| 3 | Combining CLUE-S and SWAT Models to Forecast Land Use Change and Non-point Source Pollution Impact at a Watershed Scale in Liaoning Province, China显示文摘Non-point source(NPS) pollution has become a major source of water pollution. A combination of models would provide the necessary direction and approaches designed to control NPS pollution through land use planning. In this study, NPS pollution load was simulated in urban planning, historic trends and ecological protection land use scenarios based on the Conversion of Land Use and its Effect at Small regional extent(CLUE-S) and Soil and Water Assessment Tool(SWAT) models applied to Hunhe-Taizi River Watershed, Liaoning Province, China. Total nitrogen(TN) and total phosphorus(TP) were chosen as NPS pollution indices. The results of models validation showed that CLUE-S and SWAT models were suitable in the study area. NPS pollution mainly came from dry farmland, paddy, rural and urban areas. The spatial distribution of TN and TP exhibited the same trend in 57 sub-catchments. The TN and TP had the highest NPS pollution load in the western and central plains, which concentrated the urban area and farm land. The NPS pollution load would increase in the urban planning and historic trends scenarios, and would be even higher in the urban planning scenario. However, the NPS pollution load decreased in the ecological protection scenario. The differences observed in the three scenarios indicated that land use had a degree of impact on NPS pollution, which showed that scientific and ecologically sound construction could effectively reduce the NPS pollution load in a watershed. | 6 | microRNA-218 suppresses the proliferation, invasion and promotes apoptosis of pancreatic cancer cells by targeting HMGB1显示文摘Objective: To detect the expression profiles of micro RNA-218(mi R-218) in human pancreatic cancer tissue(PCT) and cells and their effects on the biological features of human pancreatic cancer cell line PANC-1 and observe the effect of mi R-218 on the expression of the target gene high mobility group box 1(HMGB1), with an attempt to provide new treatment methods and strategies for pancreatic cancer.Methods: The expressions of mi R-218 in PCT and normal pancreas tissue as well as in various pancreatic cancer cell lines including As PC-1, Bx PC-3, and PANC-1 were determined with quantitative real-time reverse transcription polymerase chain reaction(q RT-PCR). The change of mi R-218 expression in PANC-1 cells was detected using qR T-PCT after the transfection of miR-218 mimic for 48 h. Cell Counting Kit-8(CCK-8) was applied for detecting the effect of mi R-218 on the activity of PANC-1 cells. The effects of mi R-218 on the proliferation and apoptosis of PANC-1 cells were analyzed using the flow cytometry. The effect of mi R-218 on the migration of PANC-1 cells was detected using the Trans-well migration assay. The HMGB1 was found to be a target gene of mi R-218 by luciferase reporter assay, and the effect of mi R-218 on the expression of HMGB1 protein in cells were determined using Western blotting.Results: As shown by q RT-PCR, the expressions of mi R-218 in PCT and in pancreatic cancer cell line significantly decreased when compared with the normal pancreatic tissue(NPT)(P<0.01). Compared with the control group, the miR-218 expression significantly increased in the PANC-1 group after the transfection of mi R-218 mimic for 48 h(P<0.01). Growth curve showed that the cell viability significantly dropped after the overexpression of mi R-218 in the PANC-1 cells for two days(P<0.05). Flow cytometry showed that the S-phase fraction significantly dropped after the overexpression of mi R-218(P<0.01) and the percentage of apoptotic cells significantly increased(P<0.01). As shown by the Trans-well migration assay, the enhanced mi R-218 expression was associated with a significantly lower number of cells that passed through a Transwell chamber(P<0.01). Luciferase reporter assay showed that, compared with the control group, the relative luciferase activity significantly decreased in the mi R-218 mimic group(P<0.01). As shown by the Western blotting, compared with the control group, the HMGB1 protein expression significantly decreased in the PANC-1 group after the transfection of mi R-218 mimic for 48 h(P<0.01).Conclusions: The mi R-218 expression decreases in human PCT and cell lines. mi R-218 can negatively regulate the HMGB1 protein expression and inhibit the proliferation and invasion of pancreatic cancer cells. | 11 | Gut microbiota contributes to the development of endometrial glands in gilts during the ovary-dependent period显示文摘Background:The hyper-prolificacy Meishan gilts achieved a superior endometrial gland development(EGD)than white crossbred gilts during the ovary-independent period(before 60 d of age).Then,the EGD continues under the management of ovary-derived steroid hormones that regulated by gut microbiota(after 60 d of age).However,whether Meishan gilts’superiority in EGD lasting to the ovary-dependent period(after 60 d of age)and the role of