8桑叶黄酮对α-糖苷酶活性的影响显示文摘目的:观察桑叶提取物桑叶黄酮对α-糖苷酶活性的影响及其对小鼠餐后血糖的影响。方法:实验于2005-07/10在解放军总医院营养科实验室和动物中心实验室进行。在体外实验中,设立空白组、阿卡波糖不同含量组和桑叶黄酮不同含量组(终浓度为0,7.81,15.63,31.25,62.50,125.00,250.00mg/L)。以阿卡波糖为阳性对照、以无抑制剂时α-糖苷酶活性为100%,检测桑叶总黄酮、HP-20树脂吸附黄酮和非吸附黄酮对α-糖苷酶活性的影响。在体内实验中,将昆明小鼠40只,按血糖水平随机分为4组,即淀粉组、淀粉+HP-20吸附黄酮组、蔗糖组、蔗糖+HP-20吸附黄酮组,每组10只,分别给予淀粉(3g/kg)、淀粉(3g/kg)+HP-20吸附黄酮(500mg/kg)、蔗糖(1.5g/kg)以及蔗糖(1.5g/kg)+HP-20吸附黄酮(500mg/kg)。检测并比较1h、2h血糖水平。结果:①当阿卡波糖、桑叶总黄酮含量为0,7.81,15.63,31.25,62.50,125.00,250.00mg/L时,α-糖苷酶IC50=70.43,82.11mg/L。②在相同HP-20树脂吸附黄酮浓度条件下,α-糖苷酶活性分别为100.00%,94.61%,84.90%,63.04%,26.27%,11.37%和6.76%,其IC50=59.25mg/L;在相同HP-20树脂非吸附黄酮浓度条件下,α-糖苷酶活性分别为100.00%,90.55%,88.55%,85.91%,78.00%,69.91%和56.36%,其IC50=82.86mg/L。③在给予淀粉的糖耐量实验中,HP-20树脂吸附黄酮组1h后血糖水平明显低于其淀粉对照组[(9.06±1.10),(11.13±1.45)mmol/L,t=2.1009,P=0.00209];在给予蔗糖的糖耐量实验中,HP-20树脂吸附黄酮组1h后血糖水平也明显低于其蔗糖对照组[(9.88±1.01),(12.75±1.17)mmol/L,t=2.1009,P=0.000014]。结论:体外实验提示桑叶总黄酮、HP-20树脂吸附和非吸附黄酮均具有抑制α-糖苷酶活性的作用,其中HP-20吸附黄酮对糖苷酶的抑制作用强于阿卡波糖;体内实验也证实HP-20吸附黄酮能够抑制给予淀粉和蔗糖后1h血糖的升高。薛长勇 刘英华 张荣欣 郑子新 张月红 景洪江 张永 欧阳红 Eugene Chen 2007中国组织工程研究与临床康复2007,11,21:17
9Exploring gut microbes in human health and disease:Pushing the envelope显示文摘Humans have coevolved with their microbes over thousands of years,but this relationship,is now being dramatically affected by shifts in the collective human microbiome resulting from changes in the environment and societal norms.Resulting perturbations of intestinal host-microbe interactions can lead to miscues and altered host responses that increase the risk of pathogenic processes and promote“western”disorders such as inflammatory bowel diseases,cancers,obesity,diabetes,autism,and asthma.Given the current challenges and limitations in gene therapy,approaches that can reshape the gut microbiome represent a reasonable strategy for restoring the balance between host and microbes.In this review and commentary,we highlight recent progress in our understanding of the intestinal microbiome in the context of health and diseases,focusing on mechanistic concepts that underlie the complex relationships between host and microbes.Despite these gains,many challenges lie ahead that make it difficult to close the gap between the basic sciences and clinical application.We will discuss the potential therapeutic strategies that can be used to manipulate the gut microbiota,recognizing that the promise of pharmabiotics(“bugs to drugs”)is unlikely to be completely fulfilled without a greater understanding of enteric microbiota and its impact on mammalian physiology.By leveraging the knowledge gained through these studies,we will be prepared to enter the era of personalized medicine where clinical inventions can be custom-tailored to individual patients to achieve better outcomes.Jun Sun Eugene B.Chang 2014Genes & Diseases2014,1,2:17
10Angiotensin-receptor blockers as therapy for mild-to-moderate hypertension-associated non-alcoholic steatohepatitis显示文摘AIM: To evaluate insulin resistance, cytolysis and nonalcoholic steatohepatitis (NASH) score (NAS) using the Kleiner and Brunt criteria in 54 patients with NASH and mild-to-moderate hypertension, treated with telmisartan vs valsartan for 20 mo. METHODS: All patients met the NCEP-ATP TM(standard dose 20 mg o.d., n = 28). Treatment had to be taken daily at the same hour with no concomitant medication or alcohol consumption allowed. Neither the patient nor the medical staff was aware of treatment group allocation. Paired liver biopsies obtained at inclusion (visit 1) and end of treatment (EOT) were assessed by a single blinded pathologist, not aware of patient or treatment group. Blood pressure, BMI, ALT, AST, HOMA-IR, plasma triglycerides (TG) and total cholesterol (TC) were evaluated at inclusion and every 4 mo until EOT (visit 6). RESULTS: At EOT we noticed a significant decrease in ALT levels vs inclusion in all patients and this decrease did not differ significantly in group T vs group V. HOMA-IR significantly decreased at EOT vs inclusion in all patients but in group T, the mean HOMA-IR decrease per month was higher than in group V. NAS significantly diminished at EOT in all patients with a higher decrease in group T vs group V. CONCLUSION: Angiotensin receptor blockers seem to be efficient in hypertension-associated NASH. 11Autophagy in colorectal cancer:An important switch from physiology to pathology显示文摘Colorectal cancer(CRC) remains a leading cause of cancer death in both men and women worldwide.Among the factors and mechanisms that are involved in the multifactorial etiology of CRC,autophagy is an important transformational switch that occurs when a cell shifts from normal to malignant.In recent years,multiple hypotheses have been considered regarding the autophagy mechanisms that are involved in cancer.The currently accepted hypothesis is that autophagy has dual and contradictory roles in carcinogenesis,but the precise mechanisms leading to autophagy in cancer are not yet fully defined and seem to be context dependent.Autophagy is a surveillance mechanism used by normal cells that protects them from the transformation to malignancy by removing damaged organelles and aggregated proteins and by reducing reactive oxygen species,mitochondrial abnormalities and DNA damage.However,autophagy also supports tumor formation by promoting access to nutrients that are critical to the metabolism and growth of tumor cells and by inhibiting cellular death and increasing drug resistance.Autophagy studies in CRC have focused on several molecules,mainly microtubule-associated protein 1 light chain 3,beclin 1,and autophagy related 5,with conflicting results.Beneficial effects were observed for some agents that modulate autophagy in CRC either alone or,more often,in combination with other agents.More extensive studies are needed in the future to clarify the roles of autophagy-related genes and modulators in colorectal carcinogenesis,and to develop potential beneficial agents for the prognosis and treatment of CRC.Florin Burada Elena Raluca Nicoli Marius Eugen Ciurea Daniel Constantin Uscatu Mihai Ioana Dan Ionut Gheonea 2015World Journal of Gastrointestinal Oncology2015,7,11:15
12Strategic mining options optimization:Open pit mining, underground mining or both显示文摘Near-surface deposits that extend to considerable depths are often amenable to both open pit mining and/or underground mining. This paper investigates the strategy of mining options for an orebody using a Mixed Integer Linear Programming(MILP) optimization framework. The MILP formulation maximizes the Net Present Value(NPV) of the reserve when extracted with(i) open pit mining,(ii) underground mining, and(iii) concurrent open pit and underground mining. Comparatively, implementing open pit mining generates a higher NPV than underground mining. However considering the investment required for these mining options, underground mining generates a better return on investment than open pit mining. Also, in the concurrent open pit and underground mining scenario, the optimizer prefers extracting blocks using open pit mining. 