Effect of acupotomy on chondrocyte proliferation and expression of CyclinD1,CDK4 and CDK6 in rabbits with knee osteoarthritis显示文摘Objective:The aim of this study was to investigate the mechanism of acupotomology(Apo)in the prevention of articular cartilage destruction via the promotion of chondrocyte proliferation and chondrocyte expression of cell cycle regulators,CyclinD1,CDK4 and CDK6 in a rabbit knee osteoarthritis(KOA)model.Methods:Twenty-eight rabbits were randomly divided into a control group,an OA(osteoarthritis)model group,an Apo(acupotomology)group and EA(electro-acupuncture)group(n Z 7).Improved Videman’s method was used to induce a rabbit model of KOA over 6 weeks. One week later, acupotomy and electro-acupuncture therapy was applied to animals in the respective groups and treatment lasted 4 weeks.Following these treatments, quantitative real-time PCR,immunohistochemical staining and western blotting were performed to assess the mRNA and protein levels of cell cycle regulators CyclinD1(Cell cycle protein D1),CDK4(Cyclin-dependent kinase 4)and CDK6(Cyclin-dependent kinase 6).Ethology measures and knee morphology were also compared among groups. Results:The Lequesne MG index score of morphology was increased(P<.01),and the passive range of motion(PROM)and the mRNA and protein levels of CyclinD1,CDK4,and CDK6 were significantly decreased(P<.01)in the OA model compared with the control group. The Lequesne MG index score and the morphology score were decreased in the Apo and EA group compared with the OA model group(P<.05 or P<.01),while the mRNA levels of CyclinD1,CDK4,and CDK6,and the protein levels of CDK4 were increased in the Apo and EA groups compared with the OA model group(P<.05 or P<.01).The PROM,and the protein levels of CyclinD1 and CDK6 were increased(P<.05)in the Apo group compared with the OA model group, while the PROM and the protein levels of CyclinD1 and CDK6 in the EA group were not significantly different(P>.05).Compared with the EA group, the morphology score was decreased in Apo group(P<.05).Conclusions:The mRNA levels of CyclinD1 and CDK4,and the protein level of CDK4 in chondrocytes are regulate by both Apo and EA.Apo is more effective than EA in regulating the protein levels of CyclinD1 and CDK6.According to the observed changes in morphology and cytokine levels, acupotomy can promote chondrocyte proliferation and can alleviate the destruction of articular cartilage in a model of KOA. | Yang Gao Tong Wang Wei Zhang Xiaowei Shi Shining Ma Lijuan Wang Shuai Zhou Defeng Wang Meyer Magali Changqing Guo | 2019 | Journal of Traditional Chinese Medical Sciences2019,6,3: | 2
Predicting impact forces on pipelines from deep-sea fluidized slides:A comprehensive review of key factors显示文摘Deep-sea pipelines play a pivotal role in seabed mineral resource development, global energy and resource supply provision, network communication, and environmental protection. However, the placement of these pipelines on the seabed surface exposes them to potential risks arising from the complex deep-sea hydrodynamic and geological environment, particularly submarine slides.Historical incidents have highlighted the substantial damage to pipelines due to slides.Specifically,deep-sea fluidized slides(in a debris/mud flow or turbidity current physical state),characterized by high speed, pose a significant threat.Accurately assessing the impact forces exerted on pipelines by fluidized submarine slides is crucial for ensuring pipeline safety. This study aimed to provide a comprehensive overview of recent advancements in understanding pipeline impact forces caused by fluidized deep-sea slides, thereby identifying key factors and corresponding mechanisms that influence pipeline impact forces. These factors include the velocity, density, and shear behavior of deep-sea fluidized slides,as well as the geometry,stiffness,self-weight,and mechanical model of pipelines.Additionally, the interface contact conditions and spatial relations were examined within the context of deep-sea slides and their interactions with pipelines.Building upon a thorough review of these achievements, future directions were proposed for assessing and characterizing the key factors affecting slide impact loading on pipelines.A comprehensive understanding of these results is essential for the sustainable development of deep-sea pipeline projects associated with seabed resource development and the implementation of disaster prevention measures. | Xingsen Guo Ning Fan Defeng Zheng Cuiwei Fu Hao Wu Yanjun Zhang Xiaolong Song Tingkai Nian | 2024 | International Journal of Mining Science and Technology2024,34,2: | 0
Allylation and alkylation of oxindoleketimines via imine umpolung strategy显示文摘When treated with an alkoxide base like t-BuOK in aprotic solvent,N-di phenyl methyl imino oxindoles, made conveniently through condensation of corresponding isatins with N-di phenyl methyl amine, are deprotonated to form azaallyl anions.Allylation and alkylation of this type of intermediates proceed smoothly with diverse C-electrophiles.Acidic work up finishes 3-amino-3-allyl/alkyl oxindoles. The overall transformation equals to an umpolung process at the C3 of isatins. | Mei-Hua Shen Chen Li Qing-Song Xu Bin Guo Rui Wang Xiaoqian Liu Hua-Dong Xu Defeng Xu | 2021 | Chinese Chemical Letters2021,32,7: | 0
