1OPS法玻璃化冷冻牛卵母细胞和囊胚显示文摘本文报道了利用拉细开口型细管 (Open Pulled Straw,OPS)方法进行玻璃化冷冻保存体外成熟(IVM)的牛卵母细胞及体外生产 (IVP)的牛囊胚的实验结果。实验 1的牛卵母细胞冷冻处理分 2组 ,组 1(G1 )是将卵母细胞在 IVM2 1 h时去除部分卵丘细胞后马上进行冷冻处理。组 2 (G2 )卵母细胞在 IVM6h时去除部分卵丘细胞 ,然后继续培养至 2 2 h冷冻 ;冷冻后的卵母细胞在液氮中保存 0 .5~ 2 h后解冻再继续 IVM1 h后与正常 IVM2 4h的对照组卵母细胞一起进行体外受精 (IVF)处理。实验 2则冷冻了来自IVP的 6、 7日龄的牛囊胚。结果显示 ,G1的卵母细胞 IVF后 8d的囊胚率 (2 2 .8% ,2 6/1 1 4)和孵化囊胚率 (1 4.9% ,1 7/1 1 4)都极显著地高于 G2 (分别为 2 .9% ,5 /1 71和 0 .5 % ,1 /1 71 ,P<0 .0 0 1 ) ;但两处理组的受精卵裂率差别不大 (分别为 48.2 %和 41 .5 % ,P>0 .3 )。冷冻第 6d和第 7d的体外囊胚 ,其冻后存活率和囊胚孵化率分别为 92 .7%、 89.2 %和 83 .6%、 66.7%。结果表明 ,利用谭世俭 Henrik Callesen 2002广西农业生物科学2002,21,1:19

5Low-FODMAP diet reduces irritable bowel symptoms in patients with inflammatory bowel disease显示文摘AIM To investigate the effect of a low-FODMAP diet on irritable bowel syndrome(IBS)-like symptoms in patients with inflammatory bowel disease(IBD).METHODS This was a randomised controlled open-label trial of patients with IBD in remission or with mild-to-moderate disease and coexisting IBS-like symptoms(Rome III) randomly assigned to a Low-FODMAP diet(LFD) or a normal diet(ND) for 6 wk between June 2012 andDecember 2013. Patients completed the IBS symptom severity system(IBS-SSS) and short IBD quality of life questionnaire(SIBDQ) at weeks 0 and 6. The primary end-point was response rates(at least 50-point reduction) in IBS-SSS at week 6 between groups; secondary end-point was the impact on quality of life. RESULTS Eighty-nine patients, 67(75%) women, median age 40, range 20-70 years were randomised: 44 to LFD group and 45 to ND, from which 78 patients completed the study period and were included in the final analysis(37 LFD and 41 ND). There was a significantly larger proportion of responders in the LFD group(n = 30, 81%) than in the ND group(n = 19, 46%);(OR = 5.30; 95%CI: 1.81-15.55, P < 0.01). At week 6, the LFD group showed a significantly lower median IBSSSS(median 115; inter-quartile range [IQR] 33-169) than ND group(median 170, IQR 91-288), P = 0.02. 6Feasibility of cold snare polypectomy in Japan: A pilot study显示文摘AIM: To investigate the feasibility of cold snare polypectomy(CSP) in Japan.METHODS: The outcomes of 234 non-pedunculated polyps smaller than 10 mm in 61 patients who underwent CSP in a Japanese referral center were retrospectively analyzed. The cold snare polypectomies were performed by nine endoscopists with no prior experience in CSP using an electrosurgical snare without electrocautery.RESULTS: CSPs were completed for 232 of the 234 polyps. Two(0.9%) polyps could not be removed without electrocautery. Immediate postpolypectomy bleeding requiring endoscopic hemostasis occurred in eight lesions(3.4%; 95%CI: 1.1%-5.8%), but all were easily managed. The incidence of immediate bleeding after CSP for small polyps(6-9 mm) was significantly higher than that of diminutive polyps(≤ 5 mm; 15% vs 1%, respectively). Three(5%) patients complained of minor bleeding after the procedure but required no intervention. The incidence of delayed bleeding requiringendoscopic intervention was 0.0%(95%CI: 0.0%-1.7%). In total, 12% of the resected lesions could not be retrieved for pathological examination. Tumor involvement in the lateral margin could not be histologically assessed in 70(40%) lesions.CONCLUSION: CSP is feasible in Japan. However, immediate bleeding, retrieval failure and uncertain assessment of the lateral tumor margin should not be underestimated. 7The coupling of dynamics and permeability in the hydrocarbon accumulation period controls the oil-bearing potential of low permeability reservoirs:a case study of the low permeability turbidite reservoirs in the middle part of the third member of Shahejie显示文摘The relationships between permeability and dynamics in hydrocarbon accumulation determine oilbearing potential(the potential oil charge) of low permeability reservoirs.The evolution of porosity and permeability of low permeability turbidite reservoirs of the middle part of the third member of the Shahejie Formation in the Dongying Sag has been investigated by