| 1 | Prediction of cervical lymph node metastases in papillary thyroid microcarcinoma by sonographic features of the primary site显示文摘Objective: To study the sonographic features of the primary site of papillary thyroid microcarcinoma(PTMC) for the prediction of cervical lymph node metastasis during preoperative diagnosis.Methods: A total of 710 PTMC patients between 2013 and 2016 with a diagnosis of cervical lymph node metastases were reviewed.We analyzed the sonographic features of the PTMC primary site to predict ipsilateral or central lymph node metastases in univariate and multivariate models.The ratio of abutment/perimeter of the PTMC primary site was utilized to evaluate cervical lymph node status.Results: Regarding clinical characteristics, multifocality and extrathyroidal extension were associated with cervical lymph node involvement.In the multivariate regression model, calcification and the abutment/perimeter ratio of lesions were evaluated as independent factors in level Ⅵ, ipsilateral or skip cervical lymph node metastases.The cut-off value of the ratio of abutment/perimeter of the PTMC primary site(25%) was significantly correlated with cervical lymph node metastases(P = 0.000).Conclusions: Independent sonographic features, including lesion size, lesion location, calcification, and the ratio of abutment/perimeter of the primary site, were associated with cervical lymph node metastases in PTMC patients. | Xi Wei Meng Wang Xiaoqing Wang Xiangqian Zheng Ying Li Yi Pan Yueguo Li Jiali Mu Yang Yu Dapeng Li Ming Gao Sheng Zhang | 2019 | Cancer Biology & Medicine2019,16,3: | 22 |
| 2 | Ultrasound features of extranodal extension in the metastatic cervical lymph nodes of papillary thyroid cancer:a case-control study显示文摘Objective:Extranodal extension in cervical lymph nodes is an important risk factor for the progression and prognosis of papillary thyroid cancer.The purpose of this study was to identify the common and characteristic preoperative ultrasonography features that are associated with the pathologic extranodal extension of metastatic papillary thyroid carcinoma.Methods:We retrospectively assessed and compared clinicopathologic and ultrasound features between 60 papillary thyroid cancer patients with extranodal extension and 120 control patients with papillary thyroid cancer without extranodal extension.Results:With respect to the pathological N stage and clinicopathologic features,N1b stage papillary thyroid carcinomas were more frequently found in patients who were extranodal extension-positive,in comparison with those who were extranodal extension-negative(78.3%vs.63.3%,P=0.043).Extranodal extension was detected most frequently in level VI cervical lymph nodes(48.7%).In our univariate analysis of patients with papillary thyroid carcinoma,cervical lymph nodes with extranodal extension showed higher incidences of node matting,microcalcification,cystic area,aspect ratio<2,and larger diameter than those without extranodal extension(all P<0.05).Our multivariate analysis demonstrated that node matting and cystic area were independent risk factors for the presence of extranodal extension[odds ratio(OR):4.751,95%confidence interval(CI):1.212~18.626,P=0.025;OR:2.707,95%CI:1.127~6.502,P=0.026].Conclusions:Common ultrasound features may indicate the presence of extranodal extension in patients with metastatic cervical lymph nodes of papillary thyroid carcinoma. | Jiali Mu Xiaofeng Liang Fangxuan Li Juntian Liu Sheng Zhang Jing Tian | 2018 | Cancer Biology & Medicine2018,15,2: | 7 |
| 3 | T cells expressing CD5/CD7 bispecific chimeric antigen receptors with fully human heavy-chain-only domains mitigate tumor antigen escape显示文摘Bispecific chimeric antigen receptor T-cell(CAR-T)therapies have shown promising results in clinical trials for advanced B-cell malignancies.However,it is challenging to broaden the success of bispecific CAR-T therapies to treat refractory/relapse(r/r)T-cell leukemia/lymphoma because targeting multiple T-cell-expressing antigens leads to exacerbated CAR-T cell fratricide and potential safety concerns.Fully human heavy chain variable(FHVH)antibodies that specifically target CD5 or CD7 were screened and constructed to CD5/CD7 bispecific CARs.A truncated Epidermal growth factor receptor were integrated into CAR constructs to address safety concerns.To tackle the fratricidal issue of CAR-T cells targeting T-cell-pan marker(s),CRISPR/Cas9-based CD5 and CD7 genes knockout were performed before lentiviral transduction of bispecific CARs.Functional comparison between different bispecific CAR structures:tandem CARs and dual CAR were performed in vitro and in vivo to determine the optimal construct suitable for addressing T-cell malignancy antigen escape in clinical setting.Knockout of CD5 and CD7 prevents fratricide of CD5/CD7 bispecific CAR-T cells,and FHVH-derived CD5/CD7 bispecific CAR-T cells demonstrate potent antitumor activity in vitro and in vivo.The fratricide-resistant FHVH-derived CD5/CD7 bispecific CAR-T cells have potent antitumor activity against T-cell malignancies,and tandem CARs are more effective than dual CAR in preventing tumor escape in heterogeneous leukemic cells.The meaningful clinical efficacy and safety of tandem CD5/CD7 CAR-T cells deserve to be explored urgently. | Zhenyu Dai Wei Mu Ya Zhao Jiali Cheng Haolong Lin Kedong Ouyang Xiangyin Jia Jianwei Liu Qiaoe Wei Meng Wang Chaohong Liu Taochao Tan Jianfeng Zhou | 2022 | Signal Transduction and Targeted Therapy2022,7,4: | 3 |
