| 2 | Introduction to bioimaging-based spatial multi-omic novel methods显示文摘Background: Spatial multi-omics are demonstrated to be a powerful method to assist researchers on genetic studies. In this review, bioimaging-based spatial multi-omics techniques such as seqFISH+, merFISH, integrated DNA seqFISH+, DNA merFISH, and MINA are introduced along with each technique’s probe design, development, and imaging processes.Results: seqFISH employed 4–5 fluorophores to barcode and conducted multiple rounds of hybridization, in order that mRNA can be identified through color-coding. seqFISH+ added 60 pseudo-color and distributed them equally into three channels to enhance imaging power, in order that i.e., 24,000 genes can be imaged in total. merFISH utilized 4 out 16 Hamming distance to innovatively provide a robust error-detecting method. MINA, a methodology combining merFISH (multiplexed error-robust fluorescence in situ hybridization) and chromosomal tracing, enabled multiplexed genomic architecture imaged in mammalian single cells. Optical reconstruction of chromatin architecture (ORCA) a method that could conduct DNA path tracing in nanoscale manner with kilobase resolution, an FISH variation that improved genetic resolution, enable high-precision fiducial registration and sequential imaging, and utilized Oligopaint probe to hybridize the short genomic region ranging from 2 to 10 kilobase. | 3 | Robust model predictive tracking control for the wheeled mobile robot with boundary uncertain based on linear matrix inequalities显示文摘In this study,a robust model predictive controller is designed for the trajectory tracking problem of non-holonomic constrained wheeled mobile robot based on an elliptic invariant set approach.The controller is based on a time-varying error model of robot kinematics and uses linear matrix inequalities to solve the robust tracking problem taking uncertainties into account.The uncertainties are modelled by linear fractional transform form to contain both parameter perturbations and external disturbances.The control strategy consists of a feedforward term that drives the centre of the ellipse to the reference point and a feedback term that converges the uncertain system state error to the equilibrium point.The strategy stabilises the nominal system and ensures that all states of the uncertain system remain within the ellipsoid at each step,thus achieving robust stability of the uncertain system.Finally,the robustness of the algorithm and its resistance to disturbances are verified by simulation and experiment. | Xing Gao Xin Su Aimin An Haochen Zhang | 2023 | IET Cyber-Systems and Robotics2023,5,1: | 0 |
| 4 | Blockade of integrin signaling reduces chemotherapy-induced premature senescence in collagen cultured bladder cancer cells显示文摘Background:Diminished sensitivity towards chemotherapy remains the major impediment to the clinical treatment of bladder cancer.However,the critical elements in control of chemotherapy resistance remain obscure.Methods:We adopted improved collagen gels and performed cytotoxicity analysis of doxorubicin(DOX)and mitomycin C(MMC)of bladder cancer cells in a 3D culture system.We then detected the expression of multidrug resistant gene ABCB1,dormancy-associated functional protein chicken ovalbumin upstream-transcription factor 1(COUPTF1),cell proliferation marker Ki-67,and cellular senescence marker senescence-associatedβ-galactosidase(SA-β-Gal)in these cells.We further tested the effects of integrin blockade or protein kinase B(AKT)inhibitor on the senescent state of bladder cancer.Also,we examined the tumor growth and survival time of bladder cancer mouse models given the combination treatment of chemotherapeutic agents and integrinα2β1 ligand peptide TFA(TFA).Results:Collagen gels played a repressive role in bladder cancer cell apoptosis induced by DOX and MMC.In mechanism,collagen activated the integrinβ1/AKT cascade to drive bladder cancer cells into a premature senescence state via the p21/p53 pathway,thus attenuating chemotherapy-induced apoptosis.In addition,TFA had the ability to mediate the switch from senescence to apoptosis of bladder cancer cells in xenograft mice.Meanwhile,TFA combined with chemotherapeutic drugs produced a substantial suppression of tumor growth as well as an extension of survival time in vivo.Conclusions:Based on our finding that integrinβ1/AKT acted primarily to impart premature senescence to bladder cancer cells cultured in collagen gel,we suggest that integrinβ1 might be a feasible target for bladder cancer eradication. | Linghui Deng Kun Jin Xianghong Zhou Zilong Zhang Liming Ge Xingyu Xiong Xingyang Su Di Jin Qiming Yuan Chichen Zhang Yifan Li Haochen Zhao Qiang Wei Lu Yang Shi Qiu | 2022 | Precision Clinical Medicine2022,5,2: | 0 |
