5Myo-inositol accelerates the metamorphosis from megalopa to crablet of Scylla paramamosain by modulating cuticle decomposition and reconstruction显示文摘The mud crab Scylla paramamosain is a key species in China due to its high nutritional value and great economic worth and has grown in popularity.Myo-inositol can modulate versatile physiological functions in aquatic animals.In the present study,S.paramamosain megalopa were given graded concentrations of myo-inositol(0,1,2,4,and 8 ppm)by water immersion to explore how their metamorphosis would be affected.The results showed that supplementing with myo-inositol remarkably increased transformation and survival rate from megalopa to crablet by at least 1.16 and 1.26 times,respectively.To decipher the molecular mechanism of how myo-inositol increases metamorphosis and survival rate,we further performed transcriptome-based gene expression profiling of both megalopa and crablet treated with myo-inositol.The integrative transcriptome analyses predicted that the differentially expressed genes(DEGs)were significantly enriched in chitinase activity,structural constituent of cuticle,and chitin binding,which are associated with the decomposition and reconstruction of cuticle.qPCR results confirmed that myo-inositol mediated gene expression levels of the above cuticle-related pathways.Considering the importance of the cuticle in exoskeleton formation and molting,it can be concluded that myo-inositol-induced changes in the cuticle decomposition and reconstruction might have accelerated the transformation from megalopa to crablet of S.paramamosain.Besides,numerous DEGs were significantly enriched in protein digestion and absorption,amino sugar and nucleotide sugar metabolism.It implies that myo-inositol may improve survival by regulating energy or nutrient absorption.Additionally,the accelerated metamorphosis by myo-inositol may improve survival from megalopa to crablet of S.paramamosain.Overall,this study will provide the first insights into the underlying molecular mechanisms by which myo-inositol increases metamorphosis and survival.Wenxiao Cui Hafiz Sohaib Ahmed Saqib Weifeng Gao Ruibo Wang Yang He Yang Yu Zhen Lin Qingyu Zhang Yin Zhang Shengkang Li Huaiping Zheng Yueling Zhang Mhd Ikhwanuddin Hongyu Ma 2023Aquaculture and Fisheries2023,8,6:0
6Enhancement of catalytic activity for hydrogenation of nitroaromatic by anionic metal-organic framework显示文摘Nitroaromatic hydrogenation catalysis without precious metals remains a longstanding challenge.The rate of electron transfer is the crucial factor affecting hydrogenation catalysis.Herein,an ionic Cd-based metal-organic framework(I-Cd-MOF)exhibiting a unique structure with one-dimensional(1D)opening nanochannels and good electron transfer ability was synthesized for catalyzing hydrogenation of 4-nitrophenol(4-NP).The catalytic activity of the unique I-Cd-MOF without noble metals is detected,which is higher than most reported noble metal catalysts.Remarkably,the reaction rate of I-Cd-MOF(4.28 min^(-1))is about 47.6 times higher than that of the Cd-based neutral MOF(N-Cd-MOF)with the similar crystalline structure.Liquid chromatograph mass spectrometer(LC-MS)and theoretical results demonstrate that 4-NP and five intermediates are stabilized in the channels of I-Cd-MOF,which increases the possibility of contact with H^*and H_(2)g enerated at the Cd sites.The I-Cd-MOF was extended to other nitroaromatic hydrogenation catalysis,which still displays excellent activity.More importantly,the I-MOF@Filter membrane was successfully constructed for continuous hydrogenation catalytic reactions,which maintains a high catalytic performance after 7 cycles of recycling without washing.This work fills in the application of the I-MOFs in hydrogenation catalytic reactions and provides an effective way for the rapid and green degradation of nitroaromatic compounds.Qi Wu Anyang Li Ruibo He Yaxi Wu Lei Hou Guoping Yang Wenyan Zhang Yao-Yu Wang 2024Chinese Chemical Letters2024,35,3:0
7Sonic hedgehog signaling:Alternative splicing and pathogenic role in medulloblastoma显示文摘Alternative splicing(AS)produces the different mRNA splicing bodies,which are then translated into multiple protein isoforms and participate in various biological functions.With a deeper understanding of alternative splicing through the study of transcriptomes using high-throughput sequencing-based methods,the correlation between aberrant AS and diseases triggered a great concern,especially abnormal AS and cancer.Medulloblastoma(MB)is an intracranial tumor in children.Sonic hedgehog MB(SHH-MB)accounted for approximately 30%of MB,which is associated with the activation of SHH signaling.Growing evidence shows that aberrant AS is closely related to the tumorigenesis of MB.Here,we briefly introduced the AS and its mechanism.Next,we described canonical/noncanonical hedgehog signaling and its correlation with MB.The main description focused on AS of various regulators in canonical hedgehog signaling in MB.In addition,we also described AS of various regulators in noncanonical hedgehog signaling.Meanwhile,activated hedgehog signaling also induces AS in MB.Then,we pointed out that aberrant AS of hedgehog signaling is associated with different MB subgroups.Finally,we summarized the therapeutic applications of targeted AS in cancer treatment.In summary,further understanding of AS in SHH-MB could develop therapeutic targets for splicing factors which may be a novel therapeutic strategy.Meiyu Qu Qiangqiang He Jiahao Luo Tingyu Shen Ruibo Gao Yana Xu Chengyun Xu Muhammad Qasim Barkat Ling-Hui Zeng Ximei Wu 2023Genes & Diseases2023,10,5:0
8Effect of NK cells on GVHD in H-2 haploidentical bone marrow transplantation in mice显示文摘Objective: To study the effect of natural killer (NK) cells on graft-versus-host disease (GVHD) after H-2 haploidentical bone marrow transplantation(BMT) in mice. Methods:Murine model of H-2 haploidentical BMT was established by using Balb/c(H-2d) mouse as recipient, and Balb/c(H-2d)×C57BL/6(H-2b)(H-2d/b) mouse as donor. Lethally irradiated Balb/c(H-2d) mice were transplanted with the bone marrow cells from Balb/c(H-2d)×C57BL/6(H-2b)(H-2d/b) mice containing donor spleen cells and/or NK cells. GVHD and survival rates were studied by observation of clinical manifestations and pathological changes. Results:In the group of bone marrow +spleen cells, GVHD was induced in 90% mice; but in the group plus with low amount of NK cells, GVHD was induced in 20% mice; and in the group transplanted with high amount of NK cells, GVHD was induced only in 10% mice. Compared to the group transplanted only with BM plus spleen cells, the incidences of GVHD in the latter two groups decreased significantly(P < 0.01) and the survival rates at different periods of 15, 30, 45 and 60 days increased obviously(P < 0.01).
