1Proteins Related to the Biocontrol of Pythium Damping-off in Maize with Trichoderma harzianum Rifai显示文摘Induced resistance has been evidenced as one of mechanisms of Trichoderma to control plantdiseases, however, no study showed the change of host proteomics in Trichoderma-induced resistance ofmaize against damping-off caused by Pythium ultimum Trow. The mechanism of Trichoderma harzianumRifai for controlling maize seedling disease caused by Pythium ultimum Trow was investigated firstly byproteome technique and the result suggested that T. harzianum strain T22 was not only able to promoteseedling growth but also protein accumulation. One-dimensional electrophoresis assay showed that morebands appeared on the gel with T22 or T22 combined with P. ultimum (T22 + P. ultimum) treatment than withother treatments. Enzyme assay showed that two chitinases of the root sample were more activated in thetreatments with T22 than in the other treatments without T22. Proteins in the seedling roots from the varioustreatments were separated through protein extraction and 2-D electrophoresis technique. In the seedlingsproduced from the T22-treated seeds, there were 104 up-regulated proteins and 164 down-regulated pro-teins relative to the control, and 97 and 150, respectively, after treatment with T22 + P. ultimum; however,with P. ultimum alone the values were much lower than with the other two treatments. The correlationcoefficient values were 0.72, 0.51 and 0.49 for the comparison of protein spot distribution on gel amongcontrol with T22, P. ultimum and T22 + P. ultimum, respectively. So it seemed that P. ultimum infection wasmore effective than T22 in interfering with the host proteome profile. Furthermore, analysis with MALDI-TOF-MAS showed that some important proteins associated with defensive reactions were identified in T22or T22 + P. ultimum treatments, including endochitinase, pathogenesis-related protein PRMS (pathogenesis-related maize seed), GTP-binding protein, isoflavone reductase and other proteins related to respiration. Allthose proteins are probably part of the network of resistance or development-related proteins. Interestingly,P. ultimum treatment resulted in elimination of pathogenesis-related protein PRMS on gel, and thereforedamping-off could be in part attributed to inhibition of the expression of this protein by P. ultimum infection.Some unknown proteins are also related to the defensive reaction of the host.Jie CHEN Gary E HARMAN Afio COMIS Gen-Wu CHENG 2005Journal of Integrative Plant Biology2005,47,8:8
2Coexpression of CD163 and CD141 identifies human circulating IL-10-producing dendritic cells (DC-10)显示文摘Tolerogenic dendritic cells(DCs)are key players in maintaining immunological homeostasis,dampening immune responses,and promoting tolerance.DC-10,a tolerogenic population of human IL-10-producing DCs characterized by the expression of HLA-G and ILT4,play a pivotal role in promoting tolerance via T regulatory type 1(Tr1)cells.Thus far,the absence of markers that uniquely identify DC-10 has limited in vivo studies.By in vitro gene expression profiling of differentiated human DCs,we identified CD141 and CD163 as surface markers for DC-10.The coexpression of CD141 and CD163 in combination with CD14 and CD16 enables the ex vivo isolation of DC-10 from the peripheral blood.CD14+CD16+CD141+CD163+cells isolated from the peripheral blood of healthy subjects(ex vivo DC-10)produced spontaneously and upon activation of IL-10 and limited levels of IL-12.Moreover,in vitro stimulation of allogeneic naive CD4+T cells with ex vivo DC-10 induced the differentiation of alloantigen-specific CD49b+LAG-3+Tr1 cells.Finally,ex vivo DC-10 and in vitro generated DC-10 exhibited a similar transcriptional profile,which are characterized by an anti-inflammatory and pro-tolerogenic signature.These results provide new insights into the phenotype and molecular signature of DC-10 and highlight the tolerogenic properties of circulating DC-10.These findings open the opportunity to track DC-10 in vivo and to define their role in physiological and pathological settings.Michela Comi Daniele Avancini Francesca Santoni de Sio Matteo Villa Molly Javier Uyeda Matteo Floris Daniela Tomasoni Alessandro Bulfone Maria Grazia Roncarolo Silvia Gregori 2020Cellular & Molecular Immunology2020,17,1:4
