| 3 | Anti-tumor activity of wogonin, an extract from Scutellaria baicalensis, through regulating different signaling pathways显示文摘Wogonin is a plant flavonoid compound extracted from Scutellaria baicalensis(Huang-Qin or Chinese skullcap) and has been studied thoroughly by many researchers till date for its anti-viral, anti-oxidant, anti-cancerous and neuro-protective properties. Numerous experiments conducted in vitro and in vivo have demonstrated wogonin's excellent tumor inhibitory properties. The anticancer mechanism of wogonin has been ascribed to modulation of various cell signaling pathways, including serine-threonine kinase Akt(also known as protein kinase B) and AMP-activated protein kinase(AMPK) pathways, p53-dependent/independent apoptosis, and inhibition of telomerase activity. Furthermore, wogonin also decreases DNA adduct formation with a carcinogenic compound 2-Aminofluorene and inhibits growth of drug resistant malignant cells and their migration and metastasis, without any side effects. Recently, newly synthesized wogonin derivatives have been developed with impressive anti-tumor activity. This review is the succinct appraisal of the pertinent articles on the mechanisms of anti-tumor properties of wogonin. We also summarize the potential of wogonin and its derivatives used alone or as an adjunct therapy for cancer treatment. | 5 | Pentoxifylline versus prednisolone for severe alcoholic hepatitis:A randomized controlled trial显示文摘AIM:To compare the efficacy of pentoxifylline and prednisolone in the treatment of severe alcoholic hepatitis,and to evaluate the role of different liver function scores in predicting prognosis.METHODS:Sixty-eight patients with severe alcoholic hepatitis(Maddrey score≥32)received pentoxifylline(n=34,groupⅠ)or prednisolone(n=34,groupⅡ) for 28 d in a randomized double-blind controlled study,and subsequently in an open study(with a tapering dose of prednisolone)for a total of 3 mo,and were followed up over a period of 12 mo.RESULTS:Twelve patients in groupⅡdied at the end of 3 mo in contrast to five patients in groupⅠ.The probability of dying at the end of 3 mo was higher in groupⅡas compared to groupⅠ(35.29%vs 14.71%,P=0.04;log rank test).Six patients in groupⅡ developed hepatorenal syndrome as compared to none in groupⅠ.Pentoxifylline was associated with a significantly lower model for end-stage liver disease(MELD)score at the end of 28 d of therapy(15.53±3.63 vs 17.78±4.56,P=0.04).Higher baseline Maddrey score was associated with increased mortality.CONCLUSION:Reduced mortality,improved risk-benefit profile and renoprotective effects of pentoxifylline compared with prednisolone suggest that pentoxifylline is superior to prednisolone for treatment of severe alcoholic hepatitis. | Binay Krishna De Subhabrata Gangopadhyay Deep Dutta Sumanta Das Baksi Adyapad Pani Pramit Ghosh | 2009 | World Journal of Gastroenterology2009,15,13: | 26 |
| 6 | Th17 plasticity and its changes associated with inflammatory bowel disease显示文摘CD4 T helper(Th) cell differentiation into distinct T cell subsets is critical to the normal function of the immune system. Until recently,the paradigm held that na?ve T cells differentiated into distinct subsets under the guidance of environmental cues(e.g.,cytokines) and that once polarized,these cells were committed to a particular functional state. However,the existence of transdifferentiated T cell populations,which express signature transcription factors and cytokines associated with more than one Th subset,challenges the immutability of T helper subsets and suggests that plasticity is a feature of multifaceted immune responses. How this process impacts immune dysregulation in diseases such as inflammatory bowel diseases(IBD) and the machinery that underlies this process is far from fully understood. Interleukin(IL)-17 secreting helper T(Th17) cells have been heavily implicated in tissue-specific immune pathology including murine models of IBD,human Crohn's disease and ulcerative colitis. Plasticity within this subset is suggested by the existence of IL-17 secreting cells,which,can also secrete interferon-γ,the signature cytokine for Th1 cells or,can co-express the anti-inflammatory transcription factor forkhead box p3,a signature transcription factor of regulatory T cells. | 7 | Upper-gastrointestinal bleeding secondary to peptic ulcer