1Response to Selection for Fast Growth in the Second Generation of Pacific Oyster (Crassostrea gigas)显示文摘Mass selection for fast growth was conducted in three Pacific oyster (Crassostrea gigas) stocks from China, Japan and Korea using previously established lines (CS1, JS1, and KS1). To determine whether continuous progress can be achieved by selection for growth, the progeny of three second-generation Pacific oyster lines was selected for shell height and evaluated via a 400-day farming experiment. When harvested at the end of the experiment, the selected crosses of CS2, JS2, and KS2 lines grew by 9.2%, 10.2% and 9.6% larger than the control crosses, respectively. During grow-out stage, the genetic gain of three selected lines was (10.2 ± 1.4)%, (10.4 ± 0.3)%, and (8.4 ± 1.6)%, respectively; and the corresponding realized heritability was 0.457 ± 0.143, 0.312 ± 0.071 and 0.332 ± 0.009, respectively. These results indicated that the selection for fast growth achieved steady progress in the second generation of oyster. 3Biochemical Composition and Nutritional Value of Different Shell Color Strains of Pacific Oyster Crassostrea gigas显示文摘The shell color of Pacific oyster(Crassostrea gigas) is a desirable trait; but the nutritional studies on C. gigas with different shell colors have not been conducted. Through successive selective breeding, five shell color strains of black(B), purple(P), orange(O), golden(G) and white(W) C. gigas have been developed. The aim of this study was to evaluate the chemical composition and nutritional value of five shell color strains and one commercial population with a common color. The biochemical composition including moisture, total protein, glycogen, ash, total fat, fatty acids(FA), amino acids and minerals was detected. The results indicated that the protein(50.76%–56.57%) was the major component. The content of glycogen showed a significant difference between orange shell and golden shell strains, as well as between commercial population and golden shell strain. In addition, all shell color strains contained a large amount of essential amino acids(12.20–14.15 g(100 g)^(-1)), of them leucine(2.81–3.29 g(100 g)^(-1)) and lysine(2.79–3.28 g(100 g)^(-1)) were predominant. The oysters were rich in polyunsaturated fatty acids(42.26%–45.24% of total fatty acid) with high levels of DHA(18.53%–21.16% of total fatty acid) and EPA(17.23%–18.68% of total fatty acid). Significant differences of mineral contents(Mg, Zn, Fe and Cu) were identified among the six populations. These results indicated that C. gigas with different shell colors presented rich nutritional value with high protein, glycogen, essential amino acids and polyunsaturated fatty acids. 4Relationship Between Single Nucleotide Polymorphism of Glycogen Synthase Gene of Pacific Oyster Crassostrea gigas and Its Glycogen Content显示文摘Glycogen is important not only for the energy supplementary of oysters, but also for human consumption. High glycogen content can improve the stress survival of oyster. A key enzyme in glycogenesis is glycogen synthase that is encoded by glycogen synthase gene GYS. In this study, the relationship between single nucleotide polymorphisms(SNPs) in coding regions of Crassostrea gigas GYS(Cg-GYS) and individual glycogen content was investigated with 321 individuals from five full-sib families. Single-strand conformation polymorphism(SSCP) procedure was combined with sequencing to confirm individual SNP genotypes of Cg-GYS. Least-square analysis of variance was performed to assess the relationship of variation in glycogen content of C. gigas with single SNP genotype and SNP haplotype. As a consequence, six SNPs were found in coding regions to be significantly associated with glycogen content(P < 0.01), from which we constructed four main haplotypes due to linkage disequilibrium. Furthermore, the most effective haplotype H2(GAGGAT) had extremely significant relationship with high glycogen content(P < 0.0001). 5The electromechanical features of LiNbO_(3) crystal for potential high temperature piezoelectric applications显示文摘Lithium niobate(LiNbO_(3),LN)crystal is a multi-functional material with favorable piezoelectric,nonlinear optical and electro-optic properties.In this study,the electromechanical properties of the radial extensional(RE)and the thickness extensional(TE)modes of the congruent LN are studied and the temperature dependent behaviors are revealed.The RE mode electromechanical coupling factors(kp)for the Y-and Z-oriented discs are calculated and found to be 3.8%and 24.7%,respectively,which are nearly the same as the experimental results of 3.8%and 25.2%,respectively.The maximum RE and thickness shear(TS)modes electromechanical coupling factors are obtained to be 47.6%and 68.5%for the Yx/25and Yx/167crystal cuts,respectively.The LN crystal possesses good temperature stability of the electromechanical coupling factors(RE and TE modes)from 20℃ to 500℃,where the variations of kp and kt for the Y-oriented discs are<8.0%and<1.8%,respectively.Feifei Chen Lingfeng Kong Wei Song Chao Jiang Shiwei Tian Fapeng Yu Lifeng Qin Xian Zhao 2019Journal of Materiomics2019,5,1:3
