| 1 | Matrine attenuates oxidative stress and cardiomyocyte apoptosis in doxorubicin-induced cardiotoxicity via maintaining AMPKα/UCP2 pathway显示文摘Oxidative stress and cardiomyocyte apoptosis are involved in the pathogenesis of doxorubicin(DOX)-induced cardiotoxicity. Matrine is well-known for its powerful anti-oxidant and anti-apoptotic capacities. Our present study aimed to investigate the effect of matrine on DOX-induced cardiotoxicity and try to unearth the underlying mechanisms. Mice were exposed with DOX to generate DOX-induced cardiotoxicity or normal saline as control. H9 C2 cells were used to verify the effect of matrine in vitro.DOX injection triggered increased generation of reactive oxygen species(ROS) and excessive cardiomyocyte apoptosis, which were significantly mitigated by matrine. Mechanistically, we found that matrine ameliorated DOX-induced uncoupling protein 2(UCP2) downregulation, and UCP2 inhibition bygenipin could blunt the protective effect of matrine on DOX-induced oxidative stress and cardiomyocyte apoptosis. Besides, 50-AMP-activated protein kinase α2(Ampkα2) deficiency inhibited matrine-mediated UCP2 preservation and abolished the beneficial effect of matrine in mice. Besides, we observed that matrine incubation alleviated DOX-induced H9 C2 cells apoptosis and oxidative stress level via activating AMPKα/UCP2, which were blunted by either AMPKα or UCP2 inhibition with genetic or pharmacological methods. | 3 | Gas stunning with CO_2 affected meat color,lipid peroxidation, oxidative stress, and gene expression of mitogen-activated protein kinases, glutathione S-transferases,and Cu/Zn-superoxide dismutase in the skeletal muscles of broilers显示文摘Background: Meat color and lipid peroxidation are important traits related to meat quality. CO_2 concentration is a critical factor that can affect meat quality in the commercial use of gas stunning(GS). However, the effect and mechanism of CO_2 stunning on meat color and lipid peroxidation during long-term storage remain poorly studied. We aimed to study the effects of GS methods, especial y CO_2 concentration, on meat color and meat lipid peroxidation in broilers during long-term storage at 4 °C and to explore the potential mechanism of meat color change via lipid peroxidation and the inner lipid peroxide scavenging system.Methods: Eighteen broilers were sacrificed after exposure to one of the following gas mixtures for 90 s: 40% CO_2+21% O_2+ 39% N2(G40%), 79% CO_2+ 21% O_2(G79%), or no stunning(0% CO_2, control). Meat color, serum variables,enzyme activities, and the gene expression of mitogen-activated protein kinase(MAPK), nuclear factor-erythroid2-related factor 2(Nrf2), glutathione S-transferase(GST) and superoxide dismutase(SOD) were determined.(Continued on next page)Results: The concentrations of serum triiodothyronine(T3, P = 0.03) and the ratio of serum free triiodothyronine/free thyroxine(FT3/FT4, P < 0.01) were decreased, whereas levels of serum cortisol(P < 0.01) were increased in the 40%CO_2 group compared with the control group. Additionally, the thiobarbituric acid-reactive substances(TBARS)3 d(P < 0.01) and TBARS6 d(P = 0.01) in breast meat and the TBARS3 din thigh meat(P < 0.01) were increased in the40% CO_2 group compared with the control group. Serum T3 was negatively correlated with TBARS6 dboth in the breast and thigh meat(r =-0.63, P < 0.01 and r =-0.47, P = 0.05 respectively). T3/T4 was negatively correlated with TBARS6 din the breast meat and in the thigh meat(r =-0.57, P = 0.01; and r =-0.53, P = 0.03 respectively). Compared with the control group, Lightness(L*)1 d(P = 0.03) and L*9 d(P < 0.01) were increased, whereas total chromatic aberration(E*)1 d(P = 0.05) and E*3 d(P < 0.01) were decreased in the breast meat of both the G40% and G79% groups. The values of yel owness(b*)3 d(P = 0.01), b*6 d(P < 0.01) and E*6 d(P < 0.01) in the thigh meat were lower in both the G40% and G79% groups than in the control group. In the breast muscle, the m RNA levels of c-Jun N-terminal kinase 2(JNK2, P = 0.03),GSTT1(P = 0.04), and SOD1(P = 0.05) were decreased, and the m RNA levels of JNK1(P = 0.07), Nrf2(P = 0.09), and GSTA3(P = 0.06) were slightly lower in both the G40% and G79% groups compared with the control group. However, among these genes, only the m RNA level of JNK1 was decreased in the G40% group compared with the control group and the G79% group(P = 0.03) in the thigh muscle.Conclusions: Compared with the control group, meat color quality in the breast meat was decreased, and the expression of genes in the MAPK/Nrf2/ARE(antioxidant responsive element) antioxidant pathway in breast muscle was partly suppressed by GS of both 40% and 79% CO_2. | 4 | Effects of soil nutrients and climate factors on belowground biomass in an alpine meadow in the source region of the Yangtze-Yellow rivers, Tibetan Plateau of China显示文摘Improving our knowledge of the effects of environmental factors(e.g.soil conditions,precipitation and temperature) on belowground biomass in an alpine