detailed core descriptions,thin section analyses,fluid inclusion analyses,carbon and oxygen isotope analyses,mercury injection,porosity and permeability testing,and basin modeling.The cutoff values for the permeability of the reservoirs in the accumulation period were calculated after detailing the accumulation dynamics and reservoir pore structures,then the distribution pattern of the oil-bearing potential of reservoirs controlled by the matching relationship between dynamics and permeability during the accumulation period were summarized.On the basis of the observed diagenetic features and with regard to the paragenetic sequences,the reservoirs can be subdivided into four types of diagenetic fades.The reservoirs experienced two periods of hydrocarbon accumulation.In the early accumulation period,the reservoirs except for diagenetic fades A had middle to high permeability ranging from 10 × 10^(-3) μm^2 to4207 × 10^(-3) μm^2.In the later accumulation period,the reservoirs except for diagenetic fades C had low permeability ranging from 0.015 × 10^(-3)~ μm^2 to 62 × 10^(-3) μm^2.In the early accumulation period,the fluid pressure increased by the hydrocarbon generation was 1.4-11.3 MPa with an average value of 5.1 MPa.and a surplus pressure of1.8-12.6 MPa with an average value of 6.3 MPa.In the later accumulation period,the fluid pressure increased by the hydrocarbon generation process was 0.7-12.7 MPa with an average value of 5.36 MPa and a surplus pressure of1.3-16.2 MPa with an average value of 6.5 MPa.Even though different types of reservoirs exist,all can form hydrocarbon accumulations in the early accumulation period.Such types of reservoirs can form hydrocarbon accumulation with high accumulation dynamics:however,reservoirs with diagenetic facies A and diagenetic fades B do not develop accumulation conditions with low accumulation dynamics in the late accumulation period for very low permeability.At more than 3000 m burial depth,a larger proportion of turbidite reservoirs are oil charged due to the proximity to the source rock.Also at these depths,lenticular sand bodies can accumulate hydrocarbons.At shallower depths,only the reservoirs with oil-source fault development can accumulate hydrocarbons.For flat surfaces,hydrocarbons have always been accumulated in the reservoirs around the oil-source faults and areas near the center of subsags with high accumulation dynamics.Tian Yang Ying-Chang Cao Yan-Zhong Wang Henrik Friis Beyene Girma Haile Ke-Lai Xi Hui-Na Zhang 2016Petroleum Science2016,13,2:10

9INT-767 improves histopathological features in a dietinduced ob/ob mouse model of biopsy-confirmed nonalcoholic steatohepatitis显示文摘AIM To characterize the efficacy of the dual FXR/TGR5 receptor agonist INT-767 upon histological endpoints in a rodent model of diet-induced and biopsy-confirmed non-alcoholic steatohepatitis(NASH).METHODS The effects of INT-767 on histological features of NASH were assessed in two studies using Lep^(ob/ob)(ob/ob) NASH mice fed the AMLN diet(high fat with transfat, cholesterol and fructose). In a proof-of-conceptstudy, Lep^(ob/ob)(ob/ob) NASH mice were first dosed with INT-767(3 or 10 mg/kg for 8 wk). A second ob/ob NASH study compared INT-767(3 and 10 mg/kg) to obeticholic acid(OCA)(10 or 30 mg/kg; 16 wk). Primary histological endpoints included qualitative and quantitative assessments of NASH. Other metabolic and plasma endpoints were also assessed. A comparative assessment of INT-767 and OCA effects on drug distribution and hepatic gene expression was performed in C57 Bl/6 mice on standard chow. C57 Bl/6 mice were orally dosed with INT-767 or OCA(1-30 mg/kg) for 2 wk, and expression levels of candidate genes were assessed by RNA sequencing and tissue drug levels were measured by liquid chromatography tandem-mass spectrometry.RESULTS INT-767 dose-dependently(3 and 10 mg/kg, PO, QD, 8 wk) improved qualitative morphometric scores on steatohepatitis severity, inflammatory infiltrates and fibrosis stage. Quantitative morphometric analyses revealed that INT-767 reduced parenchymal collagen area, collagen fiber density, inflammation(assessed by Galectin-3 immunohistochemistry) and hepatocyte lipid droplet area following INT-767 treatment. In a comparative study(16 wk), the FXR agonists OCA(10 and 30 mg/kg) and INT-767(3 and 10 mg/kg) both improved NASH histopathology, with INT-767 exerting greater therapeutic potency and efficacy than OCA. Mechanistic studies suggest that both drugs accumulate similarly within the liver and ileum, however, the effects of INT-767 may be driven by enhanced hepatic, but not ileal, FXR function. 