6Reproductive Cycle and Seasonal Variations in Lipid Content and Fatty Acid Composition in Gonad of the Cockle Fulvia mutica in Relation to Temperature and Food显示文摘From March 2004 to February 2005,seasonal variations in lipid content and fatty acid composition of gonad of the cockle Fulvia mutica(Reeve) were studied on the eastern coast of China in relation to the reproductive cycle and environment conditions(e.g.,temperature and food availability).Histological analysis as well as lipid and fatty acid analyses were performed on neutral and polar lipids of the gonad.Results showed that gametogenesis occurred in winter and spring at the expense of lipids previously accumulated in summer and autumn,whereas spawning occurred in summer(20.4-24.6℃).The seasonal variation in lipid content was similar to that of the mean oocyte diameter.In both neutral and polar lipids,the 20:5n-3 and 22:6n-3 levels were relatively higher than saturated fatty acids,and polyunsaturated fatty acids were abundant,with series n-3 as the predominant component.Seasonal variations in the 20:5n-3 and 22:6n-3 levels and the principal n-3 fatty acids were clearly related to the reproductive cycle.The ∑(n-3) and ∑(n-6) values were relatively high during January-May,and the associated unsaturation index was significantly higher than that in other months.The results suggest that fatty acids play an important role in the gametogenesis of F.mutica.LIU Wenguang LI Qi KONG Lingfeng 2013Journal of Ocean University of China2013,12,3:3
7Genetic Variation and Differentiation in Wide Ranging Populations of Razor Clam(Sinonovacula constricta) Inferred from AFLP Markers显示文摘The genetic variation and differentiation of the razor clam Sinonovacula constricta distributed along the coast of China were studied through amplified fragment length polymorphism(AFLP) analysis.Six primer combinations generated 193 fragments.The He values varied from 0.322 to 0.463 and the percentage of polymorphic loci ranged from 74.1% to 98.4%,which indicates a high level of genetic diversity.Cluster analysis by Nei's pairwise distance grouped all specimens by geographical origins.AMOVA consistently showed that genetic variation among populations was 8.71%,and most of the variation came from the genetic variation within populations(91.29%).Genetic differentiation among the six populations was moderate;pairwise FST ranged from 0.0282 to 0.1480,which indicated that S.constricta populations along the coast of China are genetically connected.Among all the six populations,the Beihai population is the mostly differentiated from the others,suggesting that Hainan Island and Leizhou Peninsula act as barriers to gene flow.All populations abide isolation by distance model as indicated by Mantel test,except for ZS(Zhoushan) and YQ(Yueqing) populations.Information obtained in this study will provide guidelines for conservation and fishery management of this species in the future.WANG Jingbo LI Qi KONG Lingfeng 2010Journal of Ocean University of China2010,9,3:2
8Genetic Variation Assessed with Microsatellites in Mass Selection Lines of the Pacific Oyster (Crassostrea gigas) in China显示文摘Four successive mass selection lines of the Pacific oyster, Crassostrea gigas, selected for faster growth in breeding programs in China were examined at ten polymorphic microsatellite loci to assess the level of allelic diversity and estimate the effective population size. These data were compared with those of their base population. The results showed that the genetic variation of the four generations were maintained at high levels with an average allelic richness of 18.8–20.6, and a mean expected heterozygosity of 0.902–0.921. They were not reduced compared with those of their base population. Estimated effective population sizes based on temporal variances in microsatellite frequencies were smaller to that of sex ratio-corrected broodstock count estimates. Using a relatively large number of broodstock and keeping an equal sex ratio in the broodstock each generation may have contributed to retaining the original genetic diversity and maintaining relatively large effective population size. The results obtained in this study showed that the genetic variation was not affected greatly by mass selection progress and high genetic variation still existed in the mass selection lines, suggesting that there is still potential for increasing the gains in future generations of C. gigas. 