1The effect of renal stones on serum adenosine aminohydrolase and AMP-aminohydrolase in Malaysia显示文摘Objective: To verify possible associations between adenosine aminohydrolase(ADA) and AMP-aminohydrolase(AMPDA) to E3 SUMO-protein ligase NSE2(NSMCE2) in patients with renal stones. And to isolate, purify and characterize ADA in patients with renal stones and healthy group.Methods: A total of 60 renal stones patients and 50 control were enrolled in a case-control study. The blood urea, creatinine, uric acid, protein, albumin, ADA and AMPDA were measured by colorimetric tests. The serum NSMCE2 was measured by ELISA.Results: Serum ADA, AMPDA and specii c activity of enzymes showed signii cant decrease(P < 0.05) in patients with renal stones compared to control group, mean levels of sera NSMCE2 and uric acid had a signii cant increase(P < 0.01 and P < 0.05, respectively) in patients compared to control group.Conclusions: The present study suggests that ADA, AMP deaminase and NSMCE2 can be used as a indicator to monitor the DNA damage and inl ammation disorders in the patients with kidney stones.Faridah Yusof Atheer Awad Mehde Wesen Adel Mehdi Hamid Ghazali Azlina Abd Rahman 2015Asian Pacific Journal of Tropical Biomedicine2015,5,6:3
2Identification of commonly regulated protein targets and molecular pathways in PC-3 and DU145 androgen-independent human prostate cancer cells treated with the curcumin analogue 1,5-bis(2-hydroxyphenyl)-1,4-pentadiene-3-one显示文摘Objective: To identify mutually regulated proteins in PC-3 and DU145 androgen-independent prostate cancer cell lines treated with 1,5-bis(2-hydroxyphenyl)-1,4-pentadiene-3-one(MS17), and to study the molecular pathways that contributed to the anticancer activity of MS17.Methods: PC-3 and DU145 cells were treated with 3 × EC_(50)(15 μM) concentration of MS17 for 24 h and were subjected to protein expression profiling using two-dimensional gel electrophoresis and protein identification by mass spectrometry.Selected differentially expressed proteins with significant P-value of P<0.05 and fold change over 1.5-folds were filtered through and ontologically classified.Mutually regulated proteins were ranked by fold change and identified as common protein targets of MS17.Results: Profiling data revealed that, the mutually down-regulated proteins included ACTB and ACTG associated with structural molecule activity, ACTN1 with cell cycle, ACTN4 with cell migration, HNRPK with apoptosis, PLST with morphogenesis and TERA with proteolysis.However, the expressions of CH60 and HS71 A respectively associated with response to unfolded protein demonstrated opposing regulation in PC-3 and DU145 cells.Pathway analysis of the differentially expressed proteins in PC-3 cells demonstrated the modulation of top pathways associated with cell-cell adhesion and cytoskeletal organization while in DU145 cells the pathways were associated with proteosomal degradation, regulation of electrolytes and water, regulation control of germ cells and organization of filament assembly/disassembly.Conclusions: The findings of the present study provide an understanding on the anti-tumorigenic activity of MS17 at the proteome level and warrant further research for its potential application for the management and treatment of androgen-independent prostate cancer.Kamini Citalingam Faridah Abas Nordin H.Lajis Iekhsan Othman Rakesh Naidu 2018Asian Pacific Journal of Tropical Biomedicine2018,8,9:2
3Toxicity studies for indigenous Bacillus thuringiensis isolates from Malang city,East Java on Aedes aegypti larvae显示文摘Objective:To investigate the toxicity of indigenous Bacillus thuringiensis(B.thuringiensis)isolates from Malang City for controlling Aedes aegypti(Ae.aegypti)larvae.Methods:Soil samples were taken from Purwantoro and Sawojajar sub-districts.Bacterial isolation was performed using B.thuringiensis selective media.Phenotypic characteristics of the isolates were obtained with the simple matching method.The growth and prevalence of spores were determined by the Total Plate Count method,and toxicity tests were also performed on the third instar larval stage of Ae.aegypti.The percentage of larval mortality was analysed using probit regression.The LC50was analysed by ANOVA,and the Tukey HSD interval was 95%.Results:Among the 33 selected bacterial isolates,six were obtained(PWR4-31,PWR4-32,SWJ4-2b,SWJ4-4b,SWJ-4k and SWJ5-1)that had a similar phenotype to reference B.thuringiensis.Based on the dendrogram,all of the bacterial isolates were 71%similar.Three isolates that had a higher prevalence of reference B.thuringiensis were PWR4-32,SWJ4-4b and SW5-1,of which the spore prevalence was 52.44%,23.59%,34.46%,respectively.These three indigenous isolates from Malang City successfully killed Ae.aegypti larvae.The PWR4-32 isolates were the most effective at killing the larvae.Conclusions:Six indigenous B.thuringiensis isolates among the 33 bacterial isolates found in the Sawojajar and Purwantoro sub-districts were toxic to the third instar larvae of Ae.aegypti.The PWR4-32 isolates were identical to tbe reference B.thuringiensis and had 88%phenotype similarity.The PWR4-32 isolates had the highest spore prevalence(52.44%),and the early stationary phase occurred at 36 h.The PWR4-32 isolates were the most effective at killing Ae.aegypti larvae(LC50-72 h=2.3×108 cells/mL).Zulfaidah Penata Gama Nobukazu Nakagoshi Suharjono Faridah Setyowati 2013Asian Pacific Journal of Tropical Biomedicine2013,3,2:2

10Identification of α-glucosidase inhibitors from Clinacanthus nutans leaf extract using liquid chromatography-mass spectrometry-based metabolomics and protein-ligand interaction with molecular docking显示文摘The present study used in vitro and in silico techniques, as well as the metabolomics approach to characterise α-glucosidase inhibitors from different fractions of Clinacanthus nutans. C. nutans is a medicinal plant belonging to the Acanthaceae family, and is traditionally used to treat diabetes in Malaysia. nHexane, n-hexane: ethyl acetate(1:1, v/v), ethyl acetate, ethyl acetate: methanol(1:1, v/v), and methanol fractions were obtained via partitioning of the 80% methanolic crude extract. The in vitro α-glucosidase inhibitory activity was analyzed using all the fractions collected, followed by profiling of the metabolites using liquid chromatography combined with mass spectrometry. The partial least square(PLS) statistical model was developed using the SIMCA P^+14.0 software and the following four inhibitors were obtained:(1) 4,6,8-Megastigmatrien-3-one;(2) N-Isobutyl-2-nonen-6,8-diynamide;(3) 1′,2′-bis(acetyloxy)-3′,4′-didehydro-2′-hydro-β, ψ-carotene; and(4) 22-acetate-3-hydroxy-21-(6-methyl-2,4-octadienoate)-olean-12-en-28-oic acid. The in silico study performed via molecular docking with the crystal structure of yeast isomaltase(PDB code: 3 A4 A) involved a hydrogen bond and some hydrophobic interactions between the inhibitors and protein.
