| 1 | Fluctuations in butyrate-producing bacteria in ulcerative colitis patients of North India显示文摘AIM: To study the interplay between butyrate concentration and butyrate-producing bacteria in fecal samples of ulcerative colitis (UC) patients vs control individuals. METHODS: Fecal samples were collected from 14 control individuals (hemorrhoid patients only) and 26 UC patients (severe: n = 12, moderate: n = 6, remission: n = 8), recruited by the gastroenterologist at the Department of Gastroenterology, All India Institute of Medical Sciences, New Delhi, India. Disease activity in UC patients was determined by clinical colitis activity index. We employed fluorescent in situ hybridization in combination with flow cytometry to enumerate the clostridium cluster population targeted by 16S rRNA gene probe. Major butyrate-producing species within this cluster were quantified to see if any change existed in control vs UC patients with different disease activity. This observed change was further validated by quantitative polymerase chain reaction. In addition to this,we carried out gas chromatography to evaluate the changes in concentration of major short chain fatty acids (SCFAs), namely acetate, n -butyrate, iso -butyrate, in the above samples. Student t test and Graph pad prism-6 were used to compare the data statistically. RESULTS: There was a significant decrease of Clostridium coccoides (control, 25.69% ± 1.62% vs severe, 9.8% ± 2.4%, P = 0.0001) and Clostridium leptum clusters (control, 13.74% ± 1.05% vs severe, 6.2% ± 1.8%, P = 0.0001) in fecal samples of UC patients. Furthermore, we demonstrated that some butyrateproducing members of the clostridial cluster, like Fecalibacterium prausnitzii (control, 11.66% ± 1.55% vs severe, 6.01% ± 1.6%, P = 0.0001) and Roseburia intestinalis (control, 14.48% ± 1.52% vs severe, 9% ± 1.83%, P = 0.02) were differentially present in patients with different disease activity. In addition, we also demonstrated decreased concentrations of fecal SCFAs, especially of n -butyrate (control, 24.32 ± 1.86 mmol/μL vs severe, 12.74 ± 2.75 mmol/μL, P = 0.003), iso -butyrate (control, 1.70 ± 0.41 mmol/μL vs severe, 0.68 ± 0.24 mmol/μL, P = 0.0441) and acetate (control, 39.51 ± 1.76 mmol/μL vs severe, 32.12 ± 2.95 mmol/ μL,P = 0.047), in the fecal samples of UC patients. The observed decrease of predominant butyrate producers of clostridial clusters correlated with the reduced SCFA levels in active UC patients. This was further confirmed by the restoration in the population of some butyrate producers with simultaneous increase in the level of SCFA in remission samples. | 5 | Association between obesity-related adipokines and colorectal cancer:A case-control study and meta-analysis显示文摘AIM:To examine the association between obesityrelated adipokines(adiponectin,leptin,resistin,interleukin-6(IL-6),and tumor necrosis factor-α(TNF-α)and colorectal cancer(CRC)risk.METHODS:Serum levels of adipokines were measured in 100 CRC patients and age-and sex-matched controls for the data analysis.Unconditional logistic regression models were used for estimating ORs and95%CIs related to each adipokine.For the metaanalysis,studies published before July 2013 available on Medline/PubMed and EMBASE were retrieved.The analysis included a total of 17 relevant studies(including the present case-control study):nine studies on adiponectin and eight on leptin.The effect sizes of ORs and 95%CIs were estimated using RevMan 5.1.Heterogeneity was evaluated using Cochran’s Q-test and I2 statistics.RESULTS:Among the five adipokines,only resistin levels were significantly higher in cases than in controls(P<0.001).The case-control study results showed no association between adiponectin and CRC and a negative association between leptin and CRC.However,the results of the meta-analysis showed a significant inverse association between adiponectin and CRC(OR=0.91,95%CI:0.83-1.00,P=0.04)and no association between CRC and leptin.After stratification by study design,an inverse association between adiponectin and CRC was observed in prospective studies only(OR=0.90,95%CI:0.82-0.99,P=0.03),whereas