| 1 | Iptkalim inhibits cocaine challenge—induced enhancement of dopamine levels in nucleus accumbens and striatum of rats by up—regulating Kir6.1 and Kir6.2 mRNA expression显示文摘AIM:To investigate the effect and mechanism of novel ATP-sensitive potassium channel opener (KCO) iptkalim (IPT) on acute and cocaine challenge-induced alterations in the levels of dopamine (DA) and glutamate (Glu) from nucleus accumbens (NAc), striatum, and prefrontal cortex (PFC) in rats. METHODS: The levels of DA and Glu were assayed using high performance liquid chromatography (HPLC) combined with amperometric and fluorescent detection, respectively. The mRNA levels of Kir6.1, Kir6.2, SUR1, and SUR2 were measured by semiquantitative reverse transcription polymerase chain reaction (RT-PCR). RESULTS: IPT did not affect acute cocaine (30mg/kg,ip)-induced elevations in either DA levels from NAc and striatum or Glu levels from NAc and PFC. An acute cocaine challenge (30mg/kg,ip) on d 21 after withdrawal caused an elevation in DA levels in NAc and striatum. Moreover, the same treatment also increased Gluo levels in PFC and NAc of cocaine-pretreated rats. Repeated IPT injections reversed cocaine challenge-induced DA increase in NAc and striatum. Cocaine challenge increased Kir6.1 and Kir6.2 mRNA expression in striatum and NAc and only elevate Kir6.2 expression in PFC in both cocainepretreated rats and rats pretreated with IPT plus cocaine. Moreover, expression of Kir6.1 and Kir6.2 mRNA was augmented in rats pretreated with IPT plus cocaine compared to rats pretreated with cocaine alone. No significant change was found in the SUR1 and SUR2 expression of all four groups. | 2 | A Spatiotemporal-Chaos-Based Cryptosystem Taking Advantage of Both Synchronous and Self-Synchronizing Schemes显示文摘Two-dimensional one-way coupled map lattices are used for cryptography where multiple space units producechaotic outputs in parallel. One of the outputs plays the role of driving for synchronization of the decryption sys-tem while the others perform the function of information encoding. With this separation of functions the receivercan establish a self-checking and self-correction mechanism, and enjoys the advantages of both synchronous andself-synchronizing schemes. A comparison between the present system with the system of advanced encryptionstandard (AES) is presented in the aspect of channel noise influence. | 19 | Development and validation of a nomogram to predict cardiac death after radiotherapy for esophageal cancer显示文摘Background:Patients frequently die from cardiac causes after radiotherapy for esophageal cancer.Early detection of cardiac death risk in these patients is crucial to improve clinical decision‐making and prognosis.Thus,we modeled the risk of cardiac death after irradiation for esophageal cancer.Methods:A retrospective analysis of 37,599 esophageal cancer cases treated with radiotherapy in the SEER database between 2000 and 2018 was performed.The selected cases were randomly assigned to the model development group(n=26,320)and model validation group(n=11,279)at a ratio of 7:3.We identified the risk factors most commonly associated with cardiac death by least absolute shrinkage and selection operator regression analysis(LASSO).The endpoints for model development and validation were 5‐and 10‐year survival rates.The net clinical benefit of the models was evaluated by decision curve analysis(DCA)and concordance index(C‐index).The performance of the models was further assessed by creating a receiver operating characteristic curve(ROC)and calculating the area under the curve(AUC).Kaplan‐Meier(K‐M)survival analysis was performed on the probability of death.Patients were classified according to death probability thresholds.Five‐and ten‐year survival rates for the two groups were shown using K‐M curves.Results:The major risk factors for cardiac death were age,surgery,year of diagnosis,sequence of surgery and radiotherapy,chemotherapy and a number of tumors,which were used to create the nomogram.The C‐indexes of the nomograms were 0.708 and 0.679 for the development and validation groups,respectively.DCA showed the good net clinical benefit of nomograms in predicting 5‐and 10‐year risk of cardiac death.The model exhibited moderate predictive power for 5‐and 10‐year cardiac mortality(AUC:0.833 and 0.854,respectively),and for the development and validation cohorts(AUC:0.76 and 0.813,respectively).Conclusions:Our nomogram may assist clinicians in making clinical decisions about patients undergoing radiotherapy for esophageal cancer based on early detection of cardiac death risk. | Xinfang Lv Xue Wu Kai Liu Xinke Zhao Chenliang Pan Jing Zhao Juan Chang Huan Guo Xiang Gao Xiaodong Zhi Chunzhen Ren Qilin Chen Hugang Jiang Chunling Wang Ying‐Dong Li | 2023 | Cancer Innovation2023,2,5: | 0 |
