively. The CD spectra show that the secondary structure of the purified enzyme from LaCl3-, CeCl3 -treated spinach is very dif- ferent from the control. | 9 | Clinical significance of loss of heterozygosity for M6P/IGF2R in patients with primary hepatocellular carcinoma显示文摘AIM:To investigate the relationship between loss of heterozygosity (LOH) for mannose 6-phosphate/insulin- like growth factor 2 receptor (M6P/IGF2R) and the outcomes for primary HCC patients treated with partial hepatectomy. METHODS: The LOH for M6P/IGF2R in primary HCC patients was assessed using six different gene-specific nucleotide polymorphisms. The patients studied were enrolled to undergo partial hepatectomy. RESULTS: M6P/IGF2R was found to be polymorphic in 73.3% (22/30) of the patients, and of these patients, 50.0% (11/22) had tumors showing LOH in M6P/IGF2R. Loss of heterozygosity in M6P/IGF2R was associated with significant reductions in the two year overall survival rate (24.9% vs 65.5%; P = 0.04) and the disease-free survival rate (17.8% vs 59.3%; P = 0.03). | 11 | Clinicopathological, immunohistochemical, and ultrastructural study of 13 cases of melanotic schwannoma显示文摘Background Melanotic schwannoma is a rare variant of schwannoma composed of melanin-producing cellswith ultrastructural features of schwann cells.The description of the course of the tumors differs somewhat, but itis generally considered as a benign lesion.We investigated the clinicopathologic features, immunophenotypes,and ultrastructural features of 13 patients with nonpsammomatous melanotic schwannoma (NPMS).Methods Tumor specimens of each patient were sectioned and stained with hematoxylin-eosin, Fontana-Masson, Prussian blue, and periodic acid-Schiff (PAS).Immunohistochemical markers such as S-100, Leu-7,HMB-45, Melan-A, CK, EMA, vimentin, GFAP, laminin, collagenⅣ and MIB-1 were detected with theEnvision immunohistochemical staining method.Four of the cases were observed by electron microscopy.Results Of the 13 patients, 8 were male and 5 female, aged from 11 to 92 years (mean, 38.6 years).Thetumor sites included the spinal nerve root (5 patients), cranial nerve (1), greater omentum (1), subcutaneoustissue (3), mesentery (1), bone (1) and mediastinum(1).Eleven patients were followed up for over2 years,with a mean of 5.9 years.One patient (9.1%) with a primary tumor in the greater omentum developed anotherprimary tumor of the same type in the subcutaneous tissue of the abdominal wall after the first operation.Localrecurrence of the tumor was seen in2 patients (18.2%).One patient (9.1%) showed the local recurrence andmetastasis.Seven patients (63.6%) showed no evidence of the recurrence or metastasis.Grossly, all tumorswere well-circumscribed and the gross findings were suggestive of melanin-containing tumors.The tumor wascomposed of spindled and epithelioid cells with abundant intracytoplasmic melanin pigments.Nuclei were roundand contained delicate, evenly distributed chromatins as well as small, distinct nucleoli.In some areas, thenucleoli were large and prominent.Rare mitoses were seen in mostlesions exceptthe larger omentumlesion.Thepigment was shown to be positive for the Fontana-Masson and negative for Prussian blue and PAS.Immunohistochemical staining for S-100, Leu-7, HMB-45, Melan-A, and vimentin were strongly positive.Linear immunoreactions of both laminin and collagenⅣ was detected in all patients.Ultrastructurally, numerouselongated tumor-cell processes, duplicated basement membrane and melanosomes were observed in alldevelopmental stages.Conclusions Histologically, melanotic schwannoma is a rare variant of schwannoma composed of melanin-producing cells with ultrastructural features ofschwann cells.Distinguishing between this tumor andmalignant melanoma is of paramount importance inplanning of management. Immunohistochemically,combined use of laminin and collagenⅣ is valuablein distinguishing melanotic schwannoma frommalignant melanoma. Wide local resection andadditional radiotherapy should be advocated.Furtherstudies including cytogenetic or molecular biology arestill required to better delineate melanoticschwannoma from malignant melanoma. The Cm MLO2 gene was expressed in the wild germplasm after fluorescence-labeled Agrobacterium-mediated transformation. A positive transgenic plant showed successful invasion by powdery mildew Race1. | 20 | Projected changes in mean and interannual variability of surface water over continental China显示文摘Five General Circulation Model(GCM) climate projections under the RCP8.5 emission scenario were used to drive the Variable Infiltration Capacity(VIC) hydrologic model to investigate the impacts of climate change on hydrologic cycle over continental China in the 21 st century. The bias-corrected climatic variables were generated for the Fifth Assessment Report of the Intergovernmental Panel on Climate Change(IPCC AR5) by the Inter-Sectoral Impact Model Intercomparison Project(ISIMIP). Results showed much larger fractional changes of annual mean Evapotranspiration(ET) per unit warming than the corresponding fractional changes of Precipitation(P) per unit warming across the country, especially for South China, which led to a notable decrease of surface water variability(P-E). Specifically, negative trends for annual mean runoff up to -0.33%/ year and soil moisture trends varying between -0.02% to -0.13%/year were found for most river basins across China. Coincidentally, interannual variability for both runoff and soil moisture exhibited significant positive trends for almost all river basins across China, implying an increase in extremes relative to the mean conditions. Noticeably, the largest positive trends for runoff variability and soil moisture variability, which were up to 0.41%/year and 0.90%/year, both occurred in Southwest China. In addition to the regional contrast, intra-seasonal variation was also large for the runoff mean and runoff variability changes, but small for the soil moisture mean and variability changes. Our results suggest that future climate change could further exacerbate existing water-related risks(e.g., floods and droughts) across China as indicated by the marked decrease of surface water amounts combined with a steady increase of interannual variability throughout the 21 st century.
