| 1 | Morphological and Physiological Responses of Plants to Cadmium Toxicity: A Review显示文摘Cadmium(Cd) contamination has posed an increasing challenge to environmental quality and food security. In recent years,phytoremediation has been particularly scrutinized because it is cost-effective and environmentally friendly, especially the use of metal-hyperaccumulating plants to extract or mine heavy metals from polluted soils. Under Cd stress, responses of hyperaccumulator and non-hyperaccumulator plants differ in morphological responses and physiological processes such as photosynthesis and respiration,uptake, transport, and assimilation of minerals and nitrogen, and water uptake and transport, which contribute to their ability to accumulate and detoxify Cd. This review aims to provide a brief overview of the recent progresses in the differential responses of hyperaccumulator and non-accumulator plants to Cd toxicity in terms of growth and physiological processes. | 4 | Effects of a New-Type Cleaning Agent and a Plant Growth Regulator on Phytoextraction of Cadmium from a Contaminated Soil显示文摘In China, great efforts are being made to remediate farmlands polluted by heavy metals. In this study, a soil pot experiment was conducted to examine the effects of a new-type cleaning agent, methylglycinediacetic acid(MGDA), and a plant growth regulator(PGR), diethl aminoethyl hexanoate(DA-6), on plant growth and extraction and detoxification of cadmium(Cd) by ryegrass. The results showed that foliar spray of DA-6 alone improved plant growth, with root length and shoot dry biomass increased by 38.5%–58.6% and 71.1%–89.3%, respectively, whereas addition of MGDA alone decreased root length and shoot dry biomass by 10.3%–18.6%and 9.1%–21.8%, respectively. Diethl aminoethyl hexanoate promoted the binding of Cd to cell walls and thus alleviated the toxicity of Cd and/or MGDA to plants. Applications of DA-6 and/or MGDA resulted in a significant increase in Cd extraction efficiency(P < 0.05), and the efficiency decreased in the order of MGDA + DA-6 > DA-6 > MGDA. The treatment of MGDA + DA-6 achieved 2.2%, 1.7%, and 0.8% Cd extraction efficiency by ryegrass in soils spiked with 25, 50, 100 mg Cd kg^(-1), respectively. A real-time fluorescent quantitative PCR (RFQ-PCR) method was developed for quantitative detection of T.rotula. | 6 | Estimation of growth rate of Skeletonema costatum based on relative expression amount of PCNA gene显示文摘Partial sequence of Skeletonema costatum proliferating cell nuclear antigen (PCNA) gene was obtained by reverse transcriptase PCR(RT-PCR) and 3′ rapid amplification of cDNA ends (3′-RACE) techniques. Based on the obtained PCNA and cytochrome b gene(Cyt b gene) sequences, a real-time fluorescent quantitative PCR (FQ-PCR) method was developed to detect the expression S. costatum PCNA gene, and this method was applied to study the relationship between the growth rate of S. costatum and the average expression amount of PCNA gene in a single cell. The expression amount of PCNA gene had large variation in cells collected at different culture phases, and the trend was well consistent with the growth rate, which suggested that the expression amount of PCNA gene correlated well with the cell division, and the PCNA could be a promising indicator for the S. costatum cell proliferation. | 9 | Effects of Aroclor 1254 on reproductive organ weight,sperm quality and testicular mitochondria oxidative stress of Boleophthalmus pectinirostris显示文摘Boleophthalmus pectinirostris is an amphibious economic fish and wildly distributed in the southeast coast of China. In this study, Aroclor 1254 was intraperitoneally injected into B. pectinirostris with 1, 2 and 4 μg/(g · d) for 28 d to assay the reproductive organ weight, the sperm quality (sperm concentration and motility), and the testicular mitochondrial testicular mitochondria oxidative stress. The results show that the sperm number and motility in seminal vesicles, the absolute weight of testes and seminal vesicles of B. pectinirostris treated with 2 and 4 μg/(g · d) Aroclor 1254 decreased significantly as compared to the controls (p <0.05), while those treated with 1 μg/(g · d) Aroclor 1254 had no significant effects on these indictors. For the relative weight of reproductive organs, significant reduction (p <0.05) was only observed in the seminal vesicles of B. pectinirostris treated with 4 μg/(g · d). SOD activities and GSH levels in all the Aroclor 1254 treatments were significantly lower than those of the controls (p <0.05). The activities of CAT, GPx, GR and the levels of Vit C also decreased significantly in comparison with the controls (p <0.05) at the higher dose of 2 and 4 μg/(g · d) Aroclor 1254 treatments. In addition, both H 2 O 2 level and MDA content in testicular mitochondria of B. pectinirostris had a close correlation with Aroclor 1254 dosage, and were significantly higher than the controls (p <0.05).
