| 1 | 刚地弓形虫China 1基因型成囊株在小鼠体内的动态分布显示文摘目的观察流行我国的刚地弓形虫(Toxoplasma gondii)主要基因型China 1成囊株在感染小鼠体内的动态分布和包囊形成。方法采自武汉的猫源性刚地弓形虫株(TgCtwh1,基因型为China 1,即ToxoDB#9)包囊50个(约含1×104个缓殖子)经口感染SPF级CD1雌性小鼠50只,于感染后第2、4、7、10、14、21、35、50和72天,分别大体观察小鼠健康状况并测定体重;颈椎脱臼法处死小鼠,脑组织压片观察弓形虫包囊形成时间,计算成囊率,测量包囊直径;荧光定量PCR和组织接种方法检测弓形虫在血液、心、肝、脑和淋巴结组织中的动态分布。结果实验小鼠感染后第7天体重减轻(3.650±0.252)g;第10天[(1.730±0.017)g]与第14天体重差值[(-0.390±0.554)g]比较,有统计学意义(P<0.05)。感染后第21天,在脑组织中查获包囊,成囊率为80%,直径为20~40μm;至感染后第35天,小鼠成囊率增为100%,包囊直径为50~60μm。感染后第2天,TgCtwh1虫体即出现在血液、心、肝和淋巴结组织中,拷贝数分别为3.510±0.152、4.100±0.198、4.220±0.209和4.960±0.052,感染后第4天见于脑组织(3.800±0.154);血液中的虫体在第7天达峰值(5.240±0.115),随后逐渐下降,至第35天消失;心和脑组织中的虫体分别于第14天和第10天达峰值后(5.640±0.214和5.790±0.060)维持相对稳定的水平;肝和淋巴组织中的虫体分别于第7天和第10天达峰值(5.310±0.038和6.200±0.152),此后虫体逐渐减少,至第50天转阴。结论流行中国的弓形虫China 1基因型成囊株在感染小鼠体内虫血症可持续至少21 d,感染后第21天首次在小鼠脑组织内检测到包囊。 | 王林 霍星星 陈兆武 陈鹤 余莉 宋晓蓉 Faustina Halm-Lai 罗庆礼 沈继龙 | 2012 | 中国寄生虫学与寄生虫病杂志2012,30,3: | 4 |

| 7 | 重组果糖二磷酸醛缩酶SjLAP和亮氨酸氨基肽酶SjFBPA用于日本血吸虫病的诊断和疗效考核的评价(英文)显示文摘目的评价重组亮氨酸氨基肽酶(rSjLAP)和重组果糖二磷酸醛缩酶(rSjFBPA)抗原用于诊断人血吸虫感染以及疗效考核的价值。方法异丙基-β-D-硫代半乳糖苷(IPTG)诱导pET-28a-rSjLAP/BL21和pET-28a-rSjFBPA/BL21表达目的蛋白,组氨酸标签亲和纯化柱纯化rSjLAP和rSjFBPA蛋白。88只BALB/c雌性小鼠随机分为4组,A、B和C组(各21只小鼠),D组(25只小鼠)。A、B和C组分别感染5、15和25条日本血吸虫尾蚴。D组为不感染对照组,在实验的第1天全部处死。A、B和C组在感染后第3、7、10、14、20、30和60天,分别处死小鼠3只,采眼球血制备血清,检测其抗体水平。采用单独或联合rSjLAP和rSjFBPA为抗原,ELISA法检测小鼠血清、急性血吸虫病(38份)和慢性血吸虫病患者(96份)血清中的抗体,以健康人(90份)血清为对照,同时检测华支睾吸虫病(33份)、卫氏并殖吸虫病(40份)和钩虫病患者(37份)血清,并检测急性血吸虫病患者吡喹酮治疗(60 mg/kg,2次/d×2 d)后1年的血清(36份)、慢性血吸虫病患者吡喹酮治疗(剂量,疗程同前)后1年(36份)和2年(64份)的血清。结果 BALB/c小鼠在感染后第10天,rSjLAP和rSjFBPA单独或联合使用均可检测到小鼠血清中的IgG抗体;B组(0.535±0.053,0.595±0.033,0.696±0.104)和C组(0.548±0.060,0.608±0.063,0.621±0.090)早期抗体水平明显高于A组(0.415±0.038,0.455±0.056,0.498±0.077)(P<0.05)。用rSjLAP为抗原可检测急性血吸虫病和慢性血吸虫病患者血清,阳性率分别为97.4%(37/38)和87.5%(84/96)(P>0.05);用rSjFBPA为抗原检测,其阳性率分别为94.7%(36/38)和88.5%(85/96)(P>0.05);用rSjLAP和rSjFBPA联合为抗原检测,则其阳性率分别为94.7%(36/38)和85.4%(82/96)(P>0.05)。rSjLAP或联合抗原的特异性均为96.7%(87/90),而rSjFBPA的特异性为97.8%(88/90)。给予吡喹酮治疗后,rSjLAP和rSjFBPA单独或联合使用检测急性血吸虫病血清(0.236±0.212,0.287±0.191,0.235±0.120)和慢性血吸虫病患者(0.266±0.124,0.261±0.143,0.265±0.140;0.204±0.074,0.176±0.074,0.176±0.073),抗体滴度普遍下降,与对照组(0.188±0.056,0.173±0.045,0.184±0.051)相比,差异无统计学意义(P>0.05)。rSjLAP和rSjFBPA单独为抗原检测华支睾吸虫病患者血清,交叉反应率为15.2%(5/33)和12.1%(4/33),两者联合则为9.2%(3/33)。rSjLAP检测卫氏并殖吸虫的交叉反应率为15.0%(6/40),rSjFBPA为12.5%(5/40),两种抗原联合检测为15.0%(6/40)。上述抗原单独或联合检测钩虫的交叉反应率均为8.1%(3/37)。上述各组阳性率与健康人群之间差异有统计学意义(P<0.05),显示该重组抗原在其他蠕虫检测中存在一定的交叉反应。结论用rSjFBPA和rSjLAP作为抗原的ELISA法诊断血吸虫病具有良好的敏感性和特异性。 | Faustina Halm-Lai 罗庆礼 钟政荣 宋晓蓉 陈兆武 王林 胡梅梅 储德勇 沈继龙 | 2011 | 中国寄生虫学与寄生虫病杂志2011,29,5: | 1 |