the association between leptin and CRC was inconsistent(prospective studies:OR=1.14,95%CI:1.02-1.27,P=0.02 and retrospective studies:OR=0.47,95%CI:0.29-0.74,P=0.001).The associations of resistin and TNF-αwith CRC risk were positive,but no association was observed for IL-6.CONCLUSION:Our results suggest a negative association of leptin,positive associations of resistin and TNF-α,and null associations of adiponectin and IL-6with CRC.However,further studies with larger number of prospective approaches are needed. | Rakhi Kumari Joshi Woo Jin Kim Sang-Ah Lee | 2014 | World Journal of Gastroenterology2014,20,24: | 11 |
| 6 | Marine Bacterial Biofilms in Bioremediation of Polycyclic Aromatic Hydrocarbons(PAHs) Under Terrestrial Condition in a Soil Microcosm显示文摘Polycyclic aromatic hydrocarbons(PAHs) in soil retain for a quite long period due to their hydrophobicity and aggregation properties. Biofilm-forming marine bacterial consortium(named as NCPR), composed of Stenotrophomonas acidaminiphila NCW702,Alcaligenes faecalis NCW402, Pseudomonas mendocina NR802, Pseudomonas aeruginosa N6P6, and Pseudomonas pseudoalcaligenes NP103, was used for the bioremediation of PAHs in a soil microcosm. Phenanthrene and pyrene were used as reference PAHs. Parameters that can affect PAH degradation, such as chemotaxis, solubility of PAHs in extracellular polymeric substances(EPS), and catechol2,3-dioxygenase(C23O) activity, were evaluated. P. aeruginosa N6P6 and P. pseudoalcaligenes NP103 showed chemotactic movement towards both the reference PAHs. The solubility of both the PAHs was increased with an increase in EPS concentration(extracted from all the 5 selected isolates). Significantly(P < 0.001) high phenanthrene(70.29%) and pyrene(55.54%) degradation was observed in the bioaugmented soil microcosm. The C23O enzyme activity was significantly(P < 0.05) higher in the bioaugmented soil microcosm with phenanthrene added at 173.26 ± 2.06 nmol min^(-1) mg^(-1) protein than with pyrene added at 61.80 ± 2.20 nmol min^(-1) mg^(-1) protein. The C23O activity and gas chromatography-mass spectrometer analyses indicated catechol pathway of phenanthrene metabolism. | 10 | Hepatoprotective activity of Terminalia paniculata against paracetamol induced hepatocellular damage in Wistar albino rats显示文摘Objective:To evaluate the hepatoprotective activity of Terminalia paniculata against paracetamol induced hepatic damage in rats.Methods:The plant material was shade dried, powdered and extracted with ethanol.Liv 52 and silymarin were used as standard drugs and 2%gum acacia as a control(vehicle).Alteration in the levels of biochemical markers of hepatic damage like AST,ALT,ALP and lipid peroxides were tested,and phytochemical tests were also performed.Results:Paracetamol(2 g/kg) increased the serum levels of alanine aminotransfer (ALT),aspartate aminotransferase(AST),alkaline phosphatase(ALP) and the lipid peroxides. | 11 | Life style modification improves insulin resistance and liver histology in patients with non-alcoholic fatty liver disease显示文摘AIM: To study the effect of regular aerobic exercise on insulin resistance, serum aminotransferase and liver histology in nonalcoholic fatty liver disease (NAFLD) patients. METHODS: Sixty (mean age 40.0 ± 8.5 years, 75% male) NAFLD patients were included in the study. After baseline anthropometric measurement i.e., body mass index (BMI), waist circumference (WC); all patients were advised regular aerobic exercise for 30 min/d, for at least 5 d/wk and trained to achieve around 70% of his maximal heart rate. In addition, moderately energy restricted diet was advised to patients with high BMI (> 25 kg/m 2 ). Monthly follow up was done by measuring BMI, WC, aspartate aminotransferase, and alanine aminotransferase (ALT). Insulin resistance was calculated using homeostasis model assessment (HOMA) of insulin resistance (HOMA-IR) model, at baseline and after 6mo. Insulin resistance was arbitrarily considered altered when it was ≥ 2. Liver biopsy was done in a section of patients at baseline and after 6 mo. RESULTS: Seventy percent (42/60) patients were overweight