| 1 | Transformation of hepatitis B serologic markers in babies born to hepatitis B surface antigen positive mothers显示文摘AIM: To better understand the clinical significance of hepatitis B serologic markers in babies born to hepatitis B surface antigen (HBsAg) positive mothers, the incidence of maternal serologic markers of hepatitis B via placenta and its transformation in these babies were investigated.METHODS: Mothers with positive HBsAg were selected in the third trimester of pregnancy. Their babies received immunoprophylaxis with hepatitis B immunoglobulin and hepatitis B vaccine after birth, and were consecutively followed up for hepatitis B serologic markers and HBV DNA at birth, mo 1, 4, 7, 12, and 24. RESULTS: Forty-two babies entered the study, including 16 born to hepatitis B e antigen (HBeAg)-positive HBsAg carrier mothers and 26 to HBeAg-negative HBsAg carrier mothers. Apart from four babies born to HBeAg-positive carrier mothers and demonstrated persistent positive HBeAg eventually became HBV carriers, all other babies developed anti-HBs before 12 mo of age. Among the other 12 babies born to HBeAg-positive carrier mothers,HBeAg was detected in 7 at birth, in 4 at mo 1, and in none of them thereafter. No antibody response to the transplacental HBeAg was detected. Among the babies born to HBeAg-negative carrier mothers, anti-HBe was detected 100% at birth and mo 1, in 88.5% at mo 4, in 46.2% at mo 7, in 4.2% at mo 12 and none in mo 24. Among all the immunoprophylaxis-protected babies born to either HBeAg-positive or HBeAg-negative carrier mothers, anti-HBc was detected in 100% at birth, mo 1 and mo 4, in 78.9% at mo 7, in 36.1% at mo 12 and in none at mo 24.CONCLUSION: HBeAg can pass through human placenta from mother to fetus and become undetectable before 4 mo of age, but no antibodies response to the transplacental HBeAg can be detected till mo 24 in the immunoprophylaxisprotected babies. | 2 | Influence of Temperature on Stacking Fault Energy and Creep Mechanism of a Single Crystal Nickel-based Superalloy显示文摘The influence of temperatures on the stacking fault energies and deformation mechanism of a Recontaining single crystal nickel-based superalloy during creep at elevated temperatures was investigated by means of calculating the stacking fault energy of alloy, measuring creep properties and performing contrast analysis of dislocation configuration. The results show that the alloy at 760 °C possesses lower stacking fault energy, and the stacking fault of alloy increases with increasing temperature. The deformation mechanism of alloy during creep at 760 °C is γ′ phase sheared by <110> super-dislocations, which may be decomposed to form the configuration of Shockley partials plus super-lattice intrinsic stacking fault, while the deformation mechanism of alloy during creep at 1070 °C is the screw or edge superdislocations shearing into the rafted γ′ phase. But during creep at 760 and 980 °C, some superdislocations shearing into γ′ phase may cross-slip from the {111} to {100} planes to form the K–W locks with non-plane core structure, which may restrain the dislocations slipping to enhance the creep resistance of alloy at high temperature. | 3 | An experimental investigation on acoustic emission characteristics of sandstone rockburst with different moisture contents显示文摘Rockburst occurred frequently during deep mining in China. The mechanism of rockburst is very complicated and related to many factors. In order to investigate the influence of moisture contents of rockmass on rockburst, we conducted a series of laboratory rockburst experiments of sandstone under three different moisture contents by the Modified True-Triaxial Apparatus (MTTA),in which the acoustic emission (AE) system was employed to monitor the internal damage of rock mass. A high-speed video camera was utilized to record the detail of rockburst. Based on the experimental results, the AE characteristics, such as AE count,AE energy, and AE frequency, were analyzed. The rockburst process, type, and indensity under different moisture contents were discussed. The research results show that with the increase of moisture contents, rock strength was soften, the elastic and the cumulative damage of the rock were reduced, resulting in a gradual decrease in AE cumulative counts and cumulative energy over the course of rockburst. | 5 | Infective viruses produced from full-length complementary DNA of swine vesicular disease viruses HK/70 strain显示文摘The full-length cDNA clone of swine vesicular disease virus HK/70 strain named pSVOK12 was constructed in order to study the antigenicity, replication, maturation and pathogenicity of swine vesicular disease virus. In vitro transcription RNA from pSVOK12 transfected IBRS-2 cells and the re- covered virus RNA were isolated and sequenced, then indirect hemagglutination test, indirect im- munofluorescence assays, eleectron microscope test, 50% tissue culture infecting dose (TCID50) assays and mouse virulence studies were performed to study the antigenicity and virulence of the recovered virus. The result showed that the infectious clones we ob- tained and the virus derived from pSVOK12 had the same biological properties as the parental strain HK/70. | 6 | Construction and analysis of SSH cDNA library of human vascular endothelial cells related to gastrocarcinoma显示文摘AIM: To construct subtracted cDNA libraries of human vascular endothelial cells (VECs) related to gastrocarcinoma using suppression substractive hybridization (SSH) and to analyze cDNA libraries of gastrocarcinoma and VECs in Cancer Gene Anatomy Project (CGAP) database.METHODS: Human VECs related to gastric adenocarcinoma and corresponding normal tissue were separated by magnetic beads coupled with antibody CD31 (Dynabeads CD31). A few amount of total RNA were synthesized and amplified by SMARTTM PCR cDNA Synthesis Kit. Then, using SSH and T/A cloning techniques, cDNA fragments of differentially expressed genes in human VECs of gastric adenocarcinoma were inserted into JM109 bacteria. One hundred positive bacteria clones were randomly picked and identified by colony PCR method. To analyze cDNA libraries of gastrocarcinoma and VECs in CGAP database, the tools of Library Finder,cDNA xProfiler, Digital GENE Expression Displayer (DGED),and Digital Differential Display (DDD) were used.RESULTS: Forward and reverse subtraction cDNA libraries of human VECs related to gastrocarcinoma were constructed successfully with SSH and T/A cloning techniques. However, both models are approximations to the real situation when viewpoint position is not absolutely fixed but includes a small translation, and thus distortion and blurring are sometimes present in the final registration results. | 11 | Effects of System Pressure and Heat Flux on Bubble Nucleation and Growth显示文摘Characteristics of bubble nucleation and growth are critical for its application. It is affected by several factors including viscosity, surface tension and temperature. However, the effect of pressure on bubble nucleation and growth has been underreported, although it processes significant effect on above characteristics. In this work, a micro copper electrode is etched on a slab covered with copper to produce bubble on the surface by current input. The nucleation time of bubble is measured under different heat flux and system pressures. The nucleation and growth processes are recorded with a high speed camera in order to discuss the effects of heat flux and system pressure on bubble characteristics. The experiment results indicate that the micro electrode with higher heat flux produces more thermal energy, which makes the time of bubble nucleation shorter and the speed of bubble growth faster. Higher system pressure causes the increase of the critical nucleation temperature and also baffles the bubble nucleation and growth. Bubble growth includes the stages of rapid growth and dynamic equilibrium, with the speed being from fast to slow. In the former part of rapid growth, heat flux plays a dominant role in bubble growth. While the effect of system pressure on bubble growth becomes significant in the latter part of rapid growth. Both the nucleation time and bubble growth agree well with the theoretical analysis.
