| 1 | Does combined therapy of curcumin and epigallocatechin gallate have a synergistic neuroprotective effect against spinal cord injury?显示文摘Systematic inflammatory response after spinal cord injury(SCI) is one of the factors leading to lesion development and a profound degree of functional loss. Anti-inflammatory compounds, such as curcumin and epigallocatechin gallate(EGCG) are known for their neuroprotective effects. In this study, we investigated the effect of combined therapy of curcumin and EGCG in a rat model of acute SCI induced by balloon compression. Immediately after SCI, rats received curcumin, EGCG, curcumin + EGCG or saline [daily intraperitoneal doses(curcumin, 6 mg/kg; EGCG 17 mg/kg)] and weekly intramuscular doses(curcumin, 60 mg/kg; EGCG 17 mg/kg)] for 28 days. Rats were evaluated using behavioral tests(the Basso, Beattie, and Bresnahan(BBB) open-field locomotor test, flat beam test). Spinal cord tissue was analyzed using histological methods(Luxol Blue-cresyl violet staining) and immunohistochemistry(anti-glial fibrillary acidic protein, anti-growth associated protein 43). Cytokine levels(interleukin-1β, interleukin-4, interleukin-2, interleukin-6, macrophage inflammatory protein 1-alpha, and RANTES) were measured using Luminex assay. Quantitative polymerase chain reaction was performed to determine the relative expression of genes(Sort1, Fgf2, Irf5, Mrc1, Olig2, Casp3, Gap43, Gfap, Vegf, NfκB, Cntf) related to regenerative processes in injured spinal cord. We found that all treatments displayed significant behavioral recovery, with no obvious synergistic effect after combined therapy of curcumin and ECGC. Curcumin and EGCG alone or in combination increased axonal sprouting, decreased glial scar formation, and altered the levels of macrophage inflammatory protein 1-alpha, interleukin-1β, interleukin-4 and interleukin-6 cytokines. | 2 | Epigenetic and non-epigenetic mode of SIRT1 action during oocyte meiosis progression显示文摘Background: SIRT1 histone deacetylase acts on many epigenetic and non-epigenetic targets. It is thought that SIRT1 is involved in oocyte maturation;therefore, the importance of the ooplasmic SIRT1 pool for the further fate of mature oocytes has been strongly suggested. We hypothesised that SIRT1 plays the role of a signalling molecule in mature oocytes through selected epigenetic and non-epigenetic regulation.Results: We observed SIRT1 re-localisation in mature oocytes and its association with spindle microtubules.In mature oocytes, SIRT1 distribution shows a spindle-like pattern, and spindle-specific SIRT1 action decreasesα-tubulin acetylation. Based on the observation of the histone code in immature and mature oocytes, we suggest that SIRT1 is mostly predestined for an epigenetic mode of action in the germinal vesicles(GVs) of immature oocytes. | 9 | SIRT1-dependent modulation of methylation and acetylation of histone H3 on lysine 9(H3K9)in the zygotic pronuclei improves porcine embryo development显示文摘Background: The histone code is an established epigenetic regulator of early embryonic development in mammals.The lysine residue K9 of histone H3(H3 K9) is a prime target of SIRT1, a member of NAD+-dependent histone deacetylase family of enzymes targeting both histone and non-histone substrates. At present, little is known about SIRT1-modulation of H3 K9 in zygotic pronuclei and its association with the success of preimplantation embryo development. Therefore, we evaluated the effect of SIRT1 activity on H3 K9 methylation and acetylation in porcine zygotes and the significance of H3 K9 modifications for early embryonic development.Results: Our results show that SIRT1 activators resveratrol and BML-278 increased H3 K9 methylation and suppressed H3 K9 acetylation in both the paternal and maternal pronucleus. Inversely, SIRT1 inhibitors nicotinamide and sirtinol suppressed methylation and increased acetylation of pronuclear H3 K9. Evaluation of early embryonic development confirmed positive effect of selective SIRT1 activation on blastocyst formation rate(5.2 ± 2.9% versus 32.9 ± 8.1% in vehicle control and BML-278 group, respectively; P ≤ 0.05). Stimulation of SIRT1 activity coincided with fluorometric signal intensity of ooplasmic ubiquitin ligase MDM2, a known substrate of SIRT1 and known limiting factor of epigenome