| 8 | 刚地弓形虫非洲分离株的PCR-RFLP和微卫星基因分型研究进展(英文)显示文摘刚地弓形虫(T.gondii)的遗传多样性与地理区域相关。北美和欧洲的弓形虫基因型以II型和III型为主。南美洲以非典型基因型为主。亚洲弓形虫的优势基因型主要为Chinese 1型(ToxoDB#9)和I型。目前关于非洲弓形虫基因型的研究较少。本文对源于北非、西非、东非和中非地区弓形虫的多位点PCR-RFLP和微卫星分型以及非洲地区弓形虫群体遗传结构进行了综述。从17例患者中分离出弓形虫,非典型基因型13株(76.5%),主要为Africa 1 9株(69.2%);I型1株(5.9%)以及混合基因型3株(17.6%)。从1660饲养动物中分离出314株,其中II型134株(42.7%),III型103株(32.8%),非典型基因型61株(19.4%),I型6株(1.9%)和混合基因型10株(3.2%)。人畜弓形虫基因型的比例为分别为:II型40.5%(134/331),III型31.1%(103/331),非典型基因型22.3%(74/331),I型2.1%(7/331)和混合基因型3.9%(13/331)。典型基因型占73.7%。综上可知,非洲弓形虫基因型包括典型和非典型基因型,前者在非洲人群与动物中均有较高的感染率。 | Faustina PAPPOE 王林 Dorcas OBIRI-YEBOAH 陈鹤 程维晟 王晓玮 刘芳 罗庆礼 沈继龙 | 2016 | 中国人兽共患病学报2016,32,10: | 1 |

Methods:Blood samples collected from the 193 randomly recruited participants were used for HIV screening,Plasmodium falciparum antigen screening,malaria parasite density count, CD4^+ T cell count,glutathione reductase,glutathione peroxidase and total antioxidant status measurement.Standard laboratory methods were used for the analysis.Results:The results showed that glutathione reductase,glutathione peroxidase,total antioxidant status and CD4^+ T cell count were significantly lowered in symptomatic HIV participants with and without malaria co-infection(P<0.01) in each case compared with control participants.Also,glutathione reductase,glutathione peroxidise,total antioxidant status and CD4^+ T cell count were significantly lowered in asymptomatic HIV participants with and without malaria co-infection(P<0.05) in each case,compared with control participants without malaria.Similarly,these antioxidants were significandy lowered in control participants with malaria infection(P<0.05) compared with control participants without malaria.The malaria parasite density in symptomatic HIV infected participants was negatively associated with glutathione reductase(r = -0.906,P<0.01),glutathione peroxidase(r = -0.719,P<0.01) and total antioxidant status(r = -0.824,P<0.01).Conclusions: The antioxidant activity was affected in HIV infected participants with malaria co-infeclion.
