| 1 | Crosstalk of liver immune cells and cell death mechanisms in different murine models of liver injury and its clinical relevance显示文摘BACKGROUND: Liver inflammation or hepatitis is a result of pluripotent interactions of cell death molecules, cytokines, chemokines and the resident immune cells collectively called as microenvironment. The interplay of these inflammatory mediators and switching of immune responses during hepatotoxic, viral, drug-induced and immune cell-mediated hepatitis decide the fate of liver pathology. The present review aimed to describe the mechanisms of liver injury, its relevance to human liver pathology and insights for the future therapeutic interventions.DATA SOURCES: The data of mouse hepatic models and relevant human liver diseases presented in this review are systematically collected from Pub Med, Science Direct and the Web of Science databases published in English. RESULTS: The hepatotoxic liver injury in mice induced by the metabolites of CCl4, acetaminophen or alcohol represent necrotic cell death with activation of cytochrome pathway, formation of reactive oxygen species(ROS) and mitochondrial damage. The Fas or TNF-α induced apoptotic liver injury was dependent on activation of caspases, release of cytochrome c and apoptosome formation. The Con A-hepatitis demonstrated the involvement of TRAIL-dependent necrotic/necroptotic cell death with activation of RIPK1/3. The α-Gal Cer-induced liver injury was mediated by TNF-α. The LPS-induced hepatitis involved TNF-α, Fas/Fas L, and perforin/granzyme cell death pathways. The MHV3 or Poly(I:C) induced liver injury was mediated by natural killer cells and TNF-α signaling. The necrotic ischemia-reperfusion liver injury was mediated byhypoxia, ROS, and pro-inflammatory cytokines; however, necroptotic cell death was found in partial hepatectomy. The crucial role of immune cells and cell death mediators in viral hepatitis(HBV, HCV), drug-induced liver injury, non-alcoholic fatty liver disease and alcoholic liver disease in human were discussed.CONCLUSIONS: The mouse animal models of hepatitis provide a parallel approach for the study of human liver pathology. | 3 | Effect of adipose-derived mesenchymal stem cells on hepatocellular carcinoma: In vitro inhibition of carcinogenesis显示文摘AIM To investigate the effect of adipose-derived mesenchymal stem cells(ADMSCs)and their conditioned media(CM) on hepatocellular carcinoma(HCC) cell tumorigenesis.METHODS The proliferation rate of HepG2 and PLC-PRF-5 HCC cancer cells was measured using the trypan blue exclusion method and confirmed using the cell-counting kit8(commonly known as CCK-8) assay. Apoptosis was detected by flow cytometry using annexin V-FITC. Protein and mRNA expression was quantified by ELISA and real time PCR, respectively. Migration and invasion rates were performed by Transwell migration and invasion assays. Wound healing was examined to confirm the data obtained from the migration assays.RESULTS Our data demonstrated that when co-culturing HCC cell lines with ADMSCs or treating them with ADMSC CM, the HCC cell proliferation rate was significantly inhibited and the apoptosis rate increased. The decreased proliferation rate was accompanied by an upregulation of P53 and Retinoblastoma mRNA and a downregulation of c-Myc and hTERT mRNA levels. More notably, ADMSCs and their CM suppressed the expression of the two important markers of HCC carcinogenicity, alpha-fetoprotein and Des-gamma-carboxyprothrombin. | 4 | Effectiveness of breathing exercises during the second stage of labor on labor pain and duration: a randomized controlled trial显示文摘BACKGROUND: Some research exists on the effect of non-pharmacological approaches for labor pain relief. However, there is limited information about effectiveness of breathing exercises in pregnant women to reduce maternal pain during labor.OBJECTIVE: To determine whether breathing exercises for pregnant women during the second stage of labor have beneficial effects on maternal pain, duration of labor, and the first-minute Appearance, Pulse, Grimace, Activity and Respiration(APGAR) scores. DESIGN, SETTING, PARTICIPANTS AND INTERVENTIONS: This randomized clinical trial involved 250 pregnant women, who were randomly divided into two groups: intervention group(IG; n = 125) and control group(CG; n = 125). IG received one session breathing exercise training and performed breathing exercises during the second stage of labor versus the CG that did not receive any breathing