disease:Incidence and outcomes显示文摘AIM:To evaluate the incidence,surgery,mortality,and readmission of upper gastrointestinal bleeding(UGIB)secondary to peptic ulcer disease(PUD).METHODS:Administrative databases identified all hospitalizations for UGIB secondary to PUD in Alberta,Canada from 2004 to 2010(n=7079)using the International Classification of Diseases Codes(ICD-10).A subset of the data was validated using endoscopy reports.Positive predictive value and sensitivity with 95%confidence intervals(CI)were calculated.Incidence of UGIB secondary to PUD was calculated.Logistic regression was used to evaluate surgery,in-hospital mortality,and 30-d readmission to hospital with recurrent UGIB secondary to PUD.Co-variants accounted for in our logistic regression model included:age,sex,area of residence(i.e.,urban vs rural),number of Charlson comorbidities,presence of perforated PUD,undergoing upper endoscopy,year of admission,and interventional radiological attempt at controlling bleeding.A subgroup analysis(n=6356)compared outcomes of patients with gastric ulcers to those with duodenal ulcers.Adjusted estimates are presented as odds ratios(OR)with95%CI.RESULTS:The positive predictive value and sensitivity of ICD-10 coding for UGIB secondary to PUD were85.2%(95%CI:80.2%-90.2%)and 77.1%(95%CI:69.1%-85.2%),respectively.The annual incidence between 2004 and 2010 ranged from 35.4 to 41.2 per100000.Overall risk of surgery,in-hospital mortality,and 30-d readmission to hospital for UGIB secondary to PUD were 4.3%,8.5%,and 4.7%,respectively.Interventional radiology to control bleeding was performed in 0.6%of patients and 76%of these patients avoided surgical intervention.Thirty-day readmission significantly increased from 3.1%in 2004 to 5.2%in 2010(OR=1.07;95%CI:1.01-1.14).Rural residents(OR rural vs urban:2.35;95%CI:1.83-3.01)and older individuals(OR≥65 vs<65:1.57;95%CI:1.21-2.04)were at higher odds of being readmitted to hospital.Patients with duodenal ulcers had higher odds of dying(OR=1.27;95%CI:1.05-1.53),requiring surgery(OR=1.73;95%CI:1.34-2.23),and being readmitted to hospital(OR=1.54;95%CI:1.19-1.99)when compared to gastric ulcers.CONCLUSION:UGIB secondary to PUD,particularly duodenal ulcers,was associated with significant morbidity and mortality.Early readmissions increased over time and occurred more commonly in rural areas. | Samuel Quan Alexandra Frolkis Kaylee Milne Natalie Molodecky Hong Yang Elijah Dixon Chad G Ball Robert P Myers Subrata Ghosh Robert Hilsden Sander Veldhuyzen van Zanten Gilaad G Kaplan | 2014 | World Journal of Gastroenterology2014,20,46: | 21 |
| 8 | Generalized megaviscera of lupus:Refractory intestinal pseudo-obstruction,ureterohydronephrosis and megacholedochus显示文摘Dilated dysfunction involving multiple visceral organs has been reported in patients with systemic lupus erythematosus(SLE).Chronic intestinal pseudo-obstruction(CIPO) resulting from intestinal smooth muscle damage has presented in conjunction with ureterohydronephrosis and, more rarely, biliary dilatation(megacholedochus).While the molecular pathogenesis is largely unknown, observed histo-pathologic features include widespread myositis, myocyte necrosis in the intestinal muscularis propria with subsequent atrophy and fibrosis, preserved myenteric innervations and little vasculitis.High dose immunosuppression usually results in resolution of symptoms with recovery of smooth muscle function, indicative of an autoimmune etiology.We report a patient with SLE who presented with intestinal pseudo-obstruction, ureterohydronephrosis and megacholedochus, and present images that illustrate megaviscera simultaneously involving all 3 visceral organs.Since the co-manifestation of all 3 is unusual and has been reported only once previously, wehave termed this rare clinical syndrome generalized megaviscera of lupus(GML).Although the SLE disease-activity parameters responded to aggressive immunomodulative therapy in our patient, clinical evidence of peristaltic dysfunction persisted in all involved viscera.This is a variation from the favorable outcomes reported previously in SLE patients with GML and we attribute this poor clinical outcome to disease severity and, most importantly, delayed clinical presentation.Since inflammation followed by atrophy and fibrosis are key aspects in the pathogenesis and natural history of GML, the poor response in our patient who presented late in the clinical course may be the result of ‘burnt out' inflammation with irreversible end-stage flbrosis.Thus, early recognition and timely initiation of treatment may be the key to recover visceral peristaltic function in patients with GML. | Frederick D Park Jeffrey K Lee Ganga D Madduri Pradipta Ghosh | 2009 | World Journal of Gastroenterology2009,15,28: | 20 |