15DAZL is a master translational regulator of murine spermatogenesis显示文摘Expression of DAZ-like(DAZL)is a hallmark of vertebrate germ cells,and is essential for embryonic germ cell development and differentiation,yet the gametogenic function of DAZL has not been fully characterized and most of its in vivo direct targets remain unknown.We showed that postnatal stage-specific deletion of Dazl in mouse germ cells did not affect female fertility,but caused complete male sterility with gradual loss of spermatogonial stem cells,meiotic arrest and spermatid arrest.Using the genome-wide high-throughput sequencing of RNAs isolated by cross-linking immunoprecipitation and mass spectrometry approach,we found that DAZL bound to a large number of testicular mRNA transcripts(at least 3008)at the 3′-untranslated region and interacted with translation proteins including poly(A)binding protein.In the absence of DAZL,polysome-associated target transcripts,but not their total transcripts,were significantly decreased,resulting in a drastic reduction of an array of spermatogenic proteins and thus developmental arrest.Thus,DAZL is a master translational regulator essential for spermatogenesis.Haixin Li Zhuqing Liang Jian Yang Dan Wang Hanben Wang Mengyi Zhu Baobao Geng Eugene Yujun Xu 2019National Science Review2019,6,3:11

17Diabetes mellitus as a risk factor for gastrointestinal cancer among American veterans显示文摘AIM:To assess the risk of biliary and pancreatic cancers in a large cohort of patients with type 2 diabetes mellitus(DM) .METHODS:Eligibility for this study included patients with type 2 DM(ICD-9 code 250.0) who were discharged from Department of Veteran Affairs hospitals between 1990 and 2000.Non-matched control patients without DM were selected from the same patient treatment files during the same period.Demographic information included age,sex and race.Secondary diagnoses included known risk factors based on their ICD-9 codes.By multivariate logistic regression,the occurrence of biliary and pancreatic cancer was compared between case subjects with DM and controls without DM.RESULTS:A total of 1172496 case and control subjects were analyzed.The mean age for study and control subjects was 65.8 ± 11.3 and 64.8 ± 12.6 years,respectively.The frequency of pancreatic cancer in subjects with DM was increased(0.9%) in comparison to control subjects(0.3%) with an OR of 3.22(95% CI:3.03-3.42) .The incidence of gallbladder andextrahepatic biliary cancers was increased by twofold in diabetic patients when compared to controls.The OR and 95% CI were 2.20(1.56-3.00) and 2.10(1.61-2.53) ,respectively.CONCLUSION:Our study demonstrated that patients with DM have a threefold increased risk for developing pancreatic cancer and a twofold risk for developing biliary cancer.M Mazen Jamal Eugene J Yoon Kenneth J Vega Mehrtash Hashemzadeh Kenneth J Chang 2009World Journal of Gastroenterology2009,15,42:10
18Cardiovascular dysfunction following spinal cord injury显示文摘Both sensorimotor and autonomic dysfunctions often occur after spinal cord injury(SCI). Particularly, a high thoracic or cervical SCI interrupts supraspinal vasomotor pathways and results in disordered hemodynamics due to deregulated sympathetic outflow. As a result of the reduced sympathetic activity, patients with SCI may experience hypotension, cardiac dysrhythmias, and hypothermia post-injury. In the chronic phase, changes within the CNS and blood vessels lead to orthostatic hypotension and life-threatening autonomic dysreflexia(AD). AD is characterized by an episodic, massive sympathetic discharge that causes severe hypertension associated with bradycardia. The syndrome is often triggered by unpleasant visceral or sensory stimuli below the injury level. Currently the only treatments are palliative – once a stimulus elicits AD, pharmacological vasodilators are administered to help reduce the spike in arterial blood pressure. However, a more effective means would be to mitigate AD development by attenuating contributing mechanisms, such as the reorganization of intraspinal circuits below the level of injury. 