exercise training.MAIN OUTCOME MEASURES: The effects of breathing exercises on maternal pain were determined by Visual Analogue Scale(VAS), duration of the second stage of labor, and the first-minute APGAR scores.RESULTS: The mean age of the participants was(23.2 ± 4.2)(range: 18 to 42) years. Both IG and CG had similar baseline characteristics in terms of age, education level, occupation, and smoking(P > 0.05). The mean VAS scores of IG and CG were(88.2 ± 6.3) and(90.5 ± 7.0), respectively(P < 0.001). The duration of the second stage of labor was(369.6 ± 92.0) s for IG and(440.7 ± 142.5) s for CG(P < 0.001). The mean first-minute APGAR scores were(8.84 ± 0.50) for IG and(8.73 ± 0.89) for CG(P > 0.05).CONCLUSION: Based on this study, breathing exercises with deep inhalation and exhalation in pregnant women are effective in reducing the perception of labor pain and shortening the duration of the second stage of delivery. | 6 | Reactive oxidative species enhance amyloid toxicity in APP/PS1 mouse neurons显示文摘Objective To investigate whether intracellular amyloid β (iAβ) induces toxicity in wild type (WT) and APP/PS1 mice, a mouse model of Alzheimer's disease. Methods Different forms of Aβ aggregates were microinjected into cultured WT or APP/PS1 mouse hippocampal neurons. TUNEL staining was performed to examine neuronal cell death. Reactive oxidative species (ROS) were measured by MitoSOXTM Red mitochondrial superoxide indicator. Results Crude, monomer and protofibrilAβ induced more toxicity inAPP/PS1 neurons than in WT neurons. ROS are involved in mediating the vulnerability of APP/PS1 neurons to iAβ toxicity. | 7 | Laparoscopic liver resection for hepatocellular adenoma显示文摘AIM:To investigate the role of laparoscopy in the surgical management of hepatocellular adenoma(HA). METHODS:We reviewed a prospectively collected database of consecutive patients undergoing laparoscopic liver resection for HA. RESULTS:Thirteen patients underwent fifteen pure laparoscopic liver resections for HA(male/female:3/10; median age 42 years,range 22-72 years).Two patients with liver adenomatosis required two different laparoscopic operations for ruptured adenomas.Indications for surgery were:symptoms in 12 cases,need to rule out malignancy in 2 cases and preoperative diagnosis of large HA in one case.Symptoms were related to bleeding in 10 cases,sepsis due to liver abscess following embolization of HA in one case and mass effect in one case(shoulder tip pain).Five cases with ruptured bleeding adenoma required emergency admis-sion and treatment with selective arterial embolization. Laparoscopic liver resection was then semi-electively performed.Eight patients(62%)required major hepatectomy[right hepatectomy(n=5),left hepatectomy (n=3)].No conversion to open surgery occurred.The median operative time for pure laparoscopic procedures was 270 min(range 135-360 min).The median size of the excised lesions was 85 mm(range 25-180 mm). One patient with adenomatosis developed postoperative bleeding requiring embolization.Mortality was nil. The median hospital stay was 4 d(range 1-18 d)with a median high dependency unit stay of 1 d(range 0-7 d). | 9 | Antioxidative study of Cerium Oxide nanoparticle functionalised PCL-Gelatin electrospun fibers for wound healing application显示文摘Skin wound healing involves a coordinated cellular response to achieve complete reepithelialisation.Elevated levels of reactive oxygen species(ROS)in the wound environment often pose a hindrance in wound healing resulting in impaired wound healing process.Cerium oxide nanoparticles(CeNPs)have the ability to protect the cells from oxidative damage by actively scavenging the ROS.Furthermore,matrices like nanofibers have also been explored for enhancing wound healing.In the current study CeNP functionalised polycaprolactone(PCL)-gelatin nanofiber(PGNPNF)mesh was fabricated by electrospinning and evaluated for its antioxidative potential.Wide angle XRD analysis of randomly oriented nanofibers revealed^2.6 times reduced crystallinity than pristine PCL which aided in rapid degradation of nanofibers and release of CeNP.However,bioactive composite made between nanoparticles and PCLgelatin maintained the fibrous morphology of PGNPNF upto 14 days.The PGNPNF mesh exhibited a superoxide dismutase(SOD)mimetic activity due to the incorporated CeNPs.The PGNPNF mesh enhanced proliferation of 3T3-L1 cells by^48%as confirmed by alamar blue assay and SEM micrographs of cells grown on the nanofibrous mesh.Furthermore,the PGNPNF mesh scavenged ROS,which was measured by relative DCF intensity and fluorescence microscopy;and subsequently increased the viability and proliferation of cells by three folds as it alleviated the oxidative stress.Overall,the results of this study suggest the potential of CeNP functionalised PCL-gelatin nanofibrous mesh for wound healing applications. | Hilal Ahmad Rather Ria Thakore Ragini Singh Dhwani Jhala Sanjay Singh Rajesh Vasita | 2018 | Bioactive Materials2018,3,2: | 4 |

