1Thrombopoietin induced proliferation and differentiation of fetal liver CD34^+ cells with phenotype change from hemopoiesis to neurogenesis显示文摘BACKGROUND: Previous studies have reported a neurotrophin-like motif in the N-terminal receptor binding region of the thrombopoietin (TPO) molecule, and have described localization of TPO and TPO receptor in the brain. Therefore, it is believed that TPO may be involved in regulation of neurogenesis. OBJECTIVE: To validate the effect of TPO on trans-differentiation, or differentiation from hematopoietic stem cells (HSCs) to neural stem cells (NSCs). DESIGN, TIME AND SETTING: Comparative studies were performed from March 2004 to April 2007 at the Department of Experimental Medicine, Northern Hospital, and the Department of Immunology, Fourth Military Medical University of Chinese PLA. MATERIALS: Human fetal liver (FL) was obtained from fetuses after water-balloon abortion. Gestational age ranged from 16 to 20 weeks. The study was approved by the Institutional Review Board and Ethics Committee of the Northern Hospital. TPO was kindly provided by Genentech Inc (USA). Iscove's Modified Dulbecco's Medium (IMDM) and neurobasalTM medium were purchased from Invitrogen (USA). MACS CD34 multisort kit was purchased from Miltenyi Biotec (Germany). METHODS: CD34+ cells were isolated from human FL mononuclear cells using MACS CD34 multisort kit and cultured at 1 × 105/mL in IMDM, containing TPO for 60 days with weekly changes of half of the medium. After culturing for 30 and 60 days, the TPO-induced cells were resuspended in neurobasalTM medium containing 10% fetal brain extracts and plated in an 8-well BIOCOAT poly-D-Lysine Culture Slide and cul- tured for another 7 days. MAIN OUTCOME MEASURES: Cell number, viability, phenotype and expression of hemopoiesis-related and neurogenesis-related proteins were examined by trypan blue exclusion with hemocytometer, immunoblot, immunocytochemistry and flow cytometry. RESULTS: After 60 days of induction with TPO, the cell number increased by 4.6-fold compared to the ini- tial culture. Although the proportion of the cells expressing the hemopoietic stem cell associated antigen (CD34) decreased steadily, both proportions of the cultured FL-derived CD34+ cells expressing CD41a and CD61 remained unchanged, which still accounted for 10%. Noticeably, the proportions of the cells express- ing nestin and epidermal growth factor receptor increased significantly (both > 50%), whereas the expression of more mature neural or glial proteins [microtubule-associated protein-2 (MAP2), glial fibrillary acidic pro- tein (GFAP), oligodendrocyte marker O4 (O4)] markers on the cultured fetal liver derived-CD34+ cells were at lower levels. After another 7 days incubation in neurobasal? medium, these TPO-induced cells formed neurospheres, which were labeled with nestin, and differentiated into cells with morphological characteristics of neurons, astrocytes and oligodendrocytes, which were labeled with MAP-2, GFAP, and O4, respectively. 2Bayesian analysis for the Lomax model using noninformative priors显示文摘The Lomax distribution is an important member in the distribution family.In this paper,we systematically develop an objective Bayesian analysis of data from a Lomax distribution.Noninformative priors,including probability matching priors,the maximal data information(MDI)prior,Jeffreys prior and reference priors,are derived.The propriety of the posterior under each prior is subsequently validated.It is revealed that the MDI prior and one of the reference priors yield improper posteriors,and the other reference prior is a second-order probability matching prior.A simulation study is conducted to assess the frequentist performance of the proposed Bayesian approach.Finally,this approach along with the bootstrap method is applied to a real data set.Daojiang He Dongchu Sun Qing Zhu 2023Statistical Theory and Related Fields2023,7,1:1

4Effect of dendritic cell/cytokine-induced killer cell immunobiological cancer therapy combined with adjuvant chemotherapy in patients with triple-negative breast cancer显示文摘Objective The aim of the present study was to investigate the effect of dendritic cell(DC)/cytokine-induced killer cell(CIK) immunobiological cancer therapy in patients with triple-negative breast cancer(TNBC) who underwent adjuvant chemotherapy. Methods From January 2010 to October 2013, 120 patients with postoperative TNBC were recruited and included in the study. Patients were enrolled in one of two groups according to whether they accepted DC/CIK immunobiological cancer therapy during adjuvant chemotherapy; the patients in the DC/CIK group underwent adjuvant chemotherapy combined with DC/CIK immunobiological cancer therapy, and the control group underwent adjuvant chemotherapy alone. When six cycles of adjuvant chemotherapy and six cycles of DC/CIK immunobiological cancer therapy had been completed, differences between the two groups with regard to quality of life(Qo L), immunological indicators(CD3, CD4, CD8, and NK cell levels), disease-free survival(DFS), and side effects of chemotherapy and DC/CIK treatment were evaluated.Results In the DC/CIK group, the proportion of NK cells and CD3+ and CD4+ T-cell subgroups significantly increased, and the proportion of CD8+ cells decreased when they were compared before and after DC/CIK therapy(P < 0.05). However, there were no significant changes in the control group. By the final follow-up, DFS of the treatment group and the control group was 38.4 and 34.2 months, respectively. The Qo L improved in the patients treated with chemotherapy plus DC/CIK therapy compared with the patients treated with chemotherapy alone, and the difference between groups was significant(P < 0.05). The side effects of two groups were tolerable and not significantly different between the two groups.Conclusion The DC/CIK treatment had potential benefits for patients with TNBC compared with the control group, and was not associated with any obvious side effects. 