9Linkage Disequilibrium in Wild and Cultured Populations of Pacific Oyster(Crassostrea gigas)显示文摘Linkage disequilibrium(LD) can be applied for mapping the actual genes responsible for variation of economically important traits through association mapping.The feasibility and efficacy of association studies are strongly dependent on the extent of LD which determines the number and density of markers in the studied population,as well as the experimental design for an association analysis.In this study,we first characterized the extent of LD in a wild population and a cultured mass-selected line of Pacific oyster(Crassostrea gigas).A total of 88 wild and 96 cultured individuals were selected to assess the level of genome-wide LD with 53 microsatellites,respectively.For syntenic marker pairs,no significant association was observed in the wild population;however,three significant associations occurred in the cultured population,and the significant LD extended up to 12.7 c M,indicating that strong artificial selection is a key force for substantial increase of genome-wide LD in cultured population.The difference of LD between wild and cultured populations showed that association studies in Pacific oyster can be achieved with reasonable marker densities at a relatively low cost by choosing an association mapping population.Furthermore,the frequent occurrence of LD between non-syntenic loci and rare alleles encourages the joint application of linkage analysis and LD mapping when mapping genes in oyster.The information on the linkage disequilibrium in the cultured population is useful for future association mapping in oyster.GUO Xiang LI Qi KONG Lingfeng YU Hong 2016Journal of Ocean University of China2016,15,2:2
10Seasonal Variation of the Glycogen Enzyme Activity in Diploid and Triploid Pacific Oyster Gonad During Sexual Maturation显示文摘The glycogen content and the activities of two key enzymes in glycogen metabolism, glycogen phosphorylase and gly- cogen synthetase, in the gonad of diploid and triploid Pacific oysters (Crassostrea gigas) were compared during maturation. The glycogen content in the gonad of diploids decreased with gametogenesis (by 85.7%), but the glycogen content in the gonad of trip- loids did not vary significantly. Activity of glycogen phosphorylase (GP) in the gonad of diploids decreased with gametogenesis (by 55.5%), while GP activity of triploids did not vary significantly during maturation. Activity of glycogen synthetase (GS) in the gonad of diploids increased slightly with gametogenesis, reaching a peak in June. Activity of GS declined sharply from June to July, which might be due to gonad spawning. 13Microsatellite-Centromere Mapping in Japanese Scallop(Patinopecten yessoensis) Through Half-Tetrad Analysis in Gynogenetic Diploid Families显示文摘Gene-centromere mapping is an essential prerequisite for understanding the composition and structure of genomes. Half-tetrad analysis is a powerful tool for mapping genes and understanding chromosomal behavior during meiosis. The Japanese scallop(Patinopecten yessoensis), a cold-tolerant species inhabiting the northwestern Pacific coast, is a commercially important marine bivalve in Asian countries. In this study, inheritance of 32 informative microsatellite loci was examined in 70-h D-shaped larvae of three induced meiogynogenetic diploid families of P. yessoensis for centromere mapping using half-tetrad analysis. The ratio of gynogenetic diploids was proven to be 100%, 100% and 96% in the three families, respectively. Inheritance analysis in the control crosses showed that 51 of the 53 genotypic ratios observed were in accordance with Mendelian expectations at the 5% level after Bonferroni correction. Seven of the 32 microsatellite loci showed the existence of null alleles in control crosses. The second division segregation frequency(y) of the microsatellite loci ranged from 0.07 to 0.85 with a mean of 0.38, suggesting the existence of positive interference after a single chiasma formation in some chromosomes in the scallop. Microsatellite-centromere distances ranged from 4 c M to 42 c M under the assumption of complete interference. 