| 3 | A novel quantified bitterness evaluation model for traditional Chinese herbs based on an animal ethology principle显示文摘Traditional Chinese herbs(TCH) are currently gaining attention in disease prevention and health care plans. However, their general bitter taste hinders their use. Despite the development of a variety of taste evaluation methods, it is still a major challenge to establish a quantitative detection technique that is objective, authentic and sensitive. Based on the two-bottle preference test(TBP), we proposed a novel quantitative strategy using a standardized animal test and a unified quantitative benchmark. To reduce the difference of results, the methodology of TBP was optimized. The relationship between the concentration of quinine and animal preference index(PI) was obtained. Then the PI of TCH was measured through TBP, and bitterness results were converted into a unified numerical system using the relationship of concentration and PI. To verify the authenticity and sensitivity of quantified results,human sensory testing and electronic tongue testing were applied. The quantified results showed a good discrimination ability. For example, the bitterness of Coptidis Rhizoma was equal to 0.0579 mg/mL quinine, and Nelumbinis Folium was equal to 0.0001 mg/mL. The validation results proved that the newassessment method for TCH was objective and reliable. | 4 | Xiba Granitic Pluton in the Qinling Orogenic Belt, Central China: Its Petrogenesis and Tectonic Implications显示文摘Xiba granitic pluton is located in South Qinling tectonic domain of the Qinling orogenic belt and consists mainly of granodiorite and monzogranite with significant number of microgranular quartz dioritic enclaves. SHRIMP zircon U-Pb isotopic dating reveals that the quartz dioritic enclaves formed at 214±3 Ma, which is similar to the age of their host monzogranite (218±1 Ma). The granitoids belong to high-K calc-alkaline series, and are characterized by enriched LILEs relative to HFSEs with negative Nb, Ta and Ti anomalies, and right-declined REE patterns with (La/Yb) N ratios ranging from 15.83 to 26.47 and δEu values from 0.78 to 1.22 (mean= 0.97). Most of these samples from Xiba granitic pluton exhibit εNd(t) values of 8.79 to 5.38, depleted mantle Nd model ages (T DM ) between 1.1 Ga and 1.7 Ga, and initial Sr isotopic ratios ( 87 Sr/ 86 Sr) i from 0.7061 to 0.7082, indicating a possible Meso-to Paleoproterozoic lower crust source region, with exception of samples XB01-2-1 and XB10-1 displaying higher ( 87 Sr/ 86 Sr) i values of 0.779 and 0.735, respectively, which suggests a contamination of the upper crustal materials. Quartz dioritic enclaves are interpreted as the result of rapid crystallization fractionation during the parent magmatic emplacement, as evidenced by similar age, texture, geochemical, and Sr-Nd isotopic features with their host rocks. Characteristics of the petrological and geochemical data reveal that the parent magma of Xiba granitoids was produced by a magma mingling process. The upwelling asthenosphere caused a high heat flow and the mafic magma was underplated into the bottom of the lower continent crust, which caused the partial melting of the lower continent crustal materials. This geodynamic process generated the mixing parent magma between mafic magma from depleted mantle and felsic magma derived from the lower continent crust. | 8 | Towards an Esox lucius inspired multimodal robotic fish显示文摘In this paper, we further explore multimodal locomotion via an updated robotic fish model based on Esox lucius. Besides the improved actuation properties like higher torque servomotors and powerful electronics,the robotic fish has some innovative mechanical design to pursue diverse swimming modes and superior performance. Specifically, we introduced a ±50° yawing head joint that functions as the neck for enhancing turning ability. A pair of pectoral mechanisms with two DOFs per fin is constructed to achieve 3-D swimming and to enrich multiple pectoral motions. At the control level, an improved central pattern generator(CPG) model allowing for free adjustment of the phase relationship among outputs is employed to produce rhythmic signals of multimodal swimming. Extensive experiments were carried out to examine how characteristic parameters in CPGs including amplitude, frequency, and phase lag affect the swimming performance. | 9 | A rapid reporter assay for recombinant human brain natriuretic peptide(rhBNP)by GloSensor technology显示文摘Accurate determination of biological activity is essential in quality control of recombinant human brain natriuretic peptide(rhBNP). In previous study, we successfully developed a genetically modified cell line 293GCAC3-based ELISA assay for rhBNP. But ELISA procedure is still tedious, so this study was aimed to develop a rapid and simple bioassay for rhBNP using GloSensor technology, which provides a platform of flexible luciferase-based biosensors for real-time detection of signaling events in live cells, including cGMP production. A reporter cell line 293GCAGlo-G1 was constructed by transfecting pGloSensor^(TM) 40 F plasmid into 293GCAC3. The reporter assay based on 293GCAGlo-G1 showed high precision with intraassay CV being 8.3% and inter-assay CV being 14.1%; high accuracy with 80%, 100% and 120% recovery rate being 99.2%, 102.4% and 99.0% respectively; and great linearity with R^2 of linear fitting equation being 0.99. Besides, no significant difference was found in test results of reporter assay and 293GCAC3-based ELISA assay(paired t test, p=0.630). CAR-T cells are genetically modified T cells that can specifically recognize tumor specific antigens on the surface of tumor cells, and then effectively kill tumor cells. At present, exciting results are being achieved in clinical applications of CAR-T cells for patients with hematological malignancies. The research and development of CAR-T cells for various targets and for the treatment of solid tumors have become a hot topic worldwide, so an increasing number of investigational new drug applications (INDAs) and new drug applications (NDAs) of CAR-T cell products are expected to be submitted in future. The quality control and nonclinical research of these products are of great significance in ensuring the safety and effectiveness of these products;however, they also present great challenges and difficulties. | 14 | Constructing a high-density linkage map for Gossypium hirsutum × Gossypium barbadense and identifying QTLs for lint percentage显示文摘To introgress the good fiber quality and yield from Gossypium barbadense into a commercial Upland cotton variety, a high-density simple sequence repeat(SSR) genetic linkage map was developed from a BC1F1 population of Gossypium hirsutum×Gossypium barbadense. The map comprised 2,292 loci and covered 5115.16 centi Morgan(c M) of the cotton AD genome, with an average marker interval of 2.23 c M. Of the marker order for 1,577 common loci on this new map, 90.36% agrees well with the marker order on the D genome sequence genetic map. Compared with five published high-density SSR genetic maps, 53.14% of marker loci were newly discovered in this map. Twenty-six quantitative trait loci(QTLs) for lint percentage(LP) were identified on nine chromosomes. Nine stable or common QTLs could be used for marker-assisted selection. Fifty percent of the QTLs were from G. barbadense and increased LP by 1.07%–2.41%.
