| 2 | Multiplexed Biosensing Diagnostic Platforms Detecting Autoantibodies to Tumor-Associated Antigens from Exosomes Released by CRC Cells and Tissue Samples Showed High Diagnostic Ability for Colorectal Cancer显示文摘Colorectal cancer(CRC)is the second leading cause of cancer-related death worldwide.The five-year survival rate of CRC patients depends on the stage at diagnosis,being higher than 80%when CRC is diagnosed in the early stages but lower than 10%when CRC is diagnosed in advanced stages.Autoantibodies against specific CRC autoantigens(tumor-associated antigens(TAAs))in the sera of patients have been widely demonstrated to aid in early diagnosis.Thus,we herein aim to identify autoantigens target of autoantibodies specific to CRC that possess a significant ability to discriminate between CRC patients and healthy individuals by means of liquid biopsy.To that end,we examined the protein content of the exosomes released by five CRC cell lines and tissue samples from CRC patients by means of immunoprecipitation coupled with mass spectrometry analysis.A total of 103 proteins were identified as potential autoantigens specific to CRC.After bioinformatics and meta-analysis,we selected 15 proteins that are more likely to be actual CRC autoantigens in order to evaluate their role in CRC prognosis by Western blot(WB)and immunohistochemistry(IHC).We found dysregulation at the protein level for 11 of these proteins in both tissue and plasma exosome samples from patients,along with an association of nine of these proteins with CRC prognosis.After validation,all but one showed a statistically significant high diagnostic ability to distinguish CRC patients and individuals with premalignant lesions from healthy individuals,either by luminescence Halotag-based beads,or by a multiplexed biosensing platform involving the use of magnetic microcarriers as solid support modified with covalently immobilized Halotag fusion proteins constructed for CRC detection.Taken together,our results highlight the usefulness of the approach defined here to identify the TAAs specific to chronic diseases;they also demonstrate that the measurement of autoantibody levels in plasma against the TAAs identified here could be integrated into a point-of-care(POC)device for CRC detection with high diagnostic ability. | Ana Montero-Calle Itziar Aranguren-Abeigon María Garranzo-Asensio Carmen Poves María Jesús Fernández-Aceñero Javier Martínez-Useros Rodrigo Sanz Jana Dziaková Javier Rodriguez-Cobos Guillermo Solís-Fernández Eloy Povedano Maria Gamella Rebeca Magnolia Torrente-Rodríguez Miren Alonso-Navarro Vivian de los Ríos J.Ignacio Casal Gemma Domínguez Ana Guzman-Aranguez Alberto Peláez-García JoséManuel Pingarrón Susana Campuzano Rodrigo Barderas | 2021 | Engineering2021,7,10: | 1 |

| 5 | Abscisic Acid Connects Phytohormone Signaling with RNA Metabolic Pathways and Promotes an Antiviral Response that Is Evaded by a Self- Controlled RNA Virus显示文摘A complex network of cellular receptors,RNA targeting pathways,and small-molecule signaling provides robust plant immunity and tolerance to viruses.To maximize their fitness,viruses must evolve control mechanisms to balance host immune evasion and plant-damaging effects.The genus Potyvirus comprises plant viruses characterized by RNA genomes that encode large polyproteins led by the P1 protease.A P1 autoinhibitory domain controls polyprotein processing,the release of a downstream functional RNAsilencing suppressor,and viral replication.Here,we show that P1Pro,a plum pox virus clone that lacks the P1 autoinhibitory domain,triggers complex reprogramming of the host transcriptome and high levels of abscisic acid(ABA)accumulation.A meta-analysis highlighted ABA connections with host pathways known to control RNA stability,turnover,maturation,and translation.Transcriptomic changes triggered by P1Pro infection or ABA showed similarities in host RNA abundance and diversity.Genetic and hormone treatment assays showed that ABA promotes plant resistance to potyviral infection.Finally,quantitative mathematical modeling of viral replication in the presence of defense pathways supported self-control of polyprotein processing kinetics as a viral mechanism that attenuates the magnitude of the host antiviral response.Overall,our findings indicate that ABA is an active player in plant antiviral immunity,which is nonetheless evaded by a self-controlled RNA virus. | Fabio Pasin Hongying Shan Beatriz García Maren Müller David San León Márta Ludman David H.Fresno Károly Fátyol Sergi Munné-Bosch Guillermo Rodrigo Juan Antonio García | 2020 | Plant Communications2020,1,5: | 1 |