remodeling.Conclusions: We conclude that SIRT1 modulates zygotic histone code, obviously through direct deacetylation and via non-histone targets resulting in increased H3 K9 me3. | 10 | Zinc phosphate-based nanoparticles as a novel antibacterial agent: in vivo study on rats after dietary exposure显示文摘Background: Development of new nanomaterials that inhibit or kil bacteria is an important and timely research topic. For example, financial losses due to infectious diseases, such as diarrhea, are a major concern in livestock productions around the world. Antimicrobial nanoparticles(NPs) represent a promising alternative to antibiotics and may lower antibiotic use and consequently spread of antibiotic resistance traits among bacteria, including pathogens.Results: Four formulations of zinc nanoparticles(Zn A, Zn B, Zn C, and Zn D) based on phosphates with spherical(Zn A, Zn B)or irregular(Zn C, Zn D) morphology were prepared. The highest in vitro inhibitory effect of our NPs was observed against Staphylococcus aureus(inhibitory concentration values, IC50, ranged from 0.5 to 1.6 mmol/L), fol owed by Escherichia coli(IC500.8–1.5 mmol/L). In contrast, methicil in resistant S. aureus(IC501.2–4.7 mmol/L) was least affected and this was similar to inhibitory patterns of commercial Zn O-based NPs and Zn O. After the successful in vitro testing, the in vivo study with rats based on dietary supplementation with zinc NPs was conducted. Four groups of rats were treated by 2,000 mg Zn/kg diet of Zn A, Zn B, Zn C, and Zn D, for comparison two groups were supplemented by 2,000 mg Zn/kg diet of Zn O-N and Zn O, and one group(control) was fed only by basal diet. The significantly higher(P < 0.05) Zn level in liver and kidney of al treated groups was found, nevertheless Zn NPs did not greatly influence antioxidant status of rats. However,the total aerobic and coliform bacterial population in rat feces significantly decreased(P < 0.05) in al zinc groups after 30 d of the treatment. | 14 | Zinc phosphate-based nanoparticles as alternatives to zinc oxide in diet of weaned piglets显示文摘Background: The high doses of zinc oxide(Zn O) administered orally to piglets for the prevention of diarrhea and increase of growth rate can contaminate pig farms and the surrounding environment. Therefore, there is a need to find a replacement of high doses of dietary Zn O with an equally effective alternative. In the present study, the effect of two formulations of zinc phosphate-based nanoparticles(Zn A and Zn C NPs) on growth performance,intestinal microbiota, antioxidant status, and intestinal and liver morphology was evaluated. A total of 100 weaned piglets were randomly divided into 10 equal groups with the base diet(control) or the base diet supplemented with Zn A, Zn C, or Zn O at concentrations 500, 1000, and 2000 mg Zn per kilogram of diet. Supplements were given to animals for 10 days. Fecal samples were collected on day 0, 5, 10 and 20. At the end of the treatment(day 10),three piglets from each group were sacrificed and analyzed.Results: Comparing to that of control, the significantly higher piglet weight gain was observed in all piglet groups fed with Zn A(P < 0.05). Differences in the total aerobic bacteria and coliform counts in piglet feces after NPs supplementation compared to that of control and Zn O groups were also found(P < 0.05). The majority of aerobic culturable bacteria from the feces represented Escherichia(28.57–47.62%), Enterococcus(3.85–35.71%), and Streptococcus(3.70–42.31%) spp. A total of 542 Escherichia coli isolates were screened for the virulence genes STa,STb, Stx2, F4, and F18. The substantial occurrence of E. coli virulence factors was found on day 5, mainly in fimbrillary antigen and thermostable toxins, except for piglets fed by Zn C. Zn treatment decreased Zn blood levels in piglets fed with Zn O and Zn A(500 mg/kg) and increased in Zn C(2000 mg/kg) compared to that of control(P < 0.05). The antioxidant status of piglets was affected only by Zn A. While some changes in the liver and the intestinal morphology of piglets with NPs were observed, none were serious as reflected by the normal health status and increased weigh gain performance.Conclusions: Our results indicate that Zn A NPs have a positive effect on the piglet growth performance even at the lowest concentration.
