2The zinc transporter Slc39a5 controls glucose sensing and insulin secretion in pancreatic β-cells via Sirt1- and Pgc-1α-mediated regulation of Glut2显示文摘Zinc levels are high in pancreatic β-cells, and zinc is involved in the synthesis, processing and secretion of insulin in these cells. However, precisely how cellular zinc homeostasis is regulated in pancreatic β-cells is poorly understood. By screening the expression of 14 Slc39a metal importer family member genes, we found that the zinc transporter Slc39a5 is significantly downregulated in pancreatic β-cells in diabetic db/db mice, obese ob/ob mice and high-fat diet-fed mice. Moreover,β-cell-specific Slc39a5 knockout mice have impaired insulin secretion. In addition, Slc39a5-deficient pancreatic islets have reduced glucose tolerance accompanied by reduced expression of Pgc-1α and its downstream target gene Glut2. The down-regulation of Glut2 in Slc39a5-deficient islets was rescued using agonists of Sirt1, Pgc-1α and Ppar-γ. At the mechanistic level, we found that Slc39a5-mediated zinc influx induces Glut2 expression via Sirt1-mediated Pgc-1α activation. 5Two Arabidopsis Receptor-like Cytoplasmic Kinases SZE1 and SZE2 Associate with the ZAR1-ZED1 Complex and Are Required for Effector-Triggered Immunity显示文摘Plants utilize intracellular nucleotide-binding leucine-rich repeat domain-containing receptors (NLRs) to recognize pathogen effectors and induce a robust defense response named effector-triggered immunity (ETI). The Arabidopsis NLR protein HOPZ-ACTIVATED RESISTANCE 1 (ZAR1) forms a precomplex with HOPZ-ETI-DEFICIENT 1 (ZED1),a receptor-like cytoplasmic kinase (RLCK) XII-2 subfamily member, to recognize the Pseudomonas syringae effector HopZ1 a. We previously described a dominant mutant of Arabi-dopsis ZED1, zed1-D, which displays temperature-sensitive autoimmunity in a ZAR1-dependent manner. Here, we report that the RLCKs SUPPRESSOR OF ZED1-D1 (SZE1) and SZE2 associate with the ZAR1-ZED1 complex and are required for the ZED7-D-activated autoimmune response and HopZ1a-triggered immunity. We show that SZE1 but not SZE2 has autophosphorylation activity, and that the N-terminal myristoylation of both SZE1 and SZE2 is critical for their plasma membrane localization and ZED1-D-activated autoimmunity. Furthermore, we demonstrate that SZE1 and SZE2 both interact with ZAR1 to form ja functional complex and are required for resistance against P. syringae pv. tomato DC3000 ex-pressing HopZIa. We also provide evidence that SZE1 and SZE2 interact with HopZ1a and function together with ZED1 to change the intramolecular interactions of ZAR1, leading to its activation. 18Tintinnid community throughout the Bohai Sea during the spring显示文摘As one of the important microzooplankton in marine pelagic habitats,tintinnids are classified as neritic genera,oceanic genera and cosmopolitan genera.Until now,we know little about the interaction between neritic and oceanic genera in continental shelves.Low species richness area was found in the mixing area between neritic and oceanic genera in the East China Sea.In this study,we investigated the spatial distribution of tintinnid community throughout the entire Bohai Sea in May(spring)of 2014 to find out:(1)whether the tintinnid assembalges differed in the three major bays bordering this shallow inland sea;(2)whether there was low species richness area in the Bohai Sea.Three genera and 11 species attributed to neritic and cosmopolitan taxa were reported from this region.Tintinnid abundance averaged(81±216)ind./L(0–1234 ind./L).Tintinnid community differed within the three major bays,with each characterized by different tintinnid taxa.Referring to the average abundance of three bays,tintinnids were most abundant in the Laizhou Bay((328±445)ind./L),and least abundant in the Liaodong Bay((34±57)ind./L).A low species richness area occurred in the central Bohai Sea(20–40 m isobaths),in some stations of which no tintinnid was recorded.Our study demonstrates that tintinnid community varied greatly over small spatial scales,and low species richness area was found in the mixing area between neritic and oceanic genera within this inland sea.Ying Yu Feng Zhou Wuchang Zhang Chen Wei Qunshan Wang Enjun Fang 2020Acta Oceanologica Sinica2020,39,6:0

20Targeting the LSD1-G9a-ER Stress Pathway as a Novel Therapeutic Strategy for Esophageal Squamous Cell Carcinoma显示文摘Despite recent advances in the management and treatment of esophageal squamous cell carcinoma(ESCC),the prognosis remains extremely poor,and current nonsurgical treatment options are limited.To identify new therapeutic targets,we screened a curated library of epigenetic compounds using a panel of cancer cell lines and found that coinhibiting the histone demethylase LSD1 and the histone methyltransferase G9a potently suppresses cell growth;similar results were obtained by knocking down both LSD1 and G9a expression.Importantly,we also found that inhibiting LSD1 and G9a significantly decreased tumor growth in a xenograft mouse model with ESCC cell lines.To examine the clinical relevance of these findings,we performed immunohistochemical analyses of microarray profiling data obtained from human esophageal squamous cancer tissues and found that both LSD1 and G9a are upregulated in cancer tissues compared to healthy tissues,and this increased expression was significantly correlated with poor prognosis.Mechanistically,we discovered that inhibiting LSD1 and G9a induces cell death via S-phase arrest and apoptosis,and cotargeting ER stress pathways increased this effect both in vitro and in vivo.Taken together,these findings provide compelling evidence that targeting LSD1,G9a,and ER stress-related pathways may serve as a viable therapeutic strategy for ESCC.Hongxiao Wang Zijun Song Enjun Xie Junyi Chen Biyao Tang Fudi Wang Junxia Min 2022Research2022,,3:0