or obese; 95% (57/60) had central obesity (WC > 90 cm in men, > 80 cm in women). In the 45 exercise compliant patients insulin resistance decreased from 6.4 ± 6.1 to 1.3 ± 1.0, BMI from 26.7 ± 3.3 kg/m 2 to 25.0 ± 3.3 kg/m 2 , WC from 95.7 ± 8.9 cm to 90.8 ± 7.3 cm and ALT from 84.8 ± 43.5 U/L to 41.3 ± 18.2 U/L (P < 0.01). In 15 exercise noncompliant patient's insulin resistance, BMI, WC and ALT did not show significant change at 6 mo follow up. Six of 8 patients in compliant group on repeat liver biopsy showed significant change in steatosis and necroinflammation. Nonalcoholic steatohepatitis scores improved form 5.3 ± 1.5 to 3.35 ± 1.5. The decline in insulin resistance correlated with decline in ALT (P = 0.01, r s = 0.90) and liver histology (P = 0.03, r s = 0.73). The collection comprised of accessions from 11 wild species and African cultivated rice. The germplasm was screened for BPH following standard seed box screening technique in the greenhouse. As many as 159 accessions were identified as resistant during the year 2012 based on one year screening. A selected set of BPH resistant accessions were screened again during 2013. Based on the two years screening, seven accessions of O. nivara(AA), one accession of O. officinalis(CC), seven accessions of O. australiensis(EE), five accessions of O. punctata(BB and BBCC) and nine accessions of O. latifolia(CCDD) were confirmed to be resistant to BPH. Undoped and N doped ZnO nanoparticles were synthesized via chemical precipitation approach. The prepared samples were characterized through X-ray diffraction(XRD),Transmission electron microscopy(TEM) equipped with Energy dispersive X-ray(EDAX) spectroscopy, UV–visible spectroscopy, Fourier transform infrared(FTIR) spectroscopy and micro-Raman spectroscopy(m RS). Wurtzite phase of undoped as well as 0.5–10% N doped ZnO nanoparticles was con fi rmed through characteristic XRD patterns. The particle size expansion due to N incorporation in ZnO was further revealed by TEM and EDAX analysis where 11 nm size undoped and 18–22 nm size 0.5–10% N doped ZnO(N:ZnO) nanoparticles without any impurity were ascertained. Slight blue-shift in band gap energy, as observed in our case, symbolized weak quantum con fi nement of the prepared nanoparticles. The alterations in vibrational modes of ZnO due to N incorporation, remarkably H substituting at O site and subsequently causing the passivation in N:ZnO nanoparticles, were detected through FTIR analysis. | 16 | Screening of Antioxidant Potential of Selected Barks of Indian Medicinal Plants by Multiple in vitro Assays显示文摘Objective To evaluate the antioxidant potential in herbal extract barks of five therapeutically important medicinal plants native to India, i.e. Crataeva nurvala Buch.-Ham., Buchanania lanzan Spreng., Aegle marmelos Corr., Dalbergia sissoo Roxb. ex DC., and Cedrela toona Roxb. Methods Standardized aqueous alcoholic extracts from the selected barks having different target radicals, such as superoxide radical, nitric oxide, ABTS radical, and peroxidative decomposition of phospholipids, were prepared and screened by multiple in vitro assays. These extracts were also tested for total phenolic and tannin content and correlated with antioxidant capacity. Results Total phenolic and tannin contents were found to be the highest in C. nurvala (195 GAE mg/g and 218.3 mg/g CE). SOD mimetic activity was found to be the highest in Crataeva nurvula, although all barks showed activity more than 100 units/mg extract. Lipid peroxidation inhibitory potential was found to be the highest in Crataeva nurvala (83.4% inhibition of MDA formation/10 μg extract), and also showed a comparatively high NO quenching capacity (45.5% per 10 μg extract). The highest NO quenching potential was found in Aegle marmelos (47.3% per 10 μg extract). Cedrela toona showed the lowest LPO inhibitory potential and NO quenching capacity (50.5% and 30.5%, respectively). Buchanania lanzan, a medicinal plant extensively used for inflammatory disorders and Dalbergia sissoo also showed 72.5% and 69.1% LPO inhibitory potential/10 μg extract. Trolox equivalent antioxidant