15Development of Three Multiplex PCR Primer Sets for Ark Shell(Scapharca broughtonii)and Their Validation in Parentage Assignment显示文摘Scapharca broughtonii is a commercially important and over-exploited species.In order to investigate its genetic diversity and population structure,43 novel polymorphic microsatellites were isolated and characterized.The number of alleles per locus ranged from 3 to 22 with an average of 6.93,and the observed and expected heterozygosities varied between 0.233 and 1.000,and 0.250 and 0.953,with an average of 0.614 and 0.707,respectively.Three highly informative multiplex PCRs were developed from nine of those microsatellites for S.broughtonii.We evaluated and validated these multiplex PCRs in 8 full-sib families.The average polymorphism information content(PIC) was 0.539.The frequency of null alleles was estimated as 3.13% of all the alleles segregation based on a within-family analysis of Mendelian segregation patterns.Parentage analysis of real offspring demonstrated that 100% of all offspring were unambiguously allocated to a pair of parents based on 3 multiplex sets.Those 43 microsatellite loci with high variability will be helpful for the analysis of population genetics and conservation of wild stock of S.broughtonii.The 3 sets of multiplex PCRs could be an important tool of pedigree reconstruction,population genetic analysis and brood stock management.LINing LI Qi KONG Lingfeng YU Hong 2016Journal of Ocean University of China2016,15,2:1
16Phylogenomic resolution of Imparidentia(Mollusca:Bivalvia)diversifcation through mitochondrial genomes显示文摘Despite signifcant advances in the phylogenomics of bivalves over the past decade,the higher-level phylogeny of Imparidentia(a superorder of Heterodonta)remains elusive.Here,a total of fve new mitochondrial sequences(Chama asperella,Chama limbula,Chama dunkeri,Barnea manilensis and Ctena divergens)was added to provide resolution in nodes that required additional study.Although the monophyly of Lucinida remains less clear,the results revealed the overall backbone of the Imparidentia tree and the monophyly of Imparidentia.Likewise,most relationships among the fve major Imparidentia lineages—Lucinida,Cardiida,Adapedonta,Myida and Venerida—were addressed with a well-supported topology.Basal relationships of Imparidentia recovered Lucinidae as the sister group to all remaining imparidentian taxa.Thyasiridae is a sister group to other imparidentian bivalves(except Lucinidae species)which is split into Cardiida,Adapedonta and the divergent clade of Neoheterodontei.Neoheterodontei was comprised of Venerida and Myida,the former of which now also contains Chamidae as the sister group to all the remaining venerid taxa.Moreover,molecular divergence times were inferred by calibrating nine nodes in the Imparidentia tree of life by extinct taxa.The origin of these major clades ranged from Ordovician to Permian with the diversifcation through the Palaeozoic to Mesozoic.Overall,the results obtained in this study demonstrate a better-resolved Imparidentia phylogeny based on mitochondrial genomes.Yu Wang Yi Yang Lingfeng Kong Takenori Sasaki Qi Li 2023Marine Life Science & Technology2023,5,3:1

20Mass Selection for Growth Improvement in Black Shell Line of Pacific Oyster Crassostrea gigas显示文摘The Pacific oyster,Crassostrea gigas,naturally distributing along the coast of northwest Pacific,is one of the most important bivalve species due to its high economic value and fecundity.In China,we have initiated a selective breeding program on both shell color and growth rate of C.gigas since 2010.A black shell line was obtained through four-generation family selection.In this study,mass selection for growth improvement was conducted in the sixth generation and seventh generation of black shell lines.To assess the progress of potential genetic improvement,the progeny of two generations of black shell lines were selected to evaluate their shell heights via a 450-day farming experiment.As the results,after growing for 450 days,the sixth generation and seventh generation of selected lines were 9.03% and 11.42% larger than the control lines,respectively.During the grow-out stage,the genetic gain of two generations was 8.82%±0.18% and 11.54%±0.43%,respectively;and the corresponding realized heritability was 0.45±0.04 and 0.41±0.04,respectively.These results indicated that the mass selection for shell height achieved steady progress in the two generations of C.gigas.XU Chengxun LI Qi CHONG Jindou LIU Shikai KONG Lingfeng 2019Journal of Ocean University of China2019,18,6:1
