| 3 | Inhibiting effect of antisense oligonucleotides phosphorthioate on gene expression of TIMP-1 in rat liver fibrosis显示文摘AIM To observe the inhibition of antisenseoligonucleotides (asON) phosphorthioate to thetissue inhibitors metalloproteinase-1 (TIMP-1)gene and protein expression in the liver tissue ofimmunologically induced hepatic fibrosis rats.The possibility of reversing hepatic fibrosisthrough gene therapy was observed.METHODS Human serum albumin (HSA) wasused to attack rats, as hepatic fibrosis model, inwhich asONs were used to block the gene andprotein expressing TIMP-1. According to theanalysis of modulator, structure protein, codingseries of TIMP-1 genome, we designed fourdifferent asONs. These asONs were injected intothe hepatic fibrosis models through coccygealvein. The results was observed by RT-PCR formeasuring TIMP-1 mRNA expression,immunohistochemistry and in situ hybridizationfor collagen Ⅰ, Ⅲ, special staining of collagenfiber, and electron microscopic examination.RESULTS Hepatic fibrosis could last within 363days in our modified model. The expressinglevel of TIMP-1 was high during hepatic fibrosisprocess. It has been proved by theimmunohistochemical and the electronmicroscopic examination that the asONphosphorthioate of TIMP-1 could exactly expressin vivo. The effect of colchicine wasdemonstrated to inhibit the expressing level ofmRNA and the content of collagen Ⅰ, Ⅲ in theliver of experimental hepatic fibrosis rats.However, the electron microscopy research andthe pathologic grading of hepatic fibrosisshowed that there was no significant differencebetween the treatment group and the modelgroup (P>0.05).CONCLUSION The experimental rat model ofhepatic fibrosis is one of the preferable modelsto estimate the curative effect of anti-hepaticfibrosis drugs. The asON phosphorthioate ofTIMP-1 could block the gene and proteinexpression of TIMP-1 in the liver of experimentalhepatic fibrosis rats at the mRNA level. It ispossible to reverse hepatic fibrosis, and it isexpected to study a new drug of anti-hepaticfibrosis on the genetic level. In this paper, a new form of time-dependent mild slope equation for irregular waves was derived with Fade approximation and Kubo’s time series concept. | 6 | Analysis of the delayed approach to the management of infected pancreatic necrosis显示文摘AIM: To analyze outcomes of delayed single-stage necrosectomy after early conservative management of patients with infected pancreatic necrosis (IPN) associated with severe acute pancreatitis (SAP). METHODS: Between January 1998 and December 2009, data from patients with SAP who developed IPN and were managed by pancreatic necrosectomy were analyzed. RESULTS: Fifty-nine of 61 pancreatic necrosectomies were performed by open surgery and 2 laparoscopically. In 55 patients, single-stage necrosectomy could be performed (90.2%). Patients underwent surgery at a median of 29 d (range 13-46 d) after diagnosis of acute pancreatitis. Sepsis and multiple organ failure accounted for the 9.8% mortality rate. Pancreatic fistulae (50.8%) predominantly accounted for the morbidity. The median hospital stay was 23 d, and the median interval for return to regular activities was 110 d.CONCLUSION: This series supports the concept of delayed single-stage open pancreatic necrosectomy for IPN. | 8 | NUMERICAL ANALYSIS OF THE MECHANICS OF SOIL REINFORCEMENT显示文摘Roots affect the soil stress and strain and contribute to soil reinforcement. An analysis of stress and strain in the root-soil composite is presented which combines roots, soils, and an interface element by using the Duncan-Chang E- model and a nonlinear-interface-element model. In this research, a finite element numerical analysis method was applied to simulate the stress field resulting in a composite of Pinus tabulaeformis roots and soils. Results show that roots can transmit the stress from surface soils to deep soils, and can reduce soil stress within the surface soil layer; results also show that the effect of soil reinforcement by roots is limited to surface soils. | 13 | ANALYSIS ON BUCKLING AND POSTBUCKLING OF DELAMINATION IN 3D COMPOSITES显示文摘In this paper, the problem of axisymmetric buckling and postbuckling of a circular thin-film delamination bridged by through-thickness fiber tows in 3D composites is presented. An iterativeprocedure based on Taylor’s series expansion is used to generate a family of nondimensionalized post-buckling solutions of the above problem by yon Karman’s nonlinear plate theory. Attention is fo-cused, herein, on the effects of the bridge force of through-thickness fibers on the buckling and post-buckling behavior of the delamination. It is found that fiber bridge not only increases the ability of re-sisting delamination buckling and postbuckling, but also brings on the jump of the delamination deflec-tion mode during the postbuckling phase. | 17 | DEVELOPMENT OF SOIL EROSION INDEX MODEL IN TAIWAN WATERSHEDS显示文摘With steep terrain and excessive rainfall, Taiwan is affected by severe soil erosion caused by summer typhoons and storms that bring intensive rainfall and rapid flows,. However, the actual erosion is much less than the value predicted by the USLE because the soil erosion types in Taiwan are different from those in America which has mild slope and dry weather. Developing a soil erosion index model applicable to Taiwan is the important goal of this research. Five factors, namely, soil texture, rainfall type, slope steepness, ground cover and land use, are included in this model. Soil index factor is measured by Km from the USLE model, and other index factors are calculated from local field data. The soil erosion index model (SEIM) is as follows: SE=6x10-7AIfor AI≤50 SE = 0.233AI1.8 for AI > 50 where AI is the total index value, and SE is the soil erosion quantity (ton/ha/yr).