3哈茨木霉菌 (Trichoderma harzianum)和终极腐霉菌 (Pythium ultimum)对玉米蛋白质组的影响(I)(英文)显示文摘哈茨木霉 (Trichodermaharzianum)T2 2菌株已普遍用于防治包括由终极腐霉 (Pythiumultimum)引起的苗病或根腐病在内的各种病害。玉米自交系Mo17种子经T2 2处理后播种在接种腐霉或未接种的田间土壤内 ,5d后取幼苗的根系或幼茎提取蛋白。结果表明 :在接种腐霉菌的土壤内 ,未进行T2 2处理的 5d龄幼苗长势明显比对照差 ,而经T2 2处理的幼苗长势明显比对照好。T2 2和腐霉菌复合处理及T2 2单独处理对幼苗生长影响基本相同。本研究建立了蛋白质提取和双向电泳分离技术。通过双向电泳及相应的分析软件 (PDQuestTM 2 Dsoftware)可将不同处理的幼苗自交系蛋白进行分离。T2 2菌株处理的根系产生 10 4种上游调控蛋白和 16 4种下游调控蛋白 ,T2 2与腐霉菌复合处理可产生 97种上游调控蛋白和 15 0种下游调控蛋白 ,而用腐霉菌单一处理诱导的上游或下游蛋白的数量明显少于上述 2个处理。T2 2或腐霉菌单一或复合处理的根系蛋白质组图谱与空白对照相比差异显著 ,它们与对照的蛋白质组图谱相似系数分别为 0 .72、0 .5 1和0 .4 9;T2 2与腐霉菌分别处理的蛋白质组图谱间也相差明显 ,两者的相似系数仅为 0 .6 5。进一步研究发现 ,T2 2菌体蛋白质组图谱与上述各种处理的蛋白质组图谱相似系数均很低 ,说明各种处理诱导后的幼苗?陈捷 Gary G.HARMAN Alfio COMIS 程根武 刘海南 2004植物病理学报2004,34,4:3

5Potential protective role of ACE-inhibitors and AT1 receptor blockers against levodopa-induced dyskinesias:a retrospective case-control study显示文摘Growing evidence has highlighted that angiotensin-converting enzyme(ACE)-inhibitors(ACEi)/AT1 receptor blockers(ARBs)may influence the complex interplay between dopamine and the renin-angiotensin system in the nigrostriatal pathway,thus affecting the development of levodopa-induced dyskinesia in Parkinson’s disease(PD).In the present study,we analyzed whether the use of this class of medication was associated with a reduced occurrence of levodopa-induced dyskinesia,using electronically-stored information of idiopathic PD patients enrolled at Novara University Hospital“Maggiore della Carità”.We conducted a retrospective case-control study identifying PD patients with dyskinesias(PwD;n=47)as cases.For each PwD we selected a non-dyskinetic control(NoD),nearly perfectly matched according to sex,Unified Parkinson’s Disease Rating Scale(UPDRS)part III score,and duration of antiparkinsonian treatment.Binary logistic regression was used to evaluate whether dyskinesias were associated with ACEi/ARBs use.Ninety-four PD patients were included,aged 72.18±9 years,with an average disease duration of 10.20±4.8 years and 9.04±4.9 years of antiparkinsonian treatment.The mean UPDRS part III score was 18.87±7.6 and the median HY stage was 2.In the NoD group,25(53.2%)were users and 22(46.8%)non-users of ACEi/ARBs.Conversely,in the PwD group,11(23.4%)were users and 36 non-users(76.6%)of this drug class(Pearson chi-square=8.824,P=0.003).Concerning general medication,there were no other statistically significant differences between groups.After controlling for tremor dominant phenotype,levodopa equivalent daily dose,HY 3-4,and disease duration,ACEi/ARBs use was a significant predictor of a lower occurrence of dyskinesia(OR=0.226,95%CI:0.080-0.636,P=0.005).Therefore,our study suggests that ACEi/ARBs may reduce levodopa-induced dyskinesia occurrence and,thanks to good tolerability and easy management,represent a feasible choice when dealing with the treatment of hypertension in PD patients.The study was approved by the Ethics Committee of Novara University Hospital“Maggiore della Carità”(CE 65/16)on July 27,2016.Elena Contaldi Luca Magistrelli Anna VMilner Marco Cosentino Franca Marino Cristoforo Comi 2021Neural Regeneration Research2021,16,12:2
6The Yin-Yang of osteopontin in nervous system diseases: damage versus repair显示文摘Osteopontin is a broadly expressed pleiotropic protein,and is attracting increased attention because of its role in the pathophysiology of several inflammatory,degenerative,autoimmune,and oncologic diseases.In fact,in the last decade,several studies have shown that osteopontin contributes to tissue damage not only by recruiting harmful inflammatory cells to the site of lesion,but also increasing their survival.The detrimental role of osteopontin has been indeed well documented in the context of different neurological conditions(i.e.,multiple sclerosis,Parkinson’s,and Alzheimer’s diseases).Intriguingly,recent findings show that osteopontin is involved not only in promoting tissue damage(the Yin),but also in repair/regenerative mechanisms(the Yang),mostly triggered by the inflammatory response.These two apparently discordant roles are partly related to the presence of different functional domains in the osteopontin molecule,which are exposed after thrombin or metalloproteases cleavages.Such functional domains may in turn activate intracellular signaling pathways and mediate cell-cell and cell-matrix interactions.This review describes the current knowledge on the Yin and Yang features of osteopontin in nervous system diseases.Understanding the mechanisms behind the Yin/Yang would be relevant to develop highly specific tools targeting this multifunctional protein.Giuseppe Cappellano Domizia Vecchio Luca Magistrelli Nausicaa Clemente Davide Raineri Camilla Barbero Mazzucca Eleonora Virgilio Umberto Dianzani Annalisa Chiocchetti Cristoforo Comi 2021Neural Regeneration Research2021,16,6:2
