2Angiogenic markers endoglin and vascular endothelial growth factor in gastroenteropancreatic neuroendocrine tumors显示文摘AIM:To investigate the expression and potential prognostic role of vascular endothelial growth factor(VEGF) and endoglin in gastroenteropancreatic neuroendocrine tumors(GEP-NETs) . METHODS:Microvessel density(MVD) in GEP-NETs was evaluated using endoglin and CD31 immunohistochemistry.In addition,tissue levels of endoglin and VEGF were determined in homogenates by ELISA. RESULTS:Endoglin was highly expressed on tumor endothelial cells.CD31 MVD in GEP-NETs was significantly higher compared to endoglin MVD(P<0.01) .Two-tofour-fold higher tissue levels of endoglin and VEGF were seen in tumors compared to associated normal tissue. This increased endoglin tissue expression in tumors was significantly related to tumor size(P<0.01) ,presence of metastases(P=0.04) ,and a more advanced tumor stage(P=0.02) ,whereas expression of VEGF was not. 3Exploiting DNA repair pathways for tumor sensitization,mitigation of resistance,and normal tissue protection in radiotherapy显示文摘More than half of cancer patients are treated with radiotherapy,which kills tumor cells by directly and indirectly inducing DNA damage,including cytotoxic DNA double-strand breaks(DSBs).Tumor cells respond to these threats by activating a complex signaling network termed the DNA damage response(DDR).The DDR arrests the cell cycle,upregulates DNA repair,and triggers apoptosis when damage is excessive.The DDR signaling and DNA repair pathways are fertile terrain for therapeutic intervention.This review highlights strategies to improve therapeutic gain by targeting DDR and DNA repair pathways to radiosensitize tumor cells,overcome intrinsic and acquired tumor radioresistance,and protect normal tissue.Many biological and environmental factors determine tumor and normal cell responses to ionizing radiation and genotoxic chemotherapeutics.These include cell type and cell cycle phase distribution;tissue/tumor microenvironment and oxygen levels;DNA damage load and quality;DNA repair capacity;and susceptibility to apoptosis or other active or passive cell death pathways.We provide an overview of radiobiological parameters associated with X-ray,proton,and carbon ion radiotherapy;DNA repair and DNA damage signaling pathways;and other factors that regulate tumor and normal cell responses to radiation.We then focus on recent studies exploiting DSB repair pathways to enhance radiotherapy therapeutic gain.Jac A.Nickoloff Lynn Taylor Neelam Sharma Takamitsu A.Kato 2021Cancer Drug Resistance2021,4,2:4

6More forks on the road to replication stress recovery显示文摘High-fidelity replication of DNA,and its accurate segregation to daughter cells,is critical for maintaining genome stability and suppressing cancer.DNA replication forks are stalled by many DNA lesions,activating checkpoint proteins that stabilize stalled forks.Stalled forks may eventually collapse,producing a broken DNA end.Fork restart is typically mediated by proteins initially identified by their roles in homologous recombination repair of DNA double-strand breaks(DSBs).In recent years,several proteins involved in DSB repair by non-homologous end joining(NHEJ)have been implicated in the replication stress response,including DNA-PKcs,Ku,DNA Ligase IV-XRCC4,Artemis,XLF and Metnase.It is currently unclear whether NHEJ proteins are involved in the replication stress response through indirect(signaling)roles,and/or direct roles involving DNA end joining.Additional complexity in the replication stress response centers around RPA,which undergoes significant post-translational modification after stress,and RAD52,a conserved HR protein whose role in DSB repair may have shifted to another protein in higher eukaryotes,such as BRCA2,but retained its role in fork restart.Most cancer therapeutic strategies create DNA replication stress.Thus,it is imperative to gain a better under-standing of replication stress response proteins and pathways to improve cancer therapy.Chris Allen Amanda K.Ashley Robert Hromas Jac A.Nickoloff 2011Journal of Molecular Cell Biology2011,3,1:2
