Developmentof a HCV vaccine as well as more effective HBV vaccines isan urgent task. DNA immunization provides a promisingapproach to elicit protective humoral and cellular immuneresponses against viral infection. The aim of this study is toachieve immune responses against both HCV and HBV by DNAimmunization with fusion constructs comprising various HCVE2 gene fragments fused to HBsAg gane of HBV.METHODS: C57BL/6 mice were immunized with plasmid DNAexpressing five fragments of HCV E2 fused to the gene forHBsAg respectively. After one primary and one boostingimmunizations, antibodies against HCV E2 and HBsAg weretested and subtyped in ELISA. Splenic cytokine expressionof IFN-γ and IL-10 was analyzed using an RT-PCR assay.Post-immune mouse antisera also were tested for theirability to capture HCV viruses in the serum of a hepatitis Cpatient in vitro.RESUTLTS: After immunization, antibodies against bothHBsAg and HCV E2 were detected in mouse sera, withIgG2a being the dominant immunoglobulin sub-class. High-level expression of INF-γ was deuetected in cultured splenic cells.Mouse antisera against three of the five fusion constructs wereable to capture HCV viruses in an in vitro assay.CONCLUSION: The results indicate that these fusionconstructs could efficiently elicit humoral and Th1 dominantcellular immune responses against both HBV S and HCV E2antigens in DNA-immunized mice. They thus could serve ascandidates for a bivalent vaccine against HBV and HCVinfection. | 6 | Preliminary evidence for the presence of multiple forms of cell death in diabetes cardiomyopathy显示文摘Diabetic mellitus(DM) is a common degenerative chronic metabolic disease often accompanied by severe cardiovascular complications(DCCs) as major causes of death in diabetic patients with diabetic cardiomyopathy(DCM) as the most common DCC. The metabolic disturbance in DCM generates the conditions/substrates and inducers/triggers and activates the signaling molecules and death executioners leading to cardiomyocyte death which accelerates the development of DCM and the degeneration of DCM to heart failure.Various forms of programmed active cell death including apoptosis, pyroptosis, autophagic cell death, autosis,necroptosis, ferroptosis and entosis have been identified and characterized in many types of cardiac disease.Evidence has also been obtained for the presence of multiple forms of cell death in DCM. Most importantly,published animal experiments have demonstrated that suppression of cardiomyocyte death of any forms yields tremendous protective effects on DCM. | 7 | Arabidopsis VQ10 interacts with WRKY8 to modulate basal defense against Botrytis cinerea显示文摘Recent studies in Arabidopsis have revealed that some VQ motif-containing proteins physically interact with WRKY transcription factors;however,their specific biological functions are still poorly understood.In this study,we confirmed the interaction between VQ10and WRKY8,and show that VQ10 and WRKY8 formed a complex in the plant cell nucleus.Yeast two-hybrid analysis showed that the middle region of WRKY8 and the VQ motif of VQ10 are critical for their interaction,and that this interaction promotes the DNA-binding activity of WRKY8.Further investigation revealed that the VQ10protein was exclusively localized in the nucleus,and VQ10was predominantly expressed in siliques.VQ10 expression was strongly responsive to the necrotrophic fungal pathogen,Botrytis cinerea and defense-related hormones.Phenotypic analysis showed that disruption of VQ10 increased mutant plants susceptibility to the fungal pathogen B.cinerea,whereas constitutive-expression of VQ10 enhanced resistance to B.cinerea.Consistent with these findings,expression of the defenserelated PLANT DEFENSIN1.2(PDF1.2)gene was decreased in vq10 mutant plants,after B.cinerea infection,but increased in VQ10-overexpressing transgenic plants.Taken together,our findings provide evidence that VQ10 physically interacts with WRKY8 and positively regulates plant basal resistance against the necrotrophic fungal pathogen B.cinerea. | Junqiu Chen Houping Wang Yang Li Jinjing Pan Yanru Hu Diqiu Yu | 2018 | Journal of Integrative Plant Biology2018,60,10: | 8 |

