| 1 | Establishment and Optimization of SRAP Amplification System in Lonicara caerulea L.显示文摘A single factor design was applied to optimize five factors influencing SRAP system,including Taq DNA polymerase,template DNA concentration,dNTPs,primer and Mg 2+,each at four levels. The optimal SRAP-PCR system for Lonicera caerulea L. was 20 μL SRAP-PCR amplification reaction solution containing 2.0 μL 10×PCR buffer,1.0 U Taq DNA polymerase,30 ng template DNA,0.2 mmol L -1 dNTPs,2.0 mmol L -1 Mg 2+ and 0.2 μmol L -1 primer. The suitable amplification procedure consisted of an initial denaturation at 94℃ for 5 min;denaturation at 94℃ for 1 min,annealing at 35℃ for 1 min,extension at 72℃ for 90 s and in total five cycles;denaturation at 94℃ for 1 min,annealing at 50℃ for 1 min,extension at 72℃ for 90 s and in total 35 cycles;extension at 72℃ for 8 min;preservation at 4℃. | 2 | Effects of fruit thinning on ascorbate–glutathione cycle metabolism in black currants(Ribes nigrum L.)显示文摘To elucidate mechanisms regulating ascorbic acid(AsA) biosynthesis and accumulation in the fruit and leaves of black currants,As A and the activities of key enzymes in the ascorbate–glutathione(As A–GSH) cycle were measured from fruit set to fruit ripening during fruit thinning treatments of three common commercial black currant cultivars that differed in their As A levels:'Risager'(low),'Brodtrop'(medium) and 'Adelinia'(high).Treatments were 50% fruit reduction(50% of total fruit set) by hand,control was no thinning.Fruit thinning treatment significantly increased As A content in fruit of all three cultivars from weeks 2 to 8,significantly decreased As A content in leaves from weeks 3 to 8.Dehydroascorbate reductase and monodehydroascorbate reductase activities in fruit and leaves had a similar pattern,increasing during week 2,rose until they peaked in week 4.Ascorbate peroxidase activity in fruit in the thinning treatment was slightly lower than in the control.Fruit thinning was shown to be a good model to test As A biosynthesis regulation and accumulation in black currants.The results from our study provided strong evidence that As A–GSH cycle involved in As A synthesis and accumulation in fruit. | Dong Qin Lijuan Zhao Yu Gary Gao Fangxiao Li Shulei Li Junwei Huo Shuang Lou Peng Liu | 2017 | Journal of Forestry Research2017,28,5: | 3 |
| 3 | Effects of GA3 and ABA on the respiratory pathways during the secondary bud burst in black currants显示文摘The effects of exogenous gibberellic acid(GA_3)and abscisic acid(ABA) on the total respiratory rate, percentages of total respiratory rate contributed by respiratory pathways [Embden-Meyerhof-Parnas Pathway(EMP),Pentose Phosphate Pathway(PPP), and Tricarboxylic Acid Cycle(TCA)], and conversion of starch to soluble sugars in the buds of black currants during secondary bud burst were investigated to determine the relationship between respiratory rates and secondary bud burst. ‘Adelinia', a black currant cultivar that is prone to secondary bud burst after the first harvest, was used in this study. Mature bushes of Adelinia were sprayed with 30 mg/L GA_3 and 50 mg/L ABA to manipulate bud burst. The results showed that exogenous applications of GA_3 and ABA had opposite effects on bud respiratory rate. Generally, GA_3 treatment increased the total respiratory rate and respiratory rate of the TCA and PPP, and the respiratory rates after GA_3 treatment were higher than those of control. While ABA treatment mostly decreased the total respiratory rate and the respiratory rate of TCA and PPP in buds in comparison to control. In terms of the percentage of the three respiratory rates in comparison to the total respiratory rate, GA_3 treatment significantly increased the percentage of TCA and PPP respiratory rate in comparison to the control(P \ 0.01), whereas ABA decreased the rates. GA_3 significantly increased the content of soluble sugars and decreased the starch content, while the starch content in buds after ABA treatment was significantly higher than that of the control. All results showed that PPP is a critical process for the second bud burst in black currants. While the EMP–TCA pathway is still dominant in bud respiration,provides a series of basic materials and energy(ATP). The conversion of starch to soluble sugars is essential for bud burst. | 5 | Microstructure and Mechanical Properties of an Ultrahigh-strength Titanium alloy Ti-4.5Al-5Mo-5V-6Cr-1Nb Prepared Using Laser Directed Energy Deposition and Forging:A Comparative Study显示文摘The application of titanium alloys in aerospace put forward the requirement for higher strength.Additive manu-facturing is a promising method for the efficient and economical processing of titanium alloys.However,research on the additive manufacturing of ultrahigh-strength titanium alloys is still limited.The mechanisms of microseg-regation for high alloying elements and poor plasticity are still not clear.In this study,an ultrahigh-strength titanium alloy Ti-4.5Al-5Mo-5V-6Cr-1Nb(TB18)was prepared using two methods:laser direct energy deposi-tion(LDED)and forging.The LDEDed alloy contains three zones with similar grain morphologies but different microstructure.The microsegregation of the alloy is limited due to the rapid solidification and almost eliminated after the thermal cycle and solution treatment.With stress relief treatment,the LDEDed alloy exhibits anisotropic mechanical properties.After solution and aging treatments,its ultimate strength is enhanced;however,its plas-ticity is relatively lower than that of the wrought alloy with equally high strength.The excellent balance of the strength and plasticity of the wrought alloy can be ascribed to the formation of��WGB and multiscale��laths,which provides enlightenment for optimizing the properties of the LDEDed alloy. | Junwei Yang Haibo Tang Peiyuan Wei Hongwei Gao Jiawei Wang Haixin Huo Yanyan Zhu | 2023 | Chinese Journal of Mechanical Engineering(Additive Manufacturing Frontiers)2023,2,1: | 0 |
