1Dynamics of sediment resus-pension and the conceptual schema of nutrient release in the large shallow Lake Taihu,China显示文摘On the basis of investigations in situ, it was found that mass exchange on the water-sediment interface occurred chiefly on the superficial sediment within 5-10 cm.The spatial physicochemical character of sediment was distributed uniformly. The observation of lake currents and waves indicated that the dynamic sources, which act on the interface of water and sediment, came mainly from waves under strong wind forcing, while the critical shear stresses due to the waves and currents were of the same magnitude under weak wind forcing. The critical shear stress that leads to extensive sediment resuspension was about 0.03-0.04N/m2, equivalent to a wind speed in situ up to 4 m/s. If a dynamic intensity exceeded the critical shear stress, such as a wind velocity up to 6.5 m/s, massive sediment re-suspension would be observed in the lake. Furthermore, field investigations revealed that the nutrient concentration of pore water within the sediment was far greater than that of overlaying water, which provides objective conditions for the nutrient release from sediment. According to nutrient analyses in the pore water from the superficial 5-10 cm sediments, a severe dynamic process in the Taihu Lake would bring out a peak nutrient release, i.e. a 0.12 mg/L increase of TN, and 0.005mg/L increase of TP in the lake. In the end, a general scheme of nutrient release from sediment in large shallow lakes was put forward: when the wind-driven forcing imposes on the lake, it will make the sediment resuspension. At the same time, the nutrition from the pore water will follow the sediment resuspension release to overlaying water. Because of oxidation of solid particulates when it resuspends from sediment, the disturbance of hydrodynamics will enhance thesuspension particulates absorbing nutrition. 2Direct evidence of phosphorus outbreak release from sedi-ment to overlying water in a large shallow lake caused by strong wind wave disturbance显示文摘Concentration variations of suspended solids (SS), total phosphorus (TP), dissolved total phosphorus (DTP), dissolved reactive phosphorus (SRP), and algae available phosphorus (AAP) in overlying water were ob- served during the coldest week in a year in Lake Taihu, a large shallow lake in China. Water samples at different water depths were collected with wind speeds of 8, 12, 0 and 0 m/s on 23, 24, 26 and 30 January 2004, respectively. On 23 Janu- ary 2004, SS concentration increased to 258 mg/L with a wind speed of 8 m/s lasting for 1 h. SS concentration kept increasing and reached to 507 mg/L when the strong wind lasted for 24 h and the peak value of wind-speed reached to 12 m/s on 24 January 2004. On 26 January 2004, SS concen- tration decreased to 51 mg/L with the wind speed smaller than 2 m/s lasting for about half a day. Then after five con- tinuous waveless days, SS concentration decreased to 21 mg/L on 30 January 2004. The observed results confirmed that sediments in Lake Taihu would be intensively suspended if the surface wind speed is greater than 8 m/s, and the mag- nitude of SS would increase with increasing wind-speed. Coupled with the intensive sediment suspending, concentra- tions of TP, DTP and SRP on 23 January were 0.210, 0.048 and 0.035 mg/L, respectively. And they were 0.299, 0.054 and 0.026 mg/L on 24 January, which were significantly higher than those on 26 and 30 January. SRP concentration on 23 January was twice as high as that observed on 30 January. It indicates that the strong wind may result in an outbreak re- lease of phosphorus. Moreover, AAP contents in suspended solids were 132, 97 and 226 mg/kg on 23, 24 and 26 January, respectively. Therefore, it could be estimated that this strong wind process resulted in 987 t of TP, 80 t of SRP and 167 t of AAP releasing from sediments into overlying water. Since such strong wind process is frequent in the area of Lake Taihu, dynamical release driven by wind-induced wave dis- turbance may be the main mode for internal release of phosphorus. 3Processes and mechanism of effects of sludge dredging on internal source release in lakes显示文摘Simulated research of internal loading and col- lecting and analyzing the samples from the lakes were carried out before and after dredging in polluted suburb lakes, Wuli Lake (Wuxi City) and Xuanwu Lake (Nanjing City). The research results showed that dredging can in- hibit internal loadings in a certain degree in a short term. The discrepancy of dredging effect and technical level, namely dredging quality, by different dredging methods will result in a difference of control of lake internal Ioad- ings. The internal Ioadings’ reversion will gradually ap- pear along with the bio-geochemical processes, including suspended particle precipi-tation, hydrodynamic distur- bance and microbio-transfor-mation. The reversion rate mainly depends on the dredging method and the change of interfacial processes on the new-born surface layer. The higher nutrient contents and organic matter in the sedi- ment will enhance water-sediment interfa-cial processes and nutrients regeneration. 4Growth and phosphate uptake kinetics of Microcystis aeruginosa under various environmental conditions显示文摘Growth and uptake of exogenous phosphate by Microcystis aeruginosa in batch culture under different temperature, photoperiod, and turbulence were studied by the method of phosphate isotope tracer. Relatively high temperature, long photoperiod and strong turbulence increased the cell density of M. aeruginosa in these batch cultures. The initial rapid uptake of phosphate by M. aeruginosa was independent of the temperature, photoperiod, and turbulence. Similarly, maximum exogenous phosphate uptake was not related to these environmental factors. However, elevated temperature and turbulence shortened the time, required to obtain maximum P accumulation. The growth of M. aeruginosa could alleviate the phosphorous leakage. Total amounts of exogenous phosphate uptake to M. aeruginosa and the phosphorus leakage of M. aeruginosa were significantly influenced by the growth state of M. aeruginosa closely correlated with the environmental factors. The maximum volume of exogenous phosphate uptake to M. aeruginosa was 46% of added exogenous phosphate in water with 16 hours of photoperiod. 8Inhibition of DNA restrictive endonucleases by aqueous nanoparticle suspension of methanophosphonate fullerene derivatives and its mechanisms显示文摘Aqueous nanoparticle suspension of fullerene and its derivatives are currently attracting much attention. To determine the effects of aqueous nanoparticle suspension of a mono-methanophosphonate fullerene and bis-methanophosphonate fullerene (denoted as n-MMPF and n-BMPF, respectively) on the activities of DNA restrictive endonucleases, plasmid pEGFP-N1 was cleaved at a single but differently restrictive site by EcoR I, BamH I, and isozymes Cfr9 I and Xma I, respectively. Both n-MMPF and n-BMPF inhibited the activity of EcoR I, while n-BMPF exhibited stronger inhibition than n-MMPF. Addition of n-BMPF into reaction mixtures inhibited the activities of all the four enzymes, and IC50 values for EcoR I, BamH I, Cfr9 I and Xma I were 4.3, >30, 11.7 and 8.3 μmol/L, respectively. When EcoR I was completely inhibited by n-BMPF, addition of excess amounts of pEGFP-N1 could not produce the product linear plasmid; however, increase of EcoR I amounts antagonized EcoR I inhibition of n-BMPF. Two scavengers of reactive oxygen species (ROS), mannitol and sodium azide at the concentrations of 2-10 mmol/L, did not reverse inhibition of n-BMPF, implying that this inhibition probably is not correlated to ROS.
