1Comparative transcriptome analysis during early fruit development between three seedy citrus genotypes and their seedless mutants显示文摘Identification of genes with differential transcript abundance(GDTA)in seedless mutants may enhance understanding of seedless citrus development.Transcriptome analysis was conducted at three time points during early fruit development(Phase 1)of three seedy citrus genotypes:Fallglo(Bower citrus hybrid(Citrus reticulata×C.reticulata×C.paradisi)×Temple(C.reticulata×C.sinensis)),grapefruit(C.paradisi),Pineapple sweet orange(C.sinensis),and their seedless mutants.Seed abortion in seedless mutants was observed at 26 days post anthesis(Time point 2).Affymetrix transcriptomic analysis revealed 359 to 1077 probe sets with differential transcript abundance in the comparison of seedless versus seedy fruits for each citrus genotypes and time points.The GDTA identified by 18 microarray probe sets were validated by qPCR.Hierarchical clustering analysis revealed a range of GDTA associated with development,hormone and protein metabolism,all of which may reflect genes associated with seedless fruit development.There were 14,9 and 12 genes found exhibiting similar abundance ratios in all three seedless versus seedy genotype comparisons at time point 1,2 and 3,respectively.Among those genes were genes coding for an aspartic protease and a cysteine protease,which may play important roles in seedless fruit development.New insights into seedless citrus fruit development may contribute to biotech approaches to create seedless cultivars.Shujian Zhang Qingchun Shi Ute Albrecht Robert G Shatters Jr Ric Stange Greg McCollum Shuo Zhang Chengming Fan Ed Stover 2017Horticulture Research2017,4,1:2

6Hexaacetyl-chitohexaose,a chitin-derived oligosaccharide,transiently activates citrus defenses and alters the feeding behavior of Asian citrus psyllid显示文摘Plants have a perception system triggered by pathogen and pest signals to initiate defense.These signals include evolutionarily conserved molecules from microbes and insects termed pathogen/herbivore-associated molecular patterns(PAMPs/HAMPs).Here we showed that hexaacetyl-chitohexaose(HC),an oligosaccharide from chitin,a structural component in insect exoskeletons and fungi cell walls,upregulated defense-associated genes WRKY22,GST1,RAR1,EDS1,PAL1 and NPR2,and downregulated ICS1 at 1 h after HC treatment in Sun Chu Sha mandarin leaves.The effect was transient as defense gene transcriptional changes were not observed at 18 h after the treatment.Electrical penetration graph(EPG)recordings were used to study the feeding behavior of Asian citrus psyllid(ACP)following the HC treatment.ACP is the hemipteran vector of Candidatus Liberibacter asiaticus(CLas),the pathogen associated with huanglongbing(HLB).Adult ACP displayed reduced intercellular probing,reduced xylem feeding count and duration,and increased non-probing activity on HC-treated citrus compared to controls.During an 18-h recording,percentage for total duration of xylem ingestion,phloem ingestion,intercellular probing were lower,and the percentage of nonprobing behavior was higher in HC-treated leaves than in controls.In host-selection behavior studies,HC treatment did not alter the attractiveness of citrus leaves under light or dark conditions.In addition,ACP feeding on HC-treated leaves did not show differences in mortality for up to 10 day of exposure.In summary,we report that HC induced a transient defense in citrus and an inhibitory effect on ACP feeding but did not affect host selection or the insect fitness under the tested conditions.Qingchun Shi Justin George Joseph Krystel Shujian Zhang Stephen L.Lapointe Lukasz L.Stelinski Ed Stover 2019Horticulture Research2019,6,1:0
7Mapping of QTLs and candidate genes associated with multiple phenotypic traits for Huanglongbing tolerance in citrus显示文摘Huanglongbing(HLB)is the most devastating disease for citrus worldwide.Candidatus Liberibacter asiaticus(C Las),vectored by Asian citrus psyllid(ACP,Diaphorina citri Kuwayama),is the most common pathogen causing the disease.Commercial citrus varieties are highly susceptible to HLB,whereas trifoliate orange(Poncirus trifoliata)is considered highly tolerant to HLB.An F1 segregating population and their parent trifoliate orange and sweet orange,which had been exposed to intense HLB pressure for three years,was evaluated for disease symptoms,ACP colonization,C Las titer and tree vigor repeatedly for two to three years.Trifoliate orange and sweet orange showed significant differences for most of the phenotypic traits,and the F1 population exhibited a large variation.A high-density SNP-based genetic map with 1402 markers was constructed for trifoliate orange,which