grassland is essential for understanding the consequences of carbon storage in this biome.The object of this study is to investigate the relative importance of soil nutrients and climate factors on belowground biomass in an alpine meadow in the source region of the Yangtze and Yellow rivers,Tibetan Plateau.Soil organic carbon(SOC),total nitrogen(TN) and total phosphorous(TP) contents and belowground biomass were measured at 22 sampling sites across an alpine meadow on the Tibetan Plateau.We analyzed the data by using the redundancy analysis to determine the main environmental factors affecting the belowground biomass and the contribution of each factor.The results showed that SOC,TN and TP were the main factors that influenced belowground biomass,and the contribution of SOC,TN and TP on biomass was in the range of 47.87%–72.06% at soil depths of 0–30 cm.Moreover,the combined contribution of annual mean temperature(AMT) and mean annual precipitation(MAP) on belowground biomass ranged from 0.92% to 4.10%.A potential mechanism for the differences in belowground biomass was caused by the variations in soil nitrogen and phosphorous,which were coupled with SOC.A significant correlation was observed between MAP and soil nutrients(SOC,TN and TP) at the soil depth of 0–10 cm(P<0.05).We concluded that precipitation is an important driving force in regulating ecosystem functioning as reflected in variations of soil nutrients(SOC,TN and TP) and dynamics of belowground biomass in alpine grassland ecosystems. | WANG Haiming SUN Jian LI Weipeng WU Jianbo CHEN Youjun LIU Wenhui | 2016 | Journal of Arid Land2016,8,6: | 5 |

| 7 | Application of Optical Tweezers in the Research of Molecular Interaction between Lymphocyte Function Associated Antigen-1 and Its Monoclonal Antibody显示文摘Lymphocyte function associated antigen-1 (CD11a/CD18, LFA-1) plays an important role in the structure of the immunological synapse and is required for efficient lysis of cytotoxic T lymphocytes (CTLs) and natural killer (NK) cells. To study the activation mode of LFA-1 on the NK cell surface, optical tweezers were used in the work. As an emerging technology, optical tweezers are widely used to manipulate microscopic objects and measure the forces of molecular interactions in the field of biological research. In our study, a new platform was constructed to study the single molecular behavior of receptor on cell surface using optical tweezers. Based on the platform, the interaction between an NK cell and a polystyrene microsphere coated with anti-LFA-1 antibody was observed. The result confirmed that the adhesion forces between an NK cell and a polystyrene bead were time-dependent. | 9 | Asymmetric Response of the South China Sea SST to El Nio and La Nia显示文摘The interannual variability of the sea surface temperature (SST) in the South China Sea (SCS) is investigated according to its relationship with El Nio/La Nia (EN/LN) using monthly products from ICOADS. The SCS SST bears two peaks associated with EN/LN and shows the asymmetric features. Coinciding with the mature phase of EN/LN, the first SST warming/cooling peaks in December(0)-February(1) (DJF(1)) and centers in the southern part. The major difference is in the amplitude associated with the strength of EN/LN. However, the SCS SST anomaly shows distinct difference after the mature phase of EN/LN. The EN SST warm-ing develops a mid-summer peak in June-August(1) (JJA(1)) and persists up to September-October(1), with the same amplitude of the first warming peak. Whereas the LN SST cooling peaks in May(1), it decays slowly until the end of the year, with amplitude much weaker. | 10 | NaV6O15:A promising cathode material for insertion/extraction of Mg^2+with excellent cycling performance显示文摘The rechargeable magnesium batteries(RMBs)are getting more and more attention because of their high-energy density,high-security and low-cost.Nevertheless,the high charge density of Mg^2+makes the diffusion of Mg2+in the conventional cathodes very slow,resulting in a lack of appropriate electrode materials for RMBs.In this work,we enlarge the layer spacing of V2Os by introducing Na^2+in the crystal structure to promote the diffusion kinetics of Mg^2+.The NaVeO15(NVO)synthesized by a facile method is studied as a cathode material for RMBs with the anhydrous pure Mg^2+electrolyte.As a result,the NVO not only exhibits high discharge capacity(119.2 mAh:g^-1 after 100 cycles at the current density of 20 mA:g^-1)and working voltage(above 1.6 V vs.Mg^2+/Mg),but also expresses good rate capability.Besides,the eX-situ characterizations results reveal that the Mg^2+storage mechanism in NVO is based on the intercalation and deintercalation.The density functional theory(DFT)calculation results further indicate that Mg^2+tends to occupy the semi-occupied sites of Na^+in the NVO.Moreover,the galvanostatic intermittent titration technique(GITT)demonstrates that NVO electrode has the fast diffusion kinetics of Mg^2+during discharge process ranging from 7.55×10^-13 to2.41×10^-11 cm^2·s^-1.Our work proves that the NVO is a potential cathode material for RMBs. | Dongzheng Wu Jing Zeng Haiming Hua Junnan Wu Yang Yang Jinbao Zhao | 2020 | Nano Research2020,13,2: | 2 |