| 13 | Xanthogranulomatous cholecystitis:a premalignant condition?显示文摘BACKGROUND:Xanthogranulomatous cholecystitis(XGC)is an uncommon variant of chronic cholecystitis,characterized by marked thickening of the gallbladder wall and dense local adhesions.It often mimics a gallbladder carcinoma(GBC), and may coexist with GBC,leading to a diagnostic dilemma. Furthermore,the premalignant nature of this entity is not known.This study was undertaken to assess the p53,PCNA and beta-catenin expression in XGC in comparison to GBC and chronic inflammation. METHODS:Sections from paraffin-embedded blocks of surgically resected specimens of GBC(69 cases),XGC(65), chronic cholecystitis(18)and control gallbladder(10)were stained with the monoclonal antibodies to p53 and PCNA, and a polyclonal antibody to beta-catenin.p53 expression was scored as the percentage of nuclei stained.PCNA expression was scored as the product of the percentage of nuclei stained and the intensity of the staining(1-3).A cut-off value of 80 for this score was taken as a positive result. Beta-catenin expression was scored as type of expression-membranous,cytoplasmic or nuclear staining. RESULTS:p53 mutation was positive in 52%of GBC cases and 3%of XGC,but was not expressed in chronic cholecystitis and control gallbladders.p53 expression was lower in XGC than in GBC(P<0.0001).PCNA expression was seen in 65%of GBC cases and 11%of XGC,but not in chronic cholecystitis and control gallbladders.PCNA expression was higher in GBC than XGC(P=0.0001),but there was no significant difference between the XGC,chronic cholecystitis and control gallbladder groups.Beta-catenin expression was positive in the GBC,XGC, chronic cholecystitis and control gallbladder groups.But the expression pattern in XGC,chronic cholecystitis and control gallbladders was homogenously membranous,whereas in GBC the membranous expression pattern was altered to cytoplasmic and nuclear.CONCLUSION:The expression of p53,PCNA and beta-catenin in XGC was significantly different from GBC and similar to chronic cholecystitis,thus indicating the inflammatory nature of XGC and may not support a premalignant nature of the lesion. | Mila Ghosh Puja Sakhuja Anil K Agarwal | 2011 | Hepatobiliary & Pancreatic Diseases International2011,10,2: | 11 |
| 14 | Elastic properties of chiral,anti-chiral,and hierarchical honeycombs:A simple energy-based approach显示文摘The effects of two geometric refinement strategies widespread in natural structures, chirality and self-similar hierarchy, on the in-plane elastic response of two-dimensional honeycombs were studied systematically. Simple closed-form expressions were derived for the elastic moduli of several chiral, antichiral, and hierarchical honeycombs with hexagon and square based networks. Finite element analysis was employed to validate the analytical estimates of the elastic moduli. The results were also compared with the numerical and experimental data available in the literature. We found that introducing a hierarchical refinement increases the Young's modulus of hexagon based honeycombs while decreases their shear modulus. For square based honeycombs, hierarchy increases the shear modulus while decreasing their Young's modulus. Introducing chirality was shown to always decrease the Young's modulus and Poisson's ratio of the structure. However, chirality remains the only route to auxeticity. | 17 | Sorption and Fractionation of a Peat Derived Humic Acid by Kaolinite, Montmorillonite, and Goethite显示文摘Sorption of humic acid (HA) on mineral surfaces has a profound interest regarding the fate of hydrophobic organic contaminants (HOCs) and carbon sequestration in soils. The objective of our study is to determine the fractionation behavior of HA upon sorption on mineral surfaces with varying surface properties. HA was coated sequentially on kaolinite (1:1 clay), montmorillonite (2:1 clay), and goethite (iron oxide) for four times. The