1Bone morphogenetic protein-2 gene controls tooth root development in coordination with formation of the periodontium显示文摘Formation of the periodontium begins following onset of tooth-root formation in a coordinated manner after birth. Dental follicle progenitor cells are thought to form the cementum, alveolar bone and Sharpey's fibers of the periodontal ligament (PDL). However, little is known about the regulatory morphogens that control differentiation and function of these progenitor cells, as well as the progenitor cells involved in crown and root formation. We investigated the role of bone morphogenetic protein-2 (Bmp2) in these processes by the conditional removal of the Bmp2 gene using the Sp7-Cre-EGFP mouse model. Sp7-Cre-EGFP first becomes active at E18 in the first molar, with robust Cre activity at postnatal day 0 (P0), followed by Cre activity in the second molar, which occurs after P0. There is robust Cre activity in the periodontium and third molars by 2 weeks of age. When the Bmp2 gene is removed from Sp7+ (Osterix+) cells, major defects are noted in root, cellular cementum and periodontium formation. First, there are major cell autonomous defects in root-odontoblast terminal differentiation. Second, there are major alterations in formation of the PDLs and cellular cementum, correlated with decreased nuclear factor IC (Nfic), periostin and α-SMA+ cells. Third, there is a failure to produce vascular endothelial growth factor A (VEGF-A) in the periodontium and the pulp leading to decreased formation of the microvascular and associated candidate stem cells in the Bmp2-cKOSp7-Cre-EGFP . Fourth, ameloblast function and enamel formation are indirectly altered in the Bmp2-cKOSp7-Cre-EGFP . 3The biological function of type I receptors of bone morphogenetic protein in bone显示文摘Bone morphogenetic proteins(BMPs) have multiple roles in skeletal development, homeostasis and regeneration. BMPs signal via type I and type II serine/threonine kinase receptors(BMPRI and BMPRII). In recent decades, genetic studies in humans and mice have demonstrated that perturbations in BMP signaling via BMPRI resulted in various diseases in bone, cartilage, and muscles. In this review, we focus on all three types of BMPRI, which consist of activin-like kinase 2(ALK2, also called type IA activin receptor), activinlike kinase 3(ALK3, also called BMPRIA), and activin-like kinase 6(ALK6, also called BMPRIB). The research areas covered include the current progress regarding the roles of these receptors during myogenesis,chondrogenesis, and osteogenesis. 4Expression analysis of a-smooth muscle actin and tenascin-C in the periodontal ligament under orthodontic loading or in vitro culture显示文摘α-smooth muscle actin(α-SMA) and tenascin-C are stress-induced phenotypic features of myofibroblasts. The expression levels of these two proteins closely correlate with the extracellular mechanical microenvironment. We investigated how the expression of α-SMA and tenascin-C was altered in the periodontal ligament(PDL) under orthodontic loading to indirectly reveal the intrinsic mechanical microenvironment in the PDL. In this study, we demonstrated the synergistic effects of transforming growth factor-β1(TGF-β1) and mechanical tensile or compressive stress on myofibroblast differentiation from human periodontal ligament cells(h PDLCs). The h PDLCs under higher tensile or compressive stress significantly increased their levels of α-SMA and tenascin-C compared with those under lower tensile or compressive stress. A similar trend was observed in the tension and compression areas of the PDL under continuous light or heavy orthodontic load in rats. During the time-course analysis of expression, we observed that an increase in α-SMA levels was matched by an increase in tenascin-C levels in the PDL under orthodontic load in vivo. 6全颗粒饲料对奶牛生产性能的影响显示文摘饲料中的中性洗涤纤维(NDF)含量和饲料颗粒直径是影响瘤胃功能的重要因素。试验评估了以颗粒饲料为主的奶牛瘤胃健康情况、NDF消化率和生产性能。选取8头荷斯坦母牛(336±30 d,体重346±35 kg),随机分配到交叉重复试验组中,奶牛安置于牛栏,自由采食。试验期为3周,前两周是适应期,最后一周为数据收集期。各组饲粮原料相同,但物理形式不同,分为全混合日粮(TMR)和颗粒料(直径=8 mm)。两组间物理有效NDF(pe NDF)不同,TMR组为39.8%,颗粒料组为11.8%。在试验过程中,干物质采食量(DMI)、饮水量、瘤胃反刍时间、瘤胃温度和pH情况均记录。3周后收集粪样,确定潜在的可消化NDF总消化率(pdNDF)。每阶段结束后计算平均日增重和饲料转化率。颗粒料组的DMI、DMI/体重和饮水量更高。而两组间平均日增重和饲料转化率无显著差异。颗粒料组的瘤胃反刍时间为241 min·d-1低于TMR组的507 min·d-1。饲料的不同物理外形对瘤胃温度、瘤胃pH无显著影响。TMR组的pdNDF总消化率为90.25%高于颗粒料组的86.82%。试验结果表明,动物易接受完整的颗粒饲料,DMI较高,且反刍时间减少,但瘤胃pH无显著变化。TMR组的pdNDF消化率显著增加。尽管两组奶牛在DMI和纤维消化方面表现不同,但总体表现相似,两组饲料的保存时间与其物理形式有关。在短期内,提供能满足NDF需求的完全颗粒饲料可改善瘤胃健康状况和动物生产性能,而较长时间的数据则需要进一步试验。Wang J Yang M Cao M Lin Y Che L Q Duraipandiyan V Al-Dhabi N A Fang Z F Xu S Y Feng B Liu G Wu D Bonfante E Palmonari A Mammi L Canestrari G Fustini M Formigoni A 2016饲料博览2016,0,12:2