11Helicobacter pylori infection and low serum pepsinogen I level as risk factors for gastric carcinoma显示文摘AIM: To study whether examination of CagA antibodies could increase the odds ratio for gastric cancer in a casecontrol study, and how often other serum markers of gastric cancer risk could be found in Helicobacter pylorinegative patients.METHODS: H pylori CagA and parietal cell antibodies(PCAs), and serum pepsinogen Ⅰ (SPGI) levels were compared between patients with gastric cancer and controls who received endoscopic examination due to reasons other than gastrointestinal malignancy.RESULTS: The odds ratio (OR) for gastric cancer was2.9 (95% CI 1.4-5.8) in H pylori+ patients, and 2.4 (95%CI 1.2-4.9) in CagA+ patients. When results of H pylori and CagA antibodies were combined, OR increased to5.0 (95% CI 2.5-10.0). Furthermore, if cardia cancer patients were excluded, the OR increased to 6.8 (95% CI3.1-14.8). Among patients with a low SPGI level, the OR was 12.0 (95% CI 4.1-35.3). However, the risk was significant only in the older age group. The number of patients with low SPGI was significantly higher in H pylori-/CagA+ patients as compared to other cancer patients.CONCLUSION: Examination of both H pylori and CagA antibodies increases the OR for gastric cancer in our casecontrol study. CagA antibodies are important in detecting previous H pylori infection in advanced atrophic gastritis or cancer when spontaneous decline of H pylori antibodies occurs. 14Effects of a novel bacterial phytase expressed in Aspergillus Oryzae on digestibility of calcium and phosphorus in diets fed to weanling or growing pigs显示文摘In 2 experiments, 48 weanling (initial BW: 13.5 ± 2.4 kg, Exp. 1) and 24 growing pigs (initial BW: 36.2 ± 4.0 kg, Exp. 2) were used to determine effects of a novel bacterial 6-phytase expressed in Aspergillus oryzae on the apparent total tract digestibility (ATTD) of phosphorus and calcium in corn-soybean meal diets fed to weanling and growing pigs. In Exp. 1 and 2, pigs were randomly allotted to 6 dietary treatments using a randomized complete block design and a balanced 2 period changeover design, respectively. In both experiments, 6 diets were formulated. The positive control diet was a corn-soybean meal diet with added inorganic phosphorus (Exp. 1: 0.42 and 0.86% standardized total tract digestible phosphorus and total calcium, respectively; Exp. 2: 0.32 and 0.79% standardized total tract digestible phosphorus and total calcium, respectively). A negative control diet and 4 diets with the novel phytase (Ronozyme HiPhos, DSM Nutritional Products Inc., Parsippany, NJ) added to the negative control diet at levels of 500, 1,000, 2,000, and 4,000 phytase units (FYT)/kg were also formulated. In Exp. 1, the ATTD of phosphorus was greater (P < 0.01) for the positive control diet (60.5%) than for the negative control diet (40.5%), but increased (linear and quadratic, P < 0.01) as phytase was added to the negative control diet (40.5% vs. 61.6%, 65.1%, 68.7%, and 68.0%). The breakpoint for the ATTD of phosphorus (68.4%) was reached at a phytase inclusion level of 1,016 FYT/kg. In Exp. 2, the ATTD of phosphorus was greater (P < 0.01) for the positive control diet (59.4%) than for the negative control diet (39.8%) and increased (linear and quadratic, P < 0.01) as phytase was added to the negative control diet (39.8% vs. 58.1%, 65.4%, 69.1%, and 72.8%). The breakpoint for the ATTD of phosphorus (69.1%) was reached at a phytase inclusion level of 801 FYT/kg. In conclusion, the novel bacterial 6-phytase improved the ATTD of phosphorus and calcium in both weanling and growing pigs. 18Metabolic and hepatic effects of liraglutide,obeticholic acid and elafibranor in diet-induced obese mouse models of biopsy-confirmed nonalcoholic steatohepatitis显示文摘AIM To evaluate the pharmacodynamics of compounds in clinical development for nonalcoholic steatohepatitis(NASH) in obese mouse models of biopsy-confirmedNASH.METHODS Male wild-type C57 BL/6 J mice(DIO-NASH) and Lep^(ob/ob)(ob/ob-NASH) mice were fed a diet high in trans-fat(40%), fructose(20%) and cholesterol(2%) for 30 and 21 wk, respectively. Prior to treatment, all mice underwent liver biopsy for confirmation and stratification of liver steatosis and fibrosis, using the nonalcoholic fatty liver disease activity score(NAS) and fibrosis staging system. The mice were kept on the diet and received vehicle, liraglutide(0.2 mg/kg, SC, BID), obeticholic acid(OCA, 30 mg/kg PO, QD), or elafibranor(30 mg/kg PO, QD) for eight weeks. Within-subject comparisons were performed on changes in steatosis, inflammation, ballooning degeneration, and fibrosis scores. In addition, compound effects were evaluated by quantitative liver histology, including percent fractional area of liver fat, galectin-3, and collagen 1 a1.RESULTS Liraglutide and elafibranor, but not OCA, reduced body weight in both models. Liraglutide improved steatosis scores in DIO-NASH mice only. Elafibranor and OCA reduced histopathological scores of hepatic steatosis and inflammation in both models, but only elafibranor reduced fibrosis severity. Liraglutide and OCA reduced total liver fat, collagen 1 a1, and galectin-3 content, driven by significant reductions in liver weight. The individual drug effects on NASH histological endpoints were supported by global gene expression(RNA sequencing) and liver lipid biochemistry.CONCLUSION DIO-NASH and ob/ob-NASH mouse models show distinct treatment effects of liraglutide, OCA, and elafibranor, being in general agreement with corresponding findings in clinical trials for NASH. 20CYP24A1 inhibition facilitates the anti-tumor effect of vitamin D3 on colorectal cancer cells显示文摘AIM:The effects of vitamin D3 have been investigated on various tumors, including colorectal cancer (CRC). 25-hydroxyvitamin-D3-24-hydroxylase (CYP24A1), the enzyme that inactivates the active vitamin D3 metabolite 1,25-dihydroxyvitamin D3 (1,25-D3), is considered to be the main enzyme determining the biological halflife of 1,25-D3. During colorectal carcinogenesis, the expression and concentration of CYP24A1 increases significantly, suggesting that this phenomenon could be responsible for the proposed efficacy of 1,25-D3 in the treatment of CRC. The aim of this study was to investigate the anti-tumor effects of vitamin D3 on the human CRC cell line Caco-2 after inhibition of the cytochrome P450 component of CYP24A1 activity. METHODS:We examined the expression of CYP24A1 mRNA and the effects of 1,25-D3 on the cell line Caco-2 after inhibition of CYP24A1. Cell viability and proliferation were determined by means of sulforhodamine-B staining and bromodeoxyuridine incorporation, respectively, while cytotoxicity was estimated via the lactate dehydrogenase content of the cell culture supernatant. CYP24A1 expression was measured by realtime reverse transcription polymerase chain reaction. A number of tetralone compounds were synthesized to investigate their CP24A1 inhibitory activity. RESULTS:In response to 1,25-D3, CYP24A1 mRNA expression was enhanced significantly, in a time- and dose-dependent manner. Caco-2 cell viability and proliferation were not influenced by the administration of 1,25-D3 alone, but were markedly reduced by coadministration of 1,25-D3 and KD-35, a CYP24A1-inhibiting tetralone. Our data suggest that the mechanism of action of co-administered KD-35 and 1,25-D3 does not involve a direct cytotoxic effect, but rather the inhibition of cell proliferation.
