1MiR-30a Positively Regulates the Inflammatory Response of Microglia in Experimental Autoimmune Encephalomyelitis显示文摘Multiple sclerosis(MS) is a classical inflammatory demyelinating disease of the central nervous system(CNS). Microglia are the main resident immune cells in the CNS and are closely associated with the pathogenesis of MS.In the present study, we found that mi R-30 a was highly expressed in jellyfish-like microglia in chronic active lesions of MS patients, as well as in the microglia of mice with experimental autoimmune encephalomyelitis(EAE) at the chronic phase. In vitro, the conditioned supernatant of mouse microglia overexpressing miR-30 a promoted the apoptosis of oligodendrocyte precursor cells(OPCs), and inhibited OPC differentiation. In vivo, overexpressing miR-30 a in transplanted microglia exacerbated the progression of EAE.Overexpression and knock-down experiments in primary cultured mouse microglia showed that mi R-30 a increased the expression of IL-1 b and i NOS, which are pro-inflammatory, while inhibiting the expression of Ym-1 and CD206.Mechanistically, mi R-30 a inhibited the expression of Ppargc1 b, which is the co-activator of peroxisome proliferator-activated receptor gamma, resulting in pro-inflammatory effects. 4Myt1L Promotes Differentiation of Oligodendrocyte Precursor Cells and is Necessary for Remyelination After Lysolecithin-Induced Demyelination显示文摘The differentiation and maturation of oligodendrocyte precursor cells(OPCs) is essential for myelination and remyelination in the CNS. The failure of OPCs to achieve terminal differentiation in demyelinating lesions often results in unsuccessful remyelination in a variety of human demyelinating diseases. However, the molecular mechanisms controlling OPC differentiation under pathological conditions remain largely unknown. Myt1 L(myelin transcription factor 1-like), mainly expressed in neurons,has been associated with intellectual disability, schizophrenia, and depression. In the present study, we found that Myt1 L was expressed in oligodendrocyte lineage cells during myelination and remyelination. The expression level of Myt1 L in neuron/glia antigen 2-positive(NG^(2+))OPCs was significantly higher than that in mature CC1^+ oligodendrocytes. In primary cultured OPCs,overexpression of Myt1 L promoted, while knockdown inhibited OPC differentiation. Moreover, Myt1 L was potently involved in promoting remyelination after lysolecithin-induced demyelination in vivo. Ch IP assays showed that Myt1 L bound to the promoter of Olig1 and transcriptionally regulated Olig1 expression. 5Tumor-derived neomorphic mutations in ASXL1 impairs the BAP1-ASXL1-FOXK1/K2 transcription network显示文摘Additional sex combs-like 1(ASXL1)interacts with BRCA1-associated protein 1(BAP1)deubiquitinase to oppose the polycomb repressive complex 1(PRC1)-mediated histone H2A ubiquitylation.Germline BAP1 mutations are found in a spectrum of human malignancies,while ASXL1 mutations recurrently occur in myeloid neoplasm and are associated with poor prognosis.Nearly all ASXL1 mutations are heterozygous frameshift or nonsense mutations in the middle or to a less extent the C-terminal region,resulting in the production of C-terminally truncated mutant ASXL1 proteins.How ASXL1 regulates specific target genes and how the C-terminal truncation of ASXL1 promotes leukemogen-esis are unclear.Here,we report that ASXL1 interacts with forkhead transcription factors FOXK1 and FOXK2 to regulate a subset of FOXK1/K2 target genes.We show that the C-terminally truncated mutant ASXL1 proteins are expressed at much higher levels than the wild-type protein in ASXL1 heterozygous leukemia cells,and lose the ability to interact with FOXK1/K2.Specific deletion of the mutant allele eliminates the expression of C-termi-nally truncated ASXL1 and increases the association of wild-type ASXL1 with BAP1,thereby restoring the expression of BAP1-ASXL1-FOXK1/K2 target genes,particularly those involved in glucose metabolism,oxygen sensing,and JAK-STAT3 signaling pathways.In addition to FOXK1/K2,we also identify other DNA-bind-ing transcription regulators including transcription factors(TFs)which interact with wild-type ASXL1,but not C-terminally truncated mutant.Our results suggest that ASXL1 mutations result in neomorphic alleles that contribute to leukemogenesis at least in part through dominantly inhibiting the wild-type ASXL1 from interacting with BAP1 and thereby impairing the function of ASXL1-BAP1-TF in regulating target genes and leukemia cell growth.Yu-Kun Xia Yi-Rong Zeng Meng-Li Zhang Peng Liu Fang Liu Hao Zhang Chen-Xi He Yi-Ping Sun Jin-Ye Zhang Cheng Zhang Lei Song Chen Ding Yu-Jie Tang Zhen Yang Chen Yang Pu Wang Kun-Liang Guan Yue Xiong Dan Ye 2021Protein & Cell2021,12,7:6

