| 1 | Differences in symptoms and pre-hospital delay among acute myocardial infarction patients according to ST-segment elevation on electrocardiogram: an analysis of China Acute Myocardial Infarction (CAMI) registry显示文摘Background: Approximately 70% patients with acute myocardial infarction (AMI) presented without ST-segment elevation on electrocardiogram. Patients with non-ST segment elevation myocardial infarction (NSTEMI) often presented with atypical symptoms, which may be related to pre-hospital delay and increased risk of mortality. However, up to date few studies reported detailed symptomatology of NSTEMI, particularly among Asian patients. The objective of this study was to describe and compare symptoms and presenting characteristics of NSTEMI vs. STEMI patients. Methods: We enrolled 21,994 patients diagnosed with AMI from China Acute Myocardial Infarction (CAMI) Registry between January 2013 and September 2014. Patients were divided into 2 groups according to ST-segment elevation: ST-segment elevation (STEMI) group and NSTEMI group. We extracted data on patients' characteristics and detailed symptomatology and compared these variables between two groups. Results: Compared with patients with STEMI (N=16,315), those with NSTEMI (N=5679) were older, more often females and more often have comorbidities. Patients with NSTEMI were less likely to present with persistent chest pain (54.3% vs.71.4%), diaphoresis (48.6% vs.70.0%), radiation pain (26.4% vs.33.8%), and more likely to have chest distress (42.4% vs.38.3%) than STEMI patients (all P<0.0001). Patients with NSTEMI were also had longer time to hospital. In multivariable analysis, NSTEMI was independent predictor of presentation without chest pain (odds ratio: 1.974, 95% confidence interval:1.849-2.107). Conclusions: Patients with NSTEMI were more likely to present with chest distress and pre-hospital patient delay compared with patients with STEMI. | 5 | Paleoproterozoic reworking of ancient crust in the Cathaysia Block,South China:Evidence from zircon trace elements,U-Pb and Lu-Hf isotopes显示文摘A combined study of zircon LA-ICP-MS U-Pb dating, trace elements and Hf isotope was carried out for gneissic granite from the Sanzhishu area in Jingning, SW Zhejiang Province. Nearly all the zircons separated from the granite exhibited oscillatory zoning and high Th/U ratios (>0.1). The REE profile showed a pronounced positive Ce anomaly, negative Eu anomaly and an enrichment of HREE, which are typical characteristics of magmatic zircon. Thirteen concordant or nearly concordant analytical data yielded a weighted mean 207Pb/206Pb age of 1860±13 Ma (MSWD=0.084), representing the formation age of the granite. The magmatic zircons had negative εHf(t) values of -15.6 to -10.0 and two-stage Hf model ages of 3.1 to 3.4 Ga, indicating that the granites were formed by reworking of ancient crust. The major- and trace-element data indicate that the gneissic granites are metaluminous high-K calc-alkaline rocks and exhibit the same geochemical characteristics as aluminous A-type granites, implying the emplacement of the granite in a post-orogenic extensional tectonic setting. Methods After ig administration of LBP-IV [50, 100, and 200 mg/(kg·d)] and LBP [100 mg/(kg·d)] once daily for consecutive 4 weeks to diabetic rats, the glucose and lipids in blood, m RNA expression of phosphoenolpyruvate carboxykinase(PEPCK), sterol regulatory element binding-protein-1c(SREBP-1c), and fatty acid synthase(FAS) in liver were determined. Results Ig administration of LBP and LBP-IV significantly decreased the levels of blood glucose, Hb A1 c, TC, TG, and LDL-C, as well as the hepatic m RNA expression of PEPCK, SREBP-1c, and FAS, whereas significantly increased the oral glucose tolerance of diabetic rats. | 7 | Genome Engineering in Rice Using Cas9 Variants that Recognize NG PAM Sequences显示文摘CRISPR/Cas9 genome editing relies on sgRNA-target DNA base pairing and a short downstream PAM sequence to recognize target DNA. The strict protospacer adjacent motif (PAM) requirement hinders applications of the CRISPR/Cas9 system since it restricts the targetable sites in the genomes. xCas9 and SpCas9-NG are two recently engineered SpCas9 variants that can recognize more relaxed NG PAMs, implying a great potential in addressing the issue of PAM constraint. Here we use stable transgenic lines to evaluate the efficacies of xCas9 and SpCas9-NG in performing gene editing and base editing in rice. We found that xCas9 can efficiently induce mutations at target sites with NG and GAT PAM sequences in rice. However, base editors containing xCas9 failed to edit most of the tested target sites. SpCas9-NG exhibited a robust editing activity at sites with various NG PAMs without showing any preference for the third nucleotide after NG. Moreover, we showed that xCas9 and SpCas9-NG have higher specificity than SpCas9 at the CGG PAM site. We further demonstrated that different forms of cytosine or adenine base editors containing SpCas9-NG worked efficiently in rice with broadened PAM compatibility. | 8 | Anatomy of composition