| 11 | The Geochemical and Zircon Trace Elements Characteristics of A-type Granitoids in Boziguoer, Baicheng County, Xinjiang显示文摘The Boziguoer A-type granitoids in Baicheng County,Xinjiang,belong to the northern margin of the Tarim platform as well as the neighboring EW-oriented alkaline intrusive rocks.The rocks comprise an aegirine or arfvedsonite quartz alkali feldspar syenite,an aegirine or arfvedsonite alkali feldspar granite,and a biotite alkali feldspar syenite.The major rock-forming minerals are albite,K-feldspar,quartz,arfvedsonite,aegirine,and siderophyllite.The accessory minerals are mainly zircon,pyrochlore,thorite,fluorite,monazite,bastnaesite,xenotime,and astrophyllite.The chemical composition of the alkaline granitoids show that SiO2varies from 64.55%to 72.29%with a mean value of 67.32%,Na2O+K2O is high(9.85%–11.87%)with a mean of 11.14%,K2O is 2.39%–5.47%(mean=4.73%),the K2O/Na2O ratios are 0.31–0.96,Al2O3ranges from 12.58%to 15.44%,and total FeO T is between 2.35%and 5.65%.CaO,MgO,MnO,and TiO2are low.The REE content is high and the total SREE is(263–1219)ppm(mean=776 ppm),showing LREE enrichment and HREE depletion with strong negative Eu anomalies.In addition,the chondrite-normalized REE patterns of the alkaline granitoids belong to the'seagull'pattern of the right-type.The Zr content is(113–1246)ppm(mean=594 ppm),Zr+Nb+Ce+Y is between(478–2203)ppm with a mean of 1362 ppm.Furthermore,the alkaline granitoids have high HFSE(Ga,Nb,Ta,Zr,and Hf)content and low LILE(Ba,K,and Sr)content.The Nb/Ta ratio varies from 7.23 to 32.59(mean=16.59)and the Zr/Hf ratio is 16.69–58.04(mean=36.80).The zircons are depleted in LREE and enriched in HREE.The chondrite-normalized REE patterns of the zircons are of the'seagull'pattern of the left-inclined type with strong negative Eu anomaly and without a Ce anomaly.The Boziguoer A-type granitoids share similar features with A1-type granites.The average temperature of the granitic magma was estimated at 832–839°C.The Boziguoer A-type granitoids show crust–mantle mixing and may have formed in an anorogenic intraplate tectonic setting under high-temperature,anhydrous,and low oxygen fugacity conditions. | LIU Chunhua YIN Jingwu WU Cailai SHAO Xingkun YANG Haitao XU Haiming WANG Jun | 2013 | Acta Geologica Sinica(English Edition)2013,87,6: | 2 |
| 12 | Machine-Learning-Assisted Optimization and Its Application to Antenna Designs: Opportunities and Challenges显示文摘With the rapid development of modern wireless communications and radar, antennas and arrays are becoming more complex, therein having, e.g., more degrees of design freedom, integration and fabrication constraints and design objectives. While fullwave electromagnetic simulation can be very accurate and therefore essential to the design process, it is also very time consuming, which leads to many challenges for antenna design, optimization and sensitivity analysis(SA). Recently, machine-learning-assisted optimization(MLAO) has been widely introduced to accelerate the design process of antennas and arrays. Machine learning(ML) methods, including Gaussian process regression, support vector machine(SVM) and artificial neural networks(ANNs), have been applied to build surrogate models of antennas to achieve fast response prediction. With the help of these ML methods, various MLAO algorithms have been proposed for different applications. A comprehensive survey of recent advances in ML methods for antenna modeling is first presented. Then, algorithms for ML-assisted antenna design, including optimization and SA, are reviewed. | 13 | CD49a^(+)CD49b^(+) NK cells induced by viral infection reflect an activated state of conventional NK cells显示文摘Natural killer(NK) cells are important innate effectors that play a pivotal role in the defense against tumors and infections and participate in regulating adaptive immunity. Recent studies have revealed phenotypic and functional heterogeneity of NK cells.Here, using murine models of acute and chronic lymphocytic choriomeningitis virus infection, we observed that a CD49a^(+)CD49b^(+) NK cell subset emerged in the liver and other tissues, and underwent vigorous expansion following viral infection,before progressively decreasing in cell number. These viral infection-induced CD49a^(+)CD49b^(+) NK cells displayed an activated and mature phenotype. Moreover, compared with liver-resident NK cells and conventional NK(cNK) cells, CD49a^(+)CD49b^(+) NK cells showed increased functional competence, as evidenced by higher amounts of IFN-γ production and stronger cytotoxic capabilities during viral infection. Generation of these CD49a^(+)CD49b^(+) NK cells was shown to be independent of the T-bet transcription factor. Adoptive transfer experiments revealed that c NK cells could convert into CD49a^(+)CD49b^(+) NK cells following viral infection.