19Magnetic resonance imaging in assessment of stress urinary incontinence in women: Parameters differentiating urethral hypermobility and intrinsic sphincter deficiency显示文摘AIM: To define the magnetic resonance imaging(MRI) parameters differentiating urethral hypermobility(UH) and intrinsic sphincter deficiency(ISD) in women with stress urinary incontinence(SUI).METHODS: The static and dynamic MR images of 21 patients with SUI were correlated to urodynamic(UD) findings and compared to those of 10 continent controls. For the assessment of the urethra and integrity of the urethral support structures, we applied the highresolution endocavitary MRI, such as intraurethral MRI, endovaginal or endorectal MRI. For the functional imaging of the urethral support, we performed dynamic MRI with the pelvic phased array coil. We assessed the following MRI parameters in both the patient and thevolunteer groups:(1) urethral angle;(2) bladder neck descent;(3) status of the periurethral ligaments,(4) vaginal shape;(5) urethral sphincter integrity, length and muscle thickness at mid urethra;(6) bladder neck funneling;(7) status of the puborectalis muscle;(8) pubo-vaginal distance. UDs parameters were assessed in the patient study group as follows:(1) urethral mobility angle on Q-tip test;(2) Valsalva leak point pressure(VLPP) measured at 250 cc bladder volume; and(3) maximum urethral closure pressure(MUCP). The UH type of SUI was defined with the Q-tip test angle over 30 degrees, and VLPP pressure over 60 cm H2 O. The ISD incontinence was defined with MUCP pressure below 20 cm H2 O, and VLPP pressure less or equal to 60 cm H2 O. We considered the associations between the MRI and clinical data and UDs using a variety of statistical tools to include linear regression, multivariate logistic regression and receiver operating characteristic(ROC) analysis. All statistical analyses were performed using STATA version 9.0(Stata Corp LP, College Station, TX).RESULTS: In the incontinent group, 52% have history of vaginal delivery trauma as compared to none in control group(P < 0.001). There was no difference between the continent volunteers and incontinent patients in body habitus as assessed by the body mass index. Pubovaginal distance and periurethral ligament disruption are significantly associated with incontinence; periurethral ligament symmetricity reduces the odds of incontinence by 87%. Bladder neck funneling and length of the suprapubic urethral sphincter are significantly associated with the type of incontinence on UDs; funneling reduced the odds of pure UH by almost 95%; increasing suprapubic urethral sphincter length at rest is highly associated with UH. Both MRI variables result in a predictive model for UDs diagnosis(area under the ROC = 0.944). 20中国西部地区猎隼(Falco cherrug)繁殖生物学与保护显示文摘自2001—2006年,中国科学院与英国鸟类研究中心联合对新疆准噶尔盆地和昆仑山、青海玉树地区、西藏那曲地区的猎隼(Falco cherrug)分布状况与繁殖生态进行深入调查。考察行程上万公里。卫星跟踪显示在俄罗斯、蒙古繁殖的猎隼迁往青藏高原越冬。但是,实地调查表明,在新-青-藏还有一些繁殖种群;繁殖区的海拔高度从600m,上升至5200m。根据观察,猎隼喜欢沿用旧巢,多数巢属于“垃圾巢”。因为高原缺乏合适的巢材,那些人类遗弃的皮管、铁丝、皮带、破布、汽车零件、骨片、绳子等垃圾充斥于巢穴之中。考察期间,共找到100多只猎隼成鸟、约35个猎隼巢或巢区;分布地点涉及昆仑山脉、准噶尔盆地、卡拉麦里山脉、北塔山、可可西里、唐古拉山脉等,包括新疆奇台、且末、青河、塔城,青海楚玛尔河、五道梁、曲麻菜、雁石坪、玉树,西藏安多、那曲等。猎隼巢址多位于悬崖凹陷处(21个)、河岸砂土洞(5)、桥梁(2)等处;巢高于地面7—30m。通常沿用渡鸦、大鸳、棕尾鸳、金雕的旧巢,有同步繁殖现象。附近有鼠兔、雪雀、地鸦、岩鸽、石鸡、沙鸡等活动。巢区沿着山脉或者丘陵连续分布,巢间距最近2.5km,通常4—8km或更远。在准噶尔盆地其繁殖密度1.1~1.6对/1000km^2。通常4—5月产卵,窝卵数3—5枚;5月上旬和中旬已有部分幼乌出壳。虽然在中国捕捉猎隼现象十分猖獗,但考察结果依然令人振奋。建议中国猎隼研究小组(CSRG)积极开展国际合作,通过卫星跟踪、GPS定位、电子微芯片标记(PIT)、DNA指纹图谱分析、人工猎隼栖架和巢穴招引技术等,不断完善监测手段和管理机制,为猎隼的保护与合理利用做出努力。马鸣 梅宇 Eugene Potapov 吴逸群 Andrew Dixon Dimitar Ragyov 徐峰 Nicholas C.Fox 2007干旱区地理2007,30,5:9