capacity ranged from 0.24 to 0.39 mmol/L TEAC/mg extract, indicating that all the barks tested had ABTS+· radical quenching capacity. | 18 | Synthesis,spectroscopic characterization,DNA cleavage and antibacterial studies of a novel tridentate Schiff base and some lanthanide(III) complexes显示文摘A novel potential tridentate Schiff base was prepared by condensing equimolar quantities of 2-hydroxyacetophenone and 2-aminopyrimidine in methanol.This ligand was versatile in forming a series of complexes with lanthanide ions such as La(III),Pr(III),Nd(III),Sm(III),Gd(III),Dy(III) and Yb(III).The ligand and the metal complexes were characterized through elemental analysis,molar conductance,UV-Visible,IR,1H NMR,and mass spectral studies.The spectral studies indicated that the ligand was coordinated to the metal ion in neutral tridentate fashion through the azomethine nitrogen,one of the nitrogen atoms in the pyrimidine ring and the phenolic oxygen without deprotonation.Thermal decomposition and luminescence property of lanthanum(III) complex were also examined.The X-ray diffraction patterns showed the crystalline nature of the ligand and its lanthanum(III) complex.The DNA cleavage studies of the ligand and the metal complexes were carried out and it was observed that the lanthanum(III) and neodymium(III) complexes cleaved the pUC19 DNA effectively.The ligand and the metal complexes were screened for their antibacterial activities.The metal complexes were found to be more potent bactericides than the ligand. | K.Mohanan R.Aswathy L.P.Nitha Niecy Elsa Mathews B.Sindhu Kumari | 2014 | Journal of Rare Earths2014,32,4: | 4 |
| 19 | Er^(3+)/Yb^(3+)/Li^+/Zn^(2+):Gd_2(MoO_4)_3 upconverting nanophosphors in optical thermometry显示文摘Er^(3+)-Yb^(3+)-Li^+:Gd_2(MoO_4)_3 and Er^(3+)-Yb^(3+)-Zn^(2+):Gd_2(MoO_4)_3 nanophosphors, synthesized by chemical co-precipitation technique were characterized through XRD,FESEM,dynamic light scattering(DLS),diffuse reflectance, photoluminescence, photometric and decay time analysis. The enhancement of about~28, ~149 and ~351 times in the green upconversion emission band is observed for the optimized Er^(3+)-Yb^(3+),Er^(3+)-Yb^(3+)-Li^+ and Er^(3+)-Yb^(3+)-Zn^(2+):Gd_2(MoO_4)_3 nanophosphors in comparison to the singly Er^(3+) doped nanophosphors. The electric dipole-dipole interaction is found to be responsible for the concentration quenching. The temperature dependent behaviour of the two green thermally coupled levels of the Er^(3+) ions based on the fluorescence intensity ratio technique was studied. | 20 | Effect of Low Level Subchronic Microwave Radiation on Rat Brain显示文摘Objective The present study was designed to investigate the effects of subchronic low level microwave radiation(MWR) on cognitive function,heat shock protein 70(HSP70) level and DNA damage in brain of Fischer rats.Methods Experiments were performed on male Fischer rats exposed to microwave radiation for 90 days at three different frequencies: 900,1800,and 2450 MHz.Animals were divided into 4 groups: Group I: Sham exposed,Group II: animals exposed to microwave radiation at 900 MHz and specific absorption rate(SAR) 5.953 × 10-4 W/kg,Group III: animals exposed to 1800 MHz at SAR 5.835 × 10-4 W/kg and Group IV: animals exposed to 2450 MHz at SAR 6.672 × 10-4 W/kg.All the animals were tested for cognitive function using elevated plus maze and Morris water maze at the end of the exposure period and subsequently sacrificed to collect brain tissues.HSP70 levels were estimated by ELISA and DNA damage was assessed using alkaline comet assay.Results Microwave exposure at 900-2450 MHz with SAR values as mentioned above lead to decline in cognitive function,increase in HSP70 level and DNA damage in brain.Conclusion The results of the present study suggest that low level microwave exposure at frequencies 900,1800,and 2450 MHz may lead to hazardous effects on brain. | Pravin Suryakantrao Deshmukh Kanu Megha Namita Nasare Basu Dev Banerjee Rafat Sultana Ahmed Mahesh Pandurang Abegaonkar Ashok Kumar Tripathi Pramod Kumari Mediratta | 2016 | Biomedical and Environmental Sciences2016,29,12: | 4 |
