10Substance use and substance use disorders in Africa:An epidemiological approach to the review of existing literature显示文摘The relationship between man and substances that have abuse potentials,and whose use has been associated with the development or progression of substance use disorders has continued to evolve in terms of geography,economic implications,and time.History shows that local plants with psychoactive constituents can get exported worldwide through global travel,commerce,or even conquest.Time and globalization also change people’s relationship with substances of abuse;hence,an area that was initially alien to certain substances might evolve to becoming a trafficking hub,and then a destination.A case in point is Africa where a rapidly increasing prevalence of substance use/abuse and substance use disorder among adolescents and young adults is putting enormous strain on the economy,healthcare system,and society at large.However,there appears to be a paucity of scientific literature and data on the epidemiology,risk assessment,and contributing factors to substance use and the development of substance use disorders across Africa.In this narrative review,we examine extant literature(PubMed,Google scholar,Medline)for information on the prevalence,trends,and influencers of substance use and the development of substance use disorders.This is with a view of understanding the determinants of substance use and factors that influence the development of substance use disorders in the region,and how this information can be channeled towards developing a comprehensive intervention and treatment program.Olakunle James Onaolapo Anthony Tope Olofinnade Foluso Olamide Ojo Olufunto Adeleye Joshua Falade Adejoke Yetunde Onaolapo 2022World Journal of Psychiatry2022,12,10:0
11Opening of liver mitochondrial permeability transition pore in streptozotocin-induced diabetic rats and its inhibition by methanol fraction of Ficus mucoso(Welw) root bark显示文摘Objective: Several pathologies arise from the inappropriate opening of the mitochondrial permeability transition(mPT) pore. In this regard, inhibition of mPT pore represents a cytoprotective approach to preserve mitochondrial function for treatment of diseases characterized by excessive tissue wastage such as diabetes mellitus. The aim of this study, therefore, was to study the effects of fractions of Ficus mucoso, a medicinal plant used in the traditional treatment of diabetes, on mPT pore in normal and streptozotocin(STZ)-induced diabetic rat liver.Methods: Different solvent fractions of the crude methanol extract of F. mucoso were obtained by vacuum liquid chromatography and were tested on the mPT pore. Of all the fractions tested, methanol fraction of F. mucoso(MFFM) was the most potent and was used for in vivo studies. Diabetes mellitus was induced by a single intraperitoneal injection of 60 mg/kg STZ, while treatment lasted for 14 d. In vivo, 30 male Wistar rats were divided into five groups: A, normo-glycemic control(distilled water);B, STZ(65 mg/kg;diabetic control);C, STZ + MFFM(25 mg/kg);D, STZ + MFFM(50 mg/kg);E, STZ + glibenclamide(5 mg/kg). The mPT, mitochondrial ATPase activity, lipid peroxidation and cytochrome c release were assessed spectrophotometrically while blood glucose levels were monitored using glucometer.Results: In vitro, the solvent fractions of F. mucoso, at all concentrations tested, had no effect on the mPT pore, in the absence of calcium, with no significant release of cytochrome c. Interestingly, calciumdependent pore opening was inhibited by all solvent fractions of F. mucoso, with the MFFM having the highest inhibitory effect of 83% at 3 mg/mL. Induction of opening of the mPT pore, significant(P < 0.001) enhancement of mitochondrial ATPase activity and elevated malondialdehyde(MDA) levels in STZ-induced diabetes were significantly(P < 0.001) reversed by MFFM and were comparable with the effects of glibenclamide, a standard antidiabetic drug. Also, treatment with MFFM at different doses significantly(P < 0.001) reduced high serum blood glucose compared to the diabetic control.Conclusion: F. mucoso could be useful in therapeutic management of diabetes mellitus given its ability to prevent excessive tissue wastage via inhibition of pore opening, and reduction in levels of MDA and serum blood glucose.Olubukola Titilope Oyebode Foluso Olayemi Ogunbiyi Olufunso Olabode Olorunsogo 2019Journal of Integrative Medicine2019,17,6:0

13Antiplatelet aggregation activity of some ferrocenylphenylimine compounds显示文摘Platelet hyper-aggregability triggered death and disability due to cardiovascular diseases is increasing worldwide and becoming a global concern. Therefore, it is necessary to synthesize newer drugs for the management of platelet aggregation. In this study, we investigated the antiplatelet aggregation activity of a novel series of ferrocenylimine compounds(3–10), N-(3-nitro-2-hydroxylbenzylidene)-3-ferrocenylimine(3), N-(3-bromo-2-hydroxylbenzylidene)-3-ferrocenylimine(4), N-(3-bromo-5-chlorosalicylidene)-3-ferrocenylimine(5), N-(ferrocenylformidene)-3-ferrocenylimine(6), N-(3-nitro-2-hydroxylbenzylidene)-4-ferrocenylimine(7), N-(3-bromo-2-hydroxylbenzylidene)-4-ferrocenylimine(8), N-(3-bromo-5-chlorosalicyl)-4-ferrocenylimine(9), N-(ferrocenylformidene)-4-ferrocenylimine(10) on thrombin-and ADP-induced platelet aggregation. The synthesized ferrocenylimine compounds(3–10) were found to exhibit higher antiplatelet aggregation activity than their precursors, which are 3-ferrocenylaniline(compound 1) and 4-ferrocenylaniline(compound 2).