and nature of plate convergence: Insights for alternative thoughts for terminal India-Eurasia collision显示文摘The pattern and timing of collision between India and Eurasia have long been a major concern of the international community. However, no consensus has been reached hitherto. To explore and resolve the disagreements in the Himalayan study,in this paper we begin with the methodology and basic principles for the anatomy of composition and nature of convergent margins,then followed by an effort to conduct a similar anatomy for the India-Eurasia collision. One of the most common patterns of plate convergence involves a passive continental margin, an active continental margin and intra-oceanic basins together with accreted terranes in between. The ultimate configuration and location of the terminal suture zone are controlled by the basal surface of the accretionary wedge, which may show fairly complex morphology with Z-shape and fluctuant geometry. One plausible method to determine the terminal suture zone is to dissect the compositions and structures of active continental margins. It requires a focus on various tectonic elements belonging to the upper plate, such as accretionary wedges, high-pressure(HP)-ultra-high-pressure(UHP) metamorphic rocks, Barrovian-type metamorphic rocks and basement nappes, together with superimposed forearc basins.Such geological records can define the extreme limits and the intervening surface separating active margin from the passive one,thus offering a general sketch for the surface trace of the terminal suture zone often with a cryptic feature. Furthermore, the occurrence of the cryptic suture zone in depth may be constrained by geophysical data, which, in combination with outcrop studies of HP-UHP metamorphic rocks, enables us to outline the terminal suture zone. The southern part of the Himalayan orogen records complicated temporal and spatial features, which are hard to be fully explained by the classic 'two-plate-one-ocean' template,therefore re-anatomy of the compositions and nature for this region is necessitated. Taking advantage of the methodology and basic principles of plate convergence anatomy and synthesizing previous studies together with our recent research, we may gain new insights into the evolution of the Himalayan orogeny.(1) The Yarlung-Zangbo ophiolite is composed of multiple tectonic units rather than a single terminal suture zone, and a group of different tectonic units were juxtaposed against each other in the backstop of the Gangdese forearc.(2) The Tethyan Himalayan Sequence(THS) contains mélanges with typical block-in-matrix structures, uniform southwards paleocurrents and age spectra of detrital zircons typical of Eurasia continent. All of these facts indicate that the THS belonged to Eurasia plate before the terminal collision, emplaced in the forearc of the Gangdese arc.(3) The Greater Himalayan Crystalline Complex(GHC) and Lesser Himalayan Sequence(LHS) comprise complex components including eclogites emplaced into the GHC and the upper part of the LHS. Judging from the fact that HP-UHP metamorphic rocks are exhumed and emplaced in the upper plate, the GHC and the upper part of the LHS where eclogite occur should be assigned to the upper plate, lying above the terminal subduction zone surface. It is the very surface along which the continuous subduction of the India subcontinent occurred, therefore acting as the terminal, cryptic suture. From the suture further southward, the bulk rock associations of the LHS and Sub-Himalayan Sequence(Siwalik) show little affinity of mélange, probably belonging to the foreland system of the India plate. By the anatomy of tectonic features of all the tectonic units in the Himalayan orogen as well as the ages of the subduction-accretion related deformation, we conclude that the terminal India-Eurasia collision occurred after 14 Ma, the timing of the metamorphism of the eclogites emplaced into the upper plate. The development of rifts stretching in N-S direction in Tibet and tectonic events with the transition from sinistral to dextral movements in shear zones, such as the Ailaoshan fault in East Tibet, can coordinately reflect the scale and geodynamic influence of the India-Eurasia convergence zone.By conducting a detailed anatomy of the southern Himalayas, we propose a new model for the final collision-accretion of the Himalayan orogeny. Our study indicates that the anatomy of structures, composition, and tectonic nature is the key to a better understanding of orogenic belts, which may apply to all the orogenic belts around the world. | 9 | Efficient Targeted Genome Modification in Maize Using CRISPR/Cas9 System显示文摘CRISPR(clustered regularly interspaced short palindromic repeats)/Cas9 system, which is a newly developed technology for targeted genome modification, has been successfully used in a number of species. In this study, we applied this technology to carry out targeted genome modification in maize. A marker gene Zmzb7 was chosen for targeting. The sg RNA-Cas9 construct was transformed into maize protoplasts, and