1甘薯细胞壁蔗糖转化酶基因IbCWIN家族成员鉴定及表达分析显示文摘甘薯是重要的粮食、饲料、工业原料作物和新型的生物能源作物,细胞壁蔗糖转化酶是植物源、库组织蔗糖代谢的关键酶,但关于甘薯细胞壁蔗糖转化酶基因IbCWIN家族成员的研究尚未见报道。本研究测定供试品种不同组织部位的蔗糖淀粉含量,利用生物信息学方法对IbCWIN基因家族的理化性质、保守结构域、系统进化关系、启动子作用元件、组织特异性表达模式进行分析。结果表明,甘薯茎中蔗糖含量最高,须根和叶次之,块根最低;块根淀粉含量最高,极显著高于其他部位。甘薯中含有10个IbCWIN基因,编码氨基酸442~1115个,蛋白质分子量范围49.56~124.44kD,等电点为5.0~9.1。分布在8条染色体上,都含有Glyco_32保守结构域及相同或相似的保守基序motif,属于糖基水解酶基因家族GH32。IbCWIN与木薯MeCWINV同源性高,IbCWIN基因家族启动子区域含有多种类型的顺式作用元件。qRT-PCR结果表明,IbCWIN基因家族在甘薯不同组织中均有表达且有多种表达模式,其中IbCWIN2和IbCWIN9在块根中表达量显著高于其他组织部位。本研究为下一步探索甘薯IbCWIN基因家族的功能及调控甘薯源、库关系机制提供理论指导。宋天晓 刘意 饶莉萍 Soviguidi Deka Reine Judesse 朱国鹏 杨新笋 2021作物学报2021,47,7:6
2Plant growth-promoting rhizobacteria--alleviators of abiotic stresses in soil: A review显示文摘With the continuous increase in human population,there is widespread usage of chemical fertilizers that are responsible for introducing abiotic stresses in agricultural crop lands.Abiotic stresses are major constraints for crop yield and global food security and therefore require an immediate response.The implementation of plant growth-promoting rhizobacteria(PGPR)into the agricultural production system can be a profitable alternative because of its efficiency in plant growth regulation and abiotic stress management.These bacteria have the potential to promote plant growth and to aid in the management of plant diseases and abiotic stresses in the soil through production of bacterial phytohormones and associated metabolites as well as through significant root morphological changes.These changes result in improved plant-water relations and nutritional status in plants and stimulate plants’defensive mechanisms to overcome unfavorable environmental conditions.Here,we describe the significance of plant-microbe interactions,highlighting the role of PGPR,bacterial phytohormones,and bacterial metabolites in relieving abiotic environmental stress in soil.Further research is necessary to gather in-depth knowledge on PGPR-associated mechanisms and plant-microbe interactions in order to pave a way for field-scale application of beneficial rhizobacteria,with the aim of building a healthy and sustainable agricultural system.Therefore,this review aims to emphasize the role of PGPR in growth promotion and management of abiotic soil stress with the goal of developing an eco-friendly and cost-effective strategy for future agricultural sustainability.Madhurankhi GOSWAMI Suresh DEKA 2020Pedosphere2020,30,1:6
3甘薯苯丙氨酸解氨酶基因IbPAL的克隆与表达分析显示文摘本研究基于甘薯(Ipomoea batatas(L.)Lam.)与绿原酸合成代谢相关的转录组数据克隆了苯丙氨酸解氨酶基因IbPAL,通过蛋白序列比对、系统进化分析、二级和三维蛋白结构预测、跨膜结构域以及启动子分析,对该基因的氨基酸序列和启动子序列的结构特征进行了解析;通过qRT-PCR技术分析该基因在不同甘薯品种的根、茎、茎尖和叶中的表达情况,并对其进行亚细胞定位验证。结果显示,IbPAL的CDS序列全长2130 bp,编码709个氨基酸。其氨基酸序列与已知物种的氨基酸序列同源性在80%以上。IbPAL蛋白以α螺旋和随机卷曲为主,具有1个可能的跨膜螺旋区。该基因启动子中包含多个顺式作用元件。表达模式分析结果表明,IbPAL主要在茎和茎尖中表达,其在甘薯品种‘EC16’4个组织器官中的表达量均显著高于其他品种。亚细胞定位结果显示该基因在细胞膜和细胞核上均可以表达。饶莉萍 苏文瑾 刘意 宋天晓 Soviguidi Deka Reine Judesse 杨新笋 张文英 2020植物科学学报2020,38,3:3
4Surface charge induced tuning of electrical properties of CVD assisted graphene and functionalized graphene sheets显示文摘We demonstrate a new approach to tune the electrical properties of graphene and functionalized graphene. Graphene was synthesized using thermal chemical vapour deposition(TCVD) method on copper foil using precursor gas acetylene and co-catalyst H2 gas. TCVD assisted graphene was successfully transferred onto a silicon wafer. Transferred graphene sheet was then functionalized to prepare graphene oxide(GO) and reduced graphene oxide(rGO). Different surface charge carbon nanoparticles, e.g. carbon nanoparticle with net positive charge and carbon nanoparticle with net negative charge were then immobilized on transferred graphene and functionalized graphene sheets. The functionalized graphene and charge mobilized functionalized graphene were characterized by Uv–vis spectroscopy,Fourier transformed infrared spectroscopy, scanning electron microscopy, and Raman spectroscopy. After immobilization of carbon nanomaterials, the ac electrical conductivity was found to increase due to enhancement of the surface charge, electron density, and mobility. It was observed that negative surface charge immobilized graphene and functionalized graphene show higher conductivity. 5Validity of subjective assessment as screening tool for dry eye disease and its association with clinical tests显示文摘AIM: To determine the role of subjective assessment using Mc Monnies dry eye questionnaire in diagnosing dry eye disease and its association with clinical tests.METHODS: There were 500 patients screened for dry eye using Mc Monnies dry eye questionnaire between May to October 2013 at the outpatient Department of Ophthalmology of a medical college hospital. All 500 patients were subjected to clinical tests. Dry eye was defined as having one or more symptoms often or all the time. Positive signs were if one or both eyes revealed tear film breakup time(TBUT) of ≤10s, a Schirmer test score of ≤10 mm, a Rose Bengal staining score of ≥1, a Lissamine green staining score of ≥1 or existence of meibomian gland disease(≥grade 1). Statistical analysis was performed to describe the distribution of symptoms and signs, to assess the correlations between Mc Monnies score(MS) and variable clinical signs of dry eye, and to explore the association between dry eye symptoms and variable clinical signs. Analysis was performed using software package Epi info. A Probability(P) value using Chi-square test of <0.005 was taken as significant.· RESULTS: Dry eye prevalence with symptoms(questionnaire), Schirmer test, TBUT, Rose Bengal staining and Lissamine green staining was 25.6%,15.20%, 20.80%, 23.60%, and 22.60% respectively. Among those with severe symptoms(MS>20), 75.86% had a low TBUT(<10s), 58.62% had a low Schirmer's I test(≤10 mm),86.20% had Rose Bengal staining score of ≥1, 79.31%had Lissamine green staining score of ≥1. We found statistically significant associations between positive Schirmer test and arthritis(P <0.002), dryness elsewhere(P <0.001), contact lens use(P < 0.002), systemic medication(P <0.0001), sleeping with eyes partly open(P <0.002), history of dry eyes treatment(P <0.0001),environmental factors(P <0.001), swimming(P <0.001).· CONCLUSION: Subjective assessment plays an important role in diagnosing dry eye disease.