gut microbiota in this period both remain unclear.Methods:Meishan gilts and Landrace x Yorkshire(LxY)gilts were raised under the same housing and feeding conditions until sexual maturity and then we compared their EGD and gut microbiota.Meanwhile,we transplanted fecal microbiota from Meishan gilts to L×Y gilts to explore the role of gut microbiota in EGD.We sampled plasma every 3 weeks and collected the uterus,ovary,liver,and rectal feces after the sacrifice.We then determined the hormone concentrations and expressions of the EGD-related genes.We also profiled the gut microbiota using 16S rDNA sequencing and metabolites of plasma and liver tissue using untargeted metabolomics.Finally,the correlation analysis and significant test was conducted between FMT-shifted gut microbes and EGD-related indices.Results:Meishan gilts have larger endometrial gland area(P<0.001),longer uterine horn length(P<0.01)but lighter uterine horn weight(P<0.05),a distinctive gut microbiota compared with L×Y gilts.Fecal microbiota transplantation(FMT)increased endometrial gland area(P<0.01).FMT markedly shifted the metabolite profiles of both liver and plasma,and these differential metabolites enriched in steroid hormone biosynthesis pathway.FMT increased estradiol and insulin-like growth factor 1 but decreased progesterone dynamically.FMT also increased the expression of the EGD-related genes estrogen receptor 1 gene,epithelial cadherin,and forkhead box protein A2.There is a significant correlation between FMT-shifted gut microbes and EGD-related indices.Conclusion:Sexually matured Meishan gilts achieved a superior EGD than LxY gilts.Meanwhile,gut microbiota contribute to the EGD potentially via regulating of steroid hormones during the ovary-dependent period. | Baoyang Xu Wenxia Qin Yiqin Yan Yimei Tang Shuyi Zhou Juncheng Huang Chunlin Xie Libao Ma Xianghua Yan | 2021 | Journal of Animal Science and Biotechnology2021,12,4: | 4 |

| 13 | Synthesis of Amphiphilic Starch Derivatives Using One-pot Synthesis Procedure显示文摘Amphiphilic starch derivatives with high content of functional groups were prepared from potato starch using a one-pot synthesis method with a single reaction medium for the entire procedure. Potato starch was benzylated, followed by the introduction of hydroxypropyltrimethylammonium(HPMA) moieties without the purification of intermediates. The synthesis was performed under heterogeneous conditions, leading to the formation of benzyl 2-hydroxypropyltri methylammonium starch chloride(BnHPMAS) with a total degree of substitution(DS) of up to 1.4. This process improved the efficiency of the preparation of amphiphilic starch derivatives and reduced the time and resources consumed by avoiding a separation process and purification of the intermediate compounds.The DS of BnHPMAS was in the range of 0.36 to 1.4, which could be tuned by varying the molar ratio of the reagents to repeating unit or by changing the reaction temperature, time, and medium. The structure of the amphiphilic starches was characterized using elemental analysis, size exclusion chromatography,fourier transform infrared spectroscopy(FT-IR), and nuclear magnetic resonance(NMR) spectroscopy. | 15 | Integrated analysis of multi-tissues lipidome and gut microbiome reveals microbiota-induced shifts on lipid metabolism in pigs显示文摘Lipid metabolism is very important for meat quality in pigs.Accumulating evidence shows that gut microbiota can contribute to this physiological process.However,the gut microbiota that function in lipid metabolism and adipogenesis remains unclear.Here,we compared the characteristics of fat deposition and gut microbial community between Laiwu pigs and Duroc(LandraceYorkshire)(DLY)pigs.Fecal microbiota transplantation(FMT)was performed to determine the possible impact of gut microbiota on lipid metabolism in pigs.An integrated analysis of the gut microbiome and lipidome of the small intestine,plasma,and liver was conducted to investigate the effects of FMT on host lipid metabolism.The comparative analysis of the gut microbiome showed higher abundance of Bacteroidetes(P=0.0018)while lower abundance of Firmicutes(P=0.012)in Laiwu pigs,and the microbial composition can be transferred from Laiwu pigs into DLY pigs.Transmission electron microscope and Oil red-O staining were performed to analyze the effects of FMT on lipid deposition in liver,the main target organ for lipid metabolism.The results showed that FMT significantly increased the number of lipid droplets(P=0.0035)and lipid accumulation(P=0.0026)in liver.Furthermore,integrated multi-tissues lipidome analysis demonstrated that the fatty acyls and glycerophospholipids were significantly increased(P<0.01)in intestine and liver,while glycerolipids and fatty acyls were reduced(P<0.01)in plasma.In the small intestine,FMT increased(P<0.01)the relative abundance of polyketides and prenol lipids but reduced(P<0.01)the saccharolipids.Correlation analysis revealed the potential interactions between microbiota and lipid metabolites.Together,our results indicated that the gut microbiota may regulate the lipid metabolism and enhance the accumulation of lipid droplets in the liver of pigs. | Chunlin Xie Xiaoyan Zhu Baoyang Xu Yaorong Niu Xuelei Zhang Libao Ma Xianghua Yan | 2022 | Animal Nutrition2022,,3: | 2 |

