1Underexpression of LATS1 TSG in colorectal cancer is associated with promoter hypermethylation显示文摘AIM:To investigate large tumor suppressor 1 (LATS1 ) expression, promoter hypermethylation, and microsatellite instability in colorectal cancer (CRC).METHODS:RNA was isolated from tumor tissue of 142 CRC patients and 40 colon mucosal biopsies of healthy controls. After reverse transcription, quantitative polymerase chain reaction (PCR) was performed, and LATS1 expression was normalized to expression of the ACTB and RPL32 housekeeping genes. To analyze hypermethylation, genomic DNA was isolated from 44 tumor CRC biopsies, and methylation-specific PCR was performed. Microsatellite instability (MSI) status was checked with PCR using BAT26, BAT25, and BAT40 markers in the genomic DNA of 84 CRC patients, followed by denaturing gel electrophoresis. RESULTS:Decreased LATS1 expression was found in 127/142 (89.4%) CRC cases with the average ratio of the LATS1 level 10.33 ± 32.64 in CRC patients vs 32.85 ± 33.56 in healthy controls. The lowest expression was found in Dukes' B stage tumors and G1 (welldifferentiated) cells. Hypermethylation of the LATS1 promoter was present in 25/44 (57%) CRC cases analyzed. LATS1 promoter hypermethylation was strongly associated with decreased gene expression; methylated cases showed 162× lower expression of LATS1 than unmethylated cases. Although high-grade MSI (mutation in all three markers) was found in 14/84 (17%) cases and low-grade MSI (mutation in 1-2 markers) was found in 30/84 (36%) cases, we found no association with LATS1 expression. CONCLUSION:Decreased expression of LATS1 in CRC was associated with promoter hypermethylation, but not MSI status. 3Simulation of Fluid Flow in a Channel Induced by Three Types of Fin-Like Motion显示文摘One of many interesting research activities in biofluidmechanics is dedicated to investigations of locomotion in water. Some of propulsion mechanisms observed in the underwater world are used in the development process of underwater auto-nomic vehicles(AUV). In order to characterise several solutions according to their manoeuvrability,influence on the sur-rounding fluid and energetic efficiency,a detailed analysis of fin-like movement is indispensable. In the current paper an analysis of undulatory,oscillatory and combined fin-like movements by means of numerical simulation is carried out. The conservation equation of mass and the conservation equation of momentum are solved with the Finite Volume Method(FVM) by use of the software CFX-10.0. The undulatory and oscillatory fin movements are modelled with an equation that is implemented within an additional subroutine and joined with the main solver. Numericalsimulations are carried out in the computational domain,in which one fin is fixed in a flow-through water duct. Simulations are carried out in the range of the Re number up to 105. 5Ultrabroadband wavelength-swept source based on total mode-locking of an Yb:CaF_(2)laser显示文摘We present an ultrabroadband, high-speed wavelength-swept source based on a self-modulated femtosecond oscillator. Photonic crystal fiber was pumped by a mode-locked Yb:CaF2 laser, resulting in a strong spectral broadening from 485 to 1800 nm. The pump laser cavity could be realigned in order to achieve total mode-locking of the longitudinal and transverse TEM00 and TEM01 electromagnetic modes. This led to spatial oscillations of the output beam, which induced modulation of the coupling efficiency to the fiber. Due to the fact that nonlinear spectral broadening was intensity dependent, this mechanism introduced wavelength sweeping at the fiber output. The sweeping rate could be adjusted between 7 and 21.5 MHz by changing the geometry of the pump cavity. By controlling the ratio of the transverse mode amplitudes, we were able to tune the sweeping bandwidth, eventually covering both the 1300 nm and 1700 nm bioimaging transparency windows. When compared with previously demonstrated wavelength-swept sources, our concept offers much broader tunability and higher speed. 8Fulminant liver failure following a marathon: Five case reports and review of literature显示文摘BACKGROUND The growing popularity of marathon and half-marathon runs has led to an increased number of patients presenting with exertion-induced heat stroke. Mild hepatic involvement is often observed in these patients;however, fulminant liver failure may occur in approximately 5% of all cases. Liver transplantation is a potentially curative approach for exertion-induced liver failure, although there is a lack of consensus regarding the criteria and optimal timing of this intervention. CASE SUMMARY This paper describes 5 patients (4 men and 1 woman) who were referred to the department where this study was performed with the diagnosis of exertioninduced acute liver failure. Three patients underwent liver transplantation, 1 recovered spontaneously, and 1 patient died on day 11 following the exertion. CONCLUSION Exertion-induced heat stroke may present as fulminant liver failure. These patients may recover with conservative treatment, may require liver transplantation, or may die.