indel(insertion and deletion) mutations could be detected. A mutant seedling with an expected albino phenotype was obtained from screening 120 seedlings generated from 10 callus events. Mutation efficiency in maize heterochromatic regions was also investigated. Twelve sites with different expression levels in maize centromeres or pericentromere regions were selected. The sg RNACas9 constructs were transformed into protoplasts followed by sequencing the transformed protoplast genomic DNA. The results show that the genes in heterochromatic regions could be targeted by the CRISPR/Cas9 system efficiently, no matter whether they are expressed or not. Meanwhile, off-target mutations were not found in the similar sites having no PAM(protospacer adjacent motif) or having more than two mismatches. | 14 | A Deep Learning Method for Bias Correction of ECMWF 24–240 h Forecasts显示文摘Correcting the forecast bias of numerical weather prediction models is important for severe weather warnings.The refined grid forecast requires direct correction on gridded forecast products,as opposed to correcting forecast data only at individual weather stations.In this study,a deep learning method called CU-net is proposed to correct the gridded forecasts of four weather variables from the European Centre for Medium-Range Weather Forecast Integrated Forecasting System global model(ECMWF-IFS): 2-m temperature,2-m relative humidity,10-m wind speed,and 10-m wind direction,with a forecast lead time of 24 h to 240 h in North China.First,the forecast correction problem is transformed into an image-toimage translation problem in deep learning under the CU-net architecture,which is based on convolutional neural networks.Second,the ECMWF-IFS forecasts and ECMWF reanalysis data(ERA5) from 2005 to 2018 are used as training,validation,and testing datasets.The predictors and labels(ground truth) of the model are created using the ECMWF-IFS and ERA5,respectively.Finally,the correction performance of CU-net is compared with a conventional method,anomaly numerical correction with observations(ANO).Results show that forecasts from CU-net have lower root mean square error,bias,mean absolute error,and higher correlation coefficient than those from ANO for all forecast lead times from 24 h to 240 h.CU-net improves upon the ECMWF-IFS forecast for all four weather variables in terms of the above evaluation metrics,whereas ANO improves upon ECMWF-IFS performance only for 2-m temperature and relative humidity.For the correction of the 10-m wind direction forecast,which is often difficult to achieve,CU-net also improves the correction performance. | Lei HAN Mingxuan CHEN Kangkai CHEN Haonan CHEN Yanbiao ZHANG Bing LU Linye SONG Rui QIN | 2021 | Advances in Atmospheric Sciences2021,38,9: | 15 |
| 15 | Hypoxia upregulates hypoxia inducible factor(HIF)-3α expression in lung epithelial cells: characterization and comparison with HIF-1α显示文摘The role of the hypoxia-inducible factor(HIF)subunits 1α and 2α in response to hypoxia is well established in lungepithelial cells,whereas little is known about HIF-3α with respect to transcriptional and translational regulation by hy-poxia.HIF-3α and HIF-1α are two similar but distinct basic helix-loop-helix-PAS proteins,which have been postulatedto activate hypoxia responsive genes in response to hypoxia.Here,we used quantitative real time RT-PCR and immu-noblotting to determine the activation of HIF-3α vs.HIF-1α by hypoxia.HIF-3α was strongly induced by hypoxia(1%O_2)both at the level of protein and mRNA due to an increase in protein stability and transcriptional activation,whereasHIF-1α protein and mRNA levels enhanced transiently and then decreased because of a reduction in its mRNA stabilityin A549 cells,as measured on mRNA and protein levels.Interestingly,HIF-3α and HIF-1α exhibited strikingly similarresponses to a variety of activating or inhibitory pharmacological agents.These results demonstrate that HIF-3α is ex-pressed abundantly in lung epithelial cells,and that the transcriptional induction of HIF-3α plays an important role in theresponse to hypoxia in vitro.Our findings suggest that HIF-3α,as a member of the HIF system,is complementary ratherthan redundant to HIF-1α induction in protection against hypoxic damage in alveolar epithelial cells. | Qi Fang Li Xiang Rui Wang Yue Wu Yang Han Lin | 2006 | Cell Research2006,16,6: | 14 |

| 19 | Boron neutron capture therapy for malignant melanoma: first clinical case report in China显示文摘A phase Ⅰ/Ⅱ clinical trial for treating malignant melanoma by boron neutron capture therapy(BNCT) was designed to evaluate whether the world's first in-hospital neutron irradiator(IHNI) was qualified for BNCT. In this clinical trial planning to enroll 30 patients, the first case was treated on August 19, 2014. We present the protocol of this clinical trial, the treating procedure, and the clinical outcome of this first case. Only grade 2 acute radiation injury was observed during the first four weeks after BNCT and the injury healed after treatment. No late radiation injury was found during the 24-month follow-up.