exhibited high synteny and high coverage of its reference genome.A total of 26 quantitative trait locus(QTLs)were identified in four linkage groups LG-t6,LG-t7,LG-t8 and LG-t9,of which four QTL clusters exhibit a clear co-localization of QTLs associated with different traits.Through genome-wide analysis of gene expression in response to C Las infection in‘Flying Dragon’and‘Larger-Flower DPI-50-7’trifoliate orange,85 differentially expressed genes were found located within the QTL clusters.Among them,seven genes were classified as defense or immunity protein which exhibited the highest transcriptional change after C Las infection.Our results indicate a quantitative genetic nature of HLB tolerance and identified candidate genes that should be valuable for searching for genetic solutions to HLB through breeding or genetic engineering.Ming Huang Mikeal L. 8Beneficial horticultural responses from the application of solar thermotherapy to mature Huanglongbing-affected citrus trees显示文摘The detrimental effects of Huanglongbing(HLB)on citrus are well known and so is the need for effective methods to combat this disease.Solar thermotherapy(ST),one of the management methods to help alleviate some of the negative effects of HLB,is the process of heating trees to therapeutic temperatures by encompassing the tree within a plastic structure(Visqueen)to harness the sun’s natural energy.ST was applied to mature‘Valencia’citrus trees in three locations.Tree vigor,yield,and Candidatus Liberibacter asiaticus(Las)titer in leaves were monitored for two consecutive seasons post ST treatment,while fruit and juice quality were evaluated at the end of the second season.ST promoted an increase in canopy density in most groves tested and did not have a significant effect on fruit quantity,despite the prolonged exposure of the trees to increased temperatures.Moreover,Las titer was reduced in both leaves and juice processed from affected trees post ST.With respect to fruit quality,the ratio of total soluble solids to titratable acidity was higher post ST compared to controls as was the sucrose content of the juice in most treated groves,whereas limonin was higher in juice from control trees.Principal components analysis of aroma volatiles showed significant differences between juice from treated versus control trees with volatiles imparting top-note and freshness to orange juice,such as acetaldehyde,hexanal,Z-3-hexenol and linalool,being higher in juice fromtreated trees.Taste panels confirmed that flavor differences existed,ultimately showing that ST resulted in improved juice flavor in well-managed groves.Although the effects of ST on the trees appear temporary,such outcomes emphasize its utility when used as part of an integrated management strategy for HLB-affected citrus,imparting beneficial horticultural responses with minimal to positive effects on subsequent juice flavor.Cheryl M.Armstrong Melissa S.Doud Weiqi Luo Smita Raithore Elizabeth A.Baldwin Wei Zhao Anne Plotto Jinhe Bai John A.Manthey Ed Stover Yongping Duan 2021Horticultural Plant Journal2021,7,5:0
9Rationale for reconsidering current regulations restricting use of hybrids in orange juice显示文摘Huanglongbing(HLB)is a disease that has devastated the Florida citrus industry,threatens the entire U.S.citrus industry,and globally is rapidly spreading.Florida’s citrus production is 90%sweet orange,which is quite sensitive to HLB.The heavy reliance on sweet orange for Florida citrus production makes the industry especially vulnerable to diseases that are damaging to this type of citrus.Furthermore,90%of Florida oranges are used in producing orange juice that is defined by a federal regulation known as the“orange juice standard”,specifying that at least 90%of“orange juice”must be derived from Citrus sinensis.Genomic analyses definitively reveal that sweet orange is not a true species,but just one of many introgression hybrids of C.reticulata and C.maxima,with phenotypic diversity resulting from accumulated mutations in this single hybrid,the“sweet orange”.No other fruit industry is limited by law to such a narrow genetic base.Fortunately,there are new citrus hybrids displaying reduced sensitivity to HLB,and in some cases they produce juice,alone or in blends,that consumers would recognize as“orange juice”.Reconsidering current regulations on orange juice standards may permit use of such hybrids in“orange juice”,providing greater latitude for commercialization of these hybrids,leading to higher-quality orange juice and a more sustainable Florida orange juice industry.Ed Stover Frederick G.Gmitter Jr Jude Grosser Elizabeth Baldwin Guohong Albert Wu Jinhe Bai Yu Wang Peter Chaires Juan Carlos Motamayor 2020Horticulture Research2020,7,1:0
