| 1 | Effect of Trichoderma viride biofertilizer on ammonia volatilization from an alkaline soil in Northern China显示文摘Ammonia(NH3) volatilization is one of the primary pathways of nitrogen(N) loss from soils after chemical fertilizer is applied, especially from the alkaline soils in Northern China, which results in lower efficiency for chemical fertilizers. Therefore, we conducted an incubation experiment using an alkaline soil from Tianjin(p H 8.37–8.43) to evaluate the suppression effect of Trichoderma viride(T. viride) biofertilizer on NH3 volatilization, and compared the differences in microbial community structure among all samples. The results showed that viable T. viride biofertilizer(T) decreased NH_3 volatilization by 42.21% compared with conventional fertilizer((CK), urea), while nonviable T. viride biofertilizer(TS) decreased NH_3 volatilization by 32.42%. NH_3 volatilization was significantly higher in CK and sweet potato starch wastewater(SPSW) treatments during the peak period. T. viride biofertilizer also improved the transfer of ammonium from soil to sweet sorghum. Plant dry weights increased 91.23% and 61.08% for T and TS, respectively, compared to CK. Moreover, T. viride biofertilizer enhanced nitrification by increasing the abundance of ammonium-oxidizing archaea(AOA) and ammonium-oxidizing bacteria(AOB). The results of high-throughput sequencing indicated that the microbial community structure and composition were significantly changed by the application of T. viride biofertilizer. | 4 | Dietary oregano essential oil supplementation improves intestinal functions and alters gut microbiota in latephase laying hens显示文摘Background:Dietary essential oil(EO)supplementation can exert favorable effects on gut health in broilers.However,it is unknown whether EO could improve intestinal functions,consequently beneficial for egg performance and quality in late-phase laying hens.This study was aimed to investigate the potential effects of EO on production performance,egg quality,intestinal health and ileal microbiota of hens in the late phase of production.A total of 28860-week-old Hy-line Brown laying hens were randomly divided into 4 groups and fed a basal diet(control)or basal diets supplemented with oregano EO at 100,200 and 400 mg/kg(EO100,EO200 and EO400).Results:Dietary EO supplementation resulted in a quadratic decrease(P<0.05)in feed conversion ratio with lower(P<0.05)feed conversion ratio in EO200 group than the control during weeks 9–12 and 1–12 of the trial.Compared to the control,EO addition resulted in higher(P<0.05)eggshell thickness at the end of week.4,8 and 12 and higher(P<0.05)chymotrypsin activity.There was a quadratic elevation(P<0.05)in ileal chymotrypsin and lipase activity,along with a linear increase in villus height to crypt depth ratio.Quadratic declines(P<0.05)in mRNA expression of IL-1β,TNF-α,IFN-γ and TLR-4,concurrent with a linear and quadratic increase(P<0.05)in ZO-1 expression were identified in the ileum with EO addition.These favorable effects were maximized at medium dosage(200 mg/kg)of EO addition and intestinal microbial composition in the control and EO200 groups were assessed.Dietary EO addition increased(P<0.05)the abundances of Burkholderiales,Actinobacteria,Bifidobacteriales,Enterococcaceae and Bacillaceae,whereas decreased Shigella abundance in the ileum.Conclusions:Dietary EO addition could enhance digestive enzyme activity,improve gut morphology,epithelial barrier functions and modulate mucosal immune status by altering microbial composition,thus favoring feed efficiency and eggshell quality of late-phase laying hens. | Jia Feng Mingyuan Lu Jing Wang Haijun Zhang Kai Qiu Guanghai Qi Shugeng Wu | 2022 | Journal of Animal Science and Biotechnology2022,13,1: | 6 |