Enhancement of Near-Field Radiative Heat Transfer based on High-Entropy Alloys显示文摘The enhancement of near-field radiative heat transfer(NFRHT)has now become one of the research hotspots in the fieldsof thermal management and imaging due to its ability to improve the performance of near-field thermoelectric devices and near-field imaging systems. In this paper,we design three structures(multilayer structure, nanoporous structure, and nanorod structure)based on high-entropy alloys to realize the enhancement of NFRHT.By combining stochastic electrodynamicsand Maxwell-Garnett's description of the effective medium,we calculate the radiative heat transfer under different parametersand find that the nanoporousstructure has the largest enhancement effect on NFRHT.The near-field heat transfer factor(q)of this structure(q=1.40×10^(9)W/(m^(2)·K))is three times higher than that of the planestructure(q=4.6×10^(8)W/(m^(2)·K)),and about two orders of magnitude higher than that of the SiO2plate.Thisresult providesa freshidea for the enhancement of NFRHT and will promote the application of high-entropy alloy materials in near-field heat radiation. | Shanshan DENG Ping SONG Boxi ZHANG Sen YAO Zhixin JIN Defeng GUO | 2022 | Research and Application of Materials Science2022,4,2: | 0
Realizing texture in bulk nanocrystalline Nd-Fe-B magnet via high-stress low-temperature rapid deformation显示文摘It is difficult to obtain nanoscale grain size and strong texture in hot-deformed magnets simultaneously, which is responsible for the low magnetic properties of nanocrystalline magnets. In this study,a strong texture was realized in a nanocrystalline Nd-Fe-B magnet via a high-stress low-temperature rapid deformation.Strong texture formation is ascribed to the high stress, which promotes the preferential growth of grains by increasing strain energy anisotropy. The nanocrystalline formation is ascribed to the low deformation temperature(below the melting point of the Nd-rich phase)and short deformation time. The effects of deformation temperature and applied stresses on the microstructure and magnetic properties were investigated in detail. Thus, the highest maximum energy product of 43.3 MGOe is obtained. | Bingning Wei Guosheng Zhang Ping Song Defeng Guo | 2023 | Journal of Rare Earths2023,41,5: | 0
The IDD Transcription Factors:Their Functions in Plant Development and Environmental Response显示文摘INDETERMINATE-DOMAIN proteins(IDDs)are a plant-specific transcription factor family characterized by a conserved ID domain with four zinc finger motifs.Previous studies have demonstrated that IDDs coordinate a diversity of physiological processes and functions in plant growth and development, including floral transition, plant architecture, seed and root development, and hormone signaling. In this review,we especially summarized the latest knowledge on the functions and working models of IDD members in Arabidopsis, rice, and maize, particularly focusing on their role in the regulatory network of biotic and abiotic environmental responses, such as gravity, temperature, water, and pathogens.Understanding these mechanisms underlying the function of IDD proteins in these processes is important for improving crop yields by manipulating their activity.Overall, the review offers valuable insights into the functions and mechanisms of IDD proteins in plants, providing a foundation for further research and potential applications in agriculture. | Jing Liu Defeng Shu Zilong Tan Mei Ma Huanhuan Yang Ning Guo Shipeng Li Dayong Cui | 2024 | Phyton-International Journal of Experimental Botany2024,93,1: | 0
Spectral filtering effect on multi-pulsing induced by chirped fiber Bragg grating in dispersion-managed mode-locked Yb-doped fiber lasers显示文摘We numerically and experimentally investigate the multi-pulsing mechanism in a dispersion-managed mode-locked Ybdoped fiber laser.Multi-pulsing occurs primarily owing to the inherent filtering effect of the chirped fiber Bragg grating. The spectral filtering effect restricts the spectral broadening induced by self-phase modulation and causes extra loss, leading to a decreased pump power threshold for the multi-pulsing state.Numerical simulations show that multi-pulsing emerges at a lower pump power when the spectral filter bandwidth becomes narrower. In the experiment, the spectral width increases as the net cavity dispersion approaches zero.Pulses with wider spectral widths experience more loss from the spectral filtering effect, leading to a decreased pump power threshold for multi-pulsing.Therefore,the net cavity dispersion also has an impact on the multi-pulsing threshold.Based on this conclusion,we devise a strategy to obtain single-pulsing operation with the shortest pulse width and the highest pulse energy. | Dongyu Yan Bowen Liu Defeng Zou Jie Guo Yuxi Chu Youjian Song Minglie Hu | 2021 | High Power Laser Science and Engineering2021,9,3: | 0