| 11 | Oxidation behavior of ammonium in a 3-dimensional biofilm-electrode reactor显示文摘Excess nitrogenous compounds are detrimental to natural water systems and to human health. To completely realize autohy- drogenotrophic nitrogen removal, a novel 3-dimensional biofilm-electrode reactor was designed. Titanium was electroplated with ruthenium and used as the anode. Activated carbon fiber felt was used as the cathode. The reactor was separated into two chambers by a permeable membrane. The cathode chamber was filled with granular graphite and glass beads. The cathode and cathode chamber were inhabited with domesticated biofilm. In the absence of organic substances, a nitrogen removal efciency of up to 91% was achieved at DO levels of 3.42 ± 0.37 mg/L when the applied current density was only 0.02 mA/cm2. The oxidation of ammonium in biofilm- electrode reactors was also investigated. It was found that ammonium could be oxidized not only on the anode but also on particle electrodes in the cathode chamber of the biofilm-electrode reactor. Oxidation rates of ammonium and nitrogen removal efciency were found to be afected by the electric current loading on the biofilm-electrode reactor. The kinetic model of ammonium at diferent electric currents was analyzed by a first-order reaction kinetics equation. The regression analysis implied that when the current density was less than 0.02 mA/cm2, ammonium removal was positively correlated to the current density. | 12 | Effects of Integrated Soil-Crop System Management on Soil Organic Carbon Characteristics in a Primosol in Northeast China显示文摘A synchronous increase in crop productivity, nutrient use efficiency, and soil carbon(C) sequestration is important from the point of view of food security and environmental protection. In recent years, integrated soil-crop system management(ISSM), which uses crop models and advanced nutrient management to redesign cropping systems, has been successfully demonstrated to achieve both high crop productivity and high nutrient use efficiency in China, but the effects of ISSM on soil organic C(SOC) characteristics remain unknown. In this study, the effects of current farmers' practice(FP), high-yielding practice(HY), which maximizes yields without considering costs, and ISSM on the content and chemical composition of SOC were studied in a 4-year(2009–2013) field plot experiment with maize(Zea mays L.) monoculture in an Alluvic Primosol in Northeast China. The ISSM resulted in higher soil total organic C(TOC), water-soluble organic C, easily-oxidizable organic C, particulate organic C, and humic acid C compared with HY and FP in the region. The SOC contents in aggregate size fractions generally followed a similar pattern to TOC. Compared with FP,HY decreased the mean weight diameter, geometric mean diameter, percentage of > 0.25-mm water-stable aggregates, and the stability ratio of water-stable aggregates, and increased the structure-deterioration rate and index of unstable aggregates. The opposite trend was observed between ISSM and HY. Solid-state ^(13)C nuclear magnetic resonance spectra of bulk soil showed that ISSM had higher O-alkyl C and aliphatic C/aromatic C ratio, but lower aromatic C, carbonyl C, and alkyl C/O-alkyl C and hydrophobic C/hydrophilic C ratios than HY and FP. | 15 | Motor relearning program and Bobath method improve motor function of the upper extremities in patients with stroke显示文摘BACKGROUND: In the natural evolution of cerebrovascular disease, unconscious use of affected extremity during drug treatment and daily life can improve the function of affected upper extremity partially, but it is very slow and also accompanied by the formation of abnormal mode. Therefore, functional training should be emphasized in recovering the motor function of extremity. OBJECTIVE: To observe the effects of combination of motor relearning program and Bobath method on motor function of upper extremity of patients with stroke. DESIGN: Comparison of therapeutic effects taking stroke patients as observation subjects. SETTING: Department of Neurology, General Hospital of Beijing Jingmei Group. PARTICIPANTS: Totally 120 stroke patients, including 60 males and 60 females, averaged (59±3) years, who hospitalized in the Department of Neurology, General Hospital of Beijing Jingmei Group between January 2005 and June 2006 were recruited. The involved patients met the following criteria: Stroke attack within 2 weeks; diagnosis criteria