8Cyclin-dependent Kinase 18 Promotes Oligodendrocyte Precursor Cell Differentiation through Activating the Extracellular Signal-Regulated Kinase Signaling Pathway显示文摘The correct differentiation of oligodendrocyte precursor cells(OPCs) is essential for the myelination and remyelination processes in the central nervous system.Determining the regulatory mechanism is fundamental to the treatment of demyelinating diseases. By analyzing the RNA sequencing data of different neural cells, we found that cyclin-dependent kinase 18(CDK18) is exclusively expressed in oligodendrocytes. In vivo studies showed that the expression level of CDK18 gradually increased along with myelin formation during development and in the remyelination phase in a lysophosphatidylcholine-induced demyelination model, and was distinctively highly expressed in oligodendrocytes. In vitro overexpression and interference experiments revealed that CDK18 directly promotes the differentiation of OPCs, without affecting their proliferation or apoptosis. Mechanistically, CDK18 activated the RAS/mitogen-activated protein kinase kinase1/extracellular signal-regulated kinase pathway, thus promoting OPC differentiation. 9Recent progress in the green synthesis of rare-earth doped upconversion nanophosphors for optical bioimaging from cells to animals显示文摘Rare-earth doped upconversion nanophosphors(UCNPs), which convert low energy near-infrared(NIR) photons into high energy photons such as ultraviolet, visible light and NIR light, have found various applications in optical bioimaging. In this review article, we summarize recent advances in the synthesis and applications of UCNPs achieved by us and other groups in the past few years. The approaches for the synthesis of UCNPs are presented,with an emphasis on the role of green chemistry in the advancement of this field, followed by a focused overview on their latest applications in optical bioimaging from subcellular structures through cells to living animals. 10High-gravity-assisted green synthesis of rare-earth doped calcium molybdate colloidal nanophosphors显示文摘In this work,we report an innovative route for the synthesis of rare-earth doped calcium molybdate(CaMoO4)nanophosphors by using high gravity rotating packed bed(RPB)technology and paraffin liquid as the solvent.The significant intensified mass transfer and micromixing of reactants in the RPB reactor are benefiting for homogeneous doping of rare-earth ions in the host materials,leading to nanophosphors with high quantum efficiency.The use of liquid paraffin as the solvent eliminates the safety risks associated with volatile organic compounds,increasing the potential for clean production of nanophosphors.Under excitation of deep ultraviolet(DUV)light,the CaMoO4:Na+,Eu3+nanophosphors exhibit red emission at peak wavelength of 615 nm and quantum yield of up to 35.01%.The CaMoO4:Na+,Tb3+nanophosphors exhibit green emission at peak wavelength of543 nm with quantum yield of up to 30.66%.The morphologies of the nanophosphors are tunable from nanofibers through nanorods to nanodots and the possible mechanism of controlling the formation of different nanostructures is proposed on the basis of experimental results and theoretical analysis of mesoscience.These nanophosphors are highly dispersible in organic solvents and utilized for fabricating fabrication of flexible,freestanding luminescent films based on silicone resin.We also demonstrate the red LEDs consisting of the hybrid films of CaMoO4:Na+,Eu3+nanoparticles as color-converting phosphors and DUV LEDs as illuminators,offering strong potential for future nanophosphors-basedsolid-state lighting systems.Yuan Pu Lifeng Lin Jun Liu Jiexin Wang Dan Wang 2020Chinese Journal of Chemical Engineering2020,28,6:3

12Giant and light modifiable third-order optical nonlinearity in a free-standing h-BN film显示文摘Recently,hexagonal boron nitride(h-BN)has become a promising nanophotonic platform for on-chip information devices due to the practicability in generating optically stable,ultra-bright quantum emitters.For an integrated information-processing chip,high optical nonlinearity is indispensable for various fundamental functionalities,such as all-optical modulation,high order harmonic generation,optical switching and so on.Here we study the third-order optical nonlinearity of free-standing h-BN thin films,which is an ideal platform for on-chip integration and device formation without the need of transfer.The films were synthesized by a solution-based method with abundant functional groups enabling high third-order optical nonlinearity.Unlike the highly inert pristine h-BN films synthesized by conventional methods,the free-standing h-BN films could be locally oxidized upon tailored femtosecond laser irradiation,which further enhances the third-order nonlinearity,especially the nonlinear refraction index,by more than 20 times.The combination of the free-standing h-BN films with laser activation and patterning capability establishes a new promising platform for high performance on-chip photonic devices with modifiable optical performance.Jun Ren Han Lin Xiaorui Zheng Weiwei Lei Dan Liu Tianling Ren Pu Wang Baohua Jia 2022Opto-Electronic Science2022,1,6:3

