1The Distribution of Different Clades of Seneca Valley Viruses in Guangdong Province,China显示文摘Seneca Valley virus (SVV), a newly determined etiological agent of vesicular disease in swine, causes porcine idiopathic disease and occasional acute death in piglets. Recently, an increased number of SVV infection cases have been reported in the United States (US) and China, resulting in significant economic losses to the swine industry. The first identification of SVV in China was reported in Guangdong Province, a major swine producing province. The cases of SVV were continuously reported in Guangdong in 2015 and 2016. However, the spread of SVV in Guangdong in 2017 remains unknown.In this study, we determined two new SVV strains, CH-GD-2017-1 and CH-GD-2017-2, from Guangdong. The genetic analysis suggested that the two Guangdong strains showed different characteristics to previous Guangdong strains. They showed lower nucleotide similarity with strains isolated in 2015 and 2016, and were more similar to the US strains.Phylogenetic analyses indicated that the new strains were clustered in a different clade with previous Guangdong strains.We found 28 mutated amino acids in the new strains, compared with the first Guangdong strain, SVV CH-01-2015. In the geographic analysis, we found that the US and China reported more SVV cases than other countries, and most of the SVV cases were reported in east and central China—of which, Guangdong Province is one of the major epidemic regions. 2Further Investigation on the Role of Selenium Deficiency in the Aetiology and Pathogenesis of Keshan Disease显示文摘Selenium supplements were not able to restore the ultrastructural changes in the myocardiurn of latent Keshan disease patients taken by using cardiac catheter endomyocardial biopsy. Observations on the changes of seleniurn status and the incidence of Keshan disease showed that new latent and naturally-occurring chronic cases were found in the endemic area even after selenium levels had been elevated in the residents to the levels typical in the non-endemic area. These results indicate that although selenium deficiency might be a primary pathogenetic geogen in the occurrence of Keshan disease, it is rather a conditional predisposing factor than a specific or initiative aetiologic factor for the occurrence of Keshan disease. 3ExExperimental and numerical studies on the prediction of bendability limit of QSTE340 welded tube in NC bending process显示文摘For a low carbon steel tube with small wall factor D/t and bending radius R,the over-thinning induced localized necking is one dominant failure in tube numerical control(NC) bending process,which strongly restricts the bendability limit of the tube.In addition,the deterioration of bendability of a tube is increased by the existence of the weak weld region.Therefore,an important issue is how to determinate and predict the welded tube bendability limit.In the present study,a finite element(FE) model with weld and subdivided heat affected zones under ABAQUS platform is employed to explore the deformation behaviors of welded tube NC bending.A localized necking criterion based on the critical thickness thinning is used to predict the critical principal strains,critical bending radius and burst location during the forming process.It is found that the failures always occur at the rigid supporting point of mandrel flexible balls near the tangent point at the outside of the bend,where the wall thickness of the tube is the lowest.The bending limit curves(BLCs) of the QSTE340 welded tube are obtained by shifting the standard shaped forming limit curve to the critical principal strains along the major strain axis.Comparison between the numerical and experimental results has shown that the BLC and critical bending radius predictions agree well with the experimental results.In addition,the effect of weld positions on BLC is discussed,the weld region shows an almost negligible effect on the forming limit at a non-critical location that is far away from the outside of the bend.However,when the weld is at the large tensile deformation region on the outside of the bend,a decrease of the forming limit strains is seen.YANG He REN Ning ZHAN Mei ZHANG ZhiYong QIN YaTao JIANG HaoMin DIAO KeShan CHEN XinPing 2012Science China(Technological Sciences)2012,55,8:2
4Co-transplantation of macaque autologous Schwann cells and human embryonic nerve stem cells in treatment of macaque Parkinson's disease显示文摘Objective:To investigate the therapeutic effects of co-transplantation with Schwann cells(SCs) and human embryonic nerve stem cells(NSCs) on macaque Parkinson’s disease(PD).Methods: Macaque autologous SCs and human embryonic NSCs were adopled for the treatment of macaque PD.Results:Six months after transplantation,positron emission computerized tomography showed that 18F-FP-β-CIT was significantly concentrated in the injured striatum in the cotransplanted group.Immunohistoehemieal staining of transplanted area tissue showed migration of tyrosine hydroxylase positive cells from the transplant area to the surrounding area was significantly increased in the co-transplanted group.Conclusions:Co-transplantation of SCs and NSCs could effectively cure PD in macaques.SCs harvested from the autologous peripheral nerves can avoid rejection and the ethics problems,so it is expected to be applied clinically.Ying Xia Chengchuan Jiang Zuowei Cao Keshan Shi Yang Wang 2012Asian Pacific Journal of Tropical Medicine2012,5,1:1