| 4 | NUMB enhances Notch signaling by repressing ubiquitination of NOTCH1 intracellular domain显示文摘The release and nuclear translocation of the intracellular domain of Notch receptor(NICD)is the prerequisite for Notch signaling-mediated transcriptional activation.NICD is subjected to various posttranslational modifications including ubiquitination.Here,we surprisingly found that NUMB proteins stabilize the intracellular domain of NOTCH1 receptor(N1ICD)by regulating the ubiquitin–proteasome machinery,which is independent of NUMB’s role in modulating endocytosis.BAP1,a deubiquitinating enzyme(DUB),was further identified as a positive N1ICD regulator,and NUMB facilitates the association between N1ICD and BAP1 to stabilize N1ICD.Intriguingly,BAP1 stabilizes N1ICD independent of its DUB activity but relying on the BRCA1-inhibiting function.BAP1 strengthens Notch signaling and maintains stem-like properties of cortical neural progenitor cells.Thus,NUMB enhances Notch signaling by regulating the ubiquitinating activity of the BAP1–BRCA1 complex. | Zhiyuan Luo Lili Mu Yue Zheng Wenchen Shen Jiali Li Lichao Xu Bo Zhong Ying Liu Yan Zhou | 2020 | Journal of Molecular Cell Biology2020,12,5: | 2 |

| 9 | Clinicopathological practice in the differential diagnosis of mucoepidermoid carcinoma from neoplasms with mucinous component显示文摘Background:The differential diagnosis of mucoepidermoid carcinoma(MEC)from neoplasm undergoing mucinous features brings more pitfalls to pathologists.Combining specific MAML2 gene rearrangement and histological characteristics may be the solution.Methods:Twenty-five tumors with mucinous components were selected for differential diagnosis of MEC.All the cases were detected for MAML2 gene rearrangement.The cases diagnosed as MEC were classified into four variants:classic,oncocytic,Warthin-like,and nonclassified,and they were graded using the Brandwein system.The histological characteristics of non-MECs were summarized for differential diagnosis.Univariate survival analysis was performed on MECs.Results:There were 16 MECs;62.5%were MAML2 rearranged.For the low-,intermediate-,and high-grade MECs,the rate of rearrangement was 83.3%,100%,and 28.6%,respectively.Both the oncocytic and Warthinlike MECs were MAML2 rearranged.For the classic and nonclassified MECs without MAML2 rearrangement,non-keratinized squamoid cells and distinctive mucinous cells were essential diagnostic criteria.On survival analysis,all the disease progression occurred in high-grade MECs(p=0.038).Nine cases were diagnosed as non-MECs:pleomorphic adenoma with mucinous metaplasia showed no ex-capsular involvement;metaplastic Warthin tumor appeared with overt keratinization and residual oncocytic bilayered epithelium;mix squamous cell and glandular papilloma showed an endobronchial papillary growing pattern;adenosquamous carcinoma was accompanied by squamous carcinoma in situ of the overlying mucosa.All the non-MECs were negative for MAML2 rearrangement.Conclusion:The application of combining MAML2 rearrangement and histological characteristics is helpful in the differential diagnosis between MEC and other tumors with mucinous components. | Yuelu Zhu Yan Li Lei Guo Wenbin Li Jiali Mu Haifeng Zhang Xin Li Jianming Ying Haizhen Lu | 2023 | Chronic Diseases and Translational Medicine2023,9,1: | 0 |
| 10 | Analysis of Key Physiological Characteristics of Portunus trituberculatus in Response to Short-Term Low Salinity Stress显示文摘Salinity is a significant environmental factor that can affect the survival,metamorphosis,growth and feeding of Portunus trituberculatus.In order to analyze the key physiological characteristics of P.trituberculatus in response to short-term low salinity stress,the experiments of gradually decline and recovery as well as abrupt decline in salinity were carried out.The results showed that P.trituberculatus could survive in a certain low salinity range in the short term,and salinity 12 was the lowest tolerable salinity under the present experimental conditions.The change of the hemolymph osmotic pressure displayed significant positive correlations with water salinity,and the pressure was always higher than seawater osmotic pressure.Short-term low salinity stress changed the structure and morphology of gill tissue.The expansion of gill filament ends and epithelial cell shedding were conducive to osmotic adjustment.The activities of key ion transport enzymes such as Na^(+)-K^(+)-ATPase,carbonic anhydrase and V-ATPase also changed with the osmotic regulation,while Na^(+)-K^(+)-ATPase played a dominant role.In summary,as an osmotic adjustment species,P.trituberculatus rapidly adapt to the short-term low-salinity environment by osmotic adjustment in vivo,but salinity below salinity 12 is not conducive to its survival.Our result enriched the theoretical mechanism of osmotic regulation of P.trituberculatus,providing reference for the development of aquaculture technology of P.trituberculatus. | ZHANG Xinni LIU Qi WANG Jiali GAO Gao NIU Mingming CHEN Lizhi WANG Huan WANG Fangfang WANG Chunlin MU Changkao | 2023 | Journal of Ocean University of China2023,22,3: | 0 |