unadsorbed HA fractions were characterized by elemental analysis, diffuse reflectance infrared Fourier transform spectroscopy (DRIFT), and solid state 13C nuclear magnetic resonance spectroscopy (NMR). The mineral-HA complexes were characterized by DRIFT. Polarity index [(N+O)/C] revealed higher polarity of the unadsorbed HA fractions after coating on kaolinite, reflecting that relatively higher polarity fractions of HA remain unadsorbed. Sorption of aliphatic alcohol fraction along with carbohydrate was prominent on kaolinite surface. DRIFT results of the unadsorbed HA fractions indicated more sorption of aliphatic moieties on both kaolinite and montmorillonite. DRIFT results of the unadsorbed HA fractions after sorption on kaolinite and goethite showed the sorption of the proteinaceous fractions of HA. The HA fractions obtained after coating on goethite showed significant sorption of carboxylic moieties. The results mentioned above comply reasonably well with the DRIFT spectra of the mineral-HA complexes. 13C NMR results showed higher sorption of anomeric C on kaolinite surface. Higher sorption of paraffinic fraction was observed on montmorillonite. NMR data inferred the sorption of carboxylic moieties on goethite surface. Overall, this study showed that aliphatic moieties of HA preferentially sorbed on kaolinite and montmorillonite, while carboxylic functional groups play a significant role in sorption of HA on goethite. | 19 | 超声辅助法合成的石墨烯-TiO_2复合材料声催化降解罗丹明B(英文)显示文摘A series of graphene-TiO2 composites was fabricated from graphene oxide and titanium n-butoxide(TNB) by an ultrasonic-assisted method.The structure and composition of the nanocomposites were characterized by Raman spectroscopy,BET surface area measurements,X-ray diffraction,transmission electron microscopy,and ultraviolet-visible absorption spectroscopy.The average size of the TiO2 nanoparticles on the graphene nanosheets was controlled at around 10-15 nm without using surfactant,which is attributed to the pyrolysis and condensation of dissolved TNB into TiO2 by ultrasonic irradiation.The catalytic activity of the composites under ultrasonic irradiation was determined using a rhodamine B(RhB) solution.The graphene-TiO2 composites possessed a high specific surface area,which increased the decolorization rate for RhB solution.This is because the graphene and TiO2 nanoparticles in the composites interact strongly,which enhances the photoelectric conversion of TiO2 by reducing the recombination of photogenerated electron-hole pairs. | Trisha GHOSH Chong-Yeon PARK OH Won-Chun | 2012 | 催化学报2012,33,8: | 8 |
| 20 | Detection of hepatitis B virus infection: A systematic review显示文摘AIM: To review published methods for detection of hepatitis B virus(HBV) infection.METHODS: A thorough search on Medline database was conducted to find original articles describing different methods or techniques of detection of HBV, which are published in English in last 10 years. Articles outlining methods of detection of mutants or drug resistance were excluded. Full texts and abstracts(if full text not available) were reviewed thoroughly. Manual search of references of retrieved articles were also done. We extracted data on different samples and techniques of detection of HBV, their sensitivity(Sn), specificity(Sp) and applicability.RESULTS: A total of 72 studies were reviewed. HBV was detected from dried blood/plasma spots, hepatocytes, ovarian tissue, cerumen, saliva, parotid tissue, renal tissue, oocytes and embryos, cholangiocarcinoma tissue, etc. Sensitivity of dried blood spot for detecting HBV was > 90% in all the studies. In case of seronegative patients, HBV DNA or serological markers have been detected from hepatocytes or renal tissue in many instances. Enzyme linked immunosorbent assay and Chemiluminescent immunoassay(CLIA) are most commonly used serological tests for detection. CLIA systems are also used for quantitation. Molecular techniques are used qualitatively as well as for quantitative detection. Among the molecular techniques version 2.0 of the Cobas Ampliprep/Cobas Taq Man assay and Abbott's real time polymerase chain reaction kit were found to be most sensitive with a lower detection limit of only 6.25 IU/m L and 1.48 IU/m L respectively. CONCLUSION: Serological and molecular assays are predominant and reliable methods for HBV detection.