9Editorial foreword显示文摘Statistical Theory and Related Fields is now in its thirdyear of publication. The first article in this first issue of2019 is a discussion paper by professors Bayarri, Berger,Jang, Ray, Pericchi, and Visser with the exciting topicon prior-based Bayesian information criterion for variable selection. Variable selection is a key componentfor dimension reduction in modern statistical applications in the big data era. Both Bayesian and frequentistapproaches have shown their potentials in handlingvariable selection in the past two decades, but bothface various challenges. This article shows a pioneering idea of adding the prior into the penalty term forvariable selection and having different effective sample sizes for different model parameters. The article isfollowed by a total of nine discussions written by wellknown researchers in the area of variable selection, eachof them exhibits some interesting research topics assummarised in the authors’ rejoinder. 12Bayesian analysis for quantile smoothing spline显示文摘In Bayesian quantile smoothing spline[Thompson,P.,Cai,Y.,Moyeed,R.,Reeve,D.,&Stander,J.(2010).Bayesian nonparametric quantile regression using splines.Computational Statistics and Data Analysis,54,1138-1150.],a fixed-scale parameter in the asymmetric Laplace likelihood tends to result in misleading fitted curves.To solve this problem,we propose a new Bayesian quantile smoothing spline(NBQSS),which considers a random scale parameter.To begin with,we justify its objective prior options by establishing one sufficient and one necessary condition of the posterior propriety under two classes of general priors including the invariant prior for the scale component.We then develop partially collapsed Gibbs sampling to facilitate the compu-tation.Out of a practical concern,we extend the theoretical results to NBQSS with unobserved knots.Finally,simulation studies and two real data analyses reveal three main findings.Firstly,NBQSS usually outperforms other competing curve fitting methods.Secondly,NBQSS consid-ering unobserved knots behaves better than the NBQSS without unobserved knots in terms of estimation accuracy and precision.Thirdly,NBQSS is robust to possible outliers and could provide accurate estimation.Zhongheng Cai Dongchu Sun 2021Statistical Theory and Related Fields2021,5,4:0

14OBSERVATION OF MEGAKARYOCYTOPOIESIS IN HUMAN FETUS BY IMMUNOCYTOCHEMICAL STAINING WITH ANTI-PLATELET GLYCOPROTEIN ⅡB /ⅢA MONOCLONAL ANTIBODY显示文摘Fetal liver tissues obtained from 28 human fetuses with gestation age from 3 to 6 months and fetal bone marrow from 35 human fetuses from 3 to 7 months were observed by immunochemical staining with anti-platelet GPⅡ b / Ⅲa monoclonal antibody and ABC technique. In the fetal liver, megakaryocytes were wholly located among growing fetal liver cells and near foci of hemopoiesis. Some megakaryocytes in the fetal liver were small7890- lymphoid-like megakaryocytes. The size of megakaryocytes both in the fetal liver (14.79 ± 4.52μm) and in the fetal bone marrow (16.08±7.39 μm) was small, which did not vary significantly over the gestation age ranging from 3 to 6 or 7 months. 15Fully Bayesian analysis of the relevance vector machine classification for imbalanced data problem显示文摘Relevance Vector Machine(RVM)is a supervised learning algorithm extended from Support Vector Machine based on the Bayesian sparsity model.Relevance Vector Machine classification suffers from theoretical limitations and computational inefficiency mainly because there is no closed-form solution for the posterior of the weight parameters.We propose two advanced Bayesian approaches for RVM classification,namely the Enhanced RVM and the Reinforced RVM,to perfect the theoretic framework of RVM and extend the algorithm to the imbalanced data problem,which has an arresting skew in data size between classes.First,the Enhanced RVM conducts a strict Bayesian sampling process instead of the approximation method in the original one to remedy its theoretic limitations,especially the nonconvergence of the iterations.Secondly,we conjecture that the hierarchical prior makes the Reinforced RVM achieve consistent estimations of the quantities of interest compared with the non-consistent estimations of the original RVM.Consistency is necessary for RVM classification since it makes the model more stable and localises the relevant vectors more accurately in the imbalanced data problem.The two-level prior also renders the Reinforced one competitive in the imbalanced data problem by building the inner connection of parameter dimensions and alloting a more vital relevance to the small class data weight parameter.The theoretic proofs and several numeric studies demonstrate the merits of our two proposed algorithms.Wenyang Wang Yanna Sun Keran Li Jinglin Wang Chong He Dongchu Sun 2023CAAI Transactions on Intelligence Technology2023,8,1:0