| 11 | Diagnostic value of plasminogen activity level in acute mesenteric ischemia显示文摘AIM: To investigate the changes in plasminogen activity level during mesenteric ischemia. METHODS: We performed laparotomy in 90 female Wistar-Albino rats (average weight 230 g). In sham groups (SL) (GroupsⅠand Ⅱ) the superior mesenteric artery (SMA) and vein (SMV) were explored, but not tied. In SMA groups (Groups Ⅲ and Ⅳ) the SMA was ligated, and in SMV groups (Groups Ⅴ and Ⅵ) the SMV was ligated. On re-laparatomy 2 mL of blood was drawn at 1 h in groupsⅠ, Ⅲ and Ⅴ, and at 3 h in groups Ⅱ, Ⅳ and Ⅵ. Plasminogen levels were assessed and comparisons were made between groups and within each group. RESULTS: The mean plasminogen activity in the SL group was significantly higher than SMA (25.1 ± 10.8 vs 11.8 ± 4.6, P < 0.001) or SMV (25.1 ± 10.8 vs 13.7 ± 4.4, P < 0.001) groups both at 1 h and at 3 h (29.8 ± 8.9 vs 15.1 ± 5.7, P < 0.0001; 29.8 ± 8.9 vs 14.2 ± 2.9, P < 0.0001). There were no significant differences between the values of SMA and SMV groups at 1 h (P = 0.28) and at 3 h (P = 0.71). In each group, plasminogen activity levels did not change significantly between the two measurements performed at 1 h and 3 h. | 14 | Isolation and diagnosis of Helicobacter pylori by a new method: Microcapillary culture显示文摘AIM:To investigate the performance of the microcapillary culture method(MCM) in Helicobacter pylori(H.pylori) isolation and diagnosis.METHODS:Microcapillary culture(MC),classical culture(CC),rapid urease(CLO) test,and histopathologic examination(HE) were performed with biopsy samples.Homogenized biopsy samples were loaded into capillary tubes and incubated for 48 h at 37 ℃ without providing a microaerophilic environment.Additionally,three or four loops of the homogenized sample were inoculated in a ready-to-use selective medium(Becton Dickinson,Helicobacter Agar,Modified) specific for the isolation of H.pylori and incubated at 37 ℃ in a microaerophilic atmosphere provided by Campy Gen(Becton Dickinson,Gas Pack).Bacteria reproducing in microcapillary tubes were evaluated in an inverted microscope and also were evaluated after performing a CC with the content.Results obtained by CC,CLO test,and HE were compared with those of MC.The diagnostic performances of the methods used in this study were evaluated for specificity,sensitivity,positive predictive value(PPV),negative predictive value(NPV),and CI.RESULTS:H.pylori was found positive by CLO test +HE and/or CC culture in 26 patient antrum and corpus biopsy samples.In 25(25/26) patient biopsy samples,H.pylori was isolated by MCM,whereas in only 14(14/26) patient biopsy samples,H.pylori was isolatedby CC.CLO test and HE were found positive in 17(17/26) patient biopsy samples.Comparing the results of the isolation of H.pylori by MCM,CC,CLO test,and HE,the sensitivity of the MCM was found as 96%,the specificity as 80%,the PPV as 83%,the NPV as 95%,and the 95%CI as 0.76(χ2 =31.51,P < 0.01) whereas the sensitivity of the CC was found as 54%(χ2 =19.15,P < 0.01),and the sensitivity of the CLO test and HE were found as 65%(χ2 =25.26,P < 0.01).CONCLUSION:This new microcapillary cultivation method for H.pylori has high diagnostic sensitivity compared with CC,HE,and CLO tests. | Adil M Allahverdiyev Melahat Bagirova Reyhan Caliskan Hrisi Bahar Tokman Hayat Aliyeva Gokce Unal Olga Nehir Oztel Emrah Sefik Abamor Hilal Toptas Pelin Yuksel Fatma Kalayci Mustafa Aslan Yusuf Erzin Kadir Bal Bekir S Kocazeybek | 2015 | World Journal of Gastroenterology2015,21,9: | 2 |