of cerebral hemorrhage or infarction made in the 4th National Cerebrovascular Disease Conference; confirmed by skull CT or MRI; Informed consents of therapeutic regimen were obtained. The patients were assigned into 2 groups according to their wills: rehabilitation group and control group, with 30 males and 30 females in each group. Patients in rehabilitation group averaged (59±2)years old, and those in the control group averaged (58±2)years old. METHODS: ① Patients in two groups received routine treatment in the Department of Neurology. When the vital signs of patients in the rehabilitation group were stable, individualized treatment was conducted by combined application of motor relearning program and Bobath method. Meanwhile, training of activity of daily living was performed according to the disease condition changes of patients at different phases, including the nursing and instruction of body posture, the maintenance of good extremity position, bed exercise, bedside sit up and sitting position balance, sit up exercise, dynamic and static balance exercise, walking exercise, active training and passive training. The strength, time and speed of training were increased gradually according to their physical abilities. Patients were trained 45 to 60 minutes once, 5 times a week, within 2 weeks. ② Evaluation criteria of therapeutic effect: The motor function of upper extremity was evaluated by Fugl-Meyer method on the day of beginning and end of treatment. Higher points indicated better function of upper extremity. ③ t test and paired t test were used for comparing the difference of intergroup and intragroup measurement data, respectively. MAIN OUTCOME MEASURES: Changes in Fugl-Meyer scoring of two groups before and after treatment. RESULTS: Totally 120 stroke patients participated in the final analysis. Before treatment, Fugl-Meyer scoring was close between rehabilitation group and control group [(14.47±2.38),(14.16±2.39) points, P > 0.05]; Fugl-Meyer scoring of rehabilitation group after treatment was significantly higher than that before treatment and that of control group[(37.93±2.67),(18.36±2.43) points, t =11.053, 5.408, P < 0.01]; There were no significant differences in Fugl-Meyer scoring between before treatment in the control group and control group (P > 0.05). | 18 | Investigation of role of cartilage surface polymer brush border in lubrication of biological joints显示文摘Although experimental evidence has suggested that the polymer brush border(PBB)on the cartilage surface is important in regulating fluid permeability in the contact gap,the current theoretical understanding of joint lubrication is still limited.To address this research gap,a multiscale cartilage contact model that includes PBB,in particular its effect on the fluid permeability of the contact gap,is developed in this study.Microscale modeling is employed to estimate the permeability of the contact gap.This permeability is classified into two categories:For a gap size>1μm,the flow resistance is assumed to be dominated by the cartilage roughness;for gap size<1μm,flow resistance is assumed to be dominated by the surface polymers extending beyond the collagen network of the articular cartilage.For gap sizes of less than 1μm,the gap permeability decreases exponentially with increasing aggrecan concentration,whereas the aggrecan concentration varies inversely with the gap size.Subsequently,the gap permeability is employed in a macroscale cartilage contact model,in which both the contact gap space and articular cartilage are modeled as two interacting poroelastic systems.The fluid exchange between these two media is achieved by imposing pressure and normal flux continuity boundary conditions.The model results suggest that PBB can substantially enhance cartilage lubrication by increasing the gap fluid load support(e.g.,by 26 times after a 20-min indentation compared with the test model without a PBB).Additionally,the fluid flow resistance of PBB sustains the cartilage interstitial fluid pressure for a relatively long period,and hence reduces the vertical deformation of the tissue.Furthermore,it can be inferred that a reduction in the PBB thickness impairs cartilage lubrication ability. | JinJing LIAO David W.SMITH Saeed MIRAMINI Bruce S.GARDINER Lihai ZHANG | 2022 | Friction2022,10,1: | 2 |
