| 3 | Identification of Chinese populations of Bemisia tabaci (Gennadius) by analyzing ribosomal ITS1 sequence显示文摘Spreading of Bemisia tabaci B biotype has had a large impact on the production of vegetables, ornamental and fiber crops in many countries of the world since the 1990s. This study analyzes the ribosomal ITS1 sequence (~520 bp) to distinguish genetic difference of B. tabaci from representative different geographical populations in China. | 5 | Identification and expression profiling of putative odorant-binding proteins in the malaria mosquitoes, Anopheles gambiae and A. arabiensis显示文摘Olfaction plays a major role in host-seeking behaviour of mosquitoes. An informat- ics-based genome-wide analysis of odorant-binding protein (OBP) homologues is under- taken, and 32 putative OBP genes in total in the whole genome sequences of Anopheles gam- biae are identified. Tissue-specific expression patterns of all A. gambiae OBP candidates are determined by semi-quantitative Reverse Transcription (RT)-PCR using mosquito actin gene a internal expression control standard. The results showed that 20 OBP candidates had strong expression in mosquito olfactory tissues (female antennae), which indicate that OBPs may play an important role in regulating mosquito olfactory behaviours. Species-specific expression pat- terns of all putative anopheline OBPs are also studied in two of the most important malaria vec- tors in A. gambiae complex, i.e. A. gambiae and A. arabiensis, which found 12 of the putative OBP genes examined displayed species-differential expression patterns. The cumulative relative expression intensity of the OBPs in A. arabiensis antennae was higher than that in A. gambiae (the ratio is 1441.45:1314.12), which might be due to their different host preference behaviour. While A. gambiae is a highly anthropophilic mosquito, A. arabiensis is more opportunistic (Vary- ing from anthropophilic to zoophilic). So the latter should need more OBPs to support its host selection preference. | 6 | Molecular characterization and developmental expression of the gene encoding the prothoracicotropic hormone in the beet armyworm, Spodoptera exigua显示文摘Prothoracicotropic hormone (PTTH), a neuropeptide hormone stimulating the prothoracic glands to synthesize ecdysone, plays an important role in regulating postembryonic development in insects. The cDNA encoding PTTH was isolated and sequenced from the beet armyworm, Spodoptera exigua (Spe). The deduced a?mino acid sequence is composed of a signal peptide, a peptide (65 amino acids) of un-known function, and a mature PTTH molecule (111 amino acids). The Spe-PTTH shows similarities (45.5%―70.3%) to other known PTTHs reported in Lepidoptera species, but 7 cysteine r?esidues and the hydrophobic regions were conserved. Whole-mount immunocytochemistry by using an antiserum against recombinant Helicoverpa armigera PTTH showed that Spe-PTTH was synthesized in two pairs of neurosecretory cells in the S. exigua brain. Northern blot analysis demonstrates the presence of a 1.2-kb transcript in the brain. | 7 | 思茅松毛虫的研究显示文摘思茅松毛虫是中国南部的重要松毛虫之一。以Pinus langbianensis Chu为食和以Keteleeria evelyniana为食的幼虫在形态上及发育上均有重大差异。 在安宁,幼虫主要取食Keteleeria evelyniana,每年发生1—2代。越冬后的幼虫在5—6月间化蛹而后在6—7月间发育成熟羽化。第一代幼虫在生长发育过程中产生分化:大部分幼虫生长缓慢而以幼虫越冬,其余的幼虫在四龄后生长迅速,于8月底化蛹、羽化,第二代幼虫于9月中、下旬孵化而以三龄幼虫越冬。引起分化的原因还不明了,应予深入研究。我们设想至少应追索到第一代幼虫孵化之前,因为在7月以前孵化的同一母虫的幼虫都可以产生分化。 这一害虫的化学防治试验了13种杀虫剂,每公斤“621烟剂加5%DDVP40毫升的试验在野外进行,死亡率高达90%。其他:50%phoxim 1:15000,90%Trichlorphon 1:15000,50%嘧啶硫磷1:3000,50%Cyanophos 1:5000,30%phosvel1:5000,40%伏地松1:5000,20%MTMC1:500,25%Carbaryl1:250,95%Cartap1:5000,50%Chorophenmidine1:1000,50%Garodona1:600及50%12401:3000倍均在室内进行试验,死亡率高达90%—100%。 | Teaching and Research Group of Forest Entomology | 1981 | 北京林业学院学报1981,,1: | 2 |
| 8 | Ecdysteroids in the adults and eggs of Opogona sacchari (Bojer),an invasive alien pest显示文摘In order to understand the composition and quantitative variation of ecdysteriods in the adults and eggs of Opogona sacchari (Bojer), an invasive alien pest, we analyzed the ecdysteroid composition and titers in the adult and egg of this pest. On day 4 after eclosion, the titer of ecdysteriods in the male adult was 0.080 ng/adult, much lower than 5.978 ng/adult in the female adult. During the development of ovaries, the titer of ecdysteriods was low on the first two days, and high in the late period, with the peak (10.48 ng/ovary) appearing on day 3. During the development of eggs, the titer of ecdysteriods was about 0.010 ng/egg from day 1 to day 3, and then decreased to 0.006 ng/egg on day 4. In both adults and eggs, three main components of ecdysteriods were found by identification of HPLC/RIA. | 10 | Observation on the initial inoculum source