PANC-1 cells were transfected either with HMGB1 si RNA to knock down the expression of HMGB1 or with the recombinant HMGB1 expression vector(pc DNA3.1-HMGB1) to overexpress HMGB1. The effect of ectopic expression of HMGB1 on the apoptosis of mi R-218-transfected and GEMtreated PANC-1 cells was examined by flow cytometric analysis.Results: The mi R-218 expression level was lower in GEM-resistant PANC-1 cells compared to GEMsensitive Bx PC-3 cells(P<0.05). The percentage of apoptotic PANC-1 cells was significantly increased in the mi R-218 mimic + GEM group compared to the mimic ctrl + GEM group and the normal control group(P<0.01). The HMGB1 expression level was markedly decreased in PANC-1 cells transfected with HMGB1 si RNA but was significantly increased in PANC-1 cells transfected with the recombinant HMGB1 expression vector, pc DNA3.1-HMGB1(P<0.01). | 17 | Band tillage with fertilizer application for unpuddled transplanting rice in northeast of China显示文摘Many technological innovations have been developed in agriculture for rice cultivation.Farmers still rely on intensive soil tillage,broadcasting fertilizer,excess irrigation water and labor for rice production.Rice cultivation under unpuddled condition in band-tillage(BT)is a new alternative rice cultivation system in the regions where rainfall,fresh water resources,fertilizers and labors are limited.Therefore,the concept of inter-row BT(T-shaped)with row fertilizer application in soils for unpuddled transplanted rice cultivation(T1)was carried out at Fangzheng,Harbin in the northeast of China.The improved system(T1)was compared with traditional practices(T2)in randomized complete block design with three replications.T2 was done with removal of crop residue,full tillage,broadcasting fertilizer and puddled soils by using the conventional machine.Test results showed that all parameters were satisfied and met the requirements of agronomic performances.The variations of fertilizing stability and evenness were 1.84%and 1.72%,respectively.Soil cone penetration resistance was less in T1 than T2 after tillage.Irrigation water was saved by 26.3%in treatment T1 as compared with T2 for wet land preparation,and wet soil resistance depth had no significant difference.Planting depth was significantly different between the treatments which was affected by intensive tillage and puddling in T2 as compared with T1.Soil temperature was nearly similar,but comparatively 0.23°C(on average)higher than traditional system throughout the crop growing season.Crop growth rate and grain yield increased by 8.5%and 5.3%,respectively in band till with fertilizer placed compared to conventional practices of puddled transplanting.The unit production cost(¥/kg of grain yield)was found lower by 12.3%in treatment T1 than T2.The output-input ratio was marginally greater in treatment T1;1.51)as compared to T2;1.32).Short-term findings suggest that inter-row BT system with row fertilizer application could be used in unpuddled rice farming system instead of traditional puddled rice cultivation. | Khokan Kumer Sarker Xu Chunlin Wang Xiaoyan Li Minjin Li Lianhao Liu Guiming | 2016 | International Journal of Agricultural and Biological Engineering2016,9,4: | 2 |

However,the microbiota composition that can alter the fat deposition and the meat quality of pigs remains unclear.Here, we used Laiwu (LW) pigs (a native Chinese breed with higher intramuscular fat) compared withcommercial crossbreed Duroc×(Landrace×Yorkshire) (DLY) pigs to investigate the effects of microbiotaon meat quality, especially in intramuscular fat content. A total of 32 DLY piglets were randomly allottedto 4 groups and transplanted with fecal microbiota from healthy LW pigs. The results indicated that thehigh dose of fecal microbiota transplantation (HFMT) selectively enhanced fat deposition in longissimusdorsi (P < 0.05) but decreased backfat thickness (P < 0.05) compared with control group. HFMT significantlyaltered meat color and increased feed conversation ratio (P < 0.05). Furthermore, the multi-omicsanalysis revealed that Bacteroides uniformis, Sphaerochaeta globosa, Hydrogenoanaerobacterium saccharovorans,and Pyramidobacter piscolens are the core species which can regulate lipid deposition. A total of140 male SPF C57BL/6j mice were randomly allotted into 7 groups and administrated with these 4 microbesalone or consortium to validate the relationships between microbiota and lipid deposition.Inoculating the bacterial consortium into mice increased intramuscular fat content (P < 0.05) comparedwith control mice. Increased expressions of lipogenesis-associated genes including cluster of differentiation36 (Cd36), diacylglycerol O-acyltransferase 2 (Dgat2), and fatty acid synthase (FASN) wereobserved in skeletal muscle in the mice with mixed bacteria compared with control mice.