| 5 | Associations of PGK1 promoter hypomethylation and PGK1-mediated PDHK1 phosphorylation with cancer stage and prognosis:a TCGA pan-cancer analysis显示文摘Background:Cancer cells reprogram metabolism for proliferation.Phosphoglycerate kinase 1(PGK1),as a glycolytic enzyme and newly identified protein kinase,coordinates glycolysis and mitochondrial metabolism.However,the clini-cal significance of PGK1 expression and function in cancer progression is unclear.Here,we investigated the relation-ship between the progression and prognosis of multiple cancer types and PGK1 expression and its function in the mitochondrial metabolism regulation.Methods:We performed pan-cancer analyses of PGK1 mRNA level and DNA methylation in 11,908 tumor tissues and 1582 paired normal tissues across 34 cancer types in The Cancer Genome Atlas datasets.Using specific antibodies against PGK1 S203 and PDHK1 T338 phosphorylation,we performed immunohistochemistry with tissue microarray assay in additional 818 cancer cases with 619 paired normal tissues from five cancer types.Results:The PGK1 mRNA level was significantly elevated with hypomethylation in promotor regions and associated with advanced TNM stage in 15 and four cancer types,respectively.In breast carcinoma,elevated PGK1 mRNA level and promoter hypomethylation were associated with poor prognosis.Positively correlated PGK1 S203 and PDHK1 T338 phosphorylation levels were significantly associated with short overall survival(OS)in cancers of the breast,liver,lung,stomach,and esophagus and with advanced TNM stage in breast and esophageal cancers.PGK1 pS203 and PDHK1 pT338 were also independent predictors of short OS in liver,lung,and stomach cancer.Conclusions:The elevated expression,promoter hypomethylation,and phosphorylation of PGK1 and PDHK1 were related with disease progression and short OS in diverse types of cancer.PGK1 and PDHK1 phosphorylation may be potential prognostic biomarkers. | Fei Shao Xueying Yang Wei Wang Juhong Wang Wei Guo Xiaoli Feng Susheng Shi Qi Xue Shugeng Gao Yibo Gao Zhimin Lu Jie He | 2019 | Cancer Communications2019,39,1: | 4 |

| 8 | Uterine inflammation status modulates eggshell mineralization via calcium transport and matrix protein synthesis in laying hens显示文摘This study explored the effects of uterine inflammation on eggshell mineralization,ultrastructure and mechanical properties in laying hens modified by a lipopolysaccharide(LPS)challenge or dietary essential oil(EO)addition.In trial 1,a total of 72 Hy-line Brown layers at 36 wk of age were randomly assigned to 3 treatment groups(n=8),where they were intravenously injected with phosphate buffered saline,LPS at 1 mg/kg body weight,or LPS 3 times at 24-h intervals.In trial 2,a total of 288 Hy-line Brown layers at 60 wk of age were randomly divided into 4 groups(n=8),where they were fed basal diets supplemented with EO at 0,50,100 and 200 mg/kg for 12 wk.A uterine inflammation model was constructed with LPS treatment,indicated by the elevated expression of IL-1β and TNF-α(P<0.05)and lymphocyte infiltration.Uterine inflammation caused remarkable decreases in eggshell thickness and mechanical properties with structure deteriorations(P<0.05).Uterine inflammation stimulated the expression of matrix proteins ovotransferrin(TF)and ovalbumin(OVAL),while depressing the mRNA levels of calbindin-1(CALB1)and osteopontin in uterine mucosa(P<0.05).In contrast,EO addition alleviated uterine inflammation,evidenced by depressed levels of IL-1b and IL-6(P<0.05).There was a significant elevation in shell thickness and breaking strength following EO intervention(P<0.05),and these effects were maximized at addition of 100 mg/kg.Further,EO improved shell ultrastructure including more early fusion,less type B mammillae,and increased effective thickness(P<0.05).The alleviated inflammation decreased the expression of OVAL and TF,whereas ion transport genes like CALB1 and solute carrier family 26 member 9 were upregulated(P<0.05).Our findings suggest that inflam-matory status can impact uterine functions in calcium transport and the synthesis of matrix proteins especially such as OVAL and TF,which in turn modulates calcium precipitation and ultrastructure for-mation,thereby determining eggshell mechanical properties.These findings provide a novel insight into the uterine inflammation-mediated modifications of eggshell quality. | Jia Feng Mingyuan Lu Lingling Ma Haijun Zhang Shugeng Wu Kai Qiu Yuna Min Guanghai Qi Jing Wang | 2023 | Animal Nutrition2023,,2: | 1 |
