| 1 | RAPD analysis of wild stock of penaeid shrimp (Penaeus chinensis )in Chinese coastal waters of the Huanghai Sea and coastal waters of the Bohai Sea显示文摘Genetic diversity of 32 individuals of P. chinensis in the Chinese coastal waters of the Huanghai and Bobal Seas was detected by RAPD technique. Twenty decamer primers of OPI were used for DNA amplification for each individual. The results showed that: Seventeen primers obtained reproducible fingerprints, and the bands were clear. Thirty--nine of 106 loci detected were polymorphic, amounting to 36. 8 %. | 3 | The single nucleotide polymorphisms of chicken melanocortin-4 receptor(MC4R)gene and their association analysis with carcass traits显示文摘Mutations of the melanocortin-4 receptor (MC4R) gene are associated with the appetite,obesity and growth in pig,mice and human. But little is known about the function of chicken MC4R gene. In this study,F2 chicken resource population derived from broilers crossing to Silky was screened for the polymorphisms of the MC4R gene using PCR-single strand conformation polymor-phism (PCR-SSCP) and DNA sequencing methods. Four single nucleotide polymorphisms (SNPs) sites were found. The mutation (C→T) in the 5′ regulation region of chicken MC4R gene results in one more NF-E2 and cap transcription factor binding sites in the mutation allele than in the wild allele. One missense mutation (G→A) occurs in the coding region (61nt),which changes the glycine to arginine. Moreover,in the coding region there are 2 synonymous mutations,one G→T mutation at 315nt and one C→T mutation at 336nt. Least square analysis of the SNPs and carcass traits showed that BB,DD and FF genotypes are significantly associated with body weight,carcass weight (or half carcass weight),and leg muscle weight (P<0.05 or P<0.01). But no significant association between the geno-types and abdominal fat weight is found. | 4 | Environmental Implications of Excessive Selenium: A Review显示文摘Selenium is a naturally occurring trace element that is nutritionally required in smallamounts but it can become toxic at concentrations only twice those required. The narrow mar-gin between beneficial and harmful levels has important implications for human activities thatincrease the amount of selenium in the environment. Two of these activities, disposal of fbssilfuel wastes and agricultural irrigation of arid, seleniferous soils, have poisoned fish andwildlife, and threatened public health at several locations in the United States. Research stud-ies of these episodes have generated a data base that clearly illustrates the environmental hazardof excessive selenium. It is strongly bioaccumulated by aquatic organisms and even slight in-creases in waterborne concentrations can quickly result in toxic effects such as deformed em-bryos and reproductive failure in wildlife. The selenium data base has been very beneficial indeveloping hazard assessment procedures and establishing environmentally sound water qualitycriteria. The two faces of selenium, required nutrient and Potent toxin, make it a particularlyimportant trace element in the health of both animals and man. | 5 | Establishment of trophic continuum in the food web of the Yellow Sea and East China Sea ecosystem: Insight from carbon and nitrogen stable isotopes显示文摘Stable carbon and nitrogen isotope ratios (δ13C and δ15N) are used to study the trophic structure of food web in the Yellow Sea and East China Sea ecosystem. The trophic con-tinuum of pelagic food web from phytoplankton to top preyer was elementarily established, and a trophic structure diagram in the Yellow Sea and East China Sea was outlined in combination with carbon isotopic data of benthic organisms, which is basically consistent with and makes some improvements on the simplified Yellow Sea food web and the trophic structure diagram drawn based on the biomass of main resource population during 1985―1986. | 6 | Identification of Porphyra lines using computerized DNA fingerprinting显示文摘RAPD (Randomly Amplified Polymorphic DNA) analysis was performed with filaments of 15 Porphyra lines representing four important groups (P. yezoensis, P. haitanensis, P. katadai var. Hemiphylla and P. digospermatangia). Eight stable and repeatable RAPD bands amplified with two primers, OPN-02 and OPJ-18, were selected for the construction of DNA fingerprinting. The RAPD results were scored based on the presence or absence of each of the 8 bands and then converted to computer language expressed with two digitals, 1 and 0, which represented the presence (numbered as 1) or absence (numbered as 0) of each band, respectively. Based on these results, a model DNA fingerprint and a computerized DNA fingerprint were constructed. In the constructed DNA fingerprint, each Porphyra line has its unique fingerprinting pattern and can be easily distinguished from each other. Later, a software, named as PhGI, was designed based on this DNA fingerprinting. | 7 | Analysis of tandem repeats in the genome of Chinese shrimp Fenneropenaeus chinensis显示文摘Through random sequencing, we found a total of 884000 base-pairs (bp) of random genomic sequences in the genome of Chinese shrimp (Fenneropenaeus chinensis). Using bio-soft Tandem Repeat Finder (TRF) software, 2159 tandem repeats were found, in which there were 1714 mi- crosatellites and 445 minisatellites, accounting for 79.4% and 20.6% of repeat sequences, respectively. The cumulative length of repeat sequences was found to be 116685 bp, ac- counting for 13.2% of the total DNA sequence; the cumula- tive length of microsatellites occupied 9.78% of the total DNA sequence, and that of minisatellites occupied 3.42%. In