14Impact of Enterprise Ownership and Size on Registered Hematotoxicity in Benzene-exposed Workers显示文摘Benzene is a classified as a human carcinogen that can cause myelodysplastic syndrome and acute myelogenous leukemia[1]. This compound is also a potent toxin suppressing the bone marrow. Accordingly, benzene causes serious hematological adverse effects, such as pancytopenia and aplastic anemia. In the 1920s, the carcinogenicity of benzene was identified in humans, in which it was shown to cause acute myelocytic leukemia (AML)[2]. The reports highlighting the first cases of hematopoietic disease associated with benzene appeared in China in the late 1950s[3]. Moreover, benzene exposure is likely to result in AML, having been identified as a Group Ⅰ carcinogen by the International Agency for Research on Cancer (1987)[4]. 16Constraint and Mobility Change Analysis of Rubik’s Cube-inspired Reconfigurable Joints and Corresponding Parallel Mechanisms显示文摘The current research of reconfigurable parallel mechanism mainly focuses on the construction of reconfigurable joints.Compared with the method of changing the mobility by physical locking joints,the geometric constraint has good controllability,and the constructed parallel mechanism has more configurations and wider application range.This paper presents a reconfigurable axis(rA)joint inspired and evolved from Rubik’s Cubes,which have a unique feature of geometric and physical constraint of axes of joint.The effectiveness of the rA joint in the construction of the limb is analyzed,resulting in a change in mobility and topology of the parallel mechanism.The rA joint makes the angle among the three axes inside the groove changed arbitrarily.This change in mobility is completed by the case illustrated by a 3(rA)P(rA)reconfigurable parallel mechanism having variable mobility from 1 to 6 and having various special configurations including pure translations,pure rotations.The underlying principle of the metamorphosis of this rA joint is shown by investigating the dependence of the corresponding screw system comprising of line vectors,leading to evolution of the rA joint from two types of spherical joints to three types of variable Hooke joints and one revolute joint.The reconfigurable parallel mechanism alters its topology by rotating or locking the axis of rA joint to turn all limbs into different phases.The prototype of reconfigurable parallel mechanism is manufactured and all configurations are enumerated to verify the validity of the theoretical method by physical experiments.Duanling Li Pu Jia Jiazhou Li Dan Zhang Xianwen Kong 2020Chinese Journal of Mechanical Engineering2020,33,5:2

18Zirconia quantum dots for a nonvolatile resistive random access memory device显示文摘We propose a nonvolatile resistive random access memory device by employing nanodispersion of zirconia(ZrO2) quantum dots(QDs) for the formation of an active layer. The memory devices comprising a typical sandwich structure of Ag(top)/ZrO2(active layer)/Ti(bottom) are fabricated using a facile spin-coating method. The optimized device exhibits a high resistance state/low resistance state resistance difference(about 10 Ω), a good cycle performance(the number of cycles larger than 100), and a relatively low conversion current(about 1 μA). Atomic force microscopy and scanning electron microscope are used to observe the surface morphology and stacking state of the ZrO2 active layer. Experimental results show that the ZrO2 active layer is stacked compactly and has a low roughness(Ra=4.49 nm) due to the uniform distribution of the ZrO2 QDs. The conductive mechanism of the Ag/ZrO2/Ti device is analyzed and studied, and the conductive filaments of Ag ions and oxygen vacancies are focused on to clarify the resistive switching memory behavior. 19High-gravity-assisted emulsification for continuous preparation of waterborne polyurethane nanodispersion with high solids content显示文摘In this work,we developed a continuous preparation strategy for the production of high-solidscontent waterborne polyurethane(WPU)emulsions via high-gravity-assisted emulsification in a rotating packed bed(RPB)reactor.By adjusting the experimental parameters and formula,WPU emulsions with a high solids content of 55%and alow viscosity were prepared.Preliminary applications of the high-solids-content WPU as a thermally insulating material were demonstrated.RPB emulsification is an economical and environmentally friendly production strategy because of the low energy consumption,short emulsification time,and effective devolatilization.This study demonstrated an effective method for preparation of high-solids-content WPU,moving toward commercialization and industrialization.Weihong Zhang Dan Wang Jie-Xin Wang Yuan Pu Jian-Feng Chen 2020Frontiers of Chemical Science and Engineering2020,14,6:1
