| 5 | Over-expression of WOX1 Leads to Defects in Meristem Development and Polyamine Homeostasis in Arabidopsis显示文摘In plants,the meristem has to maintain a separate population of pluripotent cells that serve two main tasks,i.e.,self-maintenance and organ initiation,which are separated spatially in meristem.Prior to our study,WUS and WUS-like WOX genes had been reported as essential for the development of the SAM.In this study,the consequences of gain of WOX1 function are described.Here we report the identification of an Arabidopsis gain-of-function mutant wox1-D,in which the expression level of the WOX1(WUSCHEL HOMEOBOX 1) was elevated and subtle defects in meristem development were observed.The wox1-D mutant phenotype is dwarfed and slightly bushy,with a smaller shoot apex.The wox1-D mutant also produced small and dark green leaves,and exhibited a failure in anther dehiscence and male sterility.Molecular evidences showed that the transcription of the stem cell marker gene CLV3 was down-regulated in the meristem of wox1-D but accumulated in the other regions,i.e.,in the root-hypocotyl junction and at the sites for lateral root initiation.The fact that the organ size and cell size in leaves of wox1-D are smaller than those in wild type suggests that cell expansion is possibly affected in order to have partially retarded the development of lateral organs,possibly through alteration of CLV3 expression pattern in the meristem.An S-adenosylmethionine decarboxylase(SAMDC) protein,SAMDC1,was found able to interact with WOX1 by yeast two-hybrid and pull-down assays in vitro.HPLC analysis revealed a significant reduction of polyamine content in wox1-D.Our results suggest that WOX1 plays an important role in meristem development in Arabidopsis,possibly via regulation of SAMDC activity and polyamine homeostasis,and/or by regulating CLV3 expression. | Yanxia Zhang Renhong Wu Genji Qin Zhangliang Chen Hongya Gu Li-Jia Qu | 2011 | Journal of Integrative Plant Biology2011,53,6: | 23 |

| 10 | Variation at the transcriptional level among Chinese natural populations of Arabidopsis thaliana in response to cold stress显示文摘The Arabidopsis 25K GeneChip (ATH1, Affymetrix) was used to make a survey of the variation of the transcriptional profiles among 5 Chinese natural populations of Arabidopsis thaliana under cold treatment. In normal growth condition, the expression level of 2.26% (513 genes in the population from Jiujiang, Jiangxi, JXjjx) to 6.52% (1482 genes in the population from Tongliang, Chongqing, CQtlx) genes was 2-fold higher than that of Col ecotype. Under cold treatment, the expression of 12.84% (2920 genes in the population from Chenggu, Shaanxi, SXcgx) to 19.46% (4426 genes in the population from Qinghe, Xinjiang, XJqhx) genes was up- or down-regulated by at least two-fold that of their controls. In general, most of up-regulated genes might be the genes essential for plant surviving at low temperature, such as genes in CBF pathway and the genes responsible for synthesizing molecules accumulated for cold tolerance. However, each natural population had some specific genes induced under cold treatment. The data indicated that some of the cold-responding genes were differentiated among the populations distributed in the natural habitats with different climate conditions. CBF3, one of the key tran-scription factor genes in cold responding pathway, showed significant differences in expression among populations. The sequence analysis indicated that the changes in its regulation region caused the dramatic difference in the expression pattern. They are important for flower pigmentation, protection against UV light and defense against phytopathogens. Many plants were found to contain multiple copies of CHS genes. This review summarizes the recent progress in the studies of the CHS-superfamily, focusing on the duplication and divergent evolution of the CHS and CHS-like genes. Comparative analyses of gene structure, ex- pression patterns and catalytic properties revealed extensive differentiation in both regulation and func- tion among duplicate CHS genes. It is also proposed that the CHS-like enzymes in the CHS-superfamily evolved from CHS at different times in various or- ganisms. | 17 | The Arabidopsis Anaphase-Promoting Complex/Cyclosome Subunit 1 is Critical for Both Female Gametogenesis and Embryogenesis显示文摘Anaphase-promoting complex/cyclosome (APC/C), a multisubunit E3 ligase, plays a critical role in cell cycle control, but the functional characterization of each subunit has not yet been completed. To investigate the function of APC1 in Arabidopsis, we analyzed four mutant alleles of APC1, and found that mutation in APC1 resulted in significantly reduced plant fertility, accumulation of cyclin B, and disrupted auxin distribution in embryos. The three mutant alleles apc1-1, apc1-2 and apc1-3 shared variable defects in female gametogenesis including degradation, abnormal nuclear number, and disrupted polarity of nuclei in the embryo sac as well as in embryogenesis, in which embryos were arrested at multiple stages. All of these defects are similar to those previously identified in apc4. The mutant apc1-4, in which the T-DNA was inserted after the transmembrane domain at the C-terminus, showed much more severe phenotypes; that is, most of the ovules were arrested at the one-nucleate female gametophyte stage (stage FG1). In the apc1 apc4 double mutants, the fertility was further reduced by one-third in apc1-1/+ apc4-1/+, and in some cases no ovules even survived in siliques of apc1-4/+ apc4-1/+. | 18 | Molecular evolution of the exon 2 of CHS genes and the possibility of its application to plant phylogenetic analysis显示文摘The exon 2 of chalcone synthase (CHS) gene is relatively conserved during evolution. In this study, three exon 2 fragments from two species in gymnosperm (Cycas panzhihuaensis, Ginkgo biloba ) and seven from four species in angiosperm (Magnolia denudata, Salix babylonica, Nymphaea tetragona, Camellia japonica) have been amplified by PCR from genomic DNA and sequenced. Together with other 73 sequences of CHS collected from EMBL database and literature, these sequences, which embrace 19 families of gymnosperm and angiosperm, have been analyzed for their phylogenetic relations by parsimony method. The result indicated that sequences from the same systematic family usually grouped together except those from Theaceae, Magnoliaceae and Nymphaeaceae. The relative rate test revealed the rate heterogeneity of CHS genes among the families. | 20 | Experimental demonstration of ultra-large-scale terahertz all-dielectric metamaterials显示文摘All-dielectric metamaterials have emerged as a promising platform for low-loss and highly efficient terahertz devices. However, existing fabrication methods have difficulty in achieving a good balance between precision and cost. Here, inspired by the nano-template-assisted self-assembly method, we develop a micro-templateassisted self-assembly(MTAS) method to prepare large-scale, high-precision, and flexible ceramic microsphere all-dielectric metamaterials with an area exceeding 900 cm × 900 cm. Free from organic solvents, vacuum, and complex equipment, the MTAS method ensures low-cost and environmentally friendly fabrication. The ceramic microsphere resonators can be readily assembled into nearly arbitrary arrangements and complex aggregates, such as dimers, trimers, quadrumers, and chains.