and dissemination of Entomophthorales-caused epizootics in populations of cereal aphids显示文摘A total number of 1092 migratory alates were trapped from air in wheat grown area of Yuanyang County, Henan Province from early April through May 2002 in order to confirm the source and dissemination of entomophthoralean inocula to cause epizootics of cereal aphids. Those included 415 Sitobion avenae, 642 Rhopalosiphum padi, 22 Metopolophium dirhodum, and 13 Schizaphis graminum. The trapped alates were daily collected and individually reared for 7 days on wheat plants in laboratory. Of those 341 alates died of fungal infection, taking 31.2% in the trapped alates. These included 224 S. avenae, 106 R. padi, 8 M. dirhodum, and 3 S. graminum. Deaths of all infected alates occurred during the first 5 days and 78.9% of the deaths occurred within the first 3 days. Individual examination under microscope proved that all deaths were attributed to entomophthoralean fungi. Of those Pandora neoaphidis accounted for 84.6%, Conidiobolus obscurus for 9.9%, and Entomophthora planchoniana for 5.5%. Four alate deaths died of cross infection of P. neoaphidis and C. conidiobolus. Based on the high infection rate of the migratory alates trapped from air and the field occurrence of epizootics in populations of cereal aphids during the trapping period, Entomophthorales-caused epizootics were likely disseminated by infected alates through their flight and colonization. | 14 | The Utility of Specific Markers Based on ITS2 Sequences for Molecular Identification and Detection of Trichogramma spp.显示文摘The technology based on specific PCR amplification using internal transcribed spacer 2 of nuclear ribosomal DNA for molecular identification and detection of Trichogramma species was studied. Firstly the ITS2s of six Trichogramma species were cloned and sequenced, and the interspecific sequence variation was analyzed. Secondly the ITS2 regions of six geographical populations of T. dendrolimi were cloned and sequenced, and the intraspecific sequence identity was analyzed. The results show that the interspecific variation and intraspecific similarity of ITS2 sequences are very suitable for designation of specific primers at specieslevel. Screening of specific primers for T. dendrolimi leads to final sensitive and stable diagnostic primers. This system lets non-specialists can not only identify adults (males and females), but also identify eggs in parasitized hosts rapidly and accurately, which is impossible by conventional methods. | 19 | Molecular Weight of Cytoplasmic Malate Dehydrogenase,Mitochondrial Malate Dehydrogenase and Lactate Dehydrogenase of a Freshwater Catfish显示文摘The multiple molecular forms of cytoplasmic malate dehydrogenase (cMDH), mitochondrial malate dehydrogenase (mMDH ) and lactate dehydrogenase (LDH ) were studied in the liver and skeletal muscle of the freshwater catfish, Clarias batrachus. There were two electrophoretically distinguishable bands (AA andBB) of cMDH and mMDH which suggests that they are apparently encoded at two gene loci (A and B) in both the tissues.However, the presence of a single band (LDH-1 ) of LDH in liver and double bands (LDH-1and LDH-2) in skeletal muscle in which LDH-2 was predominant reflects the differential expression of LDH genes in different metabolic tissues to meet the requirement of energy production. The AA isoform (74 kd) of liver cMDH was smaller than those of the AA form (110 kd) of skeletal muscle. In contrast, the BB isoform of liver (42 kd) and skeletal muscle (54 kd) were more or less similar in size. Unlike the case of cMDH, the molecular weight of AA isoform (115 kd) of liver mMDH was higher than those of the AA form (87kd) of skeletal muscle. Whereas the molecular weight of BB isoform (58 kd) of liver was in proximity to the weight of BB form (44 kd) of skeletal muscle mMDH. The size of AA isoform (74 kd) of liver cMDH was smaller, while the AA isoform (110 kd) of skeletal muscle was larger as compared to AA form of mMDH in the liver (115 kd) and skeletal muscle (87 kd). But the size of BB isoform of both the isozymes was almost equal in these metabolic tissues. The molecular weight of liver LDH-1 (96 kd) was close to the weight of LDH-1 (82 kd) in skeletal muscle. The molecular weight of skeletal muscle LDH-2 was deduced as 37 kd which is much more lower than the weight of LDH-1 in liver and skeletal muscle. The stereochemistry at 6-C and 10-C of 1 was constructed by using optically active citronellal as starting material and by the asymmetric crotylic metal reaction. In the bioassay and field tests, only la, i.e. (6R,10R)-1 was active. The other three isomers 1b (6S,10R), 1c (6R,10S) and 1d (6S,10S) were inactive.