14FoxJ1 inhibits African swine fever virus replication and viral S273R protein decreases the expression of FoxJ1 to impair its antiviral effect显示文摘African swine fever(ASF)is a highly pathogenic swine infectious disease that affects domestic pigs and wild boar,which is caused by the African swine fever virus(ASFV).ASF has caused huge economic losses to the pig industry and seriously threatens global food security and livestock health.To date,there is no safe and effective commercial vaccine against ASF.Unveiling the underlying mechanisms of ASFV-host interplay is critical for developing effective vaccines and drugs against ASFV.In the present study,RNA-sequencing,RT-qPCR and Western blotting analysis revealed that the transcriptional and protein levels of the host factor FoxJ1 were significantly down-regulated in primary porcine alveolar macrophages(PAMs)infected by ASFV.RT-qPCR analysis showed that overexpression of FoxJ1 upregulated the transcription of type I interferon and interferon stimulating genes(ISGs)induced by poly(dA:dT).FoxJ1 revealed a function to positively regulate innate immune response,therefore,suppressing the replication of ASFV.In addition,Western blotting analysis indicated that FoxJ1 degraded ASFV MGF505-2R and E165R proteins through autophagy pathway.Meanwhile,RT-qPCR and Western blotting analysis showed that ASFV S273R inhibited the expression of FoxJ1.Altogether,we determined that FoxJ1 plays an antiviral role against ASFV replication,and ASFV protein impairs FoxJ1-mediated antiviral effect by degradation of FoxJ1.Our findings provide new insights into the antiviral function of FoxJ1,which might help design antiviral drugs or vaccines against ASFV infection.Caina Ma Shasha Li Fan Yang Weijun Cao Huisheng Liu Tao Feng Keshan Zhang Zixiang Zhu Xiangtao Liu Yonghao Hu Haixue Zheng 2022Virologica Sinica2022,37,3:1

Throughout the course of history, all the social improvement and economic development are deeply concerned with soil loss and ecological environmental protection. It is now a common sense that soil and water conservation is the safety insurance for national ecology and its development. For the past long period of time, soil and water loss has been recognized as 'the No.1 killer' to the ecological environment. The nation is on the stage of the critical conditions for its development. China has massive mountain and upland area with complicated geological conditions and accelerated human destruction and serious soil and water loss. 18Insights into genetic diversity and phenotypic variations in domestic geese through comprehensive population and pan-genome analysis显示文摘Background Domestic goose breeds are descended from either the Swan goose(Anser cygnoides)or the Greylag goose(Anser anser),exhibiting variations in body size,reproductive performance,egg production,feather color,and other phenotypic traits.Constructing a pan-genome facilitates a thorough identification of genetic variations,thereby deepening our comprehension of the molecular mechanisms underlying genetic diversity and phenotypic variability.Results To comprehensively facilitate population genomic and pan-genomic analyses in geese,we embarked on the task of 659 geese whole genome resequencing data and compiling a database of 155 RNA-seq samples.By constructing the pan-genome for geese,we generated non-reference contigs totaling 612 Mb,unveiling a collection of 2,813 novel genes and pinpointing 15,567 core genes,1,324 softcore genes,2,734 shell genes,and 878 cloud genes in goose genomes.Furthermore,we detected an 81.97 Mb genomic region showing signs of genome selection,encompassing the TGFBR2 gene correlated with variations in body weight among geese.Genome-wide association studies utilizing single nucleotide polymorphisms(SNPs)and presence-absence variation revealed significant genomic associations with various goose meat quality,reproductive,and body composition traits.For instance,a gene encoding the SVEP1 protein was linked to carcass oblique length,and a distinct gene-CDS haplotype of the SVEP1 gene exhibited an association with carcass oblique length.Notably,the pan-genome analysis revealed enrichment of variable genes in the“hair follicle maturation”Gene Ontology term,potentially linked to the selection of feather-related traits in geese.A gene presence-absence variation analysis suggested a reduced frequency of genes associated with“regulation of heart contraction”in domesticated geese compared to their wild counterparts.Our study provided novel insights into gene expression features and functions by integrating gene expression patterns across multiple organs and tissues in geese and analyzing population variation.Conclusion This accomplishment originates from the discernment of a multitude of selection signals and candidate genes associated with a wide array of traits,thereby markedly enhancing our understanding of the processes underlying domestication and breeding in geese.Moreover,assembling the pan-genome for geese has yielded a comprehensive apprehension of the goose genome,establishing it as an indispensable asset poised to offer innovative viewpoints and make substantial contributions to future geese breeding initiatives.Guangliang Gao Hongmei Zhang Jiangping Ni Xianzhi Zhao Keshan Zhang Jian Wang Xiangdong Kong Qigui Wang 2024Journal of Animal Science and Biotechnology2024,15,1:0
19Larval feeding status regulates the transcript levels of genes encoding PTTH and allatoregulatory peptides in silkworm Bombyx mori显示文摘In Bombyx mori,two dorsolateral neurosecretory cells(NSCs)in each of thetwo brain lobes have been identified as prothoracicotropic hormone(PTTH)producingcells.This neuropeptide in insects stimulates the prothoracic gland for the synthesis andrelease of ecdysone,responsible for the molting events.Allatotropin(AT)and allatostatin(AST)are allatoregulatory neuropeptides that regulate juvenile hormone biosynthesis.Here,by using RT-qPCR,we showed that in B.mori,nutritional stress modulates themRNA expression of AT and AST-C(allatostain type C)in the central nervous systemconsisting of the brain lobes and all the associated ganglia.Using whole-mount in situhybridization,we showed that the feeding status of Bombyx larvae also influences the ex-pression of PTTH in the NSCs of the brain.Food deprivation significantly decreased themRNA expression levels of PTTH in larvae at active or terminal growth period.Further,we showed that insulin modulates the expression level of PTTH.However,its action wasdependent on the feeding status of the larvae.At feeding,the insulin decreased the PTTHexpression level,while at food deprivation,the insulin increased the PTTH expressionlevel.The data thus indicates that larval feeding status plays an important role in alteringthe mRNA expression levels of allatoregulatory peptide genes and PTTH.Sanathoibi D.Kh Bela Keshan 2021Insect Science2021,28,3:0
