1Molecular diagnostics in a teacup:Non-Instrumented Nucleic Acid Amplification(NINA) for rapid,low cost detection of Salmonella enterica显示文摘We report on the use of a novel non-instrumented platform to enable a Loop Mediated isothermal Amplification(LAMP) based assay for Salmonella enterica.Heat energy is provided by addition of a small amount(<150 g) of boiling water,and the reaction temperature is regulated by storing latent energy at the melting temperature of a lipid-based engineered phase change material.Endpoint classification of the reaction is achieved without opening the reaction tube by observing the fluorescence of sequence-specific FRET-based assimilating probes with a simple handheld fluorometer.At or above 22℃ ambient temperature the non-instrumented devices could maintain reactions above a threshold temperature of 61℃ for over 90 min-significantly longer than the 60 min reaction time.Using the simple format,detection limits were less than 20 genome copies for reactions run at ambient temperatures ranging from 8 to 36℃.When used with a pre-enrichment step and non-instrumented DNA extraction device,trace contaminations of Salmonella in milk close to 1 CFU/mL could be reliably detected.These findings illustrate that the non-instrumented amplification approach is a simple,viable,low-cost alternative for field-based food and agricultural diagnostics or clinical applications in developing countries.KUBOTA Ryo LABARRE Paul WEIGL Bernhard H LI Yong HAYDOCK Paul JENKINS Daniel M 2013Chinese Science Bulletin2013,58,10:4

3Durability Properties of Materials and Mixture Composition of Bitumen Stabilized Materials显示文摘Bitumen Stabilized Materials (BSMs) are widely recognized as environmentally friendly through significantly reduced energy consumption,reduced emissions and reuse of high percentage of RAP and recycled pavement materials. In order for these materials to be cost effective,their life-cycle performance needs to be proven. Durability of BSMs is a long-term phenomena that requires consideration during mix design. Time is very important variable for performance consideration under field conditions. The factors that influence durability and long-term performance of BSMs include the mineral aggregates,binder and mixture compositions. In this study,the important,but less understood aspects of durability properties of BSMs were investigated. These include wear and tear of mineral aggregates,age-hardening of binder (foamed bitumen and bitumen emulsion) and moisture susceptibility of typical mix composition. The wear and tear was investigated using Durability Mill Index (DMI) test,binder ageing was investigated for short and long-term effect,and moisture susceptibility was investigated by pulsing water pressures using new device i.e. moisture induction simulation test (MIST). The accelerated moisture conditioning simulates field conditions whereafter retained shear properties are evaluated. The results found that durability of mineral aggregates is significantly influences by the type and nature of mineral composition. The ageing of binder occurred in both short and long-term conditions,with short-term effects being dominating the foamed bitumen properties during laboratory production. Moisture susceptibility is significantly influenced by the mix compositions of aggregates blends with and without RAP,with different bitumen binder types (foamed bitumen or bitumen emulsion) and active filler type (cement or lime) and contents. The evaluation and ranking of mixes which are resistant and susceptible to moisture damage was done using retained cohesion (RC) after dry and wet monotonic triaxial testing.TWAGIRA M E JENKINS J K 2010武汉理工大学学报2010,32,17:2