| 9 | Adaptation of international coronavirus disease 2019 and breast cancer guidelines to local context显示文摘BACKGROUND The coronavirus disease 2019(COVID-19)pandemic caused by severe acute respiratory syndrome coronavirus 2(a novel coronavirus),which was first identified amid an outbreak of respiratory illness cases in Wuhan,China and declared a global health emergency,is currently considered an additional challenge in the management of patients with breast cancer(BC).Cancer patients are more vulnerable to becoming infected with severe acute respiratory syndrome coronavirus 2 and are more likely to suffer additional complications that can increase mortality.Identifying those BC patients who require more urgent therapy than others in the current situation is essential.These recommendations are based on and have been adapted from those similarly published by international scientific societies for BC management.They are divided mainly by clinical stage(early,advanced),subtype[luminal,human epidermal growth factor receptor 2(HER2),triple-negative],or type of medical treatment and setting(neoadjuvant,adjuvant,metastatic).Recommendations for HER2 and triplenegative subtypes are similar,whereas in luminal subtype there are various options of management.The objective is to adapt guidelines to local context through relevant decision-makers,avoiding duplication of efforts and optimizing use or resources.We hope that these recommendations will help medical oncologists provide the best quality care to BC patients during the COVID-19 pandemic with information tailored to our healthcare system.AIM To establish and adapt recommendations from those published by international scientific societies for BC management.METHODS The Peruvian Society of Medical Oncology developed a consensus and propose here a manuscript with recommendations for oncological medical treatment of BC during the COVID-19 pandemic.The Peruvian Society of Medical Oncology invited a panel of experts and opinion leaders on BC working in major health care systems around Peru.Panel experts selected three international clinical practice guidelines(National Comprehensive Cancer Network,European Society for Medical Oncology,Spanish Foundation Research Group in Breast Cancer),considering that these are more representative in COVID-19 management.Also,the panel agreed to include at least one European and American clinical practice guideline.RESULTS Recommendations about BC management during the COVID-19 pandemic were divided mainly by clinical stage(early,advanced),subtype(luminal,HER2,triplenegative),or type of medical treatment and setting(neoadjuvant,adjuvant,metastatic).Recommendations for HER2 and triple-negative subtypes were similar between clinical practice guidelines,whereas in luminal subtype there were various options of management.One hundred twelve recommendations were reviewed,adapted,and voted.A consensus was made in order to provide best decisions of management,avoid duplication of efforts,and optimize medical resources,considering health care system reality.These recommendations are not intended to replace clinical judgment.CONCLUSION Most of recommendations are similar,mainly in high-risk subtypes(HER2,triplenegative).Certain societies adapt them to deal with different situations involving the best decision in the management of BC patients. | Guillermo Arturo Valencia Silvia Neciosup Henry L Gómez Maria Del Pilar Benites Silvia Falcón David Moron Karin Veliz Mike Maldonado Rodrigo Auqui | 2021 | World Journal of Clinical Oncology2021,12,1: | 1 |

| 12 | Precision silviculture:use of UAVs and comparison of deep learning models for the identification and segmentation of tree crowns in pine crops显示文摘The monitoring of trees is crucial for the management of large areas of forest cultivations,but this process may be costly.However,remotely sensed data offers a solution to automate this process.In this work,we used two neural network methods named You Only Look Once(YOLO)and Mask R-CNN to overcome the challenging tasks of counting,detecting,and segmenting high dimensional Red–Green–Blue(RGB)images taken from unmanned aerial vehicles(UAVs).We present a processing framework,which is suitable to generate accurate predictions for the aforementioned tasks using a reasonable amount of labeled data.We compared our method using forest stands of different ages and densities.For counting,YOLO overestimates 8.5%of the detected trees on average,whereas Mask R-CNN overestimates a 4.7%of the trees.For the detection task,YOLO obtains a precision of 0.72 and a recall of 0.68 on average,while Mask R-CNN obtains a precision of 0.82 and a recall of 0.80.In segmentation,YOLO overestimates a 13.5%of the predicted area on average,whereas Mask R-CNN overestimates a 9.2%.The proposed methods present a cost-effective solution for forest monitoring using RGB images and have been successfully used to monitor∼146,500 acres of pine cultivations. | Manuel Ignacio Perez Bruno Karelovic Roberto Molina Rodrigo Saavedra Pierluigi Cerulo Guillermo Cabrera | 2022 | International Journal of Digital Earth2022,15,1: | 0 |

| 15 | An international survey of classification and treatment choices for group D retinoblastoma显示文摘AIM: To determine which IIRC scheme was used by retinoblastoma centers worldwide and the percentage of D eyes treated primarily with enucleation versus globe salvaging therapies as well as to correlate trends in treatment choice to IIRC version used and geographic region.METHODS: An anonymized electronic survey was offered to 115 physicians at 39 retinoblastoma centers worldwide asking about IIRC classification schemes and treatment patterns used between 2008 and 2012. Participants were asked to record which version of the IIRC was used for classification, how many group D eyes were diagnosed, and how many eyes were treated with enucleation versus globe salvaging therapies. Averages of eyes per treatment modality were calculated and stratified by both IIRC version and geographic region. Statistical significance was determined by Chi-square, ANOVA and Kruskal-Wallis tests using Prism.RESULTS: The survey was completed by 29% of physicians invited to participate. Totally 1807 D eyes were diagnosed. Regarding IIRC system, 27% of centers used the Children's Hospital of Los Angeles(CHLA) version, 33% used the Children's Oncology Group(COG) version, 23% used the Philadelphia version, and 17% were unsure. The rate for primary enucleation varied between 0 and 100% and the mean was 29%. By IIRC version, primary enucleation rates were: Philadelphia, 8%; COG, 34%; and CHLA, 37%. By geographic region, primary enucleation rates were: Latin America, 57%; Asia, 40%; Europe, 36%; Africa, 10%, US, 8%; and Middle East, 8%. However, systemic chemoreduction was used more often than enucleation in all regions except Latin America with a mean of 57% per center(P<0.0001).