| 15 | Outcomes and predictors of vitrectomy and silicone oil tamponade in retinal detachments complicated by proliferative vitreoretinopathy显示文摘AIM:To evaluate outcomes and determine factors influencing the outcomes of vitrectomy with silicone oil(SO)endotamponade for the management of rhegmatogenous retinal detachment(RRD)complicated by advanced proliferative vitreoretinopathy(PVR).METHODS:This is a retrospective,interventional case series of eyes with PVR grade C associated RRD with or without prior surgery that underwent vitreoretinal surgery and SO tamponade.Eyes with a minimum follow-up of 6mo after SO extraction were included.Eyes were classified into three PVR subgroups according to severity and extension of proliferation.The influence of several preoperative,intraoperative and postoperative factors upon the functional and anatomical outcomes was assessed using multivariate logistic regression analysis.RESULTS:A hundred and one eyes of 101 patients that met the inclusion criteria were studied.Seventy-five of 101 eyes(74.3%)had successful retinal reattachment after one operation.Increased aqueous cell and flare at the first week exam had a statistically significant association with redetachment,recurrent membrane proliferation and keratopathy.Visual acuity improvement was significantly associated with faint postoperative aqueous inflammation values,primary vitrectomy and PVR outside of the posterior pole.CONCLUSION:Although encouraging anatomical and functional outcomes are achieved after vitrectomy and SO tamponade in eyes with RRD complicated by PVR,an increase in aqueous flare or cells at the first week follow-up is most likely to result in postoperative late complications.Primary vitrectomy,PVR associated with minimal posterior pole extension and absent to mild postoperative aqueous inflammation are associated with improved post-operative final visual acuity. | Kenan Sonmez Hilal Kilinc Hekimsoy | 2022 | International Journal of Ophthalmology(English edition)2022,15,8: | 2 |

| 17 | Higher-Order Smoothing: A Novel Semantic Smoothing Method for Text Classification显示文摘It is known that latent semantic indexing(LSI) takes advantage of implicit higher-order(or latent) structure in the association of terms and documents. Higher-order relations in LSI capture 'latent semantics'. These findings have inspired a novel Bayesian framework for classification named Higher-Order Naive Bayes(HONB), which was introduced previously, that can explicitly make use of these higher-order relations. In this paper, we present a novel semantic smoothing method named Higher-Order Smoothing(HOS) for the Naive Bayes algorithm. HOS is built on a similar graph based data representation of the HONB which allows semantics in higher-order paths to be exploited. We take the concept one step further in HOS and exploit the relationships between instances of different classes. As a result, we move beyond not only instance boundaries, but also class boundaries to exploit the latent information in higher-order paths. This approach improves the parameter estimation when dealing with insufficient labeled data. | 18 | Risk factors for sudden cardiac death to determine high risk patients in specific patient populations that may benefit from a wearable defibrillator显示文摘BACKGROUND There is a high risk for sudden cardiac death(SCD) in certain patient groups that would not meet criteria for implantable cardioverter defibrillator(ICD) therapy.In conditions such as hypertrophic cardiomyopathy(HCM) there are clear risk scores that help define patients who are high risk for SCD and would benefit from ICD therapy. There are however many areas of uncertainty such as certain patients post myocardial infarction(MI). These patients are high risk for SCD but there is no clear tool for risk stratifying such patients.AIM To assess risk factors for sudden cardiac death in major cardiac disorders and to help select patients who might benefit from Wearable cardiac defibrillators(WCD).METHODS A literature search was performed looking for risk factors for SCD in patients post-MI, patients with left ventricular systolic dysfunction(LVSD), HCM, long QT syndrome(LQTS). There were 41 studies included and risk factors and the relative risks for SCD were compiled in table form.RESULTS We extracted data on relative risk for SCD of specific variables such as age,gender, ejection fraction. The greatest risk factors for SCD in post MI patients was the presence of diabetes [Hazard ratio(HR) 1.90-3.80], in patient with LVSD was ventricular tachycardia(Relative risk 3.50), in LQTS was a prolonged QTc(HR36.53) and in patients with HCM was LVH greater than 20 mm(HR 3.10). A proportion of patients currently not suitable for ICD might benefit from a WCDCONCLUSION There is a very high risk of SCD post MI, in patients with LVSD, HCM and LQTS even in those who do not meet criteria for ICD implantation. These patients may be candidates for a WCD.
