| 9 | Lipolysis supports bone formation by providing osteoblasts with endogenous fatty acid substrates to maintain bioenergetic status显示文摘Bone formation is a highly energy-demanding process that can be impacted by metabolic disorders.Glucose has been considered the principal substrate for osteoblasts,although fatty acids are also important for osteoblast function.Here,we report that osteoblasts can derive energy from endogenous fatty acids stored in lipid droplets via lipolysis and that this process is critical for bone formation.As such,we demonstrate that osteoblasts accumulate lipid droplets that are highly dynamic and provide the molecular mechanism by which they serve as a fuel source for energy generation during osteoblast maturation.Inhibiting cytoplasmic lipolysis leads to both an increase in lipid droplet size in osteoblasts and an impairment in osteoblast function.The fatty acids released by lipolysis from these lipid droplets become critical for cellular energy production as cellular energetics shifts towards oxidative phosphorylation during nutrient-depleted conditions.In vivo,conditional deletion of the ATGL-encoding gene Pnpla2 in osteoblast progenitor cells reduces cortical and trabecular bone parameters and alters skeletal lipid metabolism.Collectively,our data demonstrate that osteoblasts store fatty acids in the form of lipid droplets,which are released via lipolysis to support cellular bioenergetic status when nutrients are limited.Perturbations in this process result in impairment of bone formation,specifically reducing ATP production and overall osteoblast function. | Ananya Nandy Ron C.M.Helderman Santosh Thapa Shobana Jayapalan Alison Richards Nikita Narayani Michael P.Czech Clifford J.Rosen Elizabeth Rendina-Ruedy | 2023 | Bone Research2023,11,4: | 0 |
| 10 | Computational identification and analysis of neurodegenerative disease associated protein kinases in hominid genomes显示文摘Protein kinases play an important role in the incidence of neurodegenerative diseases.However their incidence in non-human primates is found to be very low.Small differences among the genomes might influence the disease susceptibilities.The present study deals with finding the genetic differences of protein kinases in humans and their three closest evolutionary partners chimpanzee,gorilla and orangutan for three neurodegenerative diseases namely,Alzheimer’s,Parkinson’s and Huntington’s diseases.In total 47 human protein kinases associated with three neurodegenerative diseases and their orthologs from other three nonhuman primates were identified and analyzed for any possible susceptibility factors in humans.Multiple sequence alignment and pairwise sequence alignment revealed that,18 human protein kinases including DYRK1A,RPS6KB1,and GRK6 contained significant indels and substitutions.Further phosphorylation site analysis revealed that eight kinases including MARK2 and LTK contained sites of phosphorylation exclusive to human genomes which could be particular candidates in determining disease susceptibility between human and non-human primates.Final pathway analysis of these eight kinases and their targets revealed that these kinases could have long range consequences in important signaling pathways which are associated with neurodegenerative diseases. | Saranya Jayapalan Devika Subramanian Jeyakumar Natarajan | 2016 | Genes & Diseases2016,3,3: | 0 |
| 11 | Development of a dimensionless parameter for characterization of dielectric barrier discharge devices with respect to geometrical features显示文摘Non-thermal plasma(NTP) devices produce excited and radical species that have higher energy levels than their ground state and are utilized for various applications. There are various types of NTP devices, with dielectric barrier discharge(DBD) reactors being widely used. These DBD devices vary in geometrical configuration and operating parameters, making a comparison of their performance in terms of discharge power characteristics difficult. Therefore, this study proposes a dimensionless parameter that is related to the geometrical features, and is a function of the discharge power with respect to the frequency, voltage, and capacitance of a DBD. The dimensionless parameter, in the form of a ratio of the discharge energy per cycle to the gap capacitive energy, will be useful for engineers and designers to compare the energy characteristics of devices systematically, and could also be used for scaling up DBD devices.From the results in this experiment and from the literature, different DBD devices are categorized into three separate groups according to different levels of the energy ratio. The larger DBD devices have lower energy ratios due to their lower estimated surface discharge areas and capacitive reactance. Therefore, the devices can be categorized according to the energy ratio due to the effects of the geometrical features of the DBD devices, since it affects the surface discharge area and capacitance of the DBD.