decreasing order, the 20 most abundant repeat sequence classes were as follows: AT (557), AC (471), AG (274), AAT (92), A (56), AAG (28), ATC (27), ATAG (27), AGG (18), ACT (15), C (11), AAC (11), ACAT (11), CAGA (10), AGAA (9), AGGG (7), CAAA (7), CGCA (6), ATAA (6), AGAGAA (6). Dinucleotide repeats, not only in the aspect of the number, but also in cumulative length, were the preponderant repeat type. There were few classes and low copy numbers of repeat units of the pentanucleotide repeat type, which included only three classes: AGAGA, GAGGC and AAAGA. | 10 | Kuroshio warm filament and the source of the warm water of the Tsushima Current显示文摘Characteristics and evolution of the Kuroshio frontal eddies and warm filaments are analyzed according to two series of satellite images (March 5 to 7, 1986 and April 14 to 16, 1988). The results show that the frontal eddies in the East China Sea are generated at the shelf break and move along the continental slope at a speed of 15 cm/s with the Kuroshio. The frontal eddies occur about every 10 d and evolve to be warm filaments a few hundred km in length and 30-40 km in width in the area west of the Yaku-shima. Meanwhile, the existence of the warm filament was also found in the area by analysing the hydrographic data in the area west of Kyushu during May 24-June 5, 1988.The Kuroshio warm filaments move westward opposite to the Kuroshio and then turn northward at the shelf break and become the main source of the warm water of the Tsushima Warm Current. Stock K was from the spawning stock of the western coastal waters of the Korea Peninsula, H was from the Huanghai Sea—Bohai Sea stock and S was from the health cultured stock in Jimo, Shandong Province, China. | 13 | Study on low-temperature lipase of psychrophilic bacterium 2-5-10-1 isolated from deep sea of Southern Ocean显示文摘A strain of psychrophilic bacterium, 2-5-10-1, which produces low-temperature lipase, is isolated from the deep sea of Prydz Bay in Southern Ocean. The highest lipase secretion of this strain is observed at 5 ℃ and this temperature is also for optimal growth. Tween 80 and olive oil enhance secretion of lipase. The optimal temperature and pH for lipase activity are 35 ℃ and 7.5 ℃ respectively. At 0℃, the lipase still has 37% relative enzyme activity. The lipase shows high thermolability, more than 50% activity lost after incubation at 60 ℃ for 15 min. EDTA has no effect on lipase activity, indicating the lipase activity is independent of divalent cation. To determine whether they could yield10-Hydroxy-2-decenoic acid during fermentation, gas chromatography and gas chromatography-mass spectrometry methodswere used. The results showed that the strains BH002 and BH004, were both identified as Crytococcaceae, where BH002was primarily classified into Candida for possessing the abilities. | 17 | Hepatic Histopathological Characteristics and Antioxidant Response of Phytoplanktivorous Silver Carp Intraperitoneally Injected with Extracted Microcystins显示文摘Objective To investigate the hispathological characteristics and antioxidant responses in liver of silver carp after intraperitoneal administration of microcystins (MCs) for further understanding hepatic intoxication and antioxidation mechanism in fish. Methods Phytoplanktivorous silver carp was injected intraperitoneally (i.p.) with extracted hepatotoxic microcystins (mainly MC-RR and -LR) at a dose of 1000 μg MC-LReq./kg body weight, and liver histopathological changes and antioxidant responses were studied at 1, 3, 12, 24, and 48 h, respectively, after injection. Results The damage to liver structure and the activities of hepatic antioxidant enzymes including catalase (CAT), superoxide dismutase (SOD), and glutathione peroxide (GPX) were increased in a time-dependent manner. Conclusion In terms of clinical and histological signs of intoxication and LD50 (i.p.) dose of MC-LR, silver carp appears rather resistant to MCs exposure than other fishes. | 18 | Chemical induction of settlement and metamorphosis in two Babylonia (Gastropoda) larvae显示文摘By exposure to 12 x 10 3 mol/dm3 K+ for 24 h, more than 90% of planktonic larvae of Babylonia formosae and B. areolata were effectively induced to metamorphose completely. 15 x 10 - 3 mol/dm3 K+ exerted toxic effect on larval B. areolata over a 24-h period, but produced better results of induction by 12 h exposure. Metamorphosed juveniles induced by increased by K’ could move and feed normally. Percent metamorphosis increased with average shell height for K + -induced larvae. L-DOPA, epinephrine and norepinephrine ranging from 5. 5 x 10 -- 6 to 5. 5 x 10 - 5 mol/dm3 had no inductive activity to metamorphosis in larval B. areolata, whereas the rate of metamorphosis induced by dopamine at a concentration of 10--4 mol/dm3 was approximately half less than that by 12 x 10 3 mol/dm3 K+. TEA (a K+ -channel blocker) did not inhibit metamorphosis in the two Babylonia larvae by induction of K+. | 20 | Karyotypes of three species of the marine Veneroida molluscs显示文摘The karyotypes of three species of marine Veneroida molluscs, Solen grandis Dunker, Saxidomus purpuratus Sowerby and Mactra chinensis Philippi were studied by using the adult gill tissues. The chromosomes were prepared through injecting phytohemagglutinin (PHA) and colchicine, hypotonic treatment, chopping air-dry, and squashing technology. The results show that the diploid chromosome numbers of the three Veneroida species are the same as 2n=38. The karyotype of Solen grandis is 26m+6sm+2st+4t, NF=70, the karyotype of Saxidomus purpuratus is 32m+2sm+4st/t, NF=72, and the karyotype of Mactra chinensis is 20m+16sm